1.Antimutagenic Mechanisms of Hotbed Chives, Fragrant-Flowered Garlic and Garlic Leaves

In the present paper, antimutagenic mechanisms of hotbed chives, fragrant-flowered garlic and garlic leaves were investigated by the SOS Chromotest The results showed that these vegetables could inhibit the SOS respones induced by temperature (42℃) in E coli GW1060 and GW11M (rec 441 (tif)), but they could not act on SOS network gene expression in E. coli GW2707 (lexA::Tn5), so one of their antimutagenic mechanisms is inhibitory effect on cleavage of lexA by RecA protease. Desmutagenic test results indicate that some aqueous extractions of the three vegatables can inactivate mutagens outside cells.
2.Comparative observation of C5 nerve root palsy after posterior surgical treatment of multilevel cervical spondylotic myelopathy

Objective To compare the incidence of C5 nerve root palsy after laminoplasty and laminectomy with internal fixation for treating multilevel cervical spondylotic myelopathy (MCSM).Methods From January 2008 to August 2012,98 patients with MCSM were treated with laminoplasty (47 patients,group A) or laminectomy (51 patients,group B) with internal fixation.All the patients were followed up for 13-56(26.5 ± 7.9) months.In both groups,Cobb's method was applied to measure cervical lordotic angle,and Ishihara's method was conducted to measure cervical curvature index (CCI) before and after operation.The incidence of C5 nerveroot palsy was recorded and compared.Results The incidence of C5 nerve root palsy in group A was 2.1% (1/47),while 21.6 % (11/51) in group B (x2 =5.430,P < 0.05).The JOA scores in group A and group B before and after operation and improvement rate of JOA scores had no significant difference (P> 0.05).The cervical lordotic angle and CCI in group A and group B before and after operation had no significant difference (P > 0.05).The improvement rate of CCI between two groups had no significant difference (P > 0.05).All of 11 patients with C5 nerve root palsy were group B 1,and other 40 patients were group B2.The improvement rate of CCI in group B1 was significantly higher than that in group B2 [(38.7 ± 18.3)% vs.(22.1 ± 12.1)%](t =1.772,P< 0.05).Conclusions Compared with laminoplasty,laminectomy with internal fixation has a higher incidence of C5 nerve root palsy.The C5 nerve root palsy may be associated with postoperative increase of cervical lordosis angle.Moreover,tethering of the C5 root may he one of its important pathomechanisms.
3.Isolation and Identification of Antimutagenic Constituents in Hotbed Chives

Fresh hotbed chives solutions were extracted respectively by ether, acetic ether and n-butyl alcohol and protein constituents and non-protein constituents were obtained. Ether extractions (non-protein constituents) had the strongest antirriutagenic effects by the SOS chromotest Non-protein constituents of hotbed chives were analysed further by gas chromatography-mass spectrum and 30 chemical substances were isolated and identified. Their antimutagenic substances may be dimethyl disulfide, 4-methyl-2-pyridinethione, di-2-propenyl trisulfide, 3-(allylthio)-propionic acid.
4.One-step methylation variable position analysis technology in single-tube.

OBJECTIVE:
To develop the single-tube one-step methylation variable position (MVP) analysis technology-single-tube post-digestion PCR-melting curve analysis (PDP-MCA).
METHODS:
Based on differentially methylated region (DMR) reported previously as the model, a set of primers with different melting temperatures of products in the two sides of MVP were designed. By using the FastDigest methylation-sensitive restriction enzyme (MSRE), DNA digestion, multiplex amplification, MCA detection and MCA profiles were performed in a single reaction tube. Same samples (peripheral venous blood, semen, and vaginal fluid, 5 samples each type) were tested by single-tube one step MVP and traditional MSRE-PCR MCA technology. To verify the feasibility of this method, the results were compared with that of the traditional technology. The MCA/HRM profiles of different samples were analyzed and compared.
RESULTS:
When the melting temperature of the fragments had a differential of 2 degrees C, the MCA melting peaks separated well, and MCA detection after multiplex amplification was successful. The single-tube PDP-MCA assay was developed, which integrated multiple reactions (digestion, amplification and detection) into one tube. By this method, the sample-specific profiles and data were analyzed in 2 h, which is similar to that of the traditional method. The rapid classifications of the samples were also realized.
CONCLUSION
Multiplex MVPs can be analyzed in a single closed-tube. The single-tube PDP-MCA technology is a simple, fast, and automatable method. It can be used for detection of DNA methylation variations.

6.Meta-analysis on the relationship between IL8-251 gene polymorphism and gastric cancer.

OBJECTIVETo evaluate the relationship between IL8-251 gene polymorphisms and gastric cancer.
METHODSLiteratures were reviewed and selected based on the criteria for inclusion. The Meta-analysis software, REVMAN 4.2, was applied to check the heterogeneity across the studies and calculating the pooled OR.
RESULTSTotal of 2114 cases and 2505 controls from 8 studies for IL8-251 were included. The chi(2) value was 21.48 (P = 0.003), and the pooled OR of (AA + AT) vs. TT was 1.12 (95% CI 0.90 - 1.40). Large heterogeneity was found among the studies. After the sensitivity analysis, the pooled OR of (AA + AT) vs. TT 1.21 (95% CI 1.06 - 1.39).
CONCLUSIONIL8-251-A allele might be associated with higher risk of developing gastric cancer.

