1.Mechanism of Flk-1~+ MSC alleviating injury in the mouse model of renal ischemia-reperfusion

Objective: To investigate the mechanism underlying the repairing effect of Flk-1+ MSC on injury.Methods: We established the mouse model of acute renal ischemia-reperfusion,perfused murine Flk-1+ MSC within 1 hour or after 10 days in the acute and chronic groups,respectively,determined the homing of GFP positive Flk-1+ MSCs and the percentage of proliferating cells in the injured renal tissues by immunohistochemistry,and identified chronic fibrosis after injury by Masson trichrome staining. Results: Tissue injury and inflammation were significantly alleviated in the acute group,exhibiting a good long-term recovery,but no significant improvement was observed in the chronic group.The number of PCNA positive cells reached the peak at 3 days in the acute group,as compared with 10 days in the control,indicating that MSC engraftment effected an earlier endogenous repair.GFP staining showed that most of the MSCs homed to the interstitial region and the rest to the renal tubule.Conclusion: Flk-1+ MSC has a repairing effect on renal injury via alleviating acute inflammation and launching endogenous repair,while direct differentiation from Flk-1+ MSC is not the primary mechanism of its therapeutic efficacy.
2.Repair of intestinal irradiation injury by Flk-1~+ MSCs and SP cells

Objective Comparing biological characteristics and the roles in repairing of intestinal irradiation injury of bone marrow derived Flk-1+ MSCs and small intestinal derived SP cells.Methods Phenotypes of the two cell population were determined by flow cytometery.Flk-1+ MSCs and intestinal SP cells were infused into irradiated mice,distribution and differentiation of these cells were determined.Histopathological change and fibrosis were evaluated.Results The phenotypes of Flk-1+ MSCs and intestinal SP cells were quite different.Both Flk-1+ MSCs and intestinal SP cells could home to injury tissue rapidly,participate intestinal epithelial repair,and alleviate fibrosis and collagen deposition after irradiation.SP cells mostly engrafted into intestinal epithelium,while Flk-1+ MSCs partly engrafted into interstitial region as well as epithelium.Conclusion Flk-1+ MSCs and intestinal SP cells may alleviate symptoms effectively,though their mechanisms are disparite.
3. Analysis of Diagnosis and Treatment of Gastroesophageal Reflux Disease in A Hospital

The incidence of gastroesophageal reflux disease (GERD) increased year by year and with a high risk of recurrence. The effect of proton pump inhibitors (PPIs) is not sufficient in some of the GERD patients. Aims: To investigate the status of diagnosis and treatment of GERD in a hospital. Methods: A total of 312 GERD patients from January 2017 to March 2018 at the 909th Hospital/the Affiliated Dongnan Hospital of Xiamen University were enrolled. Frequency of use of PPIs, initial therapy regimen, replacement of PPIs and dose of PPIs, use of antipsychotics, course of therapy and reexamination were retrospectively analyzed. Results: About 1/3 patients received the treatment of antipsychotics. The initial therapy was mainly PPIs+antacid or PPIs+antacid+prokinetics. PPIs was replaced in 51 patients, and the dose of PPIs was increased in 286 patients. And 54 patients received standard course of treatment. Conclusions: The treatment regimen of GERD is basically rational, however, health education should be strengthened and patient's compliance be elevated for increasing the treatment efficacy of GERD.

Objective To detect the change of exoression level of plasma microRNA‐21(miR‐21) and TGF‐β1 in cardiac remode‐lin affer acute myocardial infarction(AMI) of the pateins .Methods 200 pateints with AMI and 100 normal controls(age ,sex matched) were enrolled .Blood samples were obtained from the normal controls and patients with AMI on the 3 days ,7 days and 14 days .Real‐time PCR was developed to detect the expression of miR‐21 and TGF‐β1 in plasma .Results The expression of miR‐21 was significantly up‐regulation in the 3 days ,7 days and 14 days in MI group than that cntrol group ,0 .74 ± 0 .21 vs .2 .62 ± 0 .23 , vs .3 .67 ± 0 .25 ,vs .4 .13 ± 0 .27 up‐regulation in the 3 days ,7 days and 14 days in MI group than that cntrol group ,0 .98 ± 0 .18 vs .2 .35 ± 0 .24 ,vs .3 .67 ± 0 .25 ,vs .4 .13 ± 0 .27 ,P<0 .05 ,respectively .The expression of miR‐21 and TGF‐β1 were up‐regulation with the change of cardiac function .Positive relationship between miRNA‐21 expression and LVDd (r=0 .757 ,P<0 .05);Positive relationship between TGF‐β1 mRNA expression and LVDd(r=0 .701 ,P<0 .05) .Conclusion The expression of miR‐21 and TGF‐β1 were up‐regulation in cardiac remodelin affer AMI of the pateins ,which involved in regulation in cardiac remodelin affer AMI .
5.Role of the IGF/HGF/Gln on the culture of the intestinal organoid in vitro

