2.Effect of hypoxia on the expression of connective tissue growth factor in lung fibroblast

Objective To observe the effects of hypoxia on the expression of connective tissue growth factor(CTGF)and matrix metalloproteinase 9(MMP-9)in lung fibroblasts at different time points.Methods Fibroblasts were cultured at hypoxic condition 0,1.5,3,6,12 h respectively.CTGF mRNA and MMP-9 mRNA level were detected in each group by reverse transcription-polymerase chain reaction(RT-PCR).The concen- tration of CTGF and MMP-9 protein in fibroblasts supernatant were determined using enzyme-linked im- munosorbent assay(ELISA).Results Hypoxia stimulated fibroblasts increased the level of CTGF mRNA with- in 1.5 h,and the levels remained at a plateau up to 6 h,and then decreased by 12 h.The level of MMP-9 mRNA increased significantly within 3 h,and the levels kept the trend for increasing.ELISA revealed that the levels of both CTGF and MMP-9 protein/cell in medium conditioned by fibroblasts exposed to hypoxia were maximal at 12 h.The level of MMP in the CTGF-Ab treated groups was significantly decreased compared to the untreated groups.Conclusions These findings suggest that hypoxia stimulates fibroblasts to release CTGF as a mitogen factor,which initiates the fibrosis cascade and airway remodeling by regulating the balance of extracellular ma- trix synthesis and degradation via MMP-9 which is secreted by fibroblast cells in response to CTGF.

4.BAEP and SEP for prediction of prognosis in brain stem hemorrhage patients

ObjectiveTo investigate the correlation of brainstem auditory evoked potentials(BAEP) and somatic evoked potentials (SEP) with therapeutic outcome of brain stem hemorrhage patients.MethodsBAEP and SEP were detected in the early period of 25 caees with brain stem hemorrhage by evoked potential instrument,and were surveilled dynamically.ResultsThere was good prognosis in the patients whose BAEP and SEP were normal in the first time and repeated detection.Poor prognosis happened in ones whose BAEP and SEP were abnormal in the first time and repeated detection.The difference was significant between them(P<0.05).ConclusionCombined detection and dynamic surveillance of BAEP and SEP could predict accurately the curative result of patients with brain stem hemorrhage.
5.Expression of heat shock response protein induced by heat shock in human Tca8113 cells

Objective: To investigate the expression of heat shock response protein(HSP_ 70 ) in human squamous tongue cancer Tca8113 cells during the recovery periods of heat shock.Methods:Tca8113 cells were subjected to heat shock at 43 ℃ for 30 min, then the cells were cultured for 2,4,6,8,12,24 and 48 h respectively. 6.Survey of the influencing factors of learning adaptability in nursing student of the emergency department

Objective To investigate the influencing factors of learning adaptability of nursing student in the emergency department and come up with relevant countermeasures. Methods Two hundred nursing student in the emergency department participated in the survey. A self-designed questionnaire on their learning adaptability and the results were analyzed. Result The job category, work environment, disease of the patients, internship time, teaching method and the professional identification were the top six factors influencing their learning adaptability situation. Conclusion Such measures as strengthening the orientation of student nurses, cultivating their observation ability and communication skills, enhancing the consciousness about law, increasing practice time appropriately, improving teaching methods and ability, and strengthening sense of identity can guarantee effective internship and adaptability of the nursing student in emergency department.
7.The clinical application of standard craniotomy with large bone flap for frontal-temporal severe craniocerebral in jury patients

