AIM To investigate the role of serine/threonine protein phosphatases 1 and 2A (PP1/2A) in regulation of cell signal transduction involved in the tolerance of human umbilical vein endothelial cells (HUVEC) to hypoxia. METHODS HUVEC tolerance was established by hypoxic preconditioning. The tolerance of HUVEC was evaluated by the cell survival rate, lactic dehydrogenase (LDH) releasing and total antagonistic-oxidative capability (T-AOC). Subcellular localization of nuclear factor E2-related factor 2 (Nrf2) was determined by immunocytochemistry combined with Western blot. The expression of stress protein of heme oxygenase-1 (HO-1) was measured by Western blot. RESULTS Hypoxia 90 min decreased the survival rate and T-AOC of HUVEC significantly, increased the release of LDH in cultured HUVEC. Compared with the hypoxic group, hypoxic preconditioning (4, 8 and 24 h after hypoxia 10 min) up-regulated the tolerance against hypoxia in HUVEC, the survival rate of HUVEC and T-AOC increased and the release of LDH down-regulated when insulted with hypoxia (90 min) in HUVEC. Hypoxic preconditioning established the translocation of Nrf2 from cytoplasm to nucleus and up-regulated the expression of downstream protein HO-1. Pretreatment with okadaic acid (40 nmol·L-1), a powerful inhibitor of PP1/2A, for 10 min in hypoxic preconditioning HUVEC partly inhibited the translocation of Nrf2 from cytoplasm to nucleus and the expression of HO-1, abolished the tolerance of HUVEC established by hypoxic preconditioning. CONCLUSION PP1/2A at least partly take part in Regulation of translocation of Nrf2 and expression of HO-1, with is associated with the tolerance of HUVEC established by hypoxic preconditioning.

Objective To evaluate the protective effects of hydrotalcite,Marzulene-s,selbex,gefarnate,sucralfate and rebamipide against the gastric mucosa lesions induced by ethanol, aspirin, hydrochloric acid or prednisolone in rats. The changes in intercellular space width of gastric epithelial in rats was observed. Methods Four kinds of models were used to observe the protective effects of six agents against the gastric mucosal lesions.① Ethanol model:a total of 84 male Wistar rats were divided into 7 groups with 12 each. The rats in group 1 to 7 were orally received hydrotalcite,Marzulene-S,gefarnate,sucralfate,rebamipide or normal saline for 3 days, respectively. On day 4,the rats were given 1 ml of absolute ethanol. The length of gastric lesion were measured by ulcer index.② Aspirin model:the rats were received 300 mg/kg of aspirin and 0.1 mol/L hydrochloric acid (0.5 ml/100 g).The following procedures were as ①.③ Hydrochloric acid model:the rats were received 1 ml of 0.7 mol/L hydrochloric acid. The following procedures were as ①. ④ Prednisolone model: all groups were administrated with above 6 agents or normal saline for 5 days.During the 2nd-5th day,the rats were subcu aneously injected with prednisolone (250 mg/kg) daily. Rats were killed on 5th day,and the lesions were mcasured by ulcer index.Gastric mucosal tissue of No.1,5 and ]0 rat in the control group and the hydrotalcite group were picked up to measure the intercellular space width using transmission electron microscopy. Results In four kind of models, the ulcer index were significantly lower in rats treated with mucosal protective agents than that in the controls (P<0.05),expecially in hydrotalcite group (P<0.01).The width of intercellular space in the hydrotalcite group was significantly narrower than that in controls (P<0.05).Conclusions All of the mucosal protective agents can be against the gastric mucosal lesion induced by ethanol, hydrochloric acid, aspirin or prednisolone.Among them, the hydrotalcite is even better. The effect of hydrotalcite is further confirmed by observation of intercellular space width.

Objective To investigate the relationship between gallbladder wall-typing and the main portal vein, right portal vein hemodynamics in patients with liver cirrhosis. Methods The main portal vein, right portal vein hemodynamics and the thickness of gallbladder wall were examined in 69 patients with cirrhosis and 30 normal subjects (control group) with two-dimensional ultrasound, color Doppler ultrasound and high-frequency ultrasound. According to the gallbladder wall-typing, 69 patients with liver cirrhosis were divided into not thickened, simple thickening and bilateral subgroups. The paremeters of hemodynamics and the thickness of gallbladder wall were compared among groups. Results There was statistical difference in thickness of gallbladder wall among all 3 subgroups. Statistical difference of the diameter of main portal vein (D_(pv)) was found among the subgroups (P<0.01), except between not thickened and simple thickening subgroup. Statistical difference of the velocity of the main portal vein (V_(pv)) was found between simple thickening subgroup and control group, as well as between bilateral subgroup and the control group (P<0.01). Statistical differences of quantity of blood flow in the main portal vein (Q_(pv)) were detected between bilateral subgroup and other 2 subgroups and control group (P<0.05). No statistical difference of diameter of right portal vein (D_(rpv)) and quantity of blood flow in the right portal vein (Q_(rpv)) was found among all groups and subgroups. The velocity of the right portal vein (V_(rpv)) of bilateral subgroup was statistically different with that of the control group and the simple thickening subgroup (P<0.05). No statistical difference of Q_(rpv)/Q_(pv) existed between the patients with liver cirrhosis and the normal subjects (P>0.05). There was positive correlation between the gallbladder wall thickness and D_(pv) (r=0.886, P<0.01). Conclusion The gallbladder wall-typing has no correlation with the right portal vein hemodynamics, but is related with the main portal vein hemodynamics. The bilateral and the thickness of gallbladder wall can indirectly reflect the hemodynamic changes of main portal vein. Obstruction to flow of cystic vein is not the main hemodynamic cause for the gallbladder wall changes in patients with liver cirrhosis and portal hypertension cause, indicating that there may be some intrinsic mechanisms of this phenomenon.

Hemagglutinin (HA) contains a head domain with a high degree of variability and a relatively conserved stem region. HA is the major viral antigen on the surface of the influenza virus. To define the biologic activities of chimeric HA bearing different head domains and stem regions or their potential use, a HA chimeric gene containing the head domain of the H7 subtype virus and stem region of the H3 subtype virus was modified and expressed using a baculovirus expression vector. Then, the secreted protein was purified and its biologic activities characterized. Approximately 1.4 mg/mL cH7/3 HA could be obtained, and its molecular weight was ≈ 70 kD. The trimer form of cH7/3 protein had hemagglutination activity and could be recognized by specific antibodies. The method described here can be used for further studies on the screening of HA stem-reactive antibodies or the development of vaccines with conserved epitopes.
