1.Acupoint selection and medication rules analysis for allergic rhinitis treated with acupoint application-based on data mining technology.

To make a systematic study on acupoints selection and medication rules for allergic rhinitis treated with acupoint application, literature regarding clinical researches on acupoint application for allergic rhinitis since 2000 has been retrieved; medication and acupoints involved were preprocessed and cluster analysis technique was applied to classify the data. As a result, a total of 37 pieces of literature were included, involving 27 types of herbs and 33 acupoints. The results showed Baijiezi (Brassica alba Boiss), Xixin (Radix et Rhizoma Asari), Gansui (Radix Kansui), Yanhusuo (Corydalis), Jiangzhi (Ginger juice) and Baizhi (Radix angelicae) constituted the main herb prescription, and the layer was increased along with frequency of use. Feishu (BL 13), Dazhui (GV 14), Shenshu (BL 23), Pishu (BL 20), Gaohuang (BL 43), Fengmen (BL 12) constituted the main acupoint prescription, which were clustered into 4 classes; Feishu (BL 13) and Dazhui (GV 14) constituted the first class; Pishu (BL 20) and Shenshu (BL 23) constituted the second class; the first and second class merged into the third class, Including Feishu (BL 13), Dazhui (GV 14), Shenshu (BL 23) and Pishu (BL 20); Gaohuang (BL 43) and Fengmen (BL 12) constituted the fourth class; the fourth class and the third class constituted the whole acupoint prescription. It was showed that for the allergic rhinitis treated with acupoint application, the medication was mainly epispastic herbs with pungent nature that belonged to the lung, spleen and kidney meridian, which presented the principle of simultaneous treatment of principal and subordinate symptoms; also the medication can be selected from core layer to supporting layer based on clinical needs; for the selection of acupoints, acupoints in lung and bladder meridian were mainly selected to assist exterior and resist the pathogenic qi, which could treat allergic rhinitis fundamentally by joint use with acupoints in soleen and kidney meridian.

Clinical literature of bronchial asthma treated with acupoints application from January 2000 to March 2014 in modern periodicals databases was retrieved through computer. With cluster analysis and frequency analysis methods of data mining, acupoints selecting and medication rules of bronchial asthma treated with acupoints application were analyzed. Total 38 articles were included eventually, including 25 acupoints and 42 medicines. The results indicate that on acupoints selecting, Feishu (BL 13) is used as the main acupoint and 3 groups of bladder meridian and conception vessel acupoints are applied alternately and on medicines, Baijiezi (Brassica alba Boiss), Xixin (Radix et Rhizoma Asari), Gansui (Radix Kansui), Yanhusuo (Corydalis) and Mahuang (Radix et Rhizonma Ephedrae) are primarily adopted, epispastic medicines being the main medicines; medicines mostly belong to lung meridian, main medicines being unchanged mostly with Shengjiang as guiding drug.

3.Establishment and Application of SYBR Green I Real-Time PCR Assay for Rapid Detection of Hepatitis B Virus DNA

Objective To develop SYBR Green I real-time PCR assay for detection and identification of Hepatitis B virus. Methods Based on the sequences of Hepatitis B virus gp1 gene,primers were designed.The reaction assay and thermal cyc-ling profile were optimized.The positive standard was from recombinant clone.Both the developed assay and Zhejiang kuake biotechnology company’s assay were applied in 100 patients serum.Results The detection limit was between 5×102 copies/ml to 5×108 copies/ml with a good liner correlation and no cross reaction.The whole process just needed 2.5 h.Comparing with the company products,the sensitivity and specificity of the developed assay were 100% and 92.5% respectively.Con-clusion The established assay is rapid,simple,high sensitivity and specificity.It is not only valuable for the identification of Hepatitis B virus patients,but also provide accurate quantitative analysis for HBV patients.
4.Mutual Influence of Lead and Cadmium Ions on Bismuth Film Electrode

