1.Retrospective study of nursing care adverse events at high altitude area of Lhasa city

Objective To understand the status and features of nursing care adverse events and to offer scientific basis for improving patient safety management of nursing at high altitude area. Methods Data such as categories, causes, time distribution and other factors of nursing care adverse events were collected from January 2013 to August 2014 in the People′s Hospital of Tibet Autonomous Region were analyzed. Results Over the study period, 50 adverse events were recorded and the most common types were tube emersion(26%,13/50), pressure sores(14%,7/50) and scald(12%,6/50) respectively. The proportions of different severity of adverse events were hidden events (48%,24/50), events without bad consequence(36%,18/50), adverse events(14%,7/50) and warning events(2%,1/50) respectively. The ineffective communication, inadequate assessment, and not abiding by the rules accounted for 70%(35/50) of the causes of adverse events. 7:00-10:59 and 19:00-22:59 period was the high incidence of adverse events, respectively accounted for 32%(16/50) and 26%(13/50). Conclusions The management of nursing adverse events at high altitude area need to consider the geographical characteristics,besides, to guarantee the implementation of the system,reduce the bed / nurse ratio, improve the comprehensive quality of nurses and other measures can be taken to reduce the occurrence of adverse events.
2.Effect of epigallocatechin - 3 - gallate against oxidative stress induced by high glucose in human lens epithelium cells

Abstract?AIM: To investigate the effect of epigallocatechin-3-gallate ( EGCG ) against oxidative stress induced by high glucose in human lens epithelium ( HLE) cells.? METHODS: The HLE cell oxidative damage model induced by high concentration glucose was established, and was intervented with different concentrations of EGCG. Cell viability was determined by MTT assay, cell morphology was investigated by convert microscope, cells apoptosis was assayed by flow cytometry with Hoechst-PI staining. Moreover, the levels of super oxide dismutase ( SOD) , glutathione peroxidase ( GSH-Px) and malondialdehyde ( MDA) in supernatant were also tested after different treatment either with high concentration glucose or with different concentrations of EGCG.?RESULTS: MTT results showed that HLE cells activity increased to 50. 33%± 3. 52% and 63. 33%± 4. 63% after treated with 10 μmol/L and 100 μmol/L EGCG respectively, the difference was statistically significant compared with oxidative injury group(32. 67%±3. 10%)(P<0. 05 ); HLE cells maintained better morphology intervented with EGCG under high glucose conditions, the number of apoptotic cells reduced, SOD and GSH-Px level within HLE cells increased and MDA levels decreased.?CONCLUSION:EGCG plays its strong antioxidant effect by increasing SOD, GSH-Px content and decreasing MDA content in cells, therefore provides a reliable experimental basis for the search for effective prevention and treatment of cataract drug.
3. A study on the demands and status of the development of specialty nurse in 16 Tibet hospitals

Objectives:
To find out the demands and status in quo of the development of specialty nursing and to provide positive suggestions for the further development of specialty nursing in Tibet.
Methods:
Self-designed questionnaire was adopted to investigate 16 different level hospitals across Tibet.
Results:
Among the 16 hospitals in which there were altogether 1677 nursing staff and only 11 hospitals had cultivated specialty nurses with a number of 123 in total. The rate of specialty nurses allocated was 7.2%. 9 hospitals expressed the need to develop specialty nurses in nearly future and the number of specialty nurses needed was 577.
Conclusion
Tibet should step up the development of specialty nursing, establish and perfect the training method and management system about specialty nurses as soon as possible.
4.Therapeutic observation of tuina manipulation for lumbar intervertebral disc herniation

Objective:To observe the clinical efficacy of balance-impact tuina therapy for lumbar intervertebral disc herniation (LIDH).Methods:A total of 118 eligible LIDH patients were randomized into an observation group and a control group by the random number table method,with 59 cases in each group.The observation group was intervened by balance-impact tuina therapy,while the control group was intervened by conventional tuina therapy,both for consecutive two weeks.The scores of visual analog scale (VAS),Oswestry disability index (ODI),quality of life questionnaire-core 30 (QOL-C30) were observed before and after treatment;the relapse rate was estimated at the sixth month and twelfth month following the treatment.The data were statistically analyzed.Results:After intervention,the total effective rate was 96.6% in the observation group versus 91.5% in the control group,and the between-group difference was statistically significant (P<0.05).The VAS and ODI scores declined significantly after treatment in both groups (all P<0.05),and the observation group was markedly lower than the control group (P<0.05,P<0.01).The QOL-C30 score increased significantly after treatment in both groups (both P<0.05),and the observation group was markedly higher than the control group (P<0.05).The relapse rates at the post-treatment sixth month and twelfth month in the observation group were lower than those in the control group (P<0.05,P<0.01).Conclusion:Compared with the conventional tuina therapy,the balance-impact tuina therapy shows advantage in lessening pain,improving the function and enhancing the quality of life in the treatment of LIDH,and it has a lower relapse rate.Thus,this therapy is worth promoting in clinic.
5.Application of Laboratory Indexes in Traditional Chinese Medicine Syndrome Differentiation of Multiple Myeloma

