3.Detection of Enteroviruses and Mammalian Reoviruses by RT-PCR and Integrated Cell Culture-PCR in CPE-positive Surface Water Samples.

The environmental water samples assayed by total culturable virus assay (TCVA) were evaluated by reverse transcription-polymerase chain reaction (RT-PCR) and integrated cell culture-PCR (ICC-PCR) method for enteroviruses and reovirus since they are the usually detected virus groups by culture assays. The detection sensitivities of the TCVA, RT-PCR, and ICC-PCR were compared and the overall reliability of the detection was analyzed to confirm environmental samples for enteric viruses. A total of eight samples from different areas was analyzed by performing TCVA, RT-PCR, and ICC-PCR. Virus concentrations in surface water samples ranged from 1.03 to 47.3 most probable numbers of infectious units (MPN) per 100 liters. When primers specific for both enteroviruses and reoviruses were used in both RT-PCR and ICC-PCR, all the samples (100%) were positive for the viruses. Reoviruses were the most frequently detected ones among the samples. According to the sequence results of enteroviruses, five of the samples were contaminated by coxsackievirus type B3, and the rest by coxsackievirus type B6, echovirus type 30, or vaccine strain poliovirus type 3. It was observed that both enteroviruses and reoviruses were detected concurrently in all CPE-positive environmental water samples.

8.Integrated Cell Culture-PCR Detection of Enteroviruses and Reoviruses in Water Sources in Gyeonggi-do.

The integrated cell culture-PCR (ICC-PCR) method has been suggested as an improved method for detection of viruses in water environments. We tested 57 source waters including finished water samples in Gyeonggi-do for enteric viral contamination using total culturable virus assay (TCVA) using BGMK cells and ICC-PCR. Nineteen of the 57 source water samples (33.3%) exhibited the cytopathic effect (CPE) on BGMK cells and no finished water did exhibited CPE. Nineteen samples (33.3%) of the 57 were positive for reoviruses. For the enteroviruses, only 3 samples (5.3%) of the 57 samples showed positive results. By using ICC-PCR method, 202 flasks from source water samples were positive for enteroviruses and reoviruses. Three samples from source water were positive for both viruses. However, any flasks tested was not co-infected with two types of viruses. While the enteric viral frequencies in TCVA and ICC-PCR were similar, the viral frequency for reoviruses at first passage in two type of method was higher in ICC-PCR (94.7%) than TCVA (56.9%).

9.Isolating enterovirus and poliovirus isolated in patients with acute flaccid paralytic syndrome in 2005

Background: Paralysis has been eliminated in Vietnam in 2000. To maintain the achievement, Vietnam needs to have more than 90% of children under 1 age drinking 3 doses of OPV, controlling cases of acute flaccid paralysis and diagnosing for virology in laboratory to determine causes of the disease. Results of laboratory identifying Polio virus and enterovirus in patients with acute flaccid paralysis is evidence of successful in paralytic elimination annually in Vietnam.\r\n', u'Objectives: To of isolate polio virus and enterovirus in patients with acute flaccid paralysis suspected paralysis in 2005.\r\n', u'Subjects and methods: The study included 267 cases with acute flaccid paralysis from 29 provinces, cities in the North and six provinces in the Central of Vietnam during 2005.\r\n', u'Results:Results of surveillance for acute flaccid paralysis and indicators of samples tested in 2005 have reached the criteria regulated by the World Health Organization. 8 poliovirus strains and 56 other non-polio enterovirus strains were isolated from 538 stool specimens of above patients. All isolated poliovirus strains were confirmed as Sabin-like strains. 56 non-polio enterovirus strains included coxsaxkie A (3), conxsackieB (5), echoviruses (34), enterovirus 71 (1) and untyped enteroviruses (13). 10.Enteroviruses and poloviruses isolated in patients with acute flaccid paralytic syndrome in 2004

In 2004, 10 poliovirus strains were isolated from 4 acute flaccid paralytic cases (AFP). All these poliovirus strains were confirmed as Sabin-like strains. 66 other non-polio enterovirus strains were also isolated including 29 echoviruses, coxsackie B viruses and 30 untyped enteroviruses. These virus strains were isolated from 489 stool specimens of 245 AFP cases. The results confirmed the free status of the stable sustaining poliomyelitis in Viet nam.
