1.Risk Factors of Flow-empty Area in Carotid Artery in Doppler Power Imaging

Purpose To explore the reason which resulted in the flow-empty area in carotid artery, establishing a logistic regression model of carotid artery by power Doppler ultrasound, screening the risk factors of the occurrence of this phenomenon and analyzing their reasons. Materials and Methods Clinical data of 172 male subjects who had undergone power Doppler ultrasound were analyzed retrospectively, whether there was flow-empty area at the edge of carotid artery lumen during diastole was observed, brachial artery systolic pressure, diastolic pressure and pressure difference, carotid artery intima media thickness, and the existence of atherosclerotic plaques was measured, the age, body mass index (BMI), blood glucose, blood lipid, history of smoking and cerebrovascular accident situation was collected to establish the logistic model to analyze the risk factors resulted in flow-empty area. Results Among the 172 patients, intima media thickened in 89 cases, systolic blood pressure increased in 97 cases, diastolic blood pressure increased in 7 cases, the pulse pressure increased in 90 cases, with the age of 65 years or older in 92 cases, atherosclerotic plaque formation in 84 cases, long smoking history in 63 cases, cerebral vascular accident history in 11 cases, diabetes mellitus in 79 cases, hyperlipidemia in 101 cases, and abnormal BMI in 13 cases. Doppler energy imaging showed flow-empty area wave type in 98 cases, no flow-empty area wave type in 74 cases. 5 statistically significant variables for the occurrence of flow-empty area in carotid artery lumen during diastole were selected using logistic regression, which were carotid artery intima media thickness, BASP, the pressure difference of brachial artery, existence of atherosclerotic plaque, and the age of patients (OR=50.643, 47.248, 29.426, 32.667 and 20.514, P<0.05). Conclusion The risk factors resulted in flow-empty area in carotid artery are increased IMT, systolic BP and pressure difference, existence of atherosclerotic plaque and the age of 65 years or older. The flow-empty area provides hemodynamic information for the diagnosis of atherosclerosis, and it also plays a helping role in evaluating the degree of arteriosclerosis.
2.Effect of cyclic stretch on the proliferation of C2C12 myoblasts

BACKGROUND:Mechanical stimulation triggers a muscle tissue remodeling, during which, myoblast process is a multi-stage development process. Many signals in extracellular matrix are involved in myoblast process, and mechanical signals are considered as important external factors for muscle formation and regeneration.
Numerous studies concerning cyclic tensile stress effects on myoblast proliferation and apoptosis have been reported, but the specific mechanism underlying the mechanical stimulation in muscle formation is stil unclear. OBJECTIVE:To study the effects of cyclic stretch on proliferation of C2C12 myoblasts and its mechanism.
METHODS:C2C12 myoblasts were cultured in vitro under 10%cyclic stretch for 6, 12 and 24 hours.
RESULTS AND CONCLUSION:The results of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed that with the time of cyclic stretch, the proliferation of C2C12 myoblasts and the rate of S-phase fraction were gradual y increased and peaked at 12 hours (P<0.05), and then decreased. Western blot assay showed that the expression of nuclear transcription factorκB protein was decreased with time, which was inversely correlated to the proliferation of C2C12 myoblasts, but increased at 24 hours under the cyclic stretch. These findings indicate that the cyclic stretch can induce C2C12 myoblasts proliferation and alter the cellcycle. Nuclear transcription factorκB may be involved in this process.
3.Expression and clinical significance of BFGF and BFGFR in papillary thyroid carcinoma

