1.Effects of Food Protein Quality on Protein Metabolism of Skeletal Muscle in Young Rats

The experimental rats were fed with 5 test diets of different protein quality.RNA content and 3-methylhistidhie(3-MH) in urine were determined.The results showed that the RNA content of the skeletl muscle cells was significantly different in rais fed with different diet.A linear correlation between RNA content and amino acid score of food protein was perceived.Food with the higher protein quality,the more 3-MH in urine was excreted.A good correlation between excrstiou of 3-MH and Net Portein Utilization(NPU),an index reflecting protein quality,existed too.These suggested that protein quality regulates the balance between auabolism and catabolism,meaning the turnover of skeletal muscle proteinThe mechanism of strengthening anabolism,not reducing catabolism,can explain that high protein quality could improve growth of animals.
2.Experimental study on the mechanism of inducing apoptosis in human SPC-A1 cells by curcumin

AIM To investigate the mechanism of anti-tumor effects of curcumin on human SPC-A1 cells. METHODS Cell culture、fluorescence microscope、TUNEL method and irradiation immunity techniques were used. RESULTS ①By the action of curcumin, SPC-A1 cells grew against the wall and then peeled off gradually;fluorescence microscope showed the nucleus broke into pieces, and cell split into apoptosis bodies in different sizes; The TUNEL method further certified that apoptotic cells reached 42 67% at the concentration of 20 ?mol?L -1 curcumin at 24 h. ②The level of intracellular cAMP concentration in the SPC-A1 cells,treated with curcumin, was significantly higher than that of the control group. ③Treated with 20 ?mol?L -1 curcumin for 24 h, the expression of Caspase-8 mRNA of human SPC-A1 cells was up-regulated. CONCLUSION Curcumin can induce the apoptosis of SPC-A1 cells, and one of its molecular mechanism may be associated with the rising of the level of intracellular cAMP concentration, and up-regulating expression of Caspase-8.
3.Changes of serum procalcitonin level in patients with severe trauma and its relationship with multiple organ failure

Objective To investigate the changes of serum procalcitonin (PCT) level in patients with severe trauma,and to analyze its relationship with multiple organ failure (MODS).Methods From October 2013 to October 2018,the serum PCT levels were measured in 40 patients with severe trauma complicated with MODS (MODS group:40 cases) and 62 patients without MODS (non-MODS group:62 cases) in Yuncheng Central Hospital.The relationship between PCT and severity of trauma score (ISS),sequential organ failure (SOFA),acute physiology and chronic health status score (APACHE Ⅱ) and prognosis was analyzed.The predictive value of PCT in the prognosis of patients with MODS and severe trauma combined with MODS was analyzed by ROC.Results The serum PCT levels of the MODS group were (8.25 ± 2.35) μg/L,(7.26 ± 2.49) μg/L,(3.62 ± 1.75) μg/L on the 1 st,3rd and 7th day after admission,respectively,which were higher than those of the control group (t =7.561,9.561,8.251,all P <0.05).The serum PCT level in the MODS group increased with the increase of APACHE Ⅱ score and ISS score(F =12.108,9.173,all P < 0.05).The serum PCT level in the death group was higher than that in the survival group[(11.82 ± 3.75) μg/L vs.(4.92 ± 1.26) μg/L],and the difference was statistically significant (t =7.959,P <0.05).The area under MODS curve predicted by PCT for severe trauma patients was 0.848,with sensitivity and specificity of 80.00% (32/40),82.50% (33/40),respectively.The area under MODS curve predicted by PCT was 0.747,with sensitivity and specificity of 77.50% (31/40),72.50% (29/40),respectively.Conclusion The serum PCT level in patients with severe trauma complicated with MODS is significantly higher than that in patients without MODS,and its level is closely related to trauma,disease progression and prognosis.It is of great value to predict the occurrence and prognosis of MODS.
4.Naloxone or vagotomy does not influence centrally octreotide-induced inhibition of gastric acid secretion in rats.