Objective To explore the curative effect of standard craniotomy with large bone flap of the treatment for frontal-temporal severe craniocerebral injury patients.Methods Standard craniotomy with large bone flap was performed on 63 patients with frontal-temporal severe craniocerebral injury.All postoperative patients were followed up.The curative effect was assessed according to GOS.Results All patients were followed up for 1 year.According to the GOS,28 cases recovered well,12 cases became moderately disabled,4 cases were severely disabled,3 cases was in vegetative state and 16 cases died.The mortality rate was 25.4%.Conclusions Standard craniotomy with large bone flap is the best method of the treatment for frontal-temporal severe craniocerebral injury.Timely and thoroughly standard craniotomy with large bone flap for frontal-temporal severe craniocerebral injury patients can significantly improve the the curative effect of the patients and reduce the rate of mortality.
8.Relationship Between Serum Oxidized Low-density Lipoprotein Level and Carotid Vulnerable Plaque in Patients with Acute Cerebral Ischemic Stroke

PurposeTo investigate the relationship between the serum oxidized low-density lipoprotein (ox-LDL) level and carotid vulnerable plaque in patients with acute cerebral ischemic stroke, and to evaluate the ability of serum ox-LDL in identifying vulnerable carotid plaques.Materials and Methods 181 patients with history of acute cerebral ischemic stroke were recruited. According to the results of carotid ultrasound patients were divided into non plaque group (n=48), stable plaque group (n=38) and vulnerable plaque group (n=95). The serum total cholesterol, triglyceride, high density lipoprotein-cholesterol, low density lipoprotein-cholesterol, fasting plasma glucose and ox-LDL were measured. Carotid intima-medial thickness (CIMT), total carotid plaque area (TPA) and the characters of plaque were examined with color Doppler ultrasound.Results The serum ox-LDL and the number of patients with hypertension in plaque group were significantly higher than that in the non-plaque group (P<0.05). The serum ox-LDL and TPA in the vulnerable plaque group were significantly higher than that in the stable plaque group (P<0.05). Serum ox-LDL in patients with acute cerebral infarction was positively correlated with CIMT, serum total cholesterol and LDL-C (r1=0.154,P<0.05;r2=0.338, P<0.05;r3=0.385,P<0.05). Logistic regression analysis showed that serum ox-LDL was an independent risk factor for carotid vulnerable plaque (OR=1.038, 95%CI 1.008-1.069, P<0.01).Conclusion The serum ox-LDL is independently associated with the formation of vulnerable plaque in patients with acute cerebral infarction. This study suggests that ox-LDL can be used as a biomarker in screening for vulnerable carotid plaque in clinical practice. Carotid ultrasound combined with serology can early identify carotid artery vulnerable plaque that may lead to cerebral ischemic events.
9.Molecular Epidemiologic Study of VacA Genotypes of Helicobacter Pylori in Zhejiang Province

Objective To investigate the the distribution of Helicobacter pylori (H. pylori) vacA gentypes and the relationship between the presence of specific genotypes and clinical diseases in Zhejiang of China. Methods 262 Helicobacter pylori strains were colleted from 8 districts of Zhejiang,chromosome DNA was extracted and polymerase chain reaction (PCR) was carried out to determine the polymorphism of vacA and cagA with specific primers.PCR results were analyzed statistically according to their isolated original and clinical outcomes. Results VacA m1b, vacAm2 and vacAm1bm2 were found positive in 27.10%,4.89% and 4.20% of the 262 H.Pylori strains respectively. There was no significant difference in vacA genotypes among different districts of Zhejiang. 26.47%(27/102)of vacA m1b,66.67%(68/102) vacAm2 and 2.94%(3/102)vacAm1bm2 Helicobacter pylori strains were isolated from chronic gastritis; 29.41%(40/136)of vacA m1b,61.76%(84/136)vacAm2 and 3.68%(5/136)vacAm1bm2 Helicobacter pylori strains were isolated from Peptic Ulcer;and 16.67%(4/24)of vacA m1b,75.00%(18/24) vacAm2 and 4.17%(1/24) vacAm1bm2 Helicobacter pylori strains were isolated from gastric cancer; There was no significant difference in vacA genotypes among different clinical disease. Conclusion CagA+ and vacAs1/ m2 are predominant genotypes of H. Pylori in 8 districts of Zhejiang province. However, the relationship between vacA genotypes of H. pylori and the clinical disease can be identified in this study.
