1.The roles of NOD1 and NOD2 in tumorigenesis and development

Theinnate immunity system of human body has more and more attention for its antibacterial, antiviral, maintainingimmunehemostasis and promoting tissue damage and repair and other physiological functions.As members of NOD-like receptors(NLRs), NOD1 and NOD2 receptors are identified as intracellular pattern recognition receptors(PRRs), can be identified with molecular damage endogenous(damage-associated molecular patterns, DAMPs)and exogenous injury-molecular pathogen associated molecular(pathogen-associated molecular patterns, PAMPs), and initiation of innate and specific immune response, maintain the steady balance of body.Recently, a bunch of evidence have demonstrated that the importance of NOD1 receptor and NOD2 receptor is not limited in field of anti-infection, and the insulin resistance, kidney and liver damage recovery, cardiovascular disease and tumorigenesis are also closely related with these two receptors.So the aim of this article is to interpret the NOD1 and NOD2 general structure and function, and summarize the link between these two PRRs and tumorigenesis and finally make a clue for cancer immunotherapy.
2.Catheter Sacculus Expanding Therapy for Cricopharyngeal Achalasia:A Case Report

Objective To explore the feasibility of catheter sacculus expanding therapy for cricopharyngeal achalasia.Methods A patient cricopharyngeal achalasia after cerebral infarction treated with catheter sacculus expanding was reported.Results After 38 times expanding therapy,the patient could eat by himself without choke and whoosh.Video fluoroscopic(VF)showed that the cricopharyngeal muscle can open normally when the food went through.And there is no any aspiration.Conclusion Catheter sacculus expanding therapy can improve cricopharyngeal achalasia effectively,and can be applied easily,safe and reliable.
3.STUDY ON THE ANTIFATIGUE EFFECT OF TEA CONTAINING ACANTHOPANAX SENTICOSUS

Objective: To study the antifatigue effect of tea containing Acanthopanax senticosus.Methods: Forty eight mice were randomized into 4 groups:three groups of tea containing Acanthopanax senticosus at doses of 50, 75, 100 ml/(kg?d), and the saline group.After 4 w, muscle and liver glycogen, serum LDH activity, BUN and blood LAC were measured.Results: Three tea groups showed increase of muscle and liver glycogen,and serum LDH activity, and decrease of serum BUN and LAC in swimming mice.The 75-100 ml/(kg?d) group was the best.Conclusion: Tea containing Acanthopanax senticosus has antifatigue effect.
4.Expression of miRNA -22 in esophageal and gastric cancer tissues and cell lines

Objective To investigate the expression and clinical value of miRNA -22 in esophageal and gastric cancer tissues and cell lines.Methods Realtime -PCR was performed to detect the expression of miRNA -22 in human esophageal cancer cell lines ECA109,TE -1,TE -8,TE -13 and normal esophageal epithelial cells HEEC,48 cases of esophageal cancer tissues and matched adjacent normal tissues,human gastric cancer cell lines SGC -7901,MKN -45,NCI -N87,AGS,NUGC -3,SUN -1,normal human gastric mucosa cell line GES -1 and normal stomach fibroblastic cell NSFC,88 cases of gastric cancer tissues and matched adjacent normal tissues.Results The expression of miRNA -22 was much less in four esophageal cancer cell lines than that of immortalized normal esophageal epithelial cells HEEC,and the expression of miRNA -22 was much less in six gastric cancer cell lines than that of gastric mucosal epithelial cell line GES -1 and normal stomach fibroblastic cell line NSFC.The esophageal cancer tissues showed aberrant down regulation of miRNA -22 compared with the adjacent non -tumor tissues [(3.51 ±1.05)vs.(11.23 ±2.95),t =18.13,P <0.05].The gastric cancer tissues showed aberrant down regula-tion of miRNA -22 compared with the adjacent non -tumor tissues [(2.15 ±1.23)vs.(9.14 ±2.86),t =22.93, P <0.05].Conclusion The expression of miRNA -22 was much lower in esophageal and gastric cancer tissues and cell lines,which provided basement to nosogenesis,early diagnosis and create drug treatment of the cancers.
5.Mutation analysis of KRT1 gene in a Chinese pedigree with epidermolytic palmoplantar keratoderma