The mutual influence of lead and cadmium ions on “in-situ” bismuth film coated glassy carbon electrode was demonstrated using different concentration target ions by changing ratio between Pb2+ and Cd2+. It is found that the presence of coexisting ions can have an influence on the detection of target ions. Especially for the detection of Cd2+, the existence of Pb2+ can be beneficial for the deposition of Cd due to the more positive deposition potentials. And the analytical performances of Cd2+and Pb2+ were obtained. The detection limit is 0. 5 μg/L towards Pb2+ with a linear range from 1-80 μg/L using 60 s deposition time. The resulting calibration plots of Cd2+ are linear over the range from 1-25 and 30-200 μg/L with the detection limit of 1 μg/L using 120 s deposition time. The bismuth film was finally employed to determine the Pb2+and Cd2+in real sample with good satisfied results.
5.Dynamic changes of peripheral blood lymphocyte subsets in fever with thrombocytopenia syndrome patients

Objective The aim of this study is to dynamically investigate peripheral blood lymphocyte subsets in fever with thrombocytopenia syndrome (SFTS) patients at different stages,to evaluate the influence of these changes in the infection process.Methods Case-control study was used in the research.Twelveconfirmedthrombocytopeniasyndromevirus ( SFTSV ) infectedpatientswere enrolled.According to SFTS prevention guide issued by Chinese Ministry of Health,these patients were divided into two groups,recovery group and death group.For each group,dynamic profiles of the CD3 + T cells,CD4 + helper T cells,CD8 + cytotoxic T cell and CD3 - CD16 + CD56 + natural killer cells were tested by flow cytometry.Meanwhile, the relationshipsbetween these dynamicchanges and liver function,leukocytes,and platelets were analyzed respectively.Two independent-samples t test was used to compare the difference of the peripheral blood lymphocyte subsets count between the SFTS patients and healthy control.Small sample was analyzed by Mann-Whitney U test.Results In the early stage of infection,Th cells in peripheral blood of recovery group were significantly reduced and Th/Tc ratio was reversed.On day 5,7,9 of post infection,Th cell counts in peripheral blood were (740.9 ± 6.4),(836.2 ± 272.3 ) and ( 1083.6 ± 319.7 ) cells/μl respectively,which were significantly lower than health control ( 1351.4 ± 295.1 ) cells/μl ( t value was -2.883,-4.235,-2.145 respectively,all P <0.05).Tc cell counts were significantly more than healthy controls (690.1 ± 194.8) cells/μl through the course,which were ( 1006.3 ±356.5),(1166.4±242.4),(1102.4±245.9),(991.3±205.1) and (886.5±154.5) cells/μl on day 7,9,11,13,15 of the course (t value was 3.312,5.661,4.574,3.874,2.382,all P<0.05).NK cells were decreased significantly from the ninth day of the course.Associated with abnormal changes of cell subsets,WBC and PLT decreased significantly,and serum ALT,AST,LDH and CK etc.were higher than normal level.With the disease recovery,the abnormality above was gradually improved.In contrast,death cases showed significant decrease in T and Th cells compared with health control (P < 0.05).On day 7,8,9 of the course,the counts of total T cell were (735.9 ± 359.9),(724.9 ± 125.9),(845.3 ± 389.3) cells/μl and the counts of Th cell were ( 533.2 ± 246.9 ),( 532.1 ± 105.7 ),( 551.7 ± 86.9 ) cells/μl,significantly lower than healthy control ( 1727.9 ± 230.2 ) cells/μl and ( 1351.4 ± 295.1 ) cells/μl,with statistically differences (z value was - 2.828, - 2.342,- 2.342 and - 2.828, - 2.342, - 2.342,all P < 0.05 ).On day 7,8,9 of the course,the numbers of NK cell in death group were ( 1141.8 ± 415.5),( 1047.2 ±68.4),( 1276.3 ±545.3) cells/μl,which were significantly more than health group (470.7 ± 242.2) cells/μl,with statistically differences (z value was - 2.180,- 2.335,- 2.258,all P <0.05).Conclusions SFTSV infection can induce cell immunity damage.The changes of lymphocyte subsets are associated with clinical classification and prognosis.Significant reduction of T cell and CD4 +cell in peripheral blood are accompanied with significant increase of NK cell,which may be a pivotal indicator of poor prognosis and play an important role in making appropriate strategy in clinical treatment.( Chin J Lab Med,2012,35:826-831 )
6.Preliminary study on clinical efficacy of intra-arterial chemotherapy and intravenous chemotherapy for unilateral advanced retinoblastoma