Objective: To investigate the nutritious effects of IGF, HGF and Gln on intestinal mucosal precursor cells in vitro.Methods: Different combinations of IGF,HGF and Gln were used to observe their effects on the expansion and development of the cultured intestinal mucosal precursor cells.Results: The combination of IGF,HGF and Gln could markedly promote the expansion of cultured intestinal mucosal precursor cells.Although IGF or HGF alone could promote the expansion of intestinal mucosal precursor cells, they had little effects on the development.On the contrary,Gln alone can promote the development of intestinal mucosal precursor cells,but it had little effects on their expansion.Conclusion: IGF,HGF or Gln alone has little effects on the expansion and development of the intestinal mucosal precursor cells.When they are used together,they can efficiently provide the essential stimuli for the expansion of the cultured intestinal mucosal precursor cells in vitro and enable the recombination of the different cells and formation of intestinal mucosa-like structure in vitro.

OBJECTIVETo observe the in vivo inhibition effects of tumor necrosis factor-alpha (TNF-alpha) gene transduced tumor drainage node of lymphocytes (DNL) from tongue cancer on SCID mice transplanted tumor.
METHODS15 human tongue carcinoma models were established in SCID mice by subcutaneously injection of squamous cell carcinoma line Tca8113. TNF-alpha gene introduced DNL, combined with low dose Pinyancin (PYC), were locally injected into tumor site. The inhibition rate was determined by the weights at the 8th week after tumor dissection and fresh specimens were prepared and subject to histopathologic examination under transmission electron microscope, and in situ TUNEL was used to detect apoptosis.
RESULTSThe TNF/DNL and rIL-2 group, and the TNF/DNL and rIL-2 and PYC group both exerted a strong inhibition effect on the implanted tumor. Treated tumors of the TNF/DNL and rIL-2 and PYC group were significantly reduced in comparison with those of the TNF/DNL and rIL-2 group (P < 0.05). The apoptosis of tumor in the TNF/DNL and rIL -2 group was evidenced based on transmission electron microscope and TUNEL analysis, and the apoptosis index was higher than that of control group (P < 0.05).
CONCLUSIONLocal injection of DNL modified with TNF-alpha gene, combined with low dose PYC, exert a synergistic antitumor effect. Apoptosis may be an important mechanism of squamous cell carcinoma killed by TNF/DNL.

8.Space of Range of Motion of Index Finger and Application

Objective:To measure the space of motion of index finger with and without injury, to apply it in rehabilitation. Methods:Thirty healthy undergraduate volunteers (controls) and three undergraduate volunteers with recent index finger injury (patients) were asked to flex, extend, adduct, abduct and ring the index finger independently and combined with middle finger, twice for a motion with an interval of 30 seconds. The index finger movement trajectory was recorded with a high-definition camera, and analyzed the range of flexion, extension, adduction, abduction and ring rotation motion with MATLAB. The movement space and defect of the injured fingers were drawn. Results:The ranges of independent flexion-extension and adduction-abduction reduced 26.5% and 24.6% in the patients compared with those of the controls, respectively. The defects of ring rotation space were very different with the degrees of recovery, and the maximum loss happened on 160°, 70° and 170°. Conclusion:Loss in index fingers motion space in range and direction may help for rehabilitation assessment.
9.Research progress of needle-free injection technology

Needle-free injection technology (NFIT) refers to the drug delivery systems in which drugs are propelled as high-speed jet streams using any of the pressure source to penetrate the skin to the required depth. NFIT is a promising drug delivery system as it enables the injection of liquids, powders, and depot/projectiles, and has the advantages of preventing needle stick accidents, improving drug bioavailability, eliminating needle-phobia, increasing vaccine immunity, simplifying operations and is convenient for patients to use. NFIT and its research background, the structure and classification of needle-free jet injectors (NFJI), drugs that can be delivered using NFJI and the factors affecting the injection effect are comprehensively reviewed in this paper. The limitations and potential development directions are summarized to provide a theoretical basis for the application and development of NFIT.