Objective To analyze the mutations in keratin 1 (KRT1), KRT9 and KRT10 genes in a Chinese family with epidermolytic palmoplantar keratoderma (EPPK). Methods Clinical data were collected from a family with EPPK. Genomic DNA was extracted from the peripheral blood of 12 family members, including 6 patients and 6 unaffected members, as well as from 50 unrelated normal human controls. PCR was performed to amplify all the exons and flanking sequences of KRT1, KRT9 and KRT10 genes followed by DNA sequencing.Results A missense mutation C.1436T > C was found in the highly conserved helix termination motif of KRT1 gene of all the patients, resulting in a substitution of isoleucine by threonine at position 479 of the KRT1 protein. No mutation was found in the unaffected members or unrelated controls. Conclusions The missense mutation C.1436T > C in K.RT1 gene is likely to be the main cause of the phenotype of EPPK in this family.This is the first report of a pedigree with KRT1 gene mutation-induced EPPK in China.
6.Steroid sulfatase gene mutation in a Chinese pedigree with X-linked ichthyosis

Objective To detect the steroid sulfatase (STS) gene mutation in a Chinese pedigree with X-linked ichthyosis (XLI). Methods Genomic DNA was extracted from the peripheral blood of 3 affected patients and unaffected members in this family and 50 unrelated healthy volunteers followed by the amplification of the exon 1 and exon 10 of STS gene by PCR. Results Complete deletion of the exon 1 to 10 of STS gene was detected in all the patients in this pedigree with XLI, while no mutation was found in this gene in unaffected members of this family or normal human controls. Conclusion The complete deletion of STS gene is likely to be the main cause of the phenotype of XLI in this family.
7. Clinical practice of prophylactic nipple- sparing mastectomy and immediate reconstruction in Chinese healthy women with BRCA1/BRCA2 germline mutation

Objective: To explore the possibility of prophylactic nipple-sparing mastectomy and immediate reconstruction in healthy Chinese women with BRCA1/BRCA2 germline mutation. Methods: From January 2018 to February 2019, three healthy Chinese women with a BRCA1/ BRCA2 germline pathogenic mutation were treated at the Department of Breast Surgery, Peking University International Hospital (2 cases of BRCA1 mutation and 1 case of BRCA2 mutation); these three women (aged 34-36 years) had a strong family history of breast cancer. Thus, they underwent prophylactic nipple-sparing mastectomy and immediate reconstruction. Results: The operations were successfully completed, and no complications occurred. After a median 18-month follow-up, no breast cancer occurred in the three women; they were satisfied with the reconstructed breast's shape, and their anxiety also significantly reduced. Conclusions: Healthy Chinese women with a BRCA1/ BRCA2 germline mutation and a strong family history of breast cancer can possibly consider prophylactic nipple-sparing mastectomy and immediate reconstruction as options.
8.Three-dimentional cultured adipose mesenchymal stem cells for alveolar cleft bone repair: an in vitro study

Objective:To explore the osteogenic effect of adipose-derived mesenchymal stem cells (ADSCs) in three-dimensional culture conditions in Matrigel hydrogel to provide theoretical basis for tissue engineering bone repair of alveolar process fractures.Methods:This study was performed in the Laboratory of Guangzhou Women and Children Medical Center in June 2019. The fourth generation of ADSCs were used to adjust the cell density to 1×10 5/mL. Two-dimensional common culture media (group A) and two-dimensional osteogenic induction media (group B) were used. The ADSCs were encapsulated in hydrogel with osteogenic induction media (group C), and the proliferation of cells was detected by CCK8 method. The cell mineralization effect was detected by alizarin red staining and alkaline phosphatase activity. The expression levels of osteocalcin (OCN) and osteopontin (OPN) were detected by qRT-PCR and Western blot assay. Results:ADSCs were encapsulated in hydrogel. The results of CCK8 kit showed that ADSCs could stably proliferate under the three-dimensional culture. The alizarin red staining and alkaline phosphatase activity of ADSCs under three-dimensional culture conditions were significantly higher than those of two-dimensional condition. In the detection of Western blotting and RT-PCR, we found that the expression of osteogenic protein and mRNA (OCN and OPN) of ADSCs in three-dimensional culture was also higher than that of two-dimensional culture.Conclusions:The encapsulation of ADSCs in hydrogel does not affect the proliferative potential of stem cells, and 3D cultured stem cells can significantly enhance their osteogenesis in vitro.
9.A study on diagnostic values of combined detection of serum CA 125,HE4,SMRP for ovarian cancer