Objective:To compare the clinical efficacy and complications of intra-arterial chemotherapy (IAC) and intravenous chemotherapy (IVC) for unilateral advanced retinoblastoma (RB).Methods:A retrospective clinical study. From January 2020 to January 2021, 40 patients (40 eyes) unilateral group cT2 RB patients diagnosed at Baoding Children’s Hospital and Beijing Children’s Hospital were recruited in this study. There were 22 males (22 eyes) and 18 females (18 eyes). All were monocular. All the patients were assigned to two groups according to different treatment modalities they received: IVC group and IAC group. There were 26 eyes and 14 eyes, respectively. When the tumor invades the optic nerve, choroid, sclera, anterior chamber and iris, enucleation was performed. The globe salvage rate, tumor extraocular metastasis rate, solid tumor control rate, treatment-related complications and pathological high-risk factors after enucleation were observed. The globe salvage rate and solid tumor control rate were compared between the groups by chi square test.Results:The globe salvage rate of IAC group and IVC group were 88.5% (23/26) and 50.0% (7/14), respectively. Solid tumor control of IAC group and IVC group were 84.6% (22/26) and 42.9% (6/14), respectively. There were statistically significant differences in globe salvage rate and solid tumor control between the two groups ( χ 2=7.18, 7.56; P<0.05). Compared with IVC group, IAC group had less systemic complications, mild ocular and periocular side effects. Among 26 cases in IAC group and 14 cases in IVC group, 3 and 7 cases underwent enucleation respectively. The results of pathological examination showed that there were 2 cases and 3 cases with pathological high-risk factors in the two groups, respectively. During the follow-up period, 2 cases in IAC group had extraocular metastasis, there was no extraocular metastasis in IVC group. Conclusion:Compared with IVC, IAC has the advantages of high tumor control rate, high globe salvage rate, less and mild complications, however, there is still tumor recurrence.
7.Comparative study of NERC-300 high-frequency electric welding device in rat partial hepatectomy

Objective To contrastively study the effects of NERC-300 high frequency welding device and ultrasonic scalpel in rat partial hepatectomy.Methods Forty-eight rats were divided into the experimental group and control group(n=24).The experimental group applied NERC-300 high frequency welding instrument for conducting partial hepatectomy,while the control group used ultrasonic scalpel.The survival rate,operation time,bleeding volume,working temperature,extent of hepatic thermal damage and change of liver function on 1,3,7 d were compared between the two groups.Results The liver function recovery on 3 d in the experimental group was superior to the control group (P<0.05).The intra-operative working temperature in the experimental group was lower than that in the control group(P<0.05).The thermal damage range in the experimental group was lower than that in the control group(P<0.05).The survival rate,operation time,bleeding volume and liver function change on 1,7 d had no statistical differences between the two groups(P>0.05).Conclusion NERC-300 high frequency welding device is safe and effective for partial hepatectomy in rat.
8.Naoxin'an Capsule Alleviates Mitochondrial and Oxidative Damage in Chronic Cerebral Ischemia-induced VCI in Rats via Activating CREB/PGC-1α Pathway

