2.Effect of Schisandra chinensis lignans on neuronal apoptosis and p-AKT expression of rats in cerebral ischemia injury model.

OBJECTIVETo observe the effect of Schisandra chinensis lignans (SCL) on neuronal apoptosis and PI3K/AKT signaling pathway of rats in the cerebral ischemia injury model, and study its possible mechanism.
METHODRats were orally administered SCL high, middle and low dose groups (100, 50, 25 mg x kg(-1)) for 14 days. The cerebral ischemia injury model was established by using the suture-occluded method to rate the neurological functions. The cerebral infarction area was observed by TTC staining. The pathological changes in brain tissues were determined by HE staining. Bcl-2 and Bax expressions were detected by immunohistochemical assay. The protein expressions of p-AKT and AKT were assayed by Western blotting.
RESULTCompared with the model group, SCL high, middle and low dose groups showed reduction in the cerebral infarction area to varying degrees, improve the pathological changes in brain tissues, promote the expression of apoptin Bcl-2 and p-AKT, and inhibit the expression of apoptin Bax.
CONCLUSIONSCL shows a protective effect on rats with cerebral ischemia injury. Its mechanism may be related to the increase in p-AKT ability and antiischemic brain injury capacity and the inhibition of nerve cells.

3.Anatomical structure and histochemical localization about rhizomes of five species of Polygonatum.

OBJECTIVEThe microscopic structure and histochemical localization of the Polygonatum sibiricum, P. cytonema, P. filipes, P. zanlanscianense and P. odoratum was studied.
METHODScanning electron microscope (SEM) and digital biological microscope (DBM) were applied in the research.
RESULTVenation bundle showed in three types included collateral, amphivasal and incomplete amphivasal bundles. Morphological differences of venation bundles could be seen under the fluorescence microscope. Calcium oxalate existed in the mucilage cells looked similar to columnar crystals under SEM. In rhizome of Polygonatum, polysaccharides presented in mucilage cells, saponins and volatile oil were found in ground tissue.
CONCLUSIONMicrostructure difference of rhizomes would be used for identification of the Polygonatum plants. Polysaccharides and saponins are distributed in different cells of Polygonatum plants.

4.Expression of COL1a1 and COL3a1 in the capsule of children with developmental dislocation of the hip.

OBJECTIVEThe etiology of developmental dislocation of the hip (DDH) remains uncertain, but some research has shown that this disorder is closely related to hip joint laxity. This study examined the expression of collagens type I and III mRNA and protein in the hip capsule of children with DDH in order to investigate the roles of collagens type I and III in hip joint laxity.
METHODSNine children with DDH and nine age and gender-matched normal children (control group) were enrolled. Semiquantitative RT-PCR method was used to detect mRNA expression of COL1a1 and COL3a1 in the hip capsule. Western-Blot method was used to detect protein expression of COL1a1 and COL3a1 in the hip capsule. The quantitative analysis of the COL1a1 and COL3a1 was performed by professional image software and the results were analyzed with standard statistical methods.
RESULTSmRNA and protein expression of COL1a1 in the DDH group was significantly lower than that in the control group (P<0.01). Compared with the control group, COL1a3 mRNA expression in the DDH group decreased significantly (P<0.01), but COL1a3 protein expression was not significantly different.
CONCLUSIONSThe decreased collagen I mRNA and protein expression in the hip capsule might contribute to hip joint laxity in children with DDH. Collagen type III may not be associated with hip joint laxity in DDH.

5.A histological study of three-dimensional external zygomatic suture distraction osteogenesis.

OBJECTIVETo explore the histological change in suture of zygomatic bone for the zygomatic suture direct distraction osteogenesis.
METHODSThe zygomatic bone was distracted by 3-D external distraction appliance without osteotomy. The specimens were taken 1, 3, 5 and 8 weeks after, and then examined histologically and compared with the blank contralateral side.
RESULTSThere were lots of fibroblasts, osteoblasts and capillary vessels in the distracted suture tissues one week after distraction, and the fibers were observed to connect the sides of suture and arranged orderly. The surfaces of the expanded suture were irregular. Bone formation was active in the expanded side. The bone trabeculae were mature and oriented in the direction of distraction in the distracted sides at 3 weeks. A great amount of new woven bones were found in 5-week specimen. New bones were formed completely 8 weeks after the distraction.
CONCLUSIONSNew bone formed rapidly in the distracted side of zygomatic bone under suture distraction osteogenesis without osteotomy.

6.An experimental study of 3-dimension zygomatic suture extension for suture osteogenesis.

OBJECTIVETo explore a methods of 3-Dimension expansion of zygomatic suture in a goat model.
METHODSSeven goats were used in this study. The 3-D extensive applicator was designed and used to extend the zygomatic suture of the goats by placing it in the zygomatic bone through an infraorbital incision. Ten days after the first operation, it was gradually extended on a speed of 0.09 cm/d for 7 days. The zygomatic movement and the osteogenesis of the suture was evaluated in two weeks.
RESULTSThe zygomatic bone was extended for 0.6 cm long in average, and the osteogenesis was also shown significantly in the suture.
CONCLUSIONThe above mentioned technique could be a safe and effect method to be applied for the zygomatic extension.

7.Developmental changes of acetabular cartilage complex: an experimental study of a straight-leg swaddle model of newborn rats.