Objective To study the expression and clinical significance of BFGF factor(BFGF) and basic fibroblast growth factor receptor(BFGFR) in papillary thyroid carcinoma (PTC).Methods 80 casesof PTC and 10 cases of nodular goiter underwent surgical treatment from Mar.2001 to May.2005 in Shengjing Hospital of China Medical University.Samples were collected and immunohistochemistry staining was performed to detect the expression of BFGF and BFGFR.Results The expression rate of BFGF and BFGFR was 72.5% (58/80)vs 70.0% (56/80) in PTC,and 0.0% (0/10) vs 0.0% (0/10) in nodular goiter.The difference had statistical significance(P <0.05).The expression level of BFGF and BFGFR in PTC had no relation with sex,age or tumor size(P >0.05),but has relation with lymph node metastasis and UICC stage(P <0.05).The expression of BFGF was positively correlated with the expression of BFGFR (r =0.330,P =0.003).Conclusions BFGF and BFGFR are highly expressed in PTC but have no expression in nodular goiter or normal thyroid tissues.The expression of BFGF and BFGFR in PTC is closely related to lymph node metastasis and UICC stage,indicating the 2 may be involved in tumorigenesis,invasion,and metastasis.
4.Experimental induction of intestinal metaplasia in stomach& expression of tumor-related proteins

Objective The development and progression of intestinal metaplasia ( IM ) and expression of tumor-related proteins in dog’ s stomach were observed. Methods IM animal model was established in Beagle’ s stomach by feeding N-methyl-N’-nitro-N-trosoguanidine( MNNG ),ranitidine,and local X-irradiation. Expression of APC, p53, K-ras and bcl-2 genes in canine gastric lesion were determined by immunohistochemistry. Resullts IM canine model was successfully induced, with which sequential pathology studied. It was proved that progression of normal epithelial cells to IM cells might undergo several stages including superficial gastritis, chronic atrophic gastritis, mild focal IM, moderate,and severe IM.Aberrant bcl-2 protein was detected in atrophic gastric epithelium and abnormal expression of APC, K-ras, and bcl-2 found in IM mucosa. Conclusion Canine gastric IM was induced by the method mentioned above.The development of normal gastric mucosa to IM in dog seemed to resemble that of human being.The presence of tumor-related proteins might indicate the transformation of IM to malignancy.
5.Effect of Xuesaitong Injection on Related Indexes of Patients with Acute ST Segment Elevation Myocardial Infarction before PCI

OBJECTIVE:To explore the effects of Xuesaitong injection on related indexes of patients with acute ST segment el-evation myocardial infarction(STEMI)before percutaneous coronary intervention(PCI). METHODS:112 STEMI patients under-went PCI were analyzed retrospectively and divided into control group (48 cases) and observation group (64 cases) according to different treatment methods. Control group was given Clopidogrel sulfate tablets 300 mg and Aspirin enteric-coated tablets 300 mg orally before PCI;given conventional treatment according to patients'condition after surgery. Observation group additionally re-ceived intravenous push of Xuesaitong injection 8 mL before surgery,and Xuesaitong injection 8 mL added into Sodium chloride 250 mL intravenously,once a day after surgery,on the basis of control group. Treatment course of 2 groups lasted for 14 d. TIMI level,MPG level,the serum levels of cTnT,CKMB were observed in 2 groups before surgery,24 h after surgery;serum level of PTX-3,hs-CRP were observed before surgery,one week after surgery;LVEF,LVEDD,serum level of BNP were observed before surgery and one month after surgery;the occurrence of ADR was observed to. RESULTS:Before surgery,there was no statistical significance in TIMI level,MPG level,the serum levels of cTnT,CKMB,PTX-3 and hs-CRP,LVEF,LVEDD,serum level of BNP between 2 groups (P>0.05). 24 h after surgery,TIMI level and MPG level of 2 groups were significantly higher than be-fore,and the observation group was significantly higher than the control group;the serum levels of cTnT and CKMB in 2 groups were significantly lower than before,and the observation group was significantly lower than the control group,with statistical sig-nificance(P<0.05). One week after surgery,the serum levels of PTX-3 and hs-CRP in 2 groups were significantly lower than be-fore surgery,and the observation group was significantly lower than the control group,with statistical significance(P<0.05). One month after surgery,the level of LVEF in 2 groups were significantly higher than before surgery,and the observation group was significantly higher than the control group;LVEDD and se-rum level of BNP in 2 groups were significantly lower than be-fore surgery,and the observation group was significantly lower than the control group,with statistical significance (P<0.05). There was no statistical significance in the incidence of ADR between 2 groups(P>0.05). CONCLUSIONS:Based on convention-al treatment,Xuesaitong injection can effectively improve myocardial blood supply before PCI,decrease the level of inflammatory factor,relieve myocardial injury,improve cardiac function without increasing the incidence of ADR.