ObjectiveTo explore the effect of Naoxin'an capsule against mitochondrial dysfunction and oxidative damage in brains of rats with vascular cognitive impairment (VCI) induced by chronic cerebral ischemia. MethodA total of 150 rats were randomized into modeling group (130 rats) and sham-operated group (20 rats) with the random number table method. The two-vessel occlusion (2-VO) was employed to induce VCI in rats, and finally 87 rats developed VCI. The VCI rats were classified into model group, positive drug group (Aricept, 0.5 mg·kg-1), and low-, medium- and high-dose Naoxin'an capsule groups (0.18, 0.36, 0.72 g·kg-1, separately), with 17-18 rats in each group. The administration lasted 8 weeks. The learning and working memory of VCI rats were assayed by novel object recognition test and Y-maze test. The 8-oxoguanine (8-OxoG) level was measured based on immunofluorescence staining. Spectrophotometry was performed to determine the activity of mitochondrial respiratory chain complexes Ⅰ, Ⅱ, Ⅲ, and Ⅳ, mitochondrial swelling, mitochondrial membrane potential (MMP), and activity of pyruvate dehydrogenase (PDH) and α-ketoglutarate dehydrogenase (KGDH). The submicroscopic structure of mitochondria was observed with electron microscope. The phosphorylation of cAMP response element-binding protein (CREB) and the expression of peroxisome proliferator-activated receptor γ coactivator-1α (PGC-1α), nuclear respiratory factor 1 (NRF-1), and mitochondrial transcription factor A (mt-TFA) were determined by Western blot. The mitochondrial status Ⅳ H2O2 generation, activity of cerebral superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) content were measured with the photochemical method. ResultCompared with the sham-operated group, model group showed decrease in new object discrimination index (P<0.01) and spontaneous alternation rate in the Y-maze test (P<0.01), increase in ROS production in the brain (P<0.01), reduction in MMP and mitochondrial swelling A value (P<0.01), obvious mitochondrial swelling, vacuoles and cristae fractures in mitochondria, decrease in the level of phosphorylated CREB, expression of PGC-1α, NRF-1 and mt-TFA (P<0.01), and activity of the respiratory chain complexes Ⅰ, Ⅱ, Ⅲ, and Ⅳ (P<0.01), PDH and KGDH in the brain (P<0.01), rise of the production of H2O2 at state IV (P<0.01) and the content of MDA (P<0.01), and reduction in the activity of SOD and GSH-Px (P<0.01). Compared with the model group, each dose of Naoxin'an capsules can improve the new object discrimination index (P<0.05, P<0.01) and the rate of spontaneous alternation (P<0.05, P<0.01), decrease ROS production in the brain (P<0.01), improve the MMP and swelling A value (P<0.05, P<0.01), alleviate mitochondrial swelling and mitochondrial submicroscopic damage, elevate the phosphorylated CREB level, the expression of PGC-1α, NRF-1 and mt-TFA (P<0.05, P<0.01), and the activity of mitochondrial respiratory chain complexes Ⅲ, and Ⅳ, PDH and KGDH (P<0.01), decrease state Ⅳ H2O2 generation (P<0.01) and MDA content (P<0.05, P<0.01), and raise the activity of SOD and GSH-Px (P<0.01). ConclusionBy activating the CREB/PGC-1α pathway, Naoxin'an capsule can protect the structure and function of mitochondria, enhance the antioxidant capacity, and inhibit the mitochondrial dysfunction and oxidative damage induced by chronic cerebral ischemia, thus improving the VCI.
9.Mechanism of Dihuang Yinzi in Improving Energy Metabolism Disorder and Autophagy Injury of Astrocytes in Brain of AD Mice

ObjectiveTo explore the mechanism of Dihuang Yinzi (DHYZ)in improving astrocyte injury in the brain and regulating energy metabolism and autophagy disorder in Alzheimer's disease (AD) model mice. MethodForty male APP/PS1 transgenic mice aged four months were randomly divided into a model group and a model + DHYZ group (2.5 g·kg-1), with 20 mice in each group. Forty C57BL/6J mice with the same background and same age were randomly divided into a control group and a control + DHYZ group (2.5 g·kg-1), with 20 mice in each group. The mice in the control group and the model group were administered with an equal volume of sterilized normal saline by gavage, once a day for 150 days. Novel object recognition test and step-down test were performed to evaluate the learning and memory ability of mice. The expression of glial fibrillary acidic protein (GFAP) in astrocytes was detected by immunofluorescence and Western blot. High-performance liquid chromatography (HPLC) was used to detect adenosine triphosphate (ATP), adenosine diphosphate (ADP), and adenosine monophosphate (AMP) in brain tissues of mice, and the data obtained were used to calculate energy charge (EC) levels. The phosphorylation levels of liver kinase B1 (LKB1), adenosine 5′-monophosphate (AMP)-activated protein kinase (AMPK), UNC-51-like kinase 1 (ULK1), and mammalian target of rapamycin (mTOR) and the expression levels of autophagy-related proteins Beclin-1, microtuble-associated protein 1 light chain 3 (LC3)-Ⅱ/LC3-Ⅰ, and p62 in mouse brain were measured by Western blot. ResultCompared with the control group, the model group showed decreased novel object recognition index, shortened retention latency, increased error times in the step-down test, up-regulated protein expression of GFAP, decreased content of ATP, ADP, and EC in brain tissues, elevated AMP , increased levels of p-AMPK, p-LKB1, and p-mTOR, and protein expression of p62 , and down-regulated p-ULK1 level and protein expression of Beclin-1 and LC3-Ⅱ/LC3-Ⅰ(P<0.01), while the above experimental indexes were not significantly different in the control + DHYZ group. Compared with the model group, the model + DHYZ group showed increased novel object recognition index(P<0.05), prolonged retention latency(P<0.01), decreased error times(P<0.01) in the step-down test, reduced protein expression of GFAP(P<0.05), increased content of ATP, ADP, and EC in brain tissues (P<0.05, P<0.01), decreased AMP content(P<0.05), reduced p-AMPK, p-LKB1, and p-mTOR levels and protein expression of p62, and up-regulated p-ULK1 level and protein expression of Beclin-1 and LC3-Ⅱ/LC3-Ⅰ(P<0.01). ConclusionBy protecting astrocytes, DHYZ can improve energy metabolism and autophagy disorder in AD mice to improve the learning and memory ability of model mice.
10.Mechanism of Dihuang Yinzi in Improving Astrocyte Injury and Regulating Synaptic Structure and Function in AD Mice

