1.Resveratrol induces apoptosis in human primary gastric carcinoma cells

AIM: To investigate the apoptosis in primary gastric cancer cells induced by resveratrol, and the relation between this apoptosis and expression of bcl- 2 and bax. METHODS: In in vitro experiments, MTT assay was used to determine the cell gowth inhibitory rate. Transmission electron microscopy and TUNEL staining were used to quantitatively and qualitively detect the apoptosis of primary gastric cancer cells before and after the resveratrol treatment. Immunohistochemical staining and RT - PCR was used to detect the expression of apoptosis - regulated gene bcl - 2 and bax. RESULTS: Resveratrol inhibited the growth of primary gastric cancer cells in a dose - and time - dependent manner. Resveratrol induced primary gastric cancer cells to undergo apoptosis with typically apoptotic characteristics. TUNEL assay showed that after the treatment of primary gastric cancer cells with resveratrol for 24, 48, 72, 96 hours, the apoptotic indexs were 4.93% ± 0.19%, 16.74% ± 0.43%, 27.88% ±0.36%, 36.84% ± 1.07 % respectively. Immunohistochemical staining showed that after the treatment of primary gastric cancer cells with resveratrol for 24, 48, 72, 96 hours, the positive rates of Bcl - 2 proteins were 20.68% ± 0.49%, 10.84% ±0.33%, 6.80% ± 0.34%, 3.91% ± 0.15% and the positive rates of Bax proteins were 19.79% ± 0.98%, 30.74% ±0.85%, 40.14% ± 1.17%, 60.08% ± 1.64%. After exposed to resveratrol for 24 h, 48 h, 72 h and 96 h, the density of bcl- 2 mRNA decreased progressively with elongation of time and the density of bax mRNA increased progressively with elongation of time by RT- PCR. CONCLUSION: Resveratrol is able to induce the apoptosis in primary gastric cancer. This apoptosis may be mediated by down- regulation of Bcl- 2 and up- regulation of Bax.
2.Studies on the Change of the Content of Polysaccharides in Crude and Processed Fructus Corni

Objective To investigate the change of the content of polysaccharides in crude and processed Fructus Corni,thus to approach the process mechanism of Fructus Corni.Methods The content of polysaccharides in the crude and processed Fructus Corni was determined by phenol-sulfuric acid method.Results After processing with wine,the content of polysaccharides of Fructus Corni decreased from 10.12 %(content of total polysaccharidesbeing 51.41 %) to 5.91 %(content of total polysaccharidesbeing 53.10 %),decreased by 41.60 %as compared with that in the crude Fructus Corni.Conclusion After processing with wine,the content of polysaccharides decreases markedly.The results provide certain evidence for approaching the process mechanism of Fructus Corni.
3.Quality Standard of Yanyan Granules of Concentrated Sugar-free Type

OBJECTIVE:To establish the quality standard of Yanyan granules of concentrated sugar-free type. METHODS:Scrophularia ningpoensis,Menispermum dauricum,Radix et Rhizoma Glycyrrhizae in the granules were identified by TLC,and the content of linarin in Yanyan granules were determined by HPLC.RESULTS:The TLC spots were clear with high resolution.The linear range of linarin was 4.64~69.6 ?g(r=0.999 9) and the average recovery rate was 98.65%(RSD= 0.79%,n=6). CONCLUSION:The established standard is suitable for the quality control of the Yanyan granules of concentrated sugar-free type.

6.Detection of miRNA gene sequence variations in multiple myeloma and its significance

Objective To investigate sequence variations of 12 miRNA genes in multiple myeloma(MM) in order to find whether sequence variations in miRNA genes are associated with tumorigenesis and discuss the clinical significance of MM associated with miRNA genes mutations. Methods The miRNA gene mutations in 20 cases of MM, 4 MM-derived cell lines and 20 controls were detected by the methods of polymerase chain reaction single stranded conformation polymorphism (PCR-SSCP) and silver staining technique. Both clinical features and laboratory results were analyzed simultaneously. Results The electrophoretic patterns showed a total of three variations in miR-19a, miR-19b and miRNA-335,which were observed in 3 MM cells (15 %, 3/20). We also found variations of miRNA-335 in MM-derived cell lines KM-3and RPMI8226. However, no sequence alteration in the miRNA genes was observed in our set of controls. One of the three MM patients died, and two of them were detected mutations at the terminal stage of the disease.Conclusion A relative high frequency of miRNA gene mutation was found in MM and MM derived cell lines, which suggests possibility of a main mechanism underlying tumorigenesis. And, detecting miRNA gene mutations in MM might be benefit to evaluate the progression and prognosis of disease.

Receptors are primary determinant of viral tropism and disease pathogenesis.Heparan sulfates (HS)are ubiquitous,polyanionic carbohydrate chains linked to core proteins in cell membranes and ex- tracellular matrices of all eukaryotes.HS have also been demonstrated to function as receptors or co-receptors for a number of different viruses.To date,HS and four RGD-dependent integrins,?v?3,?v?6, ?v?1,and?v?8 have been reported to serve as receptors for Foot-and-mouth disease virus(FMDV).Different receptors may be used to interact with host cells during FMDV infection.Studies on the structure and function of receptors are very important for understanding the interaction between host cells and FMDV. Here,We mainly reviews the progress on the biological characteristics of HS and its roles in FMDV infection.
10.EDI-OCT detecting retinal and choroidal thickness in patients with pregnancy induced hypertension

AIM: To observe and compare the changes of foveal retinal and choroidal thickness in patients with pregnancy-induced hypertension (PIH) by enhanced depth imaging optical coherence tomography (EDI-OCT). METHODS: Totally 150 patients (289 eyes) diagnosed with pregnancy-induced hypertension were identified by fundus examination and EDI-OCT after mydriasis. The patients were divided into four groups for the Duke-Elder class: P1 ( vasospasm ), P2 ( angiosclerosis ), P3 (retinopathy), and control group P0(normal fundus). The average retinal nerve fiber layer thickness in each quadrant, foveal retinal and choroidal thickness were measured by EDI-OCT respectively. RESULTS: There were 36 (72 eyes, 24. 9%) of 150 women with pregnancy- induced hypertension had no retinal changes,and 114 (217 eyes, 75.1%) of 150 cases identified by clinical examinations as having retinal findings. The retinal and choroidal thickness varied with different stages of fundus changes. The RNFL thickness of central subfield(CSF),the foveal retina and choroid were significantly higher in angiosclerosis stage than that in normal fundus group (P<0. 05). The RNFL from each area, the foveal retina and choroid were all thicker in retinopathy stage than the other three groups (P<0.05). CONCLUSION: EDI-OCT is one important tool for the study of fundus changes in PIH. Findings in the morphology changes of retina and choroid with the help of EDI - OCT may indicate more about retinal microcirculation changes caused by pregnancy-induced hypertension in depth.