ObjectiveTo explore the anatomy and clinical significance of fixation combined with limited outside cut restoration gram needle internal fixation for distal radial fractures. MethodsBased on the anatomical observation and measurement of the forearm and hand. Found out the distal radial fractures fixed bracket screws and gram needle safety area and the needle into the body, and made reasonable angle respectively, to formulate standardized orientation, and applied to clinical procedures, and the clinical cases were followed up. The clinical curative effect was observed. ResultsThe place for the fixed bracket screws of the second metacarpal was the nail parts of radial dorsal partial ends. The proximal fracture for external fixation stents bolt into the nail near abductor muscle and manipulation long in the thumb short extensormuscles. Its feet side for stabbing abductor muscle and refers to a long stretch of radial side for tendon, radial side wrist short stretch muscles. The farthest distance between the lister nodule was (63.69 ±3.00) mm,nearest distance between the lister nodule was(123.64 ±5.00)mm. Gram needle do limited internal fixation security clearance for thumb long stretch tendon and refers to stretch tendons,radial side wrist length stretch tendons, thumb short extensor tendons, stretch retinaculum and this gap proximal distance lister nodule for ( 33. 52 ± 4.20) mm, remote distance lister nodule for( 8.81 ± 0. 88) mm. In clinical applications, wrist imaging recovery, function of ratings had no significant difference ( P > 0.05). But the postoperative complications of the two sets of patients had statistically significant difference ( P < 0. 05 ). ConclusionAccording to the anatomy of distal radial fractures,from the results of our research applied in fixation distal radial fractures in the operation,it had the advantages of simple operation,reset good,fixed firmly,and small trauma. It was worth of used widely in clinical.
8.Dominant Frequency Uncertainty Analysis of EEG α Activity in Pilots with Transient Ischemic Attacks

Objective To study the characteristics of EEG after tramsient ischemic attack and to offer reference for screening procedure of aircrew and astronaut selection. Method The dominant frequency uncertainty of alpha band EEG in 12 pilots(males; age 30±5) with transient ischemic episodes in middle cerebral artery(MCA) territories and in 20 normal healthy pilots was analyzed with frequency-fluctuation analysis. Result The dominant probability of the main frequency coinciding with sites affected by transient ischaemic attack(TIA) in patient pilots was higher than that in healthy pilots (P<0.01),and the dominant probability ratio logarithmic index I≥0 in all patient pilots with normal EEG, but I<0 in all healthy pilots. It was also found that not only I≥0, but the second component shifted to lower frequency(8 Hz) in patients with slight focal EEG alterations,i.e. slowing of frequency. The relative entropy values (percentage) were decreased significantly in pilots with TIA as compared with healthy pilots (P<0.05). Conclusion The dominant frequency uncertainty analysis of alpha band showed clear superiority of computerized evaluation over routine visual assessment for the diagnosis of minor cerebral ischemia. It offers not only a possibility of studying pathophysiological functional parameter, but also the reference for screening procedure in aircrew and astronaut selection.
9.Effects of activation of Src-family tyrosine kinases on human LECs apoptosis and EMT under the high glucose environment

