Thermosensitive gel with the property of LCST (Lower Critical Solution Temperature) was studied as a hot spot these years for it can be used as one supportor of drug controlled release system which can release drug at right point, right time and right quantity. With further reseaches and applications of thermosensitive gel, the modes and mechanisms of it were discovered and poited out, drug release models were also established step by step. Resesrches on drug release mechanisms and modes of thermosensitive gel will make it applied better in fields of medicine and biology.

Chemogenomics/chemical genomics has been widely used in novel drug research and discovery. Firstly, the concept of chemogenomics was introduced briefly. Secondly, we reviewed the progress of novel drug research and discovery based on forward chemogenomics, reverse chemogenomics and predictive chemogenomics. Finally, we showed progress of the research that chemogenomics has been used to novel drug research and discovery in pharmaceuticals companies.

Drugs designed to act on individual molecular targets usually can not combat multigenic diseases such as cancer, or diseases that affect multiple tissues or cell types such as diabetes. Increasingly, it is being recognised that a balanced modulation of several targets can provide a superior therapeutic effect and side effect profile compared to the action of a selective ligand. The multi-target drugs which impact multiple targets simultaneously are better at controlling complex disease systems and are less prone to drug resistance. Here, we compare the disadvantage of the selective ligands and the predominance of multi-targets drugs in detail and introduce the approaches of designing multiple ligands and the procedure of optimization particularly. A key challenge in the design of multiple ligands is attaining a balanced activity at each target of interest while simultaneously achieving a wider selectivity and a suitable pharmacokinetic profile. On this point, the multi-target approach represents a new challenge for medicinal chemists, pharmacologists, toxicologists, and biochemists.

The reduce of bioactivity and augment of the side effect of single-target drugs is generated by the multi-factorial properties of the pathogenesis of disease, which could be solved by the multi-target drugs. The problems and its solution of the design of the multi-target drugs were discussed in this paper, at the same time, the design of the multi-target drugs by pharmacophore model method is presented.

The development of new drug is not only the main driving force for the development of pharmaceutical industry, but also plays a very important role in the social development. However, with the increasing demands, new drug development is facing great difficulties in recent years. The hypothesis of highly selective single-target is meeting the challenges because of its limitations. Network pharmacology has been one of the new strategies for new drug discovery based on single-target drug research in recent years. This paper focused on the basis of network pharmacology and its research progress, discussed its development direction and application prospects, and analyzed its limitations and problems as well. The application of network pharmacology in new drug development is discussed by comparing its guidelines with those of traditional Chinese medicine theory and Effective Components Group hypothesis of Chinese medicines.

High-intensity focused ultrasound therapy is a novel, emerging, therapeutic modality that uses ultrasound waves, propagated through tissue media, as carriers of energy. This completely non-invasive technology has great potential for tumor ablation as well as hemostasis, thrombolysis and targeted drug/gene delivery. However, the application of this technology still has many drawbacks. It is expected that current obstacles to implementation will be resolved in the near future. In this review, we provide an overview of high-intensity focused ultrasound therapy from the basic physics to recent clinical studies with an interventional radiologist's perspective for the purpose of improving the general understanding of this cutting-edge technology as well as speculating on future developments.

This paper reports a new solid self-emulsifying drug delivery system--self-emulsifying microsphere prepared by membrane emulsification technology with the hydrophobic berberine hydrochloride as a model drug. Solubility test and pseudo-ternary phase diagram were employed to select the optimal prescription of liquid self-emulsifying drug delivery system. The self-emulsifying microsphere was prepared by membrane emulsification technology with the solid carrier in a gel formed by sodium alginate and calcium chloride. The results showed that the optimal prescription of liquid self-emulsifying drug delivery system was Capmul MCM/Cremophor RH40/Labrasol/1-2propanediol = 20 : 32 : 32 : 16. The solid self-emulsifying microsphere had average diameter of 10.92 microm, encapsulation efficiency of 32.57% and the droplet size of reconstituted micromulsion of 156.5 nm. Berberine hydrochloride was dispersed in microsphere in non-crystalline form. In vitro release of the self-emulsifying microsphere showed pH response characteristics. These results indicated that the self-emulsifying microsphere prepared by membrane emulsification technology might become a new dosage form for poorly water soluble drugs.
