1.Histopathological and Ultrastructural Changes of Brain Tissues after Experimental Complete Cerebral Ischemia in Rats

The selective vulnerability of brain tissues after 30 minutes of complete ischemia was observed 5, 15, and 30 minutes after ischemia and 15, 60, and 180 minutes after reperfusion in rats under optical and electron microscopies. It was found that the cerebral cortex especially the third and fifth lamina, the H1 segment of the hippocampus, and the Purkinje cells of the cerebellum were most liable to be damaged after ischemia. After reperfusion, the alterations of membrane system, nuclei and organelles were significant. The membraneous structures which contained a large amount of lipids were more severely damaged. At the same time cerebral edema was found, the severity of which was correlated with the intensity of histopathological and ultrastructural changes of the brain and the degree of blood brain barrier disruption.The findings suggest that oxygen free radicals and lipid peroxides may play an important role in the development of the damages in the brain mentioned above after reperfusion.
2.Detection and antimicrobial resistance of hypermutable Pseudomonas aeruginosa strains

Objective To investigate the isolation and antimicrobial resistance of hypermutable Pseudomonas aeruginosa strains.Methods The cultured bacteria were identified by API 20NE system.The susceptibilities of bacteria were detected by disk diffusion method and the results were confirmed with the rules of NCCLS/CLSI.The hypermutable strains were identified byhigh concentration of rifampin-resistant growth assay.Results Among analyzed 100 strains,37 strains(37.0%) were hypermutable.The resistant rates of hypermutable strains to imipenem,meropenem,cefoperazone/sulbactam,piperacillin/tazobactam,ceftazidime,cephfime,aztreonam,amikacin and ciprofloxacin were significantly higher than those of non-hypermutable strains.The multidrug resistance rates of hypermutable strains were also higher than those of non-hypermutable strains.Conclusion The hypermutable strains of Pseudomonas aeruginosa sourced from chronic respiratory tract infections dominantly.The hypermutable strains were hyper-resistant and multidrug resistant to many antibiotics,so strengthened monitoring should be encouraged to control the prevalence of hypermutable Pseudomonas aeruginosa strains.
3.Effect of Corticosterone on Morphine-induced Psychomotor Effect and Its Sensitization

Objective: To explore the role of corticosterone in morphine-induced acute psychomotor activity and behavioral sensitization.Methods: 32 male SD rats were divided into four groups. Rats were intraperitoneally given saline, morphine, corticosterone and corticosterone/morphine separately. The horizontal locomotor activity was recorded for 60min by the computer trace system automatically. The locomotor activity was measured in every two days for total seven times. 4.Reversal effect on ADM resistance in human leukaemia cell subline resistant to ADM(K562/ADM) of anti-FasL

Purpose:To explore the effect of anti-FasL on ADM resistance of K562/ADM cell subline and its possible mechanism.Methods:Control group,untreated with VER or anti-FasL ,VER-treated group,anti-FasL-treated group and anti-FasL +VER-treated group were set up. MTT、FCM、TUNEL、S-P immunechemistry and RT-PCR assay were used for detection.Results:Anti-FasL or VER treatment was capable of strengthening sensitivity of K562/ADM cells to ADM,ID 50 of anti-FasL and VER was 200ng/ml and 6?g/ml,respectively;DNA content of apoptosis from cycling analysis curve of K562/ADM cell was 7.42%,32.10%,49.29%,56.26%,respectively,and positive rate in situ apoptosis was 4.5%,36%,45%,58%,respectively,which represented a tendency of gradual increase. Anti-FasL was capable of effecting FasL and bcl-2 expression,but not Pgp.Conclusions:Anti-FasL alone or synergically with VER was capable of reversing ADM resistance of K562/ADM cell,which may be used as a novel reversor of multidrug resistance.
5.Comparision and application of detective methods for Her-2 gene in invasive breast carcinoma tissue

Objective Through comparision of Her-2 gene in invasive breast carcinoma tissues assayed by IHC, CISH and FISH respectively, to explore an optimal detective methods. MethodsIHC, CISH and FISH were used in detection for Her-2 gene of 100 cases, respectively.ResultsCISH identified no Her-2 gene amplification in IHC score negative rumors (6/6) and gene amplication in 7.1%(1/14) of IHC score (+) and in 32 % of IHC score (++) tumors(l6/50) and in 80 % of IHC score (+++) (24/80), Concordant rate between CISH and FISH was 100 %(20/20). ConclusionThere are advantage and disadvantage among IHC,CISH and FISH. CISH is an accurate and dependable methods and would be easy to apply for detection of Her-2 gene.
6.Application of sonography for arthrocentesis in the hip

