2.Effect of nimesulide on the levels of IL-8 and TNF-αin gingival cervical fluid in patients with chronic periodontitis and its clinical efficacy

Objective To investigate the clinical efficacy of nimesulide in the treatment of chronic periodontitis patients,and analyze the effect of IL-8 and TNF-αlevels in the gingival sulcus fluid.Methods 88 patients with chronic periodontitis in our hospital between June 2014 to December 2015 were divided into control group and observation group by the random number method, 44 cases in each group, the control group patients were given metronidazole treatment, observation group patients were given nimesulide treatment,two groups therapeutic effect and the incidence of adverse reactions were observed, compared two groups of patients before and after treatment in gingival sulcus fluid IL-8 and TNF-αlevel, compared sulcus bleeding index (SBI), plaque index (PLI) and clinical attachment level (CAL),before and after treatment two weeks and four weeks of probing depth (PD) of two groups.Results The total effective rate of observation group patients was 95.55%, significantly higher than the control group patients’total efficiency (81.82%) (χ2 =4.06, P=0.04), after treatment in the observation group gingival sulcus fluid IL-8 factor and TNF-αlevels were significantly lower than the control group (P<0.05), After 2 weeks and 4 weeks of treatment, PD, SBI and CAL indicators observation group were significantly lower than the control group ( P <0.05 ) , the incidence of adverse reactions of observation group patients was 4.55%, significantly lower than the incidence of adverse reactions in control group patients(22.73%)(χ2 =6.18, P=0.01).Conclusion Nimesulide in the treatment of chronic periodontitis with a highly efficient clinical efficacy, while reducing gastrointestinal side effects, etc., can effectively reduce the gingival sulcus fluid IL-8 and TNF-αlevels, ease clinical symptoms, is a highly effective treatment.
3.Ultrasound guided excision of breast benign disease

Objective To investigate the value of minimal invasion technique in treatment of breast benign disease.Methods 204 benign breast tumors of 74 women were completely excised by using Mammotome or VACORA.Results 108 masses were confirmed fibroadenoma,66 fibroadenosis,30 adenosis of breast.Postoperative followed up 3~12 months:there were 2 cases of local hematoma,5 cases of subcutaneously hematoma,1 case of recurrence,no infection.Conclusions There ale many advantages using uhrasound-guided minimal invasion technique to treat benign breast disease,including accurate localization,minimal invasion,less bleeding,easy performation,less complications and good cosmetology.
4.Circadian blood pressure analysis in hypertensive patients with chronic renal insufficiency

Objective To investigate the circadian blood pressure(BP)rhythm in hypertensive patients with chronic renal insufficiency.Methods Circadian blood pressure(BP)rhythm was studied by ambulatory 24-h monitoring among hypertensive(n=25,group B)patients with stable progression of chronic renal insufficiency,and among patients with essential hypertension(n=28,group A)without renal disease.Results ①The 24-h diastolic BP and nocturnal systolic and diastolic BP of group B were significantly higher than those of group A[(80.9±13.4)mmHg vs[70.3±15.6)mm Hg,(160.2±17.8)mm Hg vs(140.3+25.9)mm Hg,(82.6±16.1)mm Hg vs(68.8±20.2)mm Hg,P<0.05)].②Blooa pressure variability(BPV)of 24 hour systolic and diastolic BP of group B was higher than group A[(13.5±3.9)mm Hg vs(11.3±2.1)mm Hg and(9.2±1.2)mm Hg vs(8.3±1.8)mm Hg,P<0.05).BPV of nocturnal systolic and diastolic BP in group B was signitleantly higher than group A [(14.9±3.3)mm Hg vs(9.3±2.1)mm Hg and (9.7±2.4)mm Hg vs(8.0±2.2)mm Hg,P<0.01)].③The 24-h profile of BP of group A showed more non-dipper according for 64.3%(18/28),reverse-dipper according for 10.7%(3/28),but which was 40.O%(10/25)and 48.0%(12/25).Conclusion The renal patients show a disturbance in the 24-h profile of BP,higher indexes of noetttrnal BP and a greater nocturnal variability,which may be important factor for event of cardiovascular disease and renal insufficiency.
5.Probability of premature mortality due to four chronic diseases in Jiangmen City from 2012 to 2021