OBJECTIVEBy establishing a model of straight-leg swaddle of newborn rats and observing the experimental animals'hips morphologically and pathologically, this study explored the changes of gross appearance of the acetabulum and the maturity of cartilage cells in the different regions of acetabular cartilage complex.
METHODSThe legs and hips were fixed by adhesive tape for 10 days in the position of hip extension and adduction in 31 newborn Wistar rats (experimental group). The other 31 newborn rats without legs and hips treatment were used as the control group. After 10 days raising in the same condition, all the rats were sacrificed. The gross appearance, histological observations and VEGF and type X collagen immunohistochemistry were used for examining the acetabulum changes.
RESULTSA straight leg swaddle model of newborn rats was established successfully. In the experimental group the acetabulum became shallow and small and surrounded by more soft tissues. There were 49 dislocated hips (49/54) in the experimental group and 2 hips dislocated (2/60) in the control group (p<0.01). Fake acetabulum appeared in the experimental group. In the control group, the shape of the acetabulum was normol, and no fake acetabulum was found. The safranin O-fast green staining showed that the orange-red cartilage in the experimental group was wider than the control group. Immunohistochemistry observations showed VEGF and type X collagen immunoreactivities in the hypertrophic layer of the acetabular cartilage complex in the experimental group were lower than those in the control group. The percentages of VEGF positive and type X collagen positive cells in the iliac hypertrophic layer of the acetabular articular cartilage were significantly higher than those in the ischiadic ramus and the pubic branch in the experimental group.
CONCLUSIONSVEGF and type X collagen immunoreactivities in acetabular cartilage cells decrease in a straight-leg swaddle model of newborn rats. This suggests that this position might lead to dysmaturity of the acetabular cartilage cells and affect the development of the acetabulum.

8.Analysis on investigating results of Kaschin-Beck disease in Shandong province in 2008

Objective To investigate the present status of Kaschin-Beck disease(KBD) in Qingzhou at present, and provide a scientific basis for decision-making in controlling the disease. Methods According to The National Technical Scheme for Endemic Disease Control in 2007, children aged 7 to 12 were chosen to receive clinical and X-ray examination in 3 villages of 2 towns in Qingzhou. Local adults aged 16 years and older were clinically diagnosed in 10 villages of 4 towns and divided into different groups according to the clinical degrees and their ages. The Diagnostic Criteria of Kashin Beck Disease(GB 16003-1995) was carded out by clinical and X-ray diagnosis. Results Two hundred and sixty school children aged 7 to 12 were clinically diagnosed, and the patients of I degree upwards of KBD were not detected. One hundred and ninety-eight school children aged 7 to 12 were examined by X-ray, and the detectable rate was 0(0/198). In 7099 adults aged 16 years old upwards clinically tested, 502 patients of degree I and higher of KBD were detected and the detectable rate was 7.07%(502/7099). The patients distributed mainly in the population aged 36 years old and older, accounting for 99.00% (497/502). Conclusions Although KBD in children have been controlled in Qingzhou, it is still serious in adults. So the monitoring of KBD should be carried on and prevention and control for KBD in adults should be strengthened.
9.Analysis of monitoring results of Kaschin-Beck disease in Shandong province from 1996 to 2010

ObjectiveTo investigate the current status of Kaschin-Beck disease in Shandong province, and to provide a scientific basis for decision-making in controlling the disease. Methods According to the National Monitoring Program of Kaschin-Beck disease requirements, historical serious villages of Kaschin-Beck disease in Qingzhou of Shandong province were selected annually; children aged 7 to 16 were chosen to receive clinical examination and children aged 7 to 12 were taken X-ray examination. Clinical and X-ray diagnosis was carried out according to the Diagnostic Criteria of Kashin Beck Disease(GB 16003-1995). Results From 1996 to 2010, in 53 diseased villages, three thousand three hundred and eighteen school children aged 7 to 16 were clinically diagnosed, and child Kaschin-Beck disease of degree Ⅰ and above were not detected; three thousand and ninety-one school children aged 7 to 12 were examined by X-ray, forty cases were found positive, and the total positive rate was 1.29%(40/3091 ). The year with the highest positive rate was 2002, and the rate was 3.49%(13/372) ; the positive rate was 0 in 1996 and 2008. The difference of the X-ray positive rate between each year was statistically significant(x2 =31.54, P < 0.01 ). ConclusionsChild Kashin-Beck disease in Qingzhou is basically under control.Since etiology of Kashin-Beck disease is still unclear, surveillance of the disease still needs to be strengthened.
10.Model innovation of traditional Chinese medicine resources survey pilot work in Anhui province.

In order to guarantee the smooth progress of census work, complete the traditional Chinese medicine resources survey pilot tasks better, Anhui province founded the three-combined technical team organization model and practical management model. Around the objectives, integrating the professionality with the existing distribution of traditional Chinese medicine resources and the reality of technical team of the census in Anhui province. The technical team organization model combining universities, experts, locality, expressed the strength of all parties and formed efficient working groups. The establishment of responsible management, funds management and process management ensured that the Anhui census work had a high level of quality and relatively consistent progress. The result shows that the organization and management model of Anhui census technical team were the important guarantee of finishing the Anhui census work smoothly with great quality and quantity on time.
