1.Expression and Significance of TRIM24 in Uroepithelium Cell Carcinoma of Bladder

Objective To detect the expression of TRIM24 in uroepithelium cell carcinoma of bladder(BUCC)and analyze the relationship be?tween its expression and clinical pathological factors. Methods Immunohistochemical methods were employed to evaluate protein level of TRIM24 in BUCC. Results TRIM24 expression was observed in the nuclear compartments of tumor cells,while normal epithelia of bladder tissues exhibited negative results. For 72 BUCC tissues,TRIM24 overexpression(score≥4)was observed in 41.6%of cases(30/72). TRIM24 expression correlat?ed with invasive depth(P=0.029)and patients with high TRIM24 expression had high grade of BUCC(P=0.043). Conclusion There was sig?nificant over expression of TRIM24 in BUCC,which was correlated with poor differentiation and high grade of BUCC. It may become a candidate tar?get gene for the treatment of human uroepithelium cell carcinoma of bladder.

3.Effects of Xuesaitong and Danhong injection on acute cerebral infarction and the effects of serum bilirubin and uric acid

Objective To investigate the comparison of blood uric acid and bilirubin levels on patients with acute cerebral infarction and the clinical curative effect of Donhong injection.MethodsA total of 68 patients with acute cerebral infarction from our hospital neurology, according to different therapeutic methods were divided into control group and experimental group with 34 cases in each group, two groups were treated by conventional treatment, patients in the control group were treated by blood flux injection 400 mg+5% glucose injection 250 mL intravenous drip, once a day;patients in the experimental group were treated by Dan red injection 40 mL+5% glucose injection 250 mL intravenous drip, once a day,2 groups of patients were treated for 2 cycles,a cycle with 7 d.Compared the clinical efficacy, blood uric acid, bilirubin, blood homocysteine, hyperensitive C-reactive protein, serum inhibition C and adverse reactions in the two groups after treatment.ResultsCompared with the control group, the experimental group clinical total effective rate was higher after treatment (P< 0.05).Compared with the control group, the experimental group blood uric acid, blood homocysteine, hypersensitive C-reactive protein, serum inhibition C levels were lower (P< 0.05), bilirubin level was higher(P< 0.05).Adverse reactions compared between the two groups, there was no statistically significant difference.ConclusionDonhong injection in the treatment of acute cerebral infarction clinical curative effect is distinct, dacrease blood uric acid level and increase bilirubin level, improve endothelial cell damage, protect the brain tissue.
4.Optimization of Ultrasonic Extraction of Total Saponins from Marsdenia tenacissima by Box-Behnken Re-sponse Surface Method and Its Purification Technology

OBJECTIVE:To optimize the ultrasonic extraction and purification technology of total saponins from Marsdenia te-nacissima,and to prepare purified total saponins from M. tenacissima. METHODS:Using the extraction rate of total saponins from M. tenacissima as index,ethanol volume fraction,solvent-solid ratio and extraction temperature as factors,ultrasonic extraction technology of total saponins from M. tenacissima was optimized by single factor test and Box-Behnken response surface design. Us-ing the content of total saponins in elution,the effects of ethanol elution volume fraction on purification technology of total sapo-nins by D101 type macroporous adsorption resin was investigated. RESULTS:The optimal ultrasonic extraction technology was as follows as ethanol volume fraction of 85%,solvent-solid ratio of 18.5∶1 (ml/g),extraction temperature of 80 ℃. In verification test,the average extraction rate of total saponins from M. tenacissima was 4.80%(RSD=1.06%,n=3);it was close to predicted value 4.89%,and the deviation was 1.84%. The optimal purification technology was as follows as 45% ethanol as eluant,90%ethanol for purification. Verification test showed that the average content of total saponins from M. tenacissima was 62.45%(RSD=0.88%,n=3). CONCLUSIONS:The optimized technology can be used for the extraction and purification of total saponins from M. tenacissima. The technology is stable and reliable.
5.Clinical observation for low-dose azithromycin with tiotropium bromide in the treatment of stable chronic obstructive pulmonary disease

