1.Reversal for acquired resistance to EGFR inhibitors in lung cancer
Journal of International Oncology 2013;40(11):843-846
Epidermal growth factor receptor (EGFR) inhibitors targeted therapy is the forward position means for non-small cell lung cancer. However, acquired resistance to EGFR inhibitors limits the development of targeted drugs.Using existing data on drug resistance in EGFR-mutant lung cancer, this review discusses three basic approaches for overcoming resistance to EGFR-targeted therapies:intensification of EGFR inhibition, combination of EGFR inhibitors with other targeted therapies, and altering clinical management via alternate pathways.

The near infrared ( NIR ) spectra of 76 batches of the amoxicillin capsules from different manufacturers and their corresponding content powder without capsules cell were used to find some quantitative indicators to evaluate whether the piecewise direct standardization ( PDS ) algorithm succeeded in NIR quantitative model updating. 54 batches were used to construct the NIR quantitative model for capsule preparation. All the NIR spectra of amoxicillin capsules were divided into five classes by cluster analysis, and each class can be regarded as a homology sample set. The average spectrum for each homology sample set was calculated. Ten to Fifteen spectra were selected from each homology sample set as the corresponding master spectra of the PDS algorithm to correct all the NIR spectra of the amoxicillin content powder respectively. Then the corrected spectra were predicted by the constructed NIR quantitative model for amoxicillin capsules. The prediction error for each corrected powder spectrum, and the correlation coefficient between each corrected powder spectrum and the average spectrum of the corresponding homolog sample set which the PDS master spectra came from, were calculated. Finally, the relationship between the prediction error and its corresponding correlation coefficient were studied. It was found that the correction results correlated closely with the selected master spectra set in PDS algorithm. The bigger the correlation coefficient ( r) , the better the correction results. In general, when r is less than 99%, it can be judged that the PDS correction is failed. At this condition, the prediction error is often more than 5%. Therefore, the correlation coefficient between the corrected spectrum and its corresponding average spectrum of the homology sample set can be used as an indicator to evaluate the efficiency of the PDS correction.

Pigment epithelium-derived factor (PEDF) is an antiangiogenic factor which is effective in tumour inhibition in a variety of tumours and has not yet been studied in bladder tumour before. In this study the expression of PEDF, interleukin-1α (IL-1α) and -8 (IL-8) in bladder tumours was investigated. Immunohistochemistry was performed on 64 bladder tumour and 23 normal uroepithelium samples. Expression change of the factors was compared with clinicopathological parameters. Correlations between PEDF, IL-1α and IL-8 were analyzed. None of the factors was in relation to gender, tumour occurrence, and size or onset pattern. PEDF (P=0.014) and IL-1α (P=0.049) expression was down-regulated with grade progression. PEDF expression was lower in normal uroepithelium than in papillary urothelial neoplasm of low malignant potential (PUNLMP) (P=0.000) and carcinoma (P=0.009) whilst IL-1α (P=0.000 and P=0.000 respectively) and IL-8 (P=0.000 and P=0.023 respectively) expression was higher in the same grouping. PEDF expression had a negative correlation with IL-8 in PUNLMP (P=0.049, r=-0.578) as well as in tumour grouping (P=0.033, r=-0.276). Deranged expressional change of PEDF, IL-1α and IL-8 could be in relation to loss of differentiation from normal uroepithelium to papillary lesion and eventually to carcinoma.