ObjectiveTo evaluate the value of uhrasonography for arthrocentesis in the hip and the advance of the femoral head/acetabulum as the puncture location in patients without sonographically obvious joint effusion.Methods Forty-three patients diagnosed with ankylosing spondylitis or osteoarthritis were included,82 hip joints were involved and received ultrasounded-guided intra-articular injections.The puncture site was the joint space between the femoral head and the acetabulum.The times,depth,procedure time,side effects and complications of each injection were recorded.ResultsAll 82 hips could be explicitly displayed.The depths from the skin surface to the joint space were (3.4 ± 0.9)cm (left)and (3.1 ± 0.8)cm (right),respectively.Overall,100% of hip joints were punctured successfully,and 95.1% of them were succeeded with the first arthrocentesis.The procedure time was (287.9 ± 45.8) s.No side effects or complications were observed.ConclusionsThe sonographic technique could be used as a primary method of image guidance for performing hip arthrocentesis and the joint space between the femoral head and the acetabulum could be a choice of target location for the patients without joint effusion.
7.Ultrasonographic characteristics of biliary stricture after orthotopic liver transplantation

Objective To evaluate the charateristics of biliary stricture after orthotopic liver transplantation(OLT) by ultrasound. Methods Diametes,thickness,characteristic echo of bile were observed by ultrasound in 41 patients with biliary stricture after OLT,and the results were compared with those of 46 patients without biliary complication after OLT.Results The diameter of intrahepatic bile,thickness of bile,incidence of intrahepatic bile dilation or hyperechogenicity in biliary stricture patiens were higher than those in patients without biliary complication,there were significant differences between two groups(P<0.05).Conclusions The series of ultrasonographic character such as bile dilation,thickeness or hyperechogenicity of biliary wall,thin of bile duct are available to diagnose biliary stricture.
8.Investigation of the change of adiponectin level with menopause status in middle aged women and its relationship with androgen

Objective To investigate the change of adiponectin level with menopause status in women aged 40 to 65, and its relationship with androgen. Methods A cross-sectional study included woman (aged from 40 to 65) who were in hospital for routine check-up at the Sun Yat-sen Memorial Hospital from August to October in 2013. All subjects underwent laboratory examinations of adiponectin, sex hormone binding globulin (SHBG), dehydroepiandrosterone-sulfate (DHEA-S), total testosterone (TT), collected anthropometric measurements and then calculated free androgen index (FAI) and body mass index (BMI). According to their menstrual status, the subjects were divided into 4 groups: premenopausal group with 119 subjects, perimenopausal group with 60 subjects, early postmenopausal group with 62 subjects, late postmenopausal group with 64 subjects. Results (1) Adiponectin levels declined to its lowest level in menopausal transition and gradually becoming higher after menopause, which showed a U-shaped trajectory. When compared adiponectin levels in late postmenopausal group [(13 ± 5) mg/L] with those in perimenopausal [(8 ± 6) mg/L] or early postmenopausal group [(9 ± 6) mg/L], it all showed significantly difference (P<0.05). (2) Both the adiponectin levels were negatively correlated with waistline in the 4 groups (premenopausal group, r=-0.276;perimenopausal group, r=-0.334;early postmenopausal group, r=-0.211;late postmenopausal group, r=-0.218; all P<0.05). Levels of adiponectin were positively correlated with SHBG (r=0.536, P<0.05) and negatively with FAI (r=-0.363, P<0.05) in menopausal transition, while in late postmenopausal group, negatively correlated with level of DHEA-S (r=-0.450, P<0.05). When adjusted for age, BMI and waistline, the above correlations still exist. Conclusions Adiponectin levels declined to its lowest level in menopausal transition and gradually becoming higher after menopause, which showed a U-shaped trajectory during the sequential menopause status transition in middle aged women. Low level of adiponectin in menopausal transition is closely associated with the relative excess androgen occurred during this stage.
9.Application of needle aspiration cytopathology in the lymphoma diagnosis

Objective To investigate the feasibility of fine needle aspiration cytopathology in the diagosis of lymphoma.Methods To compare cytopathology with histopathology of 72 lymphoma cases and analyse immunochemistry stain of 3 cases.Results 61 of 72 cases were diagnosised as lymphoma in cytopathology,while 58 of 61 cases were diagnosised as lymphoma in histopathology.The sensitivity,specificity,accuracy,positive predictive value and negative predictive value rates of cytopathology were 89.2 % (58/65),57.1% (4/7),86.1% (62/72),95.1% (58/61),and 36.4 % (4/11) respectively.Conclusion Fine needle aspiration cytopathology is significant to the diagnosis of lymphoma.It should greatly improve the accuracy of lymphoma diagnosis and make it possible to subclassify lymphoma,combined cytopathology with other auxilary detection.
10.Research on Objective Method of Traditional Chinese Medicine Color Inspection

This study was aimed to objectify Color Inspection in Traditional Chinese Medicine (CITCM). A quantita-tive system for CITCM was designed and developed. The entire system included two parts, which were the hardware and the software. The hardware was an image acquisition device in a standard lighting condition. The software was used for digital image processing. The chromaticity of facial special region (SR) corresponding to five internal organs were calculated. The system was carried out by taking 100 samples of people. It was concluded that the experiment verified the effectiveness of the system in objective study of CITCM. It can be used as basis for the further study on CITCM.