Objective To evaluate the values of combined detection of carbohydrate antigen 125(CA125) ,human epididymis gene protein 4(HE4) ,soluble mesothelin related peptides(SMRP) in early diagnosis for ovarian cancer and the high-risk factors of ovari-an cancer .Methods 43 patients with malignant ovarian cancer were served as the malignant group ,54 patients with benign ovarian tumors as the benign group ,45 healthy women as the control group .Roche Cobas 6000 Automatic Electrochemiluminescence Immu-noassay Analyzer and enzyme-linked immunosorbent assay (ELISA) were adopted to detect serum CA125 ,HE4 ,SMRP levels . High-risk factors of ovarian cancer was subject to Logistic regression analysis .Results Serum levels of CA125 ,HE4 and SMRP of patients in malignant group were significantly higher than those in the benign group and the control group (P<0 .01) .The sensitivi-ties of CA125 ,HE4 and SMRP detection alone for ovarian cancer diagnosis were 73 .3% ,82 .8% and 70 .8% ,respectively ,while their specificities were 79 .1% ,87 .4% and 83 .3% ,respectively .The sensitivity and specificity of joint detection of CA 125 ,HE4 , SMRP for ovarian cancer diagnosis were 90 .2% and 79 .3% ,respectively .Logistic regression analysis showed that age at menarche younger than 13 years ,menstrual cycle less than 30 d and irregular menstruation were the high-risk factors of ovarian cancer ,with their odds ratio(OR) of 2 .11[95% confidence interval(CI):1 .09-4 .10 ,1 .91(95% CI:1 .21-3 .10) and 1 .57(95% CI:0 .83-2 .94) .Conclusion CA125 detection combined with HE4 or SMRP can markedly improve the diagnostic sensitivity for ovarian cancer .
10.Application of spiral computed tomography to evaluate the autogenous grafted bone in unilateral complete cleft lip and palate patients following early orthodontic tooth movement

Objective The purpose of this study was to evaluate the outcome of autogenous bone grafts in unilateral cleft lip and palate patients following early orthodontic tooth movement,and to determine the volume of new bone formation in the bone grafted region with spiral computed tomography.Methods Computed tomography scans of 12 patients were taken immediately preoperatively and at 6 months postoperatively.The patients underwent bone grafting between 9 and 13 years of age were divided into two groups based on whether postoperative orthodontic tooth movement were initiated or not.Three-dimensional models were created in each period,and the defect of alveolar cleft and volume of the newly formed bone were calculated in each patient.The roots of the moved teeth and their positions to the alveolar bone were also observed.Results The preoperative cleft width and cleft volume were not significantly different between both groups.The volume of the newly formed bone in group A was (0.98±0.23) mm3,significantly higher than that in group B,which was (0.73± 0.15) mm3.The rate of newly formed bone in group A was (72.5 ± 11.9)%,significantly higher than that in group B,which was (53.2±9.7)%.The cleft adjacent teeth could move smoothly into the bone grated area,with no root resorption observed in the computed tomography scans.Conclusions Early orthodontic tooth movement can reduce bone resorption in autogenous bone grafted unilateral cleft lip and palate patients through the observation of spiral computed tomography.It plays an active role in the bone remolding process after bone grafting.
