Objective: To observe the expression of transient receptor potential channel-1—large conductance Ca2+-activated K+channel (TRPC1-BK) signal complex in aortic smooth muscle tissue in experimental mice of atherosclerosis (AS). Methods: Our research included in 2 groups: Control group, wild C57BL/6J mice were fed by normal diet and AS group, ApoE-/- mice were fed by high fat diet to establish AS model. All animals were treated for 10 weeks, n=10 in each group. The total RNA and protein were extracted from aortic smooth muscle tissue in sacrificed mice. The mRNA and protein expressions of TRPC1, BKα and BKβ1 subunit were measured by RT-PCR, Western blot analysis and immunohistochemistry staining. Results: By RT-PCR examination, compared with Control group, AS group showed increased mRNA expression of TRPC1, P<0.05 and decreased mRNA expressions of BKα and BKβ1 subunit, both P<0.01. By Western blot analysis, compared with Control group, AS group had elevated protein expression of TRPC1, P<0.05 and reduced protein expressions of BKα and BKβ1 subunit, both P<0.01; by immunohistochemistry staining, AS group had the higher protein expression of TRPC1, P<0.01 and the lower protein expressions of BKα and BKβ1 subunit, both P<0.05. Conclusion: The mRNA and protein expressions of TRPC1-BK signal complex were affected in aortic smooth muscle tissue in AS mice, which speculated that TRPC1-BK signal complex might be become a new target for treating the relevant vascular disease.

Chinese Journal of Rehabilitation Theory and Practice 2015;21(8):967-973
Objective To compare the difference of static balance ability in different aged people, with the maximum angle of center of mass (COM) and center of pressure (COP). Methods 37 healthy people without cardiovascular and cerebrovascular diseases including 15 young people (18~40 years old), 10 middle age people (41~65 years old) and 12 old people (>65 years old) participated in this study. The maximum angle of COM-COP and the corresponding envelope areas were recorded in 4 standing posture: natural standing, close eyes standing, single leg standing and single leg standing with eyes closed. Results There was no significant difference in the maximum angle of COM-COP among 3 groups in 3 states (nature standing, close eyes standing, single leg standing with eyes closed) (P>0.05), but there was significant difference in the state of single leg standing (P<0.05) between the young and the old, and the middle-aged and the old. Conclusion The maximum angle of COM-COP in the state of single leg standing can sensitively detect the deterioration of stability of older people. It may be a new index of static balance in clinic.

Methods90 patients underwent abdominal surgery were randomly divided intothree groups with 30 cases each. Group Ⅰ :sufentanil 0. 04μg · kg-1 ·h-1 for postoperative analgesia; Group Ⅱ : sufentanil 0. 06μg · kg-1 · h-1 for postoperative analgesia; Group Ⅲ :sufentanil 0.08μg/kg · h for postoperative analgesia;PCSA started at the end of operation with catheter buried in deltoid muscle. Visual analgesia scale(VAS) ,Rasmay scale and side effects were recorded at 4,8,16,24 and 48h after operation. ResultsVAS scores decreased significantly in group Ⅱ and group Ⅲ compared with that in group Ⅰ ( all P < 0.05) ;There were no differences of Ramsay scores between groups ( all P > 0. 05) ;There were three cases of respiratory depression in group Ⅲ. ConclusionPCSA with sufentanil (0.06μg/kg) had exact analgesic effects for postoperative abdominal surgery patients with less adverse reactions.

Objective:To study the influence of metformin and paclitaxel in the proliferation and apoptosis of human ovarian cancer SKOV3 cells in vitro, and to clarify the synergistic effect of metformin and paclitaxel. Methods:The ovarian cancer SKOV3 cells were divided into blank control group, different concentrations (0.01, 0.50, 1.00, 5.00, 10.00mmol·L-1) of metformin groups and combined treatment groups(metformin combined with paclitaxel with different concentrations). The inhibitory rates of proliferation of SKOV3 cells after treated with different concentrations of metformin were detected by MTT method. The apoptotic rates of SKOV3 cells after treated with different concentrations of metformin were measured by flow cytometry. The inhibitory rates of proliferation of SKOV3 cells after treated with metformin and paclitaxel were determined by MTT method. Results:The MTT results showed that the inhibitory rates of proliferation of SKOV3 cells in different concentrations of metformin groups were increased in concentration-and timedependent manner;there were significant differences compared with blank control group (P<0.05).The flow cytometry results showed that the apoptotic rates of SKOV3 cells in different concentrations of metformin groups were increased;compared with control group, with the increasing of concentrations of metformin, the apoptotic rates of SKOV3 cells in experimental groups 48 h after treatment were increased significantly (P<0.05);the percentages of SKOV3 cells in G0/G1 phase were decreased with the increasing of metformin concentrations(P<0.05) and the percentages of SKOV3 cells in G2/M phase were increased(P<0.05).The MTT results showed that the inhibitory rate of proliferation of SKOV3 cells in 1.00 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group was higher than that in 0.50 mmol·L-1 metformin+paclitaxel group(P<0.05),and the inhibitory rates of proliferation of SKOV3 cells in combined treatment groups were higher than those in paclitaxel alone groups(P<0.05).Conclusion:Metformin inhibits the proliferation of ovarian cancer SKOV3 cells through the induction of apoptosis.Metformin can enhance the cell proliferation inhibition of paclitaxel on ovarian cancer SKOV3 cells. The combination of metformin and paclitaxel has a synergistic reaction on SKOV3 cells.

Methods:A Waters Sun Fire TM C18 column(150 mm × 4. 6 mm,5 μm) was adopted with 0. 1% ace-tic acid-methanol(60∶40) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 228nm for benzoic acid(0-18min) and 285nm for cinnamic acid (18. 1-35min). The injection volume was 10μl, and the column temperature was room temperature. Results:The linear range of benzoic acid and cinnamic acid was 1. 0-50. 0 μg·ml-1(r=0. 999 7) and 0. 2-10. 0 μg· ml-1, respectively(r=0. 999 9). The average recovery was 96. 88%(RSD=1. 6%, n=9) and 99. 35%(RSD=1. 7%, n=9), re-spectively. Conclusion:The method is accurate, fast and simple in the determination of benzoic acid and cinnamic acid in Suhexiang pills.

BACKGROUND:Hip segmentation based on CT image has been widely used in computer-assisted surgery planning, prosthesis design and finite element analysis. OBJECTIVE:To explore application effects of automatic segmentation method for hip joint based on Bayesian Decision Theory in computer-assisted hip surgery. METHODS:An accurate outer surface segmentation and extraction remain chal enging due to deformed shapes and extremely narrow inter-bone regions. In this paper, we present an automatic, fast and accurate approach for segmentation of femoral head and proximal acetabulum. The outline of the femur was segmented and extracted by contrast enhancement, thresholding algorithm and region growth algorithm. The boundaries of the bone regions are further refined based on Bayes decision rule. RESULTS AND CONCLUSION:Automatic segmentation method for hip joint based on Bayesian Decision Theory is an accurate segmentation technique for femoral head and proximal acetabulum and it can be applied in computer-assisted hip surgery and prosthesis design.
