1.Expression of Exon 5 - Deleted Estrogen Receptor mRNA in Human Uterine Leiomyoma and Myometrium.

OBJECTIVE: There is a large and increasing body of experimental and clinical data supporting the existence of variable estrogen receptors in both normal and neoplastic estrogen target tissue. Recently, exon 5 deleted estrogen receptor(D5-ER) has been identified in many estrogen dependent tumors. The purpose of this study is to assess the relationship between uterine myoma and D5-ER. METHODS: The relative amount of wild type estrogen receptor mRNA and D5-ER mRNA were examined in human myometrium and myoma tissue obtained from 12 myoma patients and 12 control subjects by RT-PCR. The correlation between WT/D5-ER ratio and age was also evaluated. RESULTS: There was no difference in expression of D5-ER mRNA not only between myoma and myometrim of the myoma patients, but also between myometrium of myoma patients and normal subjects. No significant correlation was found between D5-ER expression and age of the subjects in myometrial tissue. However, we could find negative correlation between WT/D5-ER ratio and age in myoma tissue. CONCLUSION: Upon these data, we conclude that D5-ER is a common variant mutation of estrogen receptor, which is not related with development of myoma. However, in myoma tissue, there is possibility that D5-ER may act some role in the growth of tumor.

2.Toxic Effect of Cryoprotectants on Embryo Development in a Murine Model.

OBJECTIVES: The aim of this study was to assess toxicities of cryoprotectants. METHODS: Toxicities of two cryoprotectants, dimethyl sulfoxide (DMSO) and 1,2-propanediol (PROH), were investigated using a murine embryo model. Female F-1 mice were stimulated with gonadotropin, induced ovulation with hCG and mated. Two cell embryos were collected and cultured after exposure to either DMSO or PROH. Embryo development was evaluated up to the blastocyst stage. Blastocysts were stained with bis-benzimide to evaluate the cell count and with terminal deoxynucleotidyl transferase mediated dUTP nick labeling (TUNEL) to assess apoptosis. RESULTS: The total cell count of blastocysts that were treated with DMSO at the 2-cell stage was significantly lower than that were treated with PROH (75.9+/-27.0) or the control (99.0+/-18.3) (p<0.001). On comparison of two cryoprotectant treated groups, the DMSO treated group showed a decreased cell count compared with the PROH treated group (p<0.05). Both DMSO (14.2+/-1.5) and PROH (11.2+/-1.4) treated groups showed higher apoptosis rates of cells in the blastocyst compared with the control (6.2+/-0.9, p<0.0001). In addition, the DMSO treated group showed more apoptotic cells than the PROH treated group (p<0.001). CONCLUSIONS: The potential toxicity of cryoprotectants was uncovered by prolonged exposure of murine embryos to either DMSO or PROH at room temperature. When comparing two cryoprotective agents, PROH appeared to be less toxic than DMSO at least in a murine embryo model.

Infectious keratitis is the most common serious ocular infection, and may be caused by various bacteria, fungi, viruses and parasites. The authors performed prospectively an epidemiological study to identify risk factors and causative organisms, and to evaluate clinical manifestations, methods and results of treatment in infectious keratitis under the identical protocol from April 1995 to September 1997. Logistic regression analysis [univariate analysis and multivariate analysis] was used to evaluate possible risk factors. Six hundred sixty cases of infectious keratitis reported from 19 hospitals were studied. Two hundred eighty-three organisms[247 bacteria, 32 fungi, 4 acanthamoeba] were detected in 626 eyes with infectious keratitis excluding 34 pherpetic keratitis. The Pseudomonas aeruginosa, coagulase negative staphylococcus, Staphylococcus aureus and Serratia marcescens were the major orgnisms in bacterial keratitis. Aspergillus, Fusarium and Candida were the major isolates in fungal keratitis. Contact lens wear was a risk factor for bacterial keratitis. Female, age[less than 40 years] and occupation[student, house-wife, office worker, servise] were associated with bacterial keratitis. Risk factors in herpetic keratitis were age[between 40 and 59 years] and ocular adnexal diseases. Male was associated factor with herpetic keratitis.