Objective:
To investigate the mortality of malignant tumors, diabetes, cardio-cerebrovascular diseases and chronic respiratory diseases and trends in probability of premature mortality due to these four chronic diseases in Jiangmen City, Guangdong Province from 2012 to 2021, so as to provide the evidence for perfecting the chronic disease control strategy.
Methods:
The mortality of malignant tumors, diabetes, cardio-cerebrovascular diseases and chronic respiratory diseases among household registered residents in Jiangmen City from 2012 to 2021 were collected from the Guangdong Provincial Population Death Information Registration Management System, and the crude mortality, standardized mortality by the population of the Fifth National Population Census in China in 2000 and probability of premature mortality were calculated. The trends in mortality and probability of premature mortality were analyzed using average annual percent change (AAPC), and whether achieving the targets for the probability of premature mortality due to four chronic diseases in 2025 and 2030 were evaluated.
Results:
A total of 226 012 deaths occurred due to four chronic diseases in Jiangmen City from 2012 to 2021, and the overall crude mortality and standardized mortality rates were 569.22/105 and 283.29/105, with a reduction in the probability of premature mortality from 15.04% to 12.05% (AAPC=-2.403%, Z=-7.603, P<0.001). The probability of premature mortality due to four chronic diseases decreased from 19.21% to 16.38% in males, and from 10.42% to 7.58% in females (AAPC=-1.893% and -3.085%, Z=-5.262 and -9.811, both P<0.001). The probability of premature mortality due to diabetes appeared a tendency towards a rise (AAPC=2.317%, Z=2.548, P=0.034), and the probability of premature mortality due to cardio-cerebrovascular diseases showed a tendency towards a decline (AAPC=-4.826%, Z=-13.590, P<0.001), while no significant changing trend was seen in the probability of premature mortality due to malignant tumors or chronic respiratory diseases (AAPC=-0.751% and -2.461%, Z=-1.532 and -1.730, P=0.125 and 0.122). The predicted probability of premature mortality due to four chronic diseases was 10.92% in 2025 and 9.66% in 2030 in Jiangmen City, which were both lower than the target (11.21% and 9.81%).
Conclusions
The probability of premature mortality due to four chronic diseases appeared a tendency towards a decline in Jiangmen City from 2012 to 2021, which can reach the target in 2025 and 2030. Males should be given a high priority for interventions of chronic diseases, and diabetes control should be reinforced.
6.Impact of the systematic social skills training on recovery of chronic schizophrenia patients with negative ;symptom and social skills defects

Objective To explore the impact of the systematic social skills training on recovery of chronic schizophrenia patients with negative symptom and social skills defects.Methods A total of 82 cases of long -term hospitalized patients with chronic schizophrenia were randomly divided into intervention group and control group by digital table,41 cases in each group.The two groups received psychiatric routine therapy and nursing,and general entertainment therapy,and systemic social skills training was conducted in the intervention group for 12 weeks.PANSS and SSC were respectively used to evaluate the two groups before and after the intervention.Results After 12 weeks of systematic social skills training,the total score of SSC[(24.5 ±4.5)points]in the intervention group was apparently lower than that in the control group[(29.2 ±5.2)points],and the difference was statistically significant(t =3.212, P <0.01).Compared with the control group[(20.5 ±4.3)points,(27.2 ±4.2)points,(59.1 ±9.5)points],in the intervention group the score of negative symptom mental pathology and total score of PANSS were (14.9 ± 3.7)points,(24.5 ±3.5)points,(52.8 ±7.1)points.The differences of total score between the two groups were sta-tistically significant(t =5.199,3.317,3.239,all P <0.01),while the positive symptoms score was not statistically significant(P >0.05).Compared with before intervention,in the negative symptom table,the differences of total score N2,N3,N4,N6 had statistically significant differences(t =4.533,5.413,4.501,4.668,all P <0.01).Conclusion The systematic social skills training can greatly promote chronic schizophrenia patients with negative symptom and social skills defects,improve their social skills and life qualities.
7.Connection of hepcidin genes from two fish species and their expression in Pichia pastoris.

Hepcidin are small cationic peptides with antibacterial activity expressed mainly in the liver of living organisms, and they play important roles in the host's immune response against microbial invasion and regulation of iron metabolism. Thus, they are considered to be good substitutes for traditional antibiotics. It is a good choice that the antimicrobial peptides are prepared by recombinant DNA expression. In the present study, two hepcidin mature peptide cDNAs from channel catfish (Ictalurus punctatus) (mCH) and tilapia (Oreochromis niloticus) (mTH) were connected by SOE-PCR in order to obtain more recombinant hepcidin with broad antimicrobial spectrum, and EcoR I and Not I sites were added to 5'- and 3'- ends of the fragment, respectively. The recombinant eukaryotic expression vector "pPIC9K-mCH-mTH" was successfully constructed, and transformed into Pichia pastoris GS115. The transformants containing multicopy gene insertion were selected by using different concentrations of G418 and other specific mediums, and identified by PCR for yeast genomic DNA. Expression was induced by adding 1% methanol at 30 degrees C for different times. Tricine-SDS-PAGE analysis demonstrated that the most appropriate expression time was 72 h, at which a high expression yield (77 mg/L) for the target protein was exhibited. The highly purified target protein was obtained from the fermentation supernatant by SP-Sepharose cation exchange chromatography. Bacteriostatic activity assay demonstrated that the fermentation supernatant containing the target protein and purified recombinant target protein had bacteriostatic activities against gram-positive and gram-negative bacterium. The present result provides the important initial value for industrial production of hepcidin antimicrobial peptide.