Objective To study the efficacy and safety of low-dose azithromycin with tiotropium bromide in the treatment of stable chronic obstructive pulmonary disease.Methods A hundred and ten patients were randomized into three groups:tiotropium bromide group (36 cases,group A),azithromycin with tiotropium bromide group(38 cases,group B)and control group(36 cases,group C).Patients in group A were given tiotropium bromide (18 μg,q,d )in addition to conventional treatment.The patients in group B were given lowdose azithromycin (250 mg,twice a week) in combined with tiotropium bromide.The patients in control group were given the conventional treatment only.The courses of treatment lasted for six months.Results Compared with the control group,the frequency of acute exacerbation in patients treated with azithromycin and tiotropium bromide was reduced remarkably ( 2.1 ± 0.6 and 4.9 ± 0.7,t =18.5061,P < 0.05 ).The severity of clinic symptoms ( Cough 1.3 ± 0.5 vs.2.2 ± 0.6,P < 0.05 ),expectoration ( 1.0 ± 0.2 vs.1.7 ± 0.3,P < 0.05 ),anhelation ( 1.5 ± 0.8 vs.2.1 ± 0.6,t =3.6342,P < 0.001 ) ],life quality ( 29 ± 8 vs.42 ± 11,P < 0.05 ) and six-minutes walking distance( [ 370.00 ± 14.26 ] m vs.[ 290.00 ± 12.85 ] m,P < 0.05 ) of the azithromycin with tiotropium bromide group were improved significantly when compared with control.Compared with the tiotropium bromide group,the frequency of acute exacerbation ( 2.1 ± 0.6 vs.3.2 ± 0.8,P < 0.05 ),the severity of clinic symptoms (Cough 1.3 ±0.5 vs.1.8 ±0.4,P<0.05),expectoration( 1.0 ±0.2 vs.1.3 ±0.3,P <0.05) and anhelation( 1.5 ±0.8 vs.1.9 ± 0.6,P < 0.05 ) ],life quality ( 29 ± 8 vs.36 ± 10,P < 0.05 ) and six-minutes walking distance ( [ 370.00 ± 14.26 ] m vs.[ 330.00 ± 13.76 ] m,P < 0.05 ) were improved over those of tiotropium bromide group.Conclusion The long-term low-dose azithromycin in combinned with tiotropium bromide is good and safe in treating stable COPD.Therefore,it is worth of further clinical evaluation.

Objective To observe the expression level of CD4+ CD25+ Foxp3+ regulatory T cells in the peripheral blood of acute and stable senile COPD patients ,and analyze the correlation between Treg cells and TGF‐β1 of senile COPD ,then investigate the role of Treg cells and TGF‐β1 in the onset of senile COPD .Methods Totally 26 patients with acute stage and 23 patients with stable stage were investigated as acute group and stable group ,they came from the department of geriatric of our hospital form March ,2012 to February ,2014 .Meanwhile ,20 healthy people were selected as control group .The proportion of Treg cells in pe‐ripheral blood was measured by flow cytometry method and the level of TGF‐β1 in serum was measured by ELISA .Results The percentage of Treg on peripheral blood in acute and stable groups were significantly higher than control group(P< 0 .01) ,the difference also existed in acute and stable group(P<0 .05);the level of TGF‐β1 in serum of acute group was higher than control group(P<0 .01) ,there was no correlation between the proportion of Treg and TGF‐β1(P>0 .05) .Conclusion Treg cells may be involved in the process of the pathogenesis of senile COPD and acute exacerbation .There is no correlation between the proportion of Treg cells and TGF‐β1 ,and it indicates immune disorders may exist in senile COPD patients .

9.Effect of heme oxygenase on vascular remodeling in renal hypertension

AIM: To investigate the effect of heme oxygenase on vascular remodeling in renal hypertension. METHODS: Male Wistar rats were randomly divided into sham-operated, 2K1C (two-kidney one-clip) and hemin-induced groups. Four weeks after the treatments, the thickness of aortic media and HO enzymatic activity of the aorta were determined. Immunohistochemical staining was carried out to detect protein of HO-1 in the aorta. RESULTS: The blood pressure in 2K1C renal hypertension rats started to increase two weeks after the surgery and stabled at a high level at the 4th week. Hemin, an inducer of HO-1, markedly inhibited the increase in blood pressure. Aortic medium thickness of the 2K1C rats at 4th week was 27 5% thicker than that in the sham-operated rats. The thickness of aortic medium of the hemin-induced rats was 16 1% less than that in 2K1C group. At the 4th week after operation, protein level and enzymatic activity of HO-1 in aorta were higher than that in 2K1C group compared to those in the sham-operated group. CONCLUSION: Renal hypertension caused vascular remodeling and the activation of HO-1. HO-1 induction decreased the blood pressure of renal hypertension and reduced vascular remodeling.
10.Expression of αB-crystallin in X-ray irradiated rat lenses

Objective To detect the changes of soluble αB-crystallin in X-ray irradiated rat lens,and to explore the potential role of αB-crystallin in the pathogenesis of radiation-induced cataract.Methods The radiation cataract model was established with the accelerator linear in male Sprague-Dawley(SD)rats.The rats were divided into normal control group,experimental control group and X-irradiated group(the doses were 5,15,and 25 Gy,respectively).The rats were killed at 3 morlths post-irradiation,and the lenses were carefully isolated and homogenized.The protein changes of αB-crystallin in lens supernatant were measured by Western blotting.Results Typical radiation-induced cataract was observed in 15 and 25 Gy groups,while the lenses of the normal control group,experimental control group and the 5 Gy irradiation group remained transparent.The αB-crystallin protein level was significandy decreased in a dose-dependent manner(5 Gy group:0.871±0.085;15 Gy group:0.643±0.096;25 Gy group:0.338±0.160;F=40.764,P<0.05).Conclusions The decreased expression of the molecular chaperone αB-crystallin in X-irradiated rat lens indicates its important role in the pathogenesis of irradiation cataract.