To investigate the effect of preceding naloxone injection into the third cerebroventricle or acute subdiaphragmatic vagotomy on the gastric acid secretion inhibited by the somatostatin analogue octreotide given by intracerebroventricular (icv) injection. The third ventricles were cannulated in male Wistar rats anesthetized with sodium pentobarbital. One week later, acute gastric lumen perfusion was carried out. The gastric perfusion samples were collected every 10 min and were titrated by 0.01 mol/L NaOH to neuter. On the basis of subcutaneous injection of pentagastrin (G-5, 160 micro, g/kg), icv injection of physiological saline (group A, n = 20), icv injection of octreotide (0.05 micro g) (group B, n = 20), icv injection of naloxone (2.5 micro g)+octreotide (0.05 micro g) (group C, n = 20), acute subdiaphragmatic vagotomy+ icv injection of physiological saline (group D, n = 20), or acute subdiaphragmatic vagotomy+icv injection of octreotide (0.05 micro g) (group E, n = 20) were conducted. Before and after icv injection, 1-h total acid output (TAO) was determined and compared. The experimental data were expressed in change rate (%) of TAO. The change rates (%) of TAO were 4.60% in group A, -20.35% in group B, -18.06% in group C, 5.01% in group D and -21.59% in group E, respectively. Comparison of group B or C versus group A showed that P < 0.01 and comparison between the group E versus group D showed that P < 0.01. Whereas the differences between group C and group B, group E and group B were not statistically significant (P > 0.05 for all). The results indicate that the central inhibition of gastric acid secretion by octreotide may not be mediated by the endogenous opiate substance or its receptor and the peripheral pathway for icv injection of octreotide to suppress gastric acid secretion is via extra-vagus route.
5.Examination and analysis of spectrum ECG and ECG 、vectorcardiogram in patients with coronary heart discase

Objective: To explore the significance of examination of spectrum ECG in patients with coronary heart disease (CHD). Methods: Spectrum ECG and ECG、 vectorcardiogram examination were meanwhile executed in 254 patients with coronary heart disease. Results: Diagnose coincidence rate of CHD was 66.9% by spectrum ECG and ECG、 vectorcardiogram examination. Conclusion: Spectrum ECG is a new detect method for electrocardiogram information.
6.Analysis of Application of Antihypertensive Drugs in Wuhan Xiehe Hospital

Objective:To review the application of antihypertensive drugs in Wuhan Xiehe Hospital in the period of 1996 to 1998.Methods:Statistically analyze the application of antihypertensive drugs during the period in terms of their types,sales income,DDDs*d-1 as well as the changes of daily expense on drugs.Results:During the period analyzed,the newly applied drugs are mainly ACEI drugs.The DDDs*d-1 of metoprotol,benazepril,nifedipine and nimodipine had been among the highest.The increase of DDDs*d-1 was rapid.The newly prescribed drugs were mainly oral and retard preparations.Conclusion:The Prescription of antihypertensive drugs in Wuhan Xiehe Hospital was rational and in accordance to the principles of convenience and cost-effectiveness.
7.Construction and identification of the recombinant eukaryotic expression plasmid pHsp70-hsv-tk

Objective: To construct the recombinant eukaryotic expression plasmid pHsp70-hsv-tk(pHT)for further target gene therapy research.Methods: The oligomers of primers were synthesized based on the reported sequences of hsv-tk and with the pHSV-106 plasmid as the template.Then the hsv-tk gene fragment was amplified and inserted into the pD3SX vector.E.coli DH5? was transformed with the recombinant plasmids and the bacterial colonies containing the recombinant plasmids were identified by the PCR cloning method,and the recombinant plasmids were extracted and purified.Results: The recombinant eukaryotic expression plasmid pHT was obtained.The target gene obtained by PCR amplification had the same molecular size as predicted.The recombined pD3SX plasmid contained correct recombinant hsv-tk sequences.Conclusion: The plasmid pHT was successfully constructed,which can be used for further target gene therapy research.

9.Experimental study on expanding the space of microwave coagulation in liver

Objective To explore ways for expanding the now-limited coagulation volume of percutaneous microwave coagulation therapy(PMCT) in liver in order to enhance the complete necrosis rate of large hepatic tumors with obviously fewer punctures. Methods Fresh pig livers in vitro and rabbit, pig livers in vivo with and without blood supply were coagulated using the self-designed HM-WIA microwave coagulator to make clear antenna length, microwave power, coagulation time and blood flow influence the microwave volume(MCV).Results The device enlarged the MCV by 3 times using single electrode with an antenna of 36 mm in length along with microwave power of 45-55 W and coagulation time of 15-19 min,the short axis of the coagulation area was 5.6 cm in vitro, 5.3 cm in vivo with no blood supply.The antenna 36 mm in length was proved the best in keeping the finest coagulation shape.The MCV was 2.8 times as large without blood supply as with it (P