Background Diabetic cataract is one of the major ocular complications in diabetes mellitus,including cortical cataract,nuclear cataract,subcapsular cataract and mixed cataract,and different cataractogenesis may be associated with lens epithelial cells (LECs).Subcapsular cataract is one of diabetic cataracts.Studying the biological behavior of LECs in subcapsular cataract is crucial for prevention and treatment.Objective This study was to investigate the effects of Src-family tyrosine kinases (SFKs) on apoptosis and epithelial-mesenchymal transition (EMT) of human LECs cultured by high glucose.Methods Human LECs (HLE-B3) were cultured for 24 hours with DMEM containing 5.5 mmol/L glucose (normal control group),DMEM containing 35.5 mmol/L glucose (high glucose group) and DMEM containing 35.5 mmol/L glucose + 10 μmol/L PP1,a specific inhibitor of SFKs (PP1 group).In 3,6,12 and 24 hours after culture,the apoptosis of human LECs was detected by flow cytometry assay;morphological change of human LECs was observed under the inverted microscope,and the expressions of the markers of EMT,E-cadherin and a-smooth muscle actin (α-SMA),in the cells were detected by immnofluorescence staining.In addition,the alternations of p-Src418 (active c-Src),bcl-xl,survivin,caspase-3,E-cadherin and α-SMA proteins were assayed by Western blot analysis.Results An elevated expression level of p-Src418 was found in LECs in the high glucose group and peaked 6 hours after cultured.The expressions of p-Src418(grey levels) were 0.125 ±0.036 in the high glucose group,and which was significantly higher than 0.042±0.011 in the normal control group and 0.035 ± 0.018 in the PP1 group,respectively (both at P<0.01).No remarkable differences were seen in the apoptotic rates between the high glucose group and normal control group in 6,12 and 24 hours after culture (all at P>0.05).The apoptotic rates of human LECs were(6.433±2.084)%,(10.333±2.610)% and (8.033±2.967)% in the PP1 group,which were higher than (3.233 ± 1.320) %,(3.533 ± 1.159) %,(5.733 ±0.230) % in the high glucose group and (3.133±1.170)%,(2.833±0.751)%,(3.333±1.201)% in the normal control group (all at P<0.05),however,there were significant differences in the apoptosis between the high glucose group and the normal control group (all at P>0.05).In 6 hours and 12 hours after cell culture,the expression levels of bcl-xl and survivin (grey values) in human LECs were significantly declined,but the expression of caspase-3 was increased in the PP1 group compared with the the high glucose group and the normal control group (all at P<0.05).The LECs showed slender in shape 24 hours after culture in the high glucose group,but the cell shape was close to the normal in the PP1 group.Western blot and immunofluorescence assay revealed that the expression of E-cadherin in human LECs was significantly reduced and that of α-SMA was significantly increased 6 hours after culture in the high glucose group compared with the PP1 group and the normal control group (all at P<0.05).Conclusions High glucose activates c-Src kinase of LECs in high glucose environment and therefore induces EMT and inhibits apoptosis.However,PP1 impedes the biological process of EMT and apoptosis of LECs to maintain the epithelial characteristics even under the stress of high glucose.
10.The effects of maxillary protraction in the treatment of skeletal class Ⅲ malocclusion of different skeletal maturation groups

Objective: To investigate hard tissue changes produced by maxillary protraction in patients with skeletal Class III malocclusion at different bone age,and to determine the relationship between the effect of maxillary protraction and bone age.Methods:75 subjects of skeletal Class III malocclusion with retruded maxilla were treated by maxillary protraction and rapid maxillary expansion for 6 months.Cephalometric radiographs were taken before and after treatment.The bone age of individual patients was assessed using cephalometric radiographs at the initiation of treatment on the basis of cervical vertebrace maturation indicators(CVMI).Patients were divided into three groups:Prepubertal growth peak group (CVMI Ⅰ-Ⅱ,8.2-11.3 years old), pubertal growth peak group (CVMI Ⅲ,10.2-12.3 yeasr old), and postpubertal growth peak group (CVMI Ⅳ-Ⅵ,11.5-14.5 years old).Results:(1)There was no difference in the maxillary advancement after maxillary protraction between the prepubertal growth peak and the pubertal growth peak groups;(2) in the postpubertal growth peak group, there was a decrease in maxillary skeletal advancement, whereas the dentoalveolar advancement was increased; (3) the posteroinferior rotation of mandible, the increase of lower facial height, and the eruption of maxillary molars showed no correlation with bone age.Conclusion:The importance of performing a biologic evaluation of bone age in the diagnosis and treatment planning of skeletal Class III malocclusions in individual patients must be emphasized.