8.The effect of low frequency electrical stimulation on basic fibroblast growth factor and epidermal growth factor expression and on endogenous neural stem cell proliferation in the brains of rats with acute cerebral infarction

Objective To observe the effects of low frequency electrical stimulation (LFES) on the proliferation of endogenous brain neural stem cells (NSCs) and on the expression of basic fibroblast growth factor (bFGF)and epidermal growth factor (EGF) in rats with acute cerebral infarction; to explore the therapeutic mechanism of LFES in improving neural function. Methods Fifty-four rats were randomly divided into a LFES group, a placebo stimulation group and a sham-operated group. Each group was further divided into 3rd day, 7th day and 14th day subgroups, with 6 rats in each subgroup. An acute cerebral infarction model was induced in the rats of the LFES and placebo stimulation groups by middle cerebral artery occlusion (MCAO). Three days after the operation, rats in the LFES group began LFES treatment (frequency 30 Hz, pulse width 250 μs, current intensity 3 mA, 10 min/d) ,while the placebo stimulation group was treated identically but without electricity. The rats in the sham-operated group had no special treatment. The expression of nestin positive cells in the subgranular zone of the hippocampus and the subventricular zone was detected by immunohistochemical staining. The expression of bFGF, EGF proteins and mRNA in the ischemic hemisphere was detected by Western blotting and RT-PCR analysis. A screen test was applied to evaluate motor function. Results Nestin-positive cells in the subgranular and subventricular zones of rats in the LFES group increased significantly more than in the placebo stimulation group at the 7th and 14th day. The expression of bFGF, EGF proteins and mRNA in the ischemic hemisphere was up-regulated compared to the placebo stimulation group at the 7th and 14th day. At the 14th day a difference in motor function was observed in rats in the LFES group compared with the placebo stimulation group. Conclusion LFES can promote the proliferation of endogenous brain NSCs and the expression of bFGF and EGF in rats with acute cerebral infarction. It can also improve motor function and enhance neural plasticity in the brain.
9.Effects of Different Processing Methods on the Antiosteoporotic Effective Compositions of Psoralea corylifolia L.

This study aimed at evaluating the effect of different processing methods on the contents of antiosteoporotic compositions in P.corylifolia.HPLC method adopted to analyze the contents of antiosteoporotic compositions,including coumarin (psoralen and isopsoralen),flavonoids (neobavaisoflavone,corylifolin,corylifolinin and corylifol A) and meroterpenes (bakuchiol) in P.corylifolia.The contents were analyzed before and after the processing methods.Results:The results showed that the content of coumarin,flavonoids and meroterpenes obviously changed in different processed P.Corylifolia in comparison with the crude drugs.In conclusion,it was demonstrated that the therapeutic efficacy of osteoporosis was enhanced by CP method which increased all of the three compositions.The testimony tallied with the Chinese medical theory:stir-frying with salt-water could enhance the effect of the tonification of spleen and kidney.
10.Apoptosis induced by satraplatin in human ovarian carcinoma cells A2780

AIM: To observe the growthinhibiting cell cycle-modifying and apoptosis-inducing effects of satraplatin on human ovarian carcinoma cell line A2780, and to explore its possible mechanism. METHODS: The effect of satraplatin on A2780 cells proliferation was determined using MTT, and the change in cell cycle was analyzed using PI staining. Morphologic change was visualized by fluorescence and electron microscopy. AnnexinV-FITC/PI staining multiparameter flow cytometry and immuno- histochemical TUNEL assay were used to detect apoptotic cells. The activity of caspase-3 and the effect of pan-caspase inhibitor on cell viability were measured as well. RESULTS: The growthinhibiting and apoptosis-inducing effects of satraplatin were dose-dependent and similar to those of cisplatin. Satraplatin mainly caused A2780 cell accumulation in S phase accompanied by minor accumulation in G2/M phase. Cells treated with satraplatin exhibited typical morphology of apoptosis. Satraplatin-induced increase in caspase-3 activity of A2780 cells was concentration-dependent. The viability of A2780 cells was affected by pan-caspase inhibitor z-VAD-fmk in a dose-dependent manner under certain concentration of z-VAD-fmk. CONCLUSION: Satraplatin-induced apoptosis in A2780 in vitro was observed. Caspase-dependent and independent pathways were involved in apoptosis induced by satraplatin, and the latter included caspase-3 dependent and non-caspase-3 dependent pathways.
