2.Correlations between the level of inhibition C, glycosylated hemoglobin A1c and the coronary lesions in patients with coronary heart disease

Objective To study the correlations between the levels of inhibition C (Cys-C), glycosylated hemoglobin A1c (HbA1c) and the coronary lesion. Methods 184 patients were divided into the coronary heart disease (CHD) group (n = 113) and the control group (n = 71) according to coronary angiography. The correlations between the level of Cys-C, HbA1c and Gensini score and coronary vessel lesions were investigated. Results The levels of Cys-C and HbA1c of CHD group were significantly higher than the control group (P<0.05). The levels of Cys-C and HbA1c were positively correlated with Gensini score (r = 0.452, 0.570; P < 0.05). The levels of Cys-C and HbA1c were positively correlated with coronary vessel lesions (P<0.001). Multivariate analysis showed Cys-C and HbA1c were significantly associated with Gensini score (β = 0.221, t = 2.516, P < 0.013;β=0.525, t = 6.342, P < 0.001). There was no significant correlation between the level of Cys-C and Gensini score in the subgroup A (r = 0.102, P = 0.418), and the level of Cys-C was positively correlated with Gensini score in the subgroup B (r=0.440, P<0.002). Conclusions The levels of Cys-C and HbA1c are independently associated with the severity and the vessels of the coronary stenosis. Clinically, the joint detections of the levels of Cys-C and HbA1c are a higher predictive value for coronary artery lesions.
5.MiR-150 improves cardiac fibrosis after myocardial infarction by targeting c-Myb

Objective To evaluate the role and mechanism of miR-150 in cardiac fibrosis after MI.Methods A rat model of MI was established by up-regulating miR-150 through overexpressing miR-150 lentivirus.Real-time PCR and Western blot were applied in detecting the expression of collagen 1 α 1 and α-SMA protein in infarction area border.Masson coloration was applied in measuring fibrosis.Cardiac fibroblasts were isolated and cultured.UTR was used to report the carrier and lentivirus.And c-Myb siRNA was used to verify the relationship between c-Myb and microRNA-150.Results In vivo,MiR-150 was down-regulated in myocardium border zone in 14 day and 28 day after infarction (P < 0.001,P < 0.05),and overexpressing miR-150 promoted myocardial fibrosis (P < 0.001),and inhibited the expression of collagen1α 1 and α-SMA (P < 0.01,P < 0.05).In vitro,c-Myb was the direct target gene of miR-150,and inhibited the expression of c-Myb resulting in the down regulation of collagen1α 1 and α-SMA,suggesting that the role of miR-150 was achieved by regulating c-Myb.Conclusions MiR-150 was down-regulated in myocardium border zone,and myocardial fibrosis can be improved by targeting c-Myb.
6.Effect of Nocturnal Hypertension in Primary Hypertension Patients With Early Renal Dysfunction

Objective: To investigate the effect of nocturnal hypertension in essential hypertension (EH) patients with early renal dysfunction. Methods: A total of 182 EH patients were enrolled in this study. According to weather the average night blood pressure (BP) > 120/70 mmHg, the patients were divided into 2 groups: Nocturnal hypertension group,n=89 and Control group,n=93. The levels of urine micro albumin (mALB), podocalyxin (PCX) and serum Cyst-C were examined and compared between 2 groups. Results:① For BP: the average systolic and diastolic BP in Nocturnal hypertension group were higher than those in Control group as SBP (138.05 ± 6.33) mmHg vs (102.51 ± 8.76) mmHg, DBP (84.11 ± 6.32) mmHg vs (70.03 ± 4.56) mmHg, P<0.05. ② For renal dysfunction biomarkers: several biomarker levels were higher in Nocturnal hypertension group than those in Control group as mALB (13.60 ± 0.69) mg vs (10.04 ± 0.73) mg, PCX (5.35 ± 1.69) ng/ml vs (2.05 ± 0.88) ng/ml and serum Cyst-C (1.35 ± 0.69) mg/L vs (1.02 ± 0.44) mg/L, allP<0.05.③ For correlation study: In Nocturnal hypertension group, the PCX level was positively correlated to the levels of mALB (r=0.675,P<0.05), Cyst-C (r=0.734,P<0.05), night BP (r=0.830, P<0.05) and the duration EH (r=0.688,P<0.05). Conclusion: EH patients with nocturnal hypertension have more incidences to suffer from renal dysfunction, the examination of mALB, PCX and Cyst-C are beneifciary for the early diagnosis in relevant patients.
7.Experiment research of the function of human umbilical cord blood mesenchymal stem cells in the regeneration of rat's sciatic nerve

Objective To evaluate the efectiveness of using mesenchymal stem cells(MSCs)derived from HUCB(human umbilical cord blood)to a tissue engineered bioartificial nerve on bridging a 1 0 mm sciatic nerve gap.Methods The cord blood mononuclear cells were isolated by lymphocyte separation medium,purified and expanded with MesencultTM medium and acidic environment to produce adherent layer(MSCs).Thirty SD female rats were randomly divided into three groups.Group A:Human amnion tubes were seeded by HUCBMSCs together with fibrin sealant.Group B:Human amnion tubes were seeded only with fibrin sealant.group C:autografts.9 weeks later,a series of examinations were performed which included morphological,sciatic nerve function index,weight of gastrocnemius,histological staining and immunostaining of S100.Results The HUCB-derived mononuclear cells,when seeded in specific medium,gave rise to adherent cells (MSCs).At 9 weeks after the operations,all the examinations of group A was better than group B(P<0.05).Conclusion HUCBMSCs can be isolated,purified, cultivated and expanded Mesencult~(TM) medium.HUCBMSCs can promote the nerve to regenerate in reparing the sciatic nerve gap.
9.Effects of Rosuvastatin on Production of Reactive Oxygen Species and Expressions of Periostin, Cardiotrophin-1 in Experimental Rats After Acute Myocardial Infarction

Objective: To investigate the effects of rosuvastatin on reactive oxygen species (ROS) production and periostin, cardiotrophin-1 (CT-1) expression, and to explore rosuvastatin on ventricular remodeling in experimental rats after acute myocardial infarction (AMI). Methods: A total of 45 male Wistar rats were randomly divided into 2 group, Sham operation group,n=15 and AMI group,n=30, the AMI model was established by left anterior descending coronary ligation. After 24 hours of AMI, the rats were further divided into 2 groups, AMI + rosuvastatin group, the rats received gastric rosuvastatin 1mg/(kg?d), and AMI group, the rats received gastric normal saline.n=15 in each group and all animals were treated for 6 weeks. The mRNA and protein expressions of CT-1 and periostin were examined by real time RT-PCR and immunohistochemistry, the contents of superoxide anion (O2-·) and hydroxy radical (OH·) were detected by colorimetric method among different groups. Results: Compared with Sham operation group and AMI + suvastatin group, the mRNA and protein expressions of CT-1, periostin, the contents of (O2-·), (OH·) and left heart weight index were increased in AMI group at non-infraction zone,P<005. Compared with Sham operation group, the mRNA and protein expressions of CT-1, periostin, the contents of (O2-·), (OH·) and left heart weight index were increased in AMI + suvastatin group at non-infraction zone,P<005. Compared with AMI group, the mRNA and protein expressions of CT-1 and periostin were decreased in AMI + rosuvastatin group,P<005. Conclusion: Rosuvastatin may improve ventricular remodeling via inhibiting ROS production and CT-1, periostin expression in experimental rats after AMI.
