1.Effect of Fasudil on Inflammatory Secretion of Astrocytes

Objective To observe the effect of Fasudil on inflammatory factors (TNF-α and IL-8) secreted by astro-cytes.Methods Primary rat spinal astrocytes were cultured and randomly divided into three groups :control group ,oxygen-glu-cose deprivation group (OGD group) and Fasudil group.The culture media were harvested at different time points (0 ,3 ,6 ,12 , 24 h) for detection of the concentration of TNF-αand IL-8 by ELISA.Results Compared with the control group ,the concentra-tions of TNF-αand IL-8 in OGD group were significantly increased at each time point (P<0.05);in Fasudil group ,the concen-trations of TNF-αand IL-8 increased at some time points ,but they were much lower than in OGD group (P<0.05) .Conclusion OGD can promote the secretion of TNF-αand IL-8 by astrocytes in vitro ,which can be effectively inhibited by Fasudil.It is suggested that Fasudil plays an important role in regulating the levels of TNF-αand IL-8 secreted by astrocytes.
2.Intervention effect of naloxone on scopolamine-induced impairment of spatial working memory in rats

BACKGROUND: The pathway of cholinergic nerve also with rich opium receptor in the septum-hippocampus areas is an important part for spatial learning memory. There is study that naloxone, an antagonist of opium receptor, can relieve the inhibition of activity of cholinergic neuron caused by opioid peptides in the medial septum and hippocampus, promote the release of acetylcholine (Ach)into synaptic cleft, and improve the impairment of spatial memory.OBJECTIVE: To investigate the intervention effect of naloxone on impairment of spatial working memory of rats induced by scopolamine, a blocker of cholinergic muscarinic receptor.DESIGN: A randomized and controlled trial taking experimental animals as subjects.SETTING: Geriatric Department of Wuzhongpei Memorial Hospital at Shunde District of Foshan City.MATERIALS: The experiment was completed from 2003 to 2004 at the Experimental Animal Center of Medical School of Shantou University. A total of 21 SD rats of clean grade were at random divided into three groups:normal group, scopolamine group and naloxone group (scopolamine and naloxone), with 7 in each group. Build up of Morris water maze: A circular rubber pool, 2 m diameter and 55 cm height, with 30 cm depth of water at20℃-22℃, was built up. At the center of its southeast quadrant, a cylinder platform installed, which was 12 cm in diameter and 1.5 cm below the water surface, the only place for rats to escape death. The water of the pool was covered with bits of white polythene. In the house, there were light tubes, windows and tables in fixed place served as the outside cues of the pool. On the four walls of the pool, there were several randomly-hung and eye-catching markers as inside cues.METHODS: Medicines were intraperitoneally given 30 minutes before the experiment for rats in each group, scopolamine 0.4 mg/kg and naloxone 3mg/kg in two treatment groups, the equal volume saline in normal group.Then escape latencies of rats were recorded for two successive trials through a delayed-matching\task in self-made Morris water maze. After the Morris water maze examination, all rats were put to death, the tissue of left brain was selected, conventionally treated and sliced. The expression level of cholinacetyltranslase (ChAT) in areas of hippocampus and prefrontal lobe were quantitatively assayed by immunohistochemistry and the computerized image technique. At the same time, the tissue of right brain in two rats of each group was selected, the hippocampus was immediately freed, and the change of CAI region of the hippocampus observed with electron microscope.trastructure of CA1 region of hippocampus in rats.cies of two successive trials were very significantly different in normal group[(46.4±17.7)vs (13.4±8.2)s, t =7.32, P< 0.01], and significantly different in naloxone treatment group[ (27.9±14.3)vs (9.0±3.8)s, t=3.19, P< 0.05], but not pression of ChAT in pyramidal cells in CA1 and CA3 regions of hippocampus and in neurons of prefrontal cortex of three groups was not different statisticalhippocampus in all groups was normal; However, the ultrastructure of the neuron synapsis was abnormal obviously in scopolamine group: The synaptic vesicles were markedly different in size; the vesicles obviously gathered in the part of presynaptic membrane with an enhanced density, and large and sparse in the distal part. Both presynaptic and postsynaptic membranes were indistinct and fused, synaptic cleft not clear, and postsynaptic dense materials not homogeneous, indistinct and without clear borderline.CONCLUSION: Scopolamine could impair the power of spatial working memory in rats. Naloxone could promote great release of Achin neuron synaptic vesicles into synaptic clefts, and increase the density of postsynaptic dense materials, relieve the inhibition of opioid peptide to cholinergic neuron activity in the medial septum and hippocampus, and could improve space memory injury.
3.The expression and significance of IGFBP-3 in salivary gland pleomorphic adenoma