ObjectiveTo investigate the mechanism of Dihuang Yinzi in improving astrocyte injury and protecting synaptic structure and function in the brain of Alzheimer's disease (AD) mice. MethodForty male APP/PS1 transgenic mice aged four months were randomly divided into a model group and a model + Dihuang Yinzi (0.25 g·kg-1) group, with 20 mice in each group. Forty C57BL/6J mice with the same background and same age were randomly divided into a control group and a control + Dihuang Yinzi (0.25 g·kg-1) group, with 20 mice in each group. The mice in the control + Dihuang Yinzi group and the model + Dihuang Yinzi group were administered with Dihuang Yinzi by gavage, and those in the control group and the model group received an equal volume of sterilized normal saline, once a day for 150 days. The learning and memory ability of mice was tested by the light-dark box test and Y-maze spontaneous alternation test. The content of glutamate (Glu) and glutamine (Gln) was measured by liquid chromatography-tandem mass spectrometry (LC-MS). Long-term potentiation (LTP) assay was used to detect synaptic plasticity in brain tissues. The protein expression levels of excitatory amino acid transporter 2 (EAAT2), postsynaptic density protein95 (PSD95), and synaptophysin (SYN) in brain tissues were measured by Western blot. Immunofluorescence was used to assess the localization and expression of EAAT2. Colorimetry was performed to detect Na+-K+ ATPase activity in mouse brain tissues. ResultAs compared with the control group, the model group showed shortened residence latency (P<0.01), increased number of errors (P<0.01) in the light-dark box test, reduced spontaneous alternation behaviors (P<0.01), no significant difference in the total number of arm entries in the Y-maze spontaneous alternation test, down-regulated expression of EAAT2, PSD95, and SYN (P<0.01), blunted activity of Na+-K+ ATPase (P<0.01), up-regulated Glu level (P<0.01), down-regulated Gln level (P<0.01), and reduced relative population spike (PS) amplitude and the slope of excitatory postsynaptic potential (EPSP) (P<0.05, P<0.01), while the above experimental indexes were not significantly different in the control + Dihuang Yinzi group. Compared with the model group, the model + Dihuang Yinzi group displayed prolonged residence latency (P<0.05), decreased number of errors (P<0.01) in the light-dark box test, increased spontaneous alternation behaviors (P<0.01), no significant difference in the total number of arm entries in the Y-maze spontaneous alternation test, up-regulated expression of EAAT2, PSD95, and SYN (P<0.01), potentiated activity of Na+-K+ ATPase (P<0.01), reduced Glu level (P<0.01), up-regulated Gln level (P<0.01), and increased PS amplitude and EPSP slope (P<0.01). ConclusionDihuang Yinzi can improve cognitive dysfunction in AD mice by protecting astrocytes, increasing Glu uptake to reduce its abnormal accumulation, and protecting synaptic structure and function.