7.Molecular mechanisms involved in the treatment of inflammatory bowel disease by Pulsatilla decoction.

OBJECTIVETo investigate molecular mechanisms underlying in the treatment of inflammatory bowel disease by Pulsatilla decoction.
METHODSWistar male rats were randomly divided into control group, model group, model + positive control group (mesalazine), traditional Chinese medicine treatment group, in addition, the Chinese medical treatment group was divided into middle and high dose group ( n = 8). Intragastric administration was used in the positive control group and traditional Chinese medicine treatment group. The expression of Smad7 and p-Smad3 in the colons were detected by immunohistochemistry and Western blot.
RESULTSCompared with the model group, positive medicine and traditional Chinese medicine group, especially high-dose group, could effectively inhibit the expression of Smad7, while enhancing the p-Smad3 expression.
CONCLUSIONThe activation of TGF-beta1/Smad3 signaling pathway may be the molecular mechanism underlying in the anti-inflammatory effect of inflammatory bowel disease by Pulsatilla decoction.

8.A case of peripartum cardiomyopathy with recurrent ventricular fibrillation rescued by extracorporeal membrane oxygenation

Peripartum cardiomyopathy (PPCM) often develops in the early postpartum period, especially within one month after delivery. However, in recent years, some cases of pregnancy-related cardiomyopathy with earlier or slightly later onset showed similar features with PPCM entirely. One patient with PPCM was admitted to the Department of Intensive Care Medicine of the First Affiliated Hospital of Bengbu Medical College on February 2022. After admission, the patient was exceptionally hemodynamically unstable and had repeated ventricular fibrillation. Apart from extracorporeal membrane oxygenation (ECMO) support, comprehensive treatment with cardiotonic agents and drugs to inhibit the "ventricular electric storm" was given. The cardiac structure and function were gradually improved. ECMO was successfully withdrawn on the 7th day after the onset of symptoms, and the patient was discharged after a full recovery. Early diagnosis and active intervention are the keys to improve the prognosis of patients with PPCM and avoid irreversible changes in cardiac structure.
9.Clinical trial of tiotropium combined with sulpiride in the treatment of patients with bronchial asthma

Objective To investigate the clinical effect of tiotropium combined with seretide in the treatment of patients with bronchial asthma.Methods Ninety cases of bronchial asthma were randomly divided into treatment group and control group,each group 45 cases.Control group was given seretide accuhaler.On the basis of control group,treatment group was given tiotropium inhalation powder spray treatment,18 μg each time,1 times a day.All patients were treated for 12 weeks.The pulmonary function changes,eosinophil cationic protein (ECP),interleukin-6 (IL-6),tumor necrosis factor-α (TNF-α),the clinical effect and treatment satisfaction were compared between the two groups.Results After treatment,total effective rates in control group and treatment group were 82.22% (37 cases/45 cases),97.78% (44 cases/45 cases),with significant difference (P < 0.05).After treatment,the levels of forced expiratory volume in one second (FEV1) in treatment and control groups were (1.46 ± 0.32),(1.08 ± 0.25) L,the levels of peak expiratory flow (PEF) were (4.35± 1.12),(3.77 ± 1.02) L · s-1,the levels of forced vital capacity (FVC) were(2.18 ±0.36)%,(1.71 ± 0.31) %,all with significant difference (all P < 0.05).After treatment,the levels of ECP in treatment and control groups were (160.60 ± 20.30),(289.70 ± 25.60) ng · mL-1,the levels of IL-6 were (28.90 ± 5.70),(53.70 ± 8.80) pg · mL-1,the levels of TNF-α were (10.40 ± 2.30),(17.60 ± 3.10) ng · mL-1,all with significant difference (all P < 0.05).The adverse drug reactions in treatment group were palpitations,hoarseness,with the incidence of 11.11% (5 cases/45 cases).The adverse drug reactions in control group were hoarseness,with the incidence of 8.89% (4 cases/45 cases),with no significance difference (P > 0.05).Conclusion The clinical effect of tiotropium combined with seretide in the treatment of patients with bronchial asthma is great,and can increase patients' treatment satisfaction.
10.Endothelial progenitor cell transplantation ameliorates elastin breakdown in a Kawasaki disease mouse model.

BACKGROUNDCoronary artery damage from Kawasaki disease (KD) is closely linked to the dysfunction of endothelial progenitor cells (EPCs). The aim of the present study was to evaluate the therapeutic effect of EPCs transplantation in KD model.
METHODSLactobacillus casei cell wall extract (LCWE)-induced KD model in C57BL/6 mice was established. The model mice were injected intravenously with bone marrow-derived in vitro expanded EPCs. Histological evaluation, number of circulating EPCs and the function of bone marrow EPCs were examined at day 56.
RESULTSInflammation was found around the coronary artery of the model mice after 14 days, Elastin breakdown was observed after 56 days. CM-Dil labeled EPCs incorporated into vessel repairing foci was found. At day 56, the number of peripheral EPCs in the KD model group was lower than in EPCs transplanted and control group. The functional index of bone marrow EPCs from the KD model group decreased in proliferation, adhesion and migration. Increased number of circulating EPCs and improved function were observed on the EPCs transplanted group compared with model group.
CONCLUSIONExogenously administered EPCs, which represent a novel strategy could prevent the dysfunction of EPCs, accelerate the repair of coronary artery endothelium lesion and decrease the occurrence of aneurysm.
