1.Effects of Ginkgo biloba extract on volume of cerebral infarction and expression of caspase-3 in ischemic-reperfusion rats

Objective To study the effects of Ginkgo biloba extract(EGb) on the volume of cerebral infarction and expression of caspase-3 in ischemic-reperfusion rats.Methods Setting up the brain ischemic and reperfusion model in rats by Nagasawa way.Both groups rats treated with small dose of EGb and large dose of EGb were injected intraperitoneally with EGb 50 mg/kg and 100 mg/kg at 30 min before the trial and 1 h after cerebral ischemic respectively.The infarction volume and caspase-3 expression were detected by TTC staining and immunohisto-chemistry.Then the results were compared with ischemic-reperfusion group.Results In groups treated with small dose of EGb and large dose of EGb,infarction volumes [(83.4?2.9)mm3,(37.0?2.5)mm3] were smaller obviously than that in the ischemic-reperfusion group [(138.8?6.1)mm3](all P
2.Tissue culture of shoot-tip and plantlet regeneration of Camptotheca acumunata

Object To explore the artificial propagation of the medicinal plant, Camptotheca acuminata Decne. Methods The shoot-tip was tested as the explants and cultured on culture media with different portions of hormone. Results The best medium for bud induction was the B5 basic medium with additions of 6-BA (0.2 mg/L), IBA (0.05 mg/L), and AS (afenine sulfate, 10 mg/L). While B5 with the additions of IBA (0.5 mg/L), KT (0.1 mg/L), and AS (10 mg/L) was suitable for rooting. The seedling was cultivated and grew well on the base material mixed with perlite-soil (3∶7). The survival rate of transplant was up to 96%. Conclusion The above mentioned method provides a new effective way to exploit this plant resources.
3.Clinical and imaging features of cerebral cortical vein thrombosis

Objective To investigate the clinical and imaging features of cerebral cortical vein thrombosis (CoVT). Methods The clinical data of 13 inpatients with CoVT were enroled from September 2010 to September 2015. The etiology, clinical symptoms, imaging features, and differential diagnosis of CoVT w ere summarized and analyzed. Results Among the patients, there w ere 7 females (53.85%) and 6 males (46.15%). Their age ranged from 18 to 59 years (mean 34.23 ±12.08). The most common clinical manifestations were headache (9 patients) and seizures (9 patients). The 13 patients had cerebral parenchymal involvement and they al received anticoagulant therapy. Conclusions The clinical symptoms of CoVT are various. Seizures and headache are the most common symptoms. Most of patients are accompanied by brain parenchymal lesions and venous sinus thrombosis. When patients have headache and seizures, and head images suggest hemorrhagic infarction, imaging examination of the improvement of the cerebral venous system thrombosis is recommended.
4.Progress in microRNAs associated with major avian viruses.

Recently, avian viral diseases have become one of the main models to study mechanisms of viral infections and pathogenesis. The study of regulatory relationships and mechanisms between viruses and microRNAs has also become the focus. In this review, we briefly summarize the general situations of microRNAs encoded by avian herpesviruses. Also, we analyze the regulatory relationships between tumorigenicity of avian herpesviruses and microRNAs. Additionally, the possible applications for prevention and treatment of viral diseases (such as infectious bursal disease, avian influenza and avian leucosis) using the regulatory mechanisms of microRNAs are also discussed.

5.Effect of Ellagic Acid on Myeloma SP2/0 Cells

Objective To investigate the effect of ellagic acid extracted from gallnut on multiple myeloma SP2 / 0 cell line and related mechanisms. Methods Multiple myeloma SP2 / 0 cell line was treated for 48 h with different concentrations of ellagic acid in vitro. Cell morphology,proliferation,apoptosis and cell cycle were analyzed with microscope,MTT experiment and flow cytometry,respectively. Tumor cell proliferation and apoptosis-related gene expression of COX-2 were detected by Western blotting. Results Cell cycle was arrested at the G1 phase 48 h after treatment with ellagic acid, the cell in G1 were (55. 21±3. 01)% , (64. 48 ± 0. 43)% , (75. 10 ± 2. 46)% , respectively, with significant difference as compared with control group [(34. 04±1. 74)% ,P<0. 01]. Cell suppression rate (21. 18±5. 92)% ,(44. 58±3. 43)% and (70. 15±2. 90)% ,respectively, in 20,40 and 60 μg·mL-1 ellagic acid treatment groups. Compared with the control group,the differences were significant (P<0. 01). Cell apoptosis rate was (9. 60 ± 0. 56)% , (19. 30 ± 1. 51)% and (35. 10 ± 5. 26)% , respectively, in 20,40 and 60 μg·mL-1 ellagic acid treatment groups,with significant differences as compared with the control group[(3. 23±0. 85)% ,P<0. 01]. With the increase of drug concentration,COX-2 expression was decreased. Conclusion Ellagic acid can inhibit myeloma SP2 / 0 cell proliferation and promote apoptosis.
6.Optimization of Ethanol Extraction Technology ofLichong ShengsuiDecoction by Multi-index Orthogonal Test

