Objective To evaluate the effects of 0.3 kb human alpha-fetoprotein (AFP) promotor on the expression of enhanced green fluorescent protein (EGFP) in human hepatoma cells. Methods By constructing the primer with special enzyme site, human AFP promoter was selectively amplified from human gene bank and eukaryon-expressed pAF-EGFP and pAF-DTA with AFP promotor and EGFP and with AFP promotor and DTA, respectively. pAF-EGFP was transfected into HepG2, SMMC-7721, and NIH3T3. The fluorescence was detected in order to evaluate the transfection rate. 3.The clinical observation and CT image processing of nasal septum deviation and unilateral inflammatory maxillary sinus disease

0.05). Conclusion The difference of cross sectional area in nasal cavity resulted from nasal septum deviation. Compensatory hypertrophy of inferior turbinate, uncinate process and middle turbinate in wide side resulted in the similar proportion of nasal airway of two sides of nasal cavity. However, compensatory hypertrophy lead to obstruction and stricture, accordingly, the probability of unilateral inflammatory maxillary sinus diseases in wide side of nasal cavity was higher than that in narrow side. In clinical practice, the deviated nasal septum should be corrected at the time of dealing with inflammatory maxillary sinus diseases. The normal structure and function of nasal cavity were recovered. The decompensation due to septum deviation was blocked.
4.The protective effects of estradiol on ischemic rat myocardium during the early stage of reperfusion

Ovbective To study the protective effects of 17?-estradiol on isolated ischemic rat hearts in a modified Langendorff model. Methods Thirty rats performed bilateral ovariectomies were divided equally into three groups at random. A three-phase protocol was performed: (1) 15-minutes preperfusion,(2) 30-minutes global ischemia,(3) 30-minutes reperfusion. Treatment with 0.1% DMSO (group D) and 5 ?mol/L 17?-estradiol+0.1% DMSO (group E) dissolved in K-H buffer was perfused from sixth minutes of reperfusion. Only K-H was perfused in the ischemia-reperfusion group (group I-R). The left ventricular pressure (LVP) and it's first derivative (?dp/dt max ) and coronary flow (CF) were recorded at the 15th minute of preperfusion and the 30th minute of reperfusion. Creatin phosphokinase (CPK) and lactic dehydrogenase (LDH) the coronary effluent,the content of malonaldehyde (MDA) and superoxide dismutase (SOD) activity of myocardium were measured at the 30th minute of reperfusion. Objective:Peptides were designed on the basis of high conservative regions of amino acid sequences and structures of the SEs,three-dimension structure of P72 was constructed.Methods: Bioinformatics analysis softwares such as Vector NTI 10.3, InsightII 2000,Discovery Studio 1.7 were used to analyse and predict the space structure of P72.Results:three-dimensional domains of the peptide P72 from SEA, SEB and SEC were quite similar, Peptide P72 was far away from TCRVβchain and MHC class II molecule.Conclusion:The inhibitory activity of peptide P72 may not due to binding to MHCⅡ and TCRVβchain.The exact mechanism of inhibitory activity of P72 should be explored.

Background and purpose:The reason for hepatitis B virus (HBV) with hepatocyte specificity is PreS1 enchased on the hepatitis B virus envelope (HBVE). So HBVE may have a potential application in liver targeting gene transfer. In this study, we investigated whether HBVE has the ability to target liver cancer cells. Methods:HBVE was obtained from the supernatant of Hep G 2.2.15 cells through PEG8000 system and ?-propiolactone method. The pIRS2-EGFP was packed with HBVE and resulted in the product HBVE-GFP. HBVE-GFP was transfected into HepG2, A549, HeLa and FB cells. The green fluorescent protein (GFP) expression was observed under a fluorescent microscope. The rate of GFP positive cells was determined by flow cytometer.Results:The GFP could be observed in the four groups, but the HepG2 group had a higher fluorescent intensity than the other 3 groups. Objective To observe the specific expression of diphtheria toxin A chain gene(DTA) in Ewing's sarcoma nude mice models.Methods pS2DTA plasmid was constructed by the Lus gene in pS2 plasmid was replaced with the DTA gene in pIBI30 plasmid,and transformed into DH5?, then amplified.Five nude mice were injected with 2.4?10~(7) SKES1 cells,sacrificed on week 6,and the tumor tissues were implanted onto the back of another 20 nude mice.Till 6 weeks later the diameter of the tumor up to 0.8-1.0 cm,the Ewing's sarcoma nude mice models were successfully established and randomly assigned to receive the injection of 300 ?g lipidosome+100 ?l normal saline(control group) or 300 ?g lipidosome+100 ?l normal saline+100 ?g pS2DTA (experimental group) once a day for 5 d.On day 4 after the therapy finished,4 nude mice of each group were randomly sacrificed and the DTA expression in the tissue of tumor,liver and myocardium was detected by the immunohistochemistry and RTPCR.Results The immunochemical results showed DTA expression in the tissues of control group was all negative while the DTA expression in the experimental group was negative in the tissues of myocardium and liver,positive in tumor tissue.RTPCR showed DTA expression in control group was negative in all tissues while strongly positive in tumor tissue,weakly positive in liver tissue and negative in myocardiac tissue.Conclusion The pS2DTA regulated by the EWSFLI1 fusion gene has a specific expression in transplanted Ewing's sarcoma in mice.
9.Effects of acute hypercapnia and cedilanid on diaphragmatic functions in dogs

After electrical stimulation of the phrenic nerve,the function of the diaphragm was assessed by measurement of the transdiaphragmatic pressure (PDi),establishment of the Pdi-sti-mulating frequency (F) curve,and analysis of the power spectrum on the diaphragmatic elec-tromyogram (EMGdi) at spontaneous breathing.Hypercapnia of the dogs was induced by forcing the animals to inhale a mixture of CO2 and air.The findings were as follows:(1) After acute hypercapnia,PDi rapidly and progressively decreased.The power spectrum of EMGdi was also reduced.The PDi-F curve was shifted to right.These findings suggest that the function of the diaphragm was severely damaged.(2) The decrease of pH value was the important cause of diaphragmatic fatigue.(3) The diaphragm in a static condition could reduce the damage of hypercapnia.(4) Cedi-lanid exerted certain effects on the prevention of diaphragmatic fatigue created by acute hypercapnia.

The study on the functions of the diaphragm has been carried on for 5 years.It consisted of the studies on the relationship of the tension and endurance of inspiratory muscles with pulmonary functions,the changes of diaphragmatic electromyogram in maximal ventilation volume test in normal subjects and patients with chronic obstructive pulmonary disease(COPD),the functional changes of the diaphragm in dogs with RDS and the therapeutic effects of aminophylline.the changes of diaphragmatic electromyogram in dogs with hypoxemia and hy-percapnia,determination of transdiaphragmatic pressure,the relationship of the stimulation frequency on the phrenic nerve with diaphragmtic fatigue,etc.It was found that there existed di-aphragmtic fatigue in dogs with RDS,in dogs with hypoxemia and hypercapnia,and in patients with COPD,and aminophylline,digoxin,salbutomal ?-receptor excitory agents can minimize or prevent diaphragmatic fatigue.
