1.Influences of adsorption film and worn spot under different hematocrit on the blood lubricant capacity

BACKGROUND:As a lubricant of artificial organs,lubricant capacity of blood is influenced by many factors,including hematocrit.OBJECTIVE: To explore the effect of hematocrit on blood lubricant capacity.METHODS: The lubricant capacities of four Kinds of hematocrit blood were studied by GB3142-82 lubricants-determination of bearing capacity testing.In addition,the differences of worn spots and the adsorption film formed on the steel ball under different hematocrit blood lubricating were observed by the Leica DMI 5000M material microscope.RESULTS AND CONCLUSION: ①The microshape of the adsorption films was depended on the hematocrit.②The greater hematocrit levels,the smaller worn spots formed on the steel ball,namely,increased hematocrit lead to stronger bearing capacity.
2.Protective Effects of Shuluo Sterile Powder for Injection on Injury Induced by Anoxia and Hypoglycemia in Cultured PC12 Cells

Objective To investigate the effects of the serum contained Shuluo sterile powder for injection (SLI) on injury induced by anoxia and hypoglycemia in cultured PC12 cells. Methods Cultured PC12 cells were treated with Na2S2O4 and Earle′ s liquid with glucose free to induce models of anoxia and hypoglycemia. The protective effects of the serum contained SLI on the two kinds of models were evaluated by the activity of lactate dehydrogenate (LDH) and staining with MTT assay. Results The serum contained SLI counteracted the injury induced by anoxia and hypoglycemia to various degrees and prevent the efflux of LDH. Conclusion The serum contained SLI can protect PC12 cells from injury induced by anoxia and hypoglycemia.
3.Expressions of heat shock protein in uterine cervix with different pathological changes

Objective To study the expressions of main subtypes of heat shock protein (HSP) in uterine cervix with different pathological changes. Method 478 cases of cervical biopsy specimen were divided into invasive carcinoma of cervix group (63 cases), cervical intraepithelial neoplasia group (CIN, 106 cases), chronic cervicitis group (293 cases) and normal uterine cervix (16 cases) according to pathological diagnosis. The expression levels of HSP70, HSP90α and HSP90β mRNA were detected by quantitative RT-PCR with specific complex cRNA as internal control. Results (1) The expressions of HSP70, HSP90α and HSP90β mRNA were significantly downtrend stepwise in invasive carcinoma of cervix, CIN, chronic cervicitis and normal cervix tissue. (P<0.01, respectively). (2)In the invasive carcinoma of cervix group, the expression level of HSP90β mRNA was higher in advanced stage (FIGOⅡb-Ⅳ) compared with incipient (FIGOⅠa-Ⅱa) cancer of the cervix. (P<0.05). (3)The expressions of HSP70 and HSP90β mRNA were each higher in poorly differentiated tumor than in well-differentiated tumor. (P<0.05, respectively). (4)The expression levels of all three HSP mRNA had no significant differences were observed with different histological types of cervical cancer. (P>0.05). Conclusions Heat shock protein may play some important roles in malignant transformation of cervix cell and aggravation of cervix cancer. HSP70 and HSP90α may promote cancer cell transition and proliferation, and HSP90β may participate in cell differentiation.
4.Expressions of heat shock protein in cervical cancer and precancerosis.

(4) The expression levels of all three HSP mRNA had no significant differences, it was observed with different histological types of cervical cancer (P > 0.05).

5.Effects of Yangyin Huoxue Prescription on the imbalance of Treg/Th17 and atherosclerosis in ApoE-/- mice induced by high-fat diet

Atherosclerosis(AS), characterized with the accumulation of lipids on the vessel wall, is an immune-related inflammatory disease which promotes the progression of cardiovascular diseases(CVD). The imbalance of Treg/Th17 accelerates the progression of AS. Yangyin Huoxue Prescription(YHF)is an efficient traditional Chinese medicine used in the treatment of AS, but the effects of YHF on the balance of immunity have still not been clarified. This project was designed to investigate the effects of YHF on the imbalance of Treg/Th17 and AS in ApoE-/- mice induced by high-fat diet(HFD). ApoE-/- mice were given HFD to induce AS and administered low-dose YHF(18 g/kg)or high-dose YHF(36 g/kg)for 20 weeks. Atherosclerotic plaque area was analyzed by oil red O staining. Serum lipids were measured by biochemical kits. Treg or Th17 cells in peripheral blood were detected by flow cytometry. mRNA and protein expression of Foxp3 and RORγt of aortas were determined by qRT-PCR, Western blot and immunohistochemistry. Splenic CD4+T cells of mice were isolated and activated by anti-CD3/CD28, and then treated with lipopolysaccharide(LPS)and YHF. The expression of mRNA and protein of Foxp3 and RORγt were detected by qRT-PCR and immunofluorescence. It was found that YHF reduced the plaque area, decreased lipid level and increased the ratio of Treg cells in peripheral blood. Moreover, YHF increased mRNA or protein expression of Foxp3 in aortas in vivo or CD4+T cells in vitro while decreasing mRNA or protein expression of RORγt. These results suggested that YHF can regulate the imbalance of Treg/Th17 in ApoE-/- mice induced by HFD, and reduce the inflammatory stimulation of LPS on CD4+T cells, thereby improving AS.
6.Effects of Mycobacterium tuberculosis HSP65 on Treg/Th17 immune balance in ApoE-knockout mice

To investigate the effects of Mycobacterium tuberculosis heat shock protein 65(HSP65)on Treg/Th17 immune balance in ApoE-knockout(ApoE-/-)mice, ApoE-/- mice with a high-cholesterol diet were immunized with M. tuberculosis HSP65. Sera were obtained for measurement of anti-HSP65 antibodies by ELISA; the effect of administration of different antigens was investigated, respectively, using flow cytometry analysis on the number of CD4+CD25+Foxp3+Tregs and CD4+IL-17+ Th17; the production of cytokines(IL-10, TGF-β1, IL-17 and IL-21)by these cells were determined by ELISA; total plasma cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol(HDL-C)and low-density lipoprotein cholesterol(LDL-C)levels were detected by biochemical autoanalyzer. Atherosclerotic lesions were measured by lipid deposition stained with oil red O. The results demonstrated that the levels of anti-HSP65 IgG antibodies were increased significantly in Mycobacterium tuberculosis HSP65-treated ApoE-/- mice, revealed obvious decrease in Treg number, Treg related cytokines(IL-10, TGF-β1)levels and significant increase in Th17 number, Th17 related cytokines(IL-17 and IL-21)levels, the levels of TC, TG, HDL-C and LDL-C did not change between groups, while the atherosclerotic lesions significantly increased. Results indicate that M. tuberculosis HSP65 could interrupt the Th17/Treg immune balance in ApoE-/- mice, suggesting a potential role in the formation and progression of atherosclerosis.
7.Research progress of preventing thrombosis in blood pump of ventricular assist devices

Blood pump is the core component of artificial ventricular assist device, and thrombosis is a severe complication of blood pump in clinical application. Methods of controlling and reducing thrombosis include materials surface modification, structure and parameters optimization of blood pump, and others. The typical symptoms of thrombosis and the hazard of various types of blood pump, the formation mechanism and primary factors for thrombosis, and the simulation prediction models for thrombosis were reviewed in this paper.