Bones are stained into red color with feeding madder, but we do not know whether the fed madder can change the bone biomechanical properties and bone mineral contents in animals. In this research, we established a rat model with feeding madder. The bone biomechanical properties were detected by universal material mechanics, bone mineral contents were detected by inductively coupled plasma mass spectrometry and spectrometer, and red color material in bone was analyzed by high performance liquid chromatography. The results showed that bone biomechanical parameters in femur diaphysis in the 10% and 15% group rats were significantly higher than those in the control group after feeding madder for 6 months. The level of calcium, magnesium and zinc in femur diaphysis in 10% and 15% group rats were higher than those in the control group after feeding madder for 6 months. However, it was shown that the kidney congestion and hyperemia and the level of blood urea nitrogen and creatinine in the 15% group rats were significantly different compared to those in the control group rats after feeding madder for 6 months. The red colored material in bone is related to alizarin analyzed with high-performance liquid chromatography. The conclusion could be drawn that feeding 10% madder in diet was not toxic to the rats fed for 6 months, and it could improve bone biomechanical properties and increase bone mineral elements.

Chinese Journal of Practical Nursing 2017;33(29):2294-2298
Objective To investigate behaviors of nurses on pressure ulcer prevention, exploring the related factors of pressure ulcer prevention behaviors. Methods With cross-sectional survey,a self-made questionnaire of nurse′s pressure ulcer prevention behaviors was used to investigate nurses′behaviors. Results The overall score of behavior was(43.97±8.95)points,standard score from high to low were risk assessment (6.98 ± 1.69) points, incontinence care (6.66 ± 1.51) points, Health education (9.61 ± 2.50) points, position and reposition (9.21 ± 2.19) points, supporting surface (11.51 ± 3.20) points. Different department (Z=-2.379, P<0.05), job title (χ2=11.601, P<0.01), past experiences of learning pressure ulcer knowledge(Z=2.474,P<0.05),latest learning time(Z=-2.399,P<0.05),caring number of pressure ulcer patients (χ2=17.338, P<0.01) had different scores. Multiple linear regression analysis of behavior indicated that recent education time from now was the influential factor, which can explain 15.5% of the variation in prevention behavior. Conclusions Nurses′ pressure ulcer prevention behaviors need to be improved, with nurse whose recent studying time was less than one year showing better preventive behavior. Nurse′s behaviors got worse when they didn′t get education for a long time, which indicated that the frequency of training should be enhanced.

Journal of Clinical Surgery 2023;31(12):1185-1190
Objective To investigate the expression, function and prognostic value of Ras-related C3 botulinum toxin substrate 2(RAC2)in breast cancer. Methods The expression profile of RAC2 mRNA was investigated in the TCGA-BRCA cohort, the prognostic value was analyzed using the Kaplan-Meier algorithm, the potential molecular function was investigated by gene set enrichment analysis, the relationship between RAC2 and immune infiltration was analyzed by Estimate and Cibersort algorithms, and the association between Rac2 and mutation spectrum was evaluated by maftools software package. The RNA-seq software package"pRRophetic"was used to predict the effect of chemotherapy drugs. In the validation cohort,Immunohistochemistry(IHC)staining was performed on tumor pathological sections of patients.Kaplan-Meier survival analysis and cox regression analysis were used to analyze the relationship between RAC2 and the prognosis of breast cancer. Results Bioinformatics analysis showed that the expression of RAC2 was up-regulated in estrogen receptor(ER)positive BC.In the Pam50 molecular subtype of BC,its expression decreased in the order of normal-like group, triple negative group,HER2 overexpression group,Luminal A group and Luminal B group.BC patients with high RAC2 expression had better overall survival(OS)and recurrence-free survival(RFS).The results of the validation cohort confirmed that RAC2 can be used as an independent predictor of BC prognosis.RAC2 was involved in the process of immune infiltration of tumor cells, and its high expression was mainly related to the mutations of CCDC168,VPS13C and AHNAK genes. Its up-regulation increased the drug sensitivity of doxorubicin, cisplatin, paclitaxel and gemcitabine. In terms of metabolism,it was negatively correlated with glycolysis and pentose phosphate pathway(PPP),and positively correlated with fatty acid oxidation(FAO)and glutamine metabolism. Conclusion High expression of RAC2 is associated with a good prognosis of BC and plays a role in the metabolism and immune infiltration.Further study on the molecular mechanism of RAC2 may provide new strategies for the prognosis and treatment of BC.
