1.Essence of professor SHI Xin-de's experience in application of persicae ramulus-associated prescriptions.

Persicae Ramulus decoction, as the first prescription in Treatise on febrile Diseases, has been recommended by physicians of successive generations. It is also the general prescription for harmonizing yingfen and weifen, yin and yang, qi and blood. Although it only consists of five herbal medicines, it has a wider range of application and more categorized formulas than other prescriptions. Though Persicae Ramulus decoction was originally formulated to treat taiyang apoplexy, it has functions beyond the treatment of exopathic diseases. This formula is also effective in treating internal diseases, surgical diseases, gynecologic diseases, paediatric diseases, etc. KE Yun-bo praised it as the No. 1 formula among ZHANG Zhong-jin's formulas as well as the general prescription for harmonizing Yin and Yang, yingfen and weifen, resolving fleshy exterior and inducing perspiration. Professor SHI Xin-de has been expert at treating intractable diseases by using Persicae Ramulus-associated prescriptions, such as Xiaojianzhong decoction and Baohe pill for children's chronic eczema, Persicae Ramulus and Puerariae Lobatae Radix decoction and Yupingfeng powder for chronic nephritis, and Persicae Ramulus and Longgu Muli decoction for insomnia. Instead of being restricted to Chinese or Western disease names, he prescribed appropriate formulas according to syndromes, thereby achieving a good efficacy.

2.Contracted services of general practitioner improve management of primary hypertension in community

Two hundred and forty patients with primary hypertension in Weifang Community were randomly sampled as contract group and another 240 hypertensive patients were sampled as control group.Patients in contract group signed service contract with general practitioners (GPs),who provided individualized service for hypertension management.Patients in control group receive conventional outpatient service for hypertension management.The clinical data of patients before and one year after contracted services were documented and compared with those in control group.The results showed that in contract group the average systolic pressure decreased from 134.60 mm Hg (1 mm Hg =0.133 kPa) to 130.62 mm Hg (P =0.000),reaching-the-target-BP rate increased from 56.3% to 85.5% (P =0.000),serum total cholesterol and triglyceride decreased significantly one year after receiving contracted services (P =0.000) ; while these indicators were not significantly changed in control group (all P > 0.05).The results indicate that the contracted services of GPs can improve the effectiveness of hypertension management in community.

5.Inhibitory effect of miR-204 on corneal epithelial cell proliferation in vitro

Background As the main cellular constituent of corneal epithelium,corneal epithelial cells play critical roles in regulating and controlling the migration, proliferation and differentiation of cells during the repair of damage.MicroRNA (miRNA) is endogenously expressed small non-coding RNAs, which participates in a variety of biological processes.Previous studies demonstrated that miR-204 is highly expressed in normal corneal epithelium,but its function is still unclear.Objective This study was to investigate the function and mechanism of miR-204 in corneal epithelial cell proliferation.Methods Corneal epithelial tissue was collected during the corneal refractive surgery from the patients with refractive error under the informed consent, and human corneal epithelial cells(HCECs) were cultured and passaged.The relative expressing levels of miR-204 mRNA in the normal corneal epithelium and HCECs were detected by real time quantitative PCR.Cultured HCECs were evenly divided into three groups.The liposome with miR-204 mimic was transfected into the cells of the miR-204 mimic group, and blank liposome was transfected in the cells of the positive control group,and regularly cultured cells served as the normal control group.Cell proliferation capability was evaluated by colony-forming assay, and the percentage of the cells in different cell cycles was analyzed by flow cytometry.Western blot assay was employed to detect the expression levels of p-RB, E2F1 ,p27,CyclinA, CDC2, p-CDC2 and CDK2 proteins in the cells.Results The relative expression levels of miR-204 mRNA were 1.077 ±0.268 in the normal corneal epithelium and 0.041 ±0.018 in the HCECs, showing a significant difference between them (t =7.700,P<0.001).The cloning cell number was evidently decreased in the miR-204 mimic group in comparison with the positive control group and normal control group.The percentage of cells in the G1 phase was 47.75% in the miR-204 mimic group, which was significantly higher than 37.23% in the positive control group and 40.72% in the normal control group.The expression levels of E2F1 and p27 proteins in the cells were elevated (t=14.87,25.11;both at P<0.01) and those of CDK2 and p-CDC2 proteins were decreased (t=5.39,10.65;both at P<0.01) in the miR-204 mimic group in comparison with the positive control group.Conclusions The overexpression of miR-204 in the normal corneas probably is associated with non-proliferation status of corneal epithelial cells.Transfection of miR-204 into corneal epithelial cells can inhibit the proliferation of corneal epithelial cells probably by up-regulating the expression of E2F1 and p27 and suppressing the expression of CDK2/CyclinA and p-CDC2/CyclinA,which lead to cell arrest in G1 phase.
6.HP1γ promotes the development of ovarian cancer and is modulated by miR-30b

Objective To investigate the effect of HP1γ on the development of ovarian cancer. Methods RT-qPCR was used to deteched the level of HP1γin ovarian cancer samples. The proliferation of A2780 and SKOV3 cell line was detected by CCK8 assay. RNAi was used to knock down the expression of HP1γ mRNA. Results The level of HP1γwas higher in ovarian cancer than that in the normal tissue. Mir-30b may inhibit the proliferation of ovarian cancer cell. Conclusion HP1γpromotes the proliferation and progress of ovarian cancer cells.

9.Analytical methods with qualitative fingerprint and quantitative measurement of effective components of Cortex Moutan

Objective To establish a qualitative and quanti ta tive method with RP-HPLC for controlling the quality of Cortex Moutan. Methods The experimental conditions of the RP-HPLC method wer e as follows: planetsil C 18 column (150 mm?4.6 mm, 5 ?m), mobile phase of methanol-water-acetic acid ( 44∶56∶0.1), flow rate at 1.0 mL?min -1, detection wavelength at 240 nm and room temperature. The qualitative fingerprint of Cortex Moutan and the quantitative measurement of paeonol wer e carried out under the chromatographic conditions mentioned above. Res ults Eleven comparatively stable peaks were detected among 13 differ ent samples, from which 10 common peaks used as index peaks for qualitative iden tification were conformed by correlating the peak areas. At the same time, the c ontents of paeonol in samples range from 1.32% to 2.78%. Conclusion The analytical method with qualitative fingerprint of Cortex Moutan and quantitative measurement of paeonol can effectively be used to control the quality of Cortex Moutan.
10.Determination of Organic Acids in Cosmetics by Ion-exchange Chromatography Suppressed Conductivity Detection

0.999).The relative standard derivations (RSDs) were 0.2%-5.0%, the rates of recovery were 91.1%-108.5%. The detection limits for analysis ranged from 1.3 mg/kg to10 mg/kg. Conclusion The method presented in the paper is sensitive, accurate and suitable for the determination of organic acids cosmetic samples.