Objective To optimize the ethanol extraction technology ofLichong ShengsuiDecoction.Methods An L9(34) orthogonal test was used in the study. The extraction rates of calycosin glycoside, icariin, baohuosideⅠ, ginsenoside Rg1, ginsenoside Re, ginsenoside Rb1, astragaloside, and ginsenoside Rd were set as indexes. The influence of ethanol concentration, extraction time, extraction temperature, and amount of ethanol on the yield of Lichong ShengsuiDecoction were detected by comprehensive scoring method.Results The optimal ethanol extraction technology forLichong ShengsuiDecoction was soaking for 2 h with ten times of 60% ethanol and then reflux extracting for two times; extraction time was 1.5 h each time at 80℃.ConclusionThe optimal extraction technology is efficient, stable and feasible, which can provides data for the further study ofLichong ShengsuiDecoction.
7.Analysis of clinical and imaging characteristics of bilateral medial medullary infarction

Objective To investigate the clinical and imaging characteristics of bilateral medial medullary infarction. Methods The clinical data of 3 patients with bilateral medial medullary infarction admitted to Shengjing Hospital of China Medical University were analyzed retrospectively. The related literature was reviewed. Results Three patients in this group were all males. Their main clinical manifestations were quadriplegia, dysarthria, and paresthesia, and 1 of them complicated with respiratory failure. One patient was suspected of having Guillain-Barre syndrome. The hyperintensities of heart-shape,Y- shape,and V- shape were its imaging features of typical MRI diffusion weighted imaging. Conclusion Bilateral medial medullary infarction is a rare posterior circulation ischemic lesion in clinical practice. Its early symptoms are not typical and easy to be misdiagnosed and missed. MRI diffusion weighted imaging is its main imaging examination method.
8.Expression and Purification of an N?terminal Fragment of the Cav1.2 Calcium Channel and Characterization of Its Interaction with Calmodulin

Objective To investigate a method for the purification of the N?terminal peptide fragment(NT)of the myocardial calcium channel Cav1.2,and characterize its interaction with calmodulin(CaM). Methods EscherichiacoliBL?21 cells were transformed with plasmid pGEX?6p?3/NT harboring the NT?GST fusion gene. The cells harboring pGEX?6p?3/NT were cultured and protein expression was induced with isopropyl?β?D?thiogalactoside(IPTG). Then,the GST?NT fusion protein was purified by using glutathione Sepharose 4B(GS?4B)beads. GST was cleaved off with the PreScission protease,and SDS?PAGE was performed to detect the purity and relative molecular weight of the purified peptide. Further, GST pull?down assay was performed to characterize the interaction of the NT peptide with CaM. Results SDS?PAGE analysis showed that the NT peptide was successfully purified,with high purity. Results of the GST pull?down assay showed that the NT peptide could interact with CaM. Conclusion This study establishes a method for the purification of the NT peptide and lays the foundation for further research on the interaction partners and biological functions of NT.
9.Vector Construction,Protein Expression,Purification and Identification of Calmodulin Mg2+Binding Site Mutants

Objective To construct plasmid vectors of calmodulin(CaM)Mg2+binding site mutants,and to express,purify and identify the mutant proteins. Methods Three kinds of cDNAs coding for the mutated CaM were cloned into pGEX?6P?3 plasmid vectors. These recombinant plasmids were transfected into Escherichia coli BL21 to express GST fusion proteins of CaM mutants. The fusion proteins were purified with Glutathione?Sep?harose 4B beads and PreScission protease. Results Both enzyme digestion analysis and DNA sequence identification proved the successful con?struction of the CaM mutant plasmids. SDS?PAGE results showed the high purity of each CaM mutant protein. The concentrations of three CaM mu?tants were around 1.0 mg/mL. Conclusion Prokayotic expression vectors of CaM Mg2+binding site mutants were successfully developed,and the eli?gible CaM mutant proteins were obtained. This study provided an important basis for further study on CaM’s biological function.
10.Osteoprotegerin gene polymorphism is not associated with the risk of large atherosclerotic stroke in Han population in Liaoning, China

Objective To investigate the correlation between the gene polymorphism of osteoprotegerin genes rs3134069,rs2073617 and rs2073618 and the onset risk of large artery atherosclerotic stroke (LAA) in the Han population in Liaoning,China.Methods Using the case-control study design,213 patients with LAA were selected as case group according to the TOAST etiology classification,and 197 health subjects in the same period were selected as control group.Rs3134069,rs2073617,and rs2073618 were genotyped using the modified multiplex ligase reaction technique.Multivariate logistic regression analysis was used to determine the independent correlation between gene polymorphism and LAA.Results There were no significant differences in the genotypes and alsle distribution frequencies of OPG genes rs3134069,rs2073617 and rs2073618 between the case group and the control group (all P >0.05).The dominant and recessive models also showed that there were no significant differences (all P>0.05).Conclusion The polymorphisms of OPG genes rs3134069,rs2073617 and rs2073618 were not significantly associated with LAA susceptibility in Han population of Liaoning,China.The presence of each allele could not constitute a risk factor for the onset of LAA.
