1.Analysis of Allergens Detection in Asthmatic Infants and Young Children

Objective To analyze the allergens detection in asthmatic infants and young children. Methods The specific IgE test and allergens screening test were conducted in 243 infants and young children with whooping diseases including asthmatic bronchitis, asthmatic pneumonia and asthma by skin prick test and Pharmacia UniCAP test. Results 1. Food allergen was the allergen detection most frequently in infants(P = 0.03). The proportion of aeroallergens increased significantly with age (positive rate:67. 2%). Milk and egg white was the main food allergen detection in infants and young children,and the main aeroallergen detection was house dust mite. 2. The allergen - specific IgE test using UniCAP test system was more accurate than skin prick test. Conclusions The infants are more susceptible to food hypersensitivity; the increasing opportunity of aeroallergen exposure may lead to a higher prevalence of aeroallergens in young children. UniCAP system is more accurate than other methods.
2.Relationship between estrogen receptor and cisplatin resistance in human ovarian cancer SKOV3 cells

Objective To investigate the relationship between estrogen receptor and cisplatin resistance in ovarian cancer SKOV3 cells.Methods A total of 30 mice were inoculated 4 × 106 ovarian cancer SKOV3 cells to establish the xenograft tumor model with human ovarian cancer SKOV3 cells.And they were randomly divided to control group and test group with 15 cases per group.Control group was given 0.9% NaC1,intraperitoneal injection,once a week,a total of 5 times.Test group was given cisplatin 20 mg · kg-1,intraperitoneal injection,once a week,a total of 5 times.Then the ovarian cancer SKOV3 cells were collected from two groups.The expression of estrogen receptor in ovarian cancer SKOV3 cells was detected by immunocytochemistry and flow cytometry.MTT assay and flow cytometry were used to detect the effect of proliferation and apoptosis in ovarian cancer SKOV3 cells with different concentrations of cisplatin (0,1,5,10,15,20 μg · mL-1) combined with ICI182 (0,1,5,10,20 μmol · L-1) or alone.Results In control and test groups,the immune cell chemistry score (HIS score) were (10.04 ± 2.93),(6.73 ±2.37)points;the expression of estrogen receptor were 231.39 ± 15.33,205.81 ± 12.50 with significant difference(P > 0.05).ICI182 had anti-proliferative effect on the control and test groups with dose-dependent and time-dependent manners.The inhibition rate of proliferation significantly increased by ICI182 combined with cisplatin (P <0.01).After treated with 10 μmol · L-1 ICI182 + 10 μg · mL-1 cisplatin in the test group and control group,the inhibition rates were (70.07-± 0.64) % and (82.13 ± 0.87) % with significant difference (P < 0.01).Conclusion Estrogen receptor is involved in the process of drug resistance,and it is possible to reverse the cisplatin resistance of ovarian cancer cells by inhibiting estrogen receptor protein by ICI182.
3.Renal Hemodynamics in Patients with End Stage Renal Disease

Objective To investigate the relationship between refractory hypertension and renal hemodynamics in end stage renal diseases (ESRD) patients.Methods ESRD patients were classified into:patients with refractory hypertension (group A) and patients with normal blood pressure(group B).Renal hemodynamic indices were ex- amined by duplex ultrasonography.Fasting serum lipid (TC,TG,HDL-C,LDL-C,Lp(a),ox-LDL) and serum parathyroid hormane (PTH) were determined in all patients.Results Significant differences were found in renal hemodynamic indices such as peak systolic velocity (PSV),mean flow velocity (MV),pulsatility index (PI),renal- aortic ratio (RAR) and in clinical index such as Lp(a) and ox-LDL between the two group.Refractory hyperten- sion patients had lower renal hemodynamic indices and higher Lp(a) and ox-LDL levels than in patients with con- trolled BP.Logistic regression analysis revealed that refractory hypertension was related with PSV,EDV,Pl and RAR,but not relevant with sex,age,dialysis time,hematocrit,BUN,creatinine,TC,TG,HDL-C,LDL-C, PTH,MV and RI.Conclusion Atherosclerotic renal artery stenosis and severe disorder in renal hemodynamics is likely the cause for refractory hypertention in ESRD patients.The rise of serum Lp(a) and ox-LDL might acceler- ate renal artery atherosclerosis.
4.Application of nutrition guidance in maintenance of hemodialysis patients

Objective To evaluate the nutritional status and nursing nutritional guidance in maintenance of hemodialysis patients(MHD).Methods 48 cases of MHD were selected and randomly divided into two groups:experimental group(n=23)and control group(n=25).Control group was siven routine nursing.Experimental group waft given routine nursing plus nutritional guidance.Results After six months nutritional guidance,the nutritional status of the experimental group are better significantly than that of the control group(P<0.05).Conclusions Nutritional guidance Call significantly improve the nutritional status of the MHD.
5.Expression of endotoxin receptors on hepatic stellate cells during their activation.