Objective:To investigate the expression of insulin-like growth factor binding protein 3 (IGFBP-3) in salivary pleomorphic adenoma(SPA).Methods:The expression of IGFBP-3 protein in 40 cases of SPA(group SPA),40 of normal glandular tissue(group N) and 10 of salivary gland malignant tumor(group CA) was detected by Western blot.The expression of IGFBP-3 mRNA in 50 cases of SPA,50 of salivary gland normal tissue and 10 of CA was detected by qRT-PCR.Results:The expression(A value) of IGFBP-3 protein in group N,SPA and CA was 8.54 ± 3.95,4.78 ± 2.07,3.63 ± 2.27 respectively.The expression ration of IGFBP-3 mRNA of group N vs SPA or CA,P < 0.05;SPA vs CA,P > 0.05 (SPA/N was 0.654 ± 0.387,CA/N:0.452 ± 0.229) respectively,but showed no significance difference between SPA and the CA groups(P > 0.05).Difference of IGFBP-3 mRNA expression was observed with different envelope infiltration of SPA (P < 0.05),no significant difference was observed in different age,gender or relapse groups.Conclusion:IGFBP-3 Low expression of IGFBP-3 in pleomorphic adenomas may reduce the antagonism of IGF-1R,causing the proliferation of tumor cells and promote tumor formation.
4.The expression and significance of DNMT1 gene in oral squamous cell carcinoma

The expression of DNA methyltranferase 1 (DNMT1 )mRNA and protein in 20 controls of normal oral mucosa tissue and 43 cases of oral squamous cell carcinoma(OSCC)was detected by Real-Time PCR and immunohistochemical staining and Western blot respectively. DNMTl mRNA CT values in OSCC and the controls were 0.958 6 ±0.986 6 and 0.459 5 ±0.525 8 respectively(Z =-2.028,P <0.05), The positive expression of DNMT1 protein in OSCC and the controls was 87% and 25% respectively(P <0.05).DNMT1 may play a role in the development of OSCC.
5.The effect of acupuncture combined with auricular application for puerperal hypogalactia

Objective To observe the effect of acupuncture and auricular application in the treatment of puerperal hypogalactia.Methods 300 patients with puerperal hypogalactia,according to the principle of random, balanced,together,were divided into 3 groups:Chinese herbal medicine group and acupuncture combined with auricular group and massage group,each group in 1 00cases.Collected patients'informed consent,and obtained the hospital ethics committee approval.The clinical curative effect was observed.Results After 3 days,in the Chinese herbal medicine group 34 cases recovered,24 cases were markedly effective,efficient in 1 5 cases,27 cases were invalid.In the massage group 7 cases recovered,1 3 cases had marked effect,1 5 cases were effective,ineffective in 65 cases.In the acupuncture combined with auricular group 73 cases cured,1 3 cases were markedly effective,effective in 9 cases, 5 cases were ineffective.The total effective rate between the acupuncture combined with auricular group and Chinese herbal medicine group had significant difference(χ2 =9.600,P <0.01 ).The total effective rate between the acupuncture combined with auricular group and the massage group had significant difference (χ2 =1 3.400,P <0.01 ).The total effective rate between Chinese herbal medicine group and massage group had significant difference (χ2 =6.400,P <0.01 ).Conclusion Acupuncture and auricular application has obvious curative effect for patients with postpartum hypogalactia.

Objective To research the best extracting technology of simiaoyongan decoction.Methods As the total contents of chlorogenic acid and harpagoside to be evaluation indicator, adopted L9 (34 ) orthogonal design to select out the best extraction.The main determination was by HPLC method.Results Accounted by the total contents of the two composi-tions, the influential effects of the experimental results were extraction solvent >reflux times >the volume of extraction>the circumfluence time.Conclusion The best extracting technology is adding 10-fold times of 50% alcohol, percolating 1 hours, and three times.

Objective To study the changes of arterial blood lactate(ABL) and serum enzymes activities and thier correlation.Method Lactate dehydrogenase(LDH),alkaline phosphatase(AKP) and creatine kinase(CK) in serum were determined in sixty-four critical patients in the intensive care unit(ICU) at admission immediately,following 24h,48h and 72h,respectively.The patients was divided into three groups:favorable,survival and death groups.Results Before treatment,ABL level was(3.9?2.5,2.9?1.3 and 3.8?2.3)mmol/L in favorable,survival and death groups respectively.Although ABL in favorable and survival groups were decreased gradually to normal level during 24~72h,the death group showed that the decline was very slowly it still persistent elevated in 72h.There was significanct difference between the death group and the favorable and survival groups(P

10.Quantitative analysis of the patterns of heat shock protein 70 expression induced by Hantaan virus infection in vitro

Objective To explore the pattern and quantify the heat shock protein HSP)70 and HSP70 mRNA in Vero-E6 cells after infection with Hantann virus(HTNV).Methods The expres- sion of HSP70 and change of its mRNA level were detected by immunocytochemical staining,nucleic acid hybridization in situ and RT-PCR.Results In situ hybridization and RT-PCR were used to eval- uate the level of HSP70 mRNA during Hantaan 76-118 infection.HSP70 mRNA increased 0.5 h after infection,reached its peak by 12 h and gradually declined to steady state level by 72 h(vs.sham infec- ted group,P<0.05).The expression of HSP70 protein induced by Hantaan 76-118 infection was e- valuated by quantitative immunocytochemical staining.HSP70 increased 0.5 h after infection,reached its peak by 12 h and decreased at 72 h after infection(vs.sham infected group,P<0.05).Conclu- sions HSP70 can be induced directly by HTNV infection at both mRNA and protein levels,It pro- vides a basis for the further study of the pathogenesis,prevention and treatment of hemorrhagic fever with renal syndrome(HFRS).
