2.Effects of alcoholic extracts of seven traditional Chinese medicines and psoralen on tyrosinase in human YUGEN8 melanoma cell

Objective To explore the effects of alcoholic extracts of traditional Chinese medicines on the post-translational processing and trafficking of tyrosinase.Methods Human YUGEN8 amelanotic melanoma cells were grown in vitro;the cells were incubated with one of the seven traditional Chinese medicines,including Rhizoma Chuanxiong and psoralen.Protein analysis with Western blot,enzymolysis with endoglycosidase H (Endo H),and subcellular localization with laser confocal microscopy were per- formed.The expression,maturity and export from endoplasmic reticulum (ER) of tyrosinase in the treated cells were compared with those in the untreated controls.Results Compared with controls,an approximate- ly 80-kDa,Endo H-resistant tyrosinase doublet,which represented mature glycoform of tyrosinase,was in- creased in melanocytes treated with Semen Cuscutae,and in those treated with Semen Persicae.Within those cells,tyrosinase was distributed outside ER resident protein calnexin.Conclusion Both Semen Cus- cutae and Semen Persicae could induce tyrosinase maturation,stability and export from ER to distal site.
3.Photochemotherapy for Psoriasis.

The beneficial effects of sunlight and ultraviolet light for the treatment, of psoriasis vulgaris has long been recognized. Parrish et al were the first to introduce a controlled light source, which allowed the delivery of a known quantity of light energy to the patients skin after the patient had ingested a photosensitizer, 8-methoxypsoralen (methoxsalen) two hours previous to UVA exposure. This successful treatment has become known as PUVA (psoralen and UVA). PUVA photochernotherapy denotes a systemic approach that is based on the interaction of light and a photoactive drug. Parrish et al have described successful treatment of psoriasis with PUVA and other investigators alI over the world have confirmed their beneficial results. The rationale of photochemotherapy in psoriasis is the inhibition of the increased DNA synthesis within the psoriatic lesions by the interaction of psoralen molecule and light energy in the UVA range(320-400nm). The present study was undertaken to evaluate the therapeutic effectiveness of PUVA photochernotherapy combining orally administered methoxsalen and UAA exposure using Dermatron UVA phototherapy system in patients with wi5espread psoriasis. A total of 15 patients with psoriasis over more than 30% of their bodies were selected at the department of dermatology, National Medical Center through March 1978 to September 1978. Oral administration of methoxsalen, 20 to 50 mg doses according to body weight, was followed by exposure to a high-intensity longwave ultraviolet light emitting a continuous spectrum between 320 and 400nm (peak emission, 350 - 365nm). Initial UVA exposure times were based on each patients minimal phototoxicity dose (MPD) and exposure times were increased at each treatment by 1 - 2 minutes to maintain minimal erythema. Frequency of treatment was two to three times weekly. Once the psoriatic lesions had, cleared completely the patient was placed on a maintenance schedule. If psoriasis recurred the frequency of treatment was increased. The results were as follows: l. 5 patients were dropped out due to unknown reasons. 2. 7 cases showed clearing of over 95, of the lesions (Response 5'). Mean number of PU VA therapy was 16. 7. During 1 4 months follow-up period, 4 cases remained free of psoriasis but 3 cases couldn't be followed. 3. Immediate side effects of PUVA were temporary and generally mild. No significant changes in laboratory screenings were noted. Side effects of therapy included transient nausea(33.3%), dizziness(26.7%), erythema.(66.7%), pruritus (60%), increased pigrnentation(100 %). 4. Comparison of pre-and post-PUVA therapy in biopsy specimens by light microscopy revealed histologic improvement evidenced by diminished acanthosis and parakeratosis, and regeneration of granular layers in all of 8 cases. Glycogen accumulation in the affected epiderrnis was significantly decreased or absent after PUVA therapy in all cases.

4.The experimental study on radioguided surgery in lung carcinoma using ~(18)F-FDG

Objective To investigate the effectiveness of radio gu ided surgery (RGS) in lung carcinoma by using ~18F-FDG. Methods Forty mice bearing lung adenocarcinoma were divided randomly into two gro ups according to presence or absence of the lung metastasis. A dose of 200?l (1 00?Ci) of ~18F-FDG was injected via the tail vein. The external radioimmu nography was performed after injection. All viscerae and tumor were detected i n vivo by means of a hand-held gammaray-detecting probe (GDP), and the radio active distribution was analyzed with the well-gammaray detector. Resul ts The tumor images in mice were clear, and the images were best at two hours after injection. The values of %ID/g of tumor and heart obtained by well -gammaray detector were higher than those of other organs. Ratios of tumor/norm al tissue (T/NT) except cardiac muscle being detected by GDP ranged from 3.71 t o 13.57. 5.The effects of molecules of the surface of colon carcinoma cells

Objective To investigate the effects of the HLA-Ⅰ and co-stimulators CD_(80)and CD_(86)on HT-29 cells surface, after heat killed Bifidobacterium and HT-29 cells were mingled culturing for 12 hours. Methods The HLA-Ⅰ and co-stimulators CD_(80)and CD_(86)on HT-29 cells surface were measured by Flow Cytometer. Results Heat killed Bifidobacterium can improve the expressions of HLA-1, CD_(80)and CD_(86)on HT-29 cells significantly, the livability of colon carcinoma cell was more than 90 %. Conclusion Heat killed Bifidobacterium have potential antitumorous features.
6.ENDOSONOGRAPHIC FEATURES OF MEDIASTINAL LYMPH NODES IN LUNG CANCER PATIENTS

The purpose of this study was to evaluate the features of mediastinal lymph nodes in lung cancer patients during endoscopic ultrasonography(EUS), and to establish the clinical method for the preoperative staging of lung cancer using EUS. The EUS patterns of 103 lymph nodes in 21 patients with lung cancer were evaluated. Confirmation of benign and malignant lymph nodes was obtained by surgical resection and pathology. The following features were compared between benign and malignant lymph nodes: short axis , long axis, distinct margins and the shape of nodes. The results showed that all of the lymph nodes were hypoechoic. Both the short axis greater than 1.0cm and distinct margins or round shape independently predicted malignant invasion ( P