OBJECTIVETo investigate the role of endotoxin receptor expression in the activation of hepatic stellate cells (HSCs).
METHODSHSCs were isolated from normal rats and the expression of endotoxin receptors on quiet HSCs and in vitro activated HSCs was determined using RT-PCR and immunocytochemical staining methods. A rat model of liver fibrosis and cirrhosis was established. The expressions of CD14 and alpha-SMA in liver tissues were detected by immunohistochemical staining.
RESULTSFreshly isolated HSCs had a low level of CD14 mRNA expression and no expression of TLR4 mRNA was detected. The in vitro activated HSCs had increased expressions of CD14 mRNA and TLR4 mRNA and LPS up-regulated the expression of endotoxin receptors. Immunocytochemical staining showed cytoplasmic and nucleolus staining for CD14 in the cultured HSCs. LPS played a further role on CD14 protein expression. In the development of liver fibrosis, the number of CD14-positive cells in the livers was increased and these cells were distributed along the sinusoids. In the later stage of liver fibrosis, the CD14-positive cells were gathered in the fibrotic septae, which also contained alpha-SMA positive cells.
CONCLUSIONThe activated HSCs expressed endotoxin receptors. The endotoxin receptors may be involved in the role in which HSCs played in the inflammatory process and liver fibrosis development.

6.An enzyme-linked immunosorbent assay for determining serum anti-themocyte globulin concentration.

OBJECTIVETo establish an enzyme-linked immunosorbent assay (ELISA) for determining anti-themocyte globulin (ATG) levels in serum samples.
METHODSThe microplate was coated with mouse anti-rabbit IgG monoclonal antibody, and sheep anti-rabbit polyclonal antibody conjugated with HRP was used as the second antibody for detecting the serum ATG levels in patients undergoing allogeneic hematopoietic stem cell transplantation.
RESULTSThe optimal concentration of the coating antibody and dilution ratios of the serum samples and IgG-HRP conjugate were 0.2 microg/ml, 1:40 and 1:2500, respectively. The lower sensitivity limit of the assay was 31.25 ng/ml for ATG detection. A linear relationship was established within the concentration range from 40 to 1000 ng/ml, with the coefficients of variation of 7.91 within assay and 5.22 between assays, respectively. Seven patients undergoing stem cell transplantation with ATG pretreatment showed gradually decreased concentration of ATG, and after 90 days ATG could still be detected.
CONCLUSIONThe sandwich ELISA we established provides a specific and sensitive method for quantitative measurement of ATG in the clinical setting. In patients undergoing stem cell transplantation with ATG pretreatment, the ATG concentration gradually decreases but remains detectable 90 days after the administration.

7.The effect of microRNA-145 on immune-inflammatory response of foam cells by targeting CD40

Objective To investigate the effect of miRNA-145 (miR-145) on immuno-inflammatory reaction of foam cells by targeting CD40. Methods Mouse macrophage cell line RAW 264.7 cells cultured in vitro were randomly divided into model group (non-transfected), miR-145 mimics group (transfected miR-145 mimics), miR-145 inhibitor group (transfected miR-145 inhibitor) and silencing CD40 sequence group (transfected siCD40). Then oxidized low density lipoprotein (ox-LDL) was used to stimulate for 24 h to establish immune inflammatory damage cell model. Quantitative real-time polymerase chain reaction (RT-qPCR) and Western blot assay were used to detect the levels of CD 40 mRNA and protein of each group. ELISA was used to detect the levels of inflammatory factors interleukin (IL)-1, IL-6 and tumor necrosis factor (TNF) -α in cell supernatant. Results Compared with model group, the levels of CD40 mRNA, CD40 protein and IL-1, IL-6, TNF-αwere all significantly decreased in miR-145 mimics group (P<0.01). After transfected with miR-145 inhibitor, the above indexes were all significantly increased than those of model group and miR-145 mimics group (P<0.01). After transfected with CD40 siRNA, the levels of CD40 mRNA, CD40 protein and IL-1, IL-6, TNF-αwere all obviously decreased compared with those of miR-145 inhibitor group (P<0.01). Conclusion MiR-145 can regulate the immune inflammatory process of foam cells through the target gene CD40, inhibit the activation of CD40/CD40L signaling pathway and inhibit inflammatory response.
8.Meta analysis on clinical effectiveness of Chinese medicine physiotherapy combined with tropicamide eye drops for pseudomyopia