Objective To study the relationship between traditional Chinese medicine (TCM)syndrome differentiation of multiple myeloma(MM)and laboratory indexes of MM,thus to supply evidence for the syndrome differentiation of MM. Methods A total of 59 MM patients were enrolled in the study. TCM syndrome differentiation was performed. And we also collected the laboratory indexes of serum protein electrophoresis (SPE),immunofixation electrophoresis (IFE),blood urea nitrogen (BUN),creatinine (CREA),serum beta 2 microglobulin,serum calcium(Ca),hemoglobin(Hb),Bence Jones protein,flow cytometric-immunophenotyping(FCM) and bone marrow cytology. The differences of the laboratory indexes in various syndrome types of MM were analyzed. Results No significant differences were found in myeloma cell morphology, M protein content, IFE typing results, Bence Jones protein positive rate and the concentration of Ca of various syndrome types of MM (P >0.05). However,the concentrations of BUN,CREA,serum beta 2 microsglobulin,Hb,and the positive rates of CD19- and CD56+ were significantly different in various syndrome types of MM (P < 0.05 or P < 0.01). Moreover,Hb,BUN,CREA,and serum beta 2 microspheres were significantly correlated with each other(P<0.05 or P < 0.01). Conclusion Hb, BUN, CREA, blood beta 2 microglobulin and FCM detection can be sued for the auxiliary diagnosis of syndrome types of MM, and for preliminary demonstration of illness severity and prognosis. The results will provide the basis for the treatment of MM with traditional Chinese medicine and western medicine.
6.Animal experimental study of the treatment of periapical disease with recombinant human bone morphogenetic protein-2 composite.

OBJECTIVETo investigate the effect of the treatment of periapical diseases with recombinant human netic protein (rhBMP-2) composite in dog models so as to provide basis for its clinical application.
METHODSThe endotoxin-bacteria mixed fluid was injected into each root canal of the experimental teeth of dogs and the animal periapical were built. The rhBMP-2 composite was mixed by rhBMP-2, TCP, collagen and metronidazole and used in the treatment cal diseases in dog models as root apex screen. The imageology and pathology research were went on.
RESULTSThe effective rate of the experimental group was remarkably higher than that of the carrier group and the control group with remarkable ference (P < 0.05). The quality of the repair of tissue was obviously better than the two other groups.
CONCLUSIONThe rhBMP-2 composite is a promising biological root-canal filling material.

8.Clinical study on correlativity of stagnation of the liver-qi with tenderness response of auricular points.

OBJECTIVETo probe into the correlativity of TCM syndromes to auricular points.
METHODSSixty-three cases of typical liver-qi stagnation syndrome and 52 healthy persons were divided into an observation group and a control group, respectively. The auricular points on both side were respectively detected by auricular point pressure pain method with spring pressure bar and a pressure of 50 g, and then the number of auricular points with positive response on both sides were respectively calculated and the positive rates between the two groups were compared.
RESULTSCan (liver), Shidao (esophagus), Sanjiao (triple energy), Pi (spleen), Yidan (pancreas and gall bladder), Tingzhong (center of superior concha), Wei (stomach), Shenmen had high tenderness positive rates, with significant differences between the liver-qi stagnation syndrome group and the healthy person group. The correlativity were Gan>Shidao>Sanjiao>Pi>Yidan>Tingzhong>Wei>Shenmen.
CONCLUSIONTCM syndromes and auricular points have correlativity.

9.Enrichment of breast cancer stem cells using a keratinocyte serum-free medium.

BACKGROUNDKeratinocyte serum-free medium (K-SFM) is a defined medium used to support the growth of primary keratinocytes and embryonic stem cell. The aim of this research was to optimize enrichment of breast cancer stem cells (CSCs) using K-SFM.
METHODSA K-SFM was used to enrich CSCs from two breast cancer cell lines and a primary culture of breast cancer. RPMI-1640 supplemented with 10% fetal calf serum (FCS) was used as a control. CSCs were identified with flow cytometry using CD44(+)/CD24(-) as molecular markers. The expression of a variety of CSC markers (Oct-4, ABCG2, Nanog, N-cadherin, and E-cadherin) was analyzed with real-time PCR.
RESULTSMuch higher percentage of CSCs was achieved with K-SFM: 17.3% for MCF-7 cells, 17.4% for SKBR-3, and 20.0% for primary breast cancer culture. Less than 1% CSC was achieved using RPMI-1640 supplemented with 10% FCS. In comparison to the CSCs obtained with RPMI-1640, CSCs in the K-SFM expressed higher levels of Oct-4, ABCG2, Nanog and N-cadherin, and lower level of E-cadherin.
CONCLUSIONK-SFM is an optimal culture medium to maintain and to enrich breast CSCs.

10.Construction of sRNA-deficient and-overexpressing strains of Yersinia pestis

Objective To construct small RNA deletion and overexpression strains with a length of less than 100 nt in Yersinia pestis.Methods Deletion mutants of the target sRNAs were constructed by increasing the length of homologous regions.Meanwhile, the high copy plasmid pBAD/HisA was modified into an inducible transcriptional vector as an sRNA-overexpression plasmid by using QuikChange lightning site-directed mutagenesis kit .The presence , size, and transcription-al initiation sites of the indicated sRNA were predicted by transcriptome sequencing , primer extension , and previous stud-ies.The full-length DNA fragments of target sRNAs were transformed into the transcriptional vector .The overexpressing strains of sRNAs were identified by Northern Blot .Results and Conclusion Four sRNAs deletion mutants of sR01, sR02, sR03 and HmsA and three sRNAs overexpression mutants MicF , HmsA and CpxQ were successfully constructed .A method of construction of sRNA deficient and overexpressing strains of Y.pestis has been quickly and efficiently established by λ-Red homologous recombination technology and QuikChange ? lightning site-directed mutagenesis kit.