AIM:To evaluate the curative effect of Chinese medicine physiotherapy combined with tropicamide eye drops to treat pseudomyopia in children and adolescent. METHODS: We collected randomized controlled trials from CNKI, CBM, Wanfang database, PubMed, EMBASE and Cochrane Library in 2000-2015, and the improved Jadad scale was used to evaluate the methodology of the literature, and the data was extracted. The Review Manager 5. 3 statistical software was used for meta analysis. RESULTS: A total of 5 articles were included in the analysis, with a total sample size of 836 cases. The curative effect of experimental group on pseudomyopia was better than that of control group,and the differences are statistically significant (Z=6.39,P<0.01). CONCLUSION: Compared with topiramine eye drops alone, combined with traditional Chinese medicine therapy for treating pseudomyopia in children and adolescent is more effective,and is safe and reliable.

10.Role of dendritic cells in the pathogenesis of severe pneumonia in children.

OBJECTIVESevere pneumonia is one of the common severe diseases in children. Increasing evidences show that immune response greatly contribute to severe pneumonia. Dendritic cells (DC) are the important antigen presenting cells in the lung. To study the role of dendritic cells in development of severe pneumonia in children, the authors measured the number of mature DC in bronchoalveolar lavage fluid (BALF), and evaluated the relationship among IL-12, pro-inflammatory cytokines and clinical scores.
METHODSThe following 3 groups of children were enrolled in this study: severe pneumonia group: 27 children with severe pneumonia treated between November 2002 and May 2003 in PICU; mild pneumonia group: 30 children with mild pneumonia in department of pulmonology; control group: 29 children without pneumonia but receiving ventilator treatment for chest surgery. Mature DC in BALF was determined in severe pneumonia group and the control group on the day of tracheal intubation for mechanical ventilation. Acute lung injury scores and severe disease scores were evaluated in children with severe pneumonia and mild pneumonia. All children's serum levels of TNF-alpha, IL-6 and IL-12 were measured by using ELISA within 24 hours after admission. SPSS version 11.5 was used for statistical analysis.
RESULTS(1) The percent of mature DC in children with severe pneumonia was significantly higher when compared with the control group on the first day after ventilation [14.2 (3.9 - 51.8)] vs. [1.3 (0.2 - 22.5)] (Z = 5.44, P < 0.01). (2) In severe pneumonia group, the concentration of serum IL-12 [117.0 (79.9 - 159.4) ng/L], TNF-alpha [90.6 (52.2 - 185.9) ng/L], IL-6 [128.7 (73.3 - 793.8) ng/L] were significantly higher than those in mild pneumonia group where the values were [71.6 (19.4 - 196.8)], [26.6 (2.5 - 113.9)], and [39.9 (7.8 - 82.5)] (P < 0.01), and the control group [6.4 (12.2 - 92.0)], [6.4 (1.8 - 91.9)], and [23.0 (6.4 - 54.2)] (P < 0.01). Serum IL-12, TNF-alpha and IL-6 levels in children with mild pneumonia were higher than those of control group (P < 0.01). (3) The percent of mature DC was increased with the serum level of IL-12 (r = 0.48, P < 0.01), TNF-alpha (r = 0.58, P < 0.01), IL-6 (r = 0.51, P < 0.01) and lung injury scores (r = 0.39, P < 0.05), but it did not correlate with severe disease scores (r = -0.11, P > 0.05).
CONCLUSIONSThere is excessive production of pro-inflammatory cytokines and over-stimulation of lung dendritic cells in children with severe pneumonia. Over-stimulation of lung dendritic cells, the increased serum levels of IL-12, TNF-alpha, IL-6 and the severity of pneumonia may suggest that DC plays an important role in pathogenesis of severe pneumonia in children.
