1.Pay attention to the implementation and management of the laboratory medicine research projects

In recent years,with increasing investment on laboratory medicine scientific research in China,the project management,implementation,evaluation system and quality improvement become more and more important for research managers and executives.In this paper,the main problems of project execution and implementation and accordingly proposed some constructive suggestions were analysed,in the aspects of the leading role of principal investigator,periodical discussion,peer guidance,scientific progress management,effective using of project funds and the establishment of appropriate reward and punishment mechanisms.It is hoped that these suggestions can be used to achieve success in laboratory medicine research project.
2.Meta analysis of the level of plasma D-dimer in patients with ovarian cancer

Objective To evaluate the correlation between plasma D dimer levels and ovarian cancer, as well as its pathologic staging.Methods According to the inclusion and exclusion criteria , articles published about the correlation between plasma D dimer levels and human ovarian cancer were searched by means of PubMed , Embase, CNKI and WanFang Data from their inception to December 31th , 2013.In addition , references′list of the included articles and their related citations in Pubmed were also reviewed for additional articles.After quality assessment of all studies , Meta-analysis and ANOVA were performed with RevMan 5.2 and SAS 9.1 for statistic analysis.Results A total of 13 articles were identified, including 679 cases of ovarian cancers , 685 cases of benign controls and 477 cases of healthy controls.Meta-analysis revealed that:(1) Plasma D dimer levels in ovarian cancer groups were higher than that in benign control groups ( Z =8.61, P <0.000 01 ) and healthy control groups ( Z =6.76, P <0.000 01);(2) Plasma D dimer levels in benign control groups were higher than that in healthy controls (Z=3.15, P=0.002); (3) Plasma D dimer levels in advanced ovarian cancer FIGO (Ⅲ-Ⅳ) were higher than that in early cancer FIGO (Ⅰ-Ⅱ) (Z=3.38, P=0.000 9);(4)But there were no difference between relapse groups and remission groups (Z=1.19, P=0.23).ANOVA revealed that:(1) There were significant difference of D dimer levels among ovarian cancer groups or benign groups , healthy control groups; ( 2 ) Significant increasing of D dimer levels was revealed by Pairwise ANOVA from healthy controls, benign controls to ovarian cancer.Conclusions Plasma D dimer levels were high in patients with ovarian cancer , especially in FIGO (Ⅲ-Ⅳ) ovarian cancer.No difference of D dimer levels were investigated between relapse phase and remission phase.
3.Study on the effect of the active phase of Doula delivery ball apparatus combined with correct occipitoposterior

Objective To investigate the effect of Doula delivery ball apparatus combined with correct occipitoposterior.Methods Four hundred primiparas whose dilatation of cervix of 3 cm and position of foetus was occipitopostenor position, were randomly divided into observation group and control group according their wishes, and each group of 200 cases.Doula delivery ball was used for the observation group,while the free body position, the traditional delivery mode was the control group.The vaginal delivery rate, degree of pain, postpartum 2 h bleeding volume and neonatal asphyxia rate were compared between the two groups.Results The degree of pain Ⅲ degree was 4.0% (8/200) ,lower than the control group(50.0% (100/200), x2 =11.55,P<0.01).The vaginal delivery rate in observation group was 84.5% (169/200), higher than that in control group (60.5% (121/200);x2 =28.905,P=0.001).Postpartum 2H bleeding was (156.3±53.2) ml, lower than the control group((268.1±54.6) ml, t =20.70, P<0.01).The Postpartum bleeding rate was 0.5% (1/200), lower than that in control group (4.0% (8/200) ,P =0.018).Conclusion Doula delivery ball is applied to the first active stage of labor can effectively relieve pain, promote the safety of natural childbirth.
4.Serum amyloid A induces the formation of NETs

Objective To explore whether serum amyloid A (SAA) can induce the formation of neutrophil extracellular traps(NETs)in neutrophils in vitro. Methods A stable method for inducing NETs formation in vitro was established, in-cluding isolation of peripheral blood neutrophils, cell culture, and NETs formation and observation. The neutrophils were iso-lated from peripheral blood of healthy volunteers. And cells were cultured in vitro and classified into three groups:negative control (NC) group, SAA group and lipopolysaccharide (LPS) group. Following the distinct stimulation in three groups, NETs formation was observed and its percentage was calculated. The concentration of hinstone (h) 3 in supernatant was detected by ELISA. Results The purification and vitality of isolated neutrophils were both more than 95%. The nuclei of neutrophils lost their shape and spread, NETs formation was found. More NETs formation was found in SAA group than that in NC group (P < 0.05). Moreover, the concentration of h3 in supernatant was significantly higher in SAA group than that in NC group (P<0.05). Conclusion SAA can induce the formation of NETs in vitro.
5.Increased expression of calreticulin promotes angiogenesis involved in the pathogenesis of rheumatoid arthritis

Objective Calreticulin (CRT)is a multifunctional protein of endoplasmic reticulum implicated in the pathogenesis of rheumatoid arthritis(RA). The present study was undertaken to determine whether CRT was involved in an?giogenesis events in RA. Methods Serum CRT levels were measured by enzyme-linked immnuosorbent assay(ELISA)in 106 patients with established RA, 75 osteoarthritis(OA)and 80 healthy controls(HC). CRT levels in synovial fluid were al?so measured in 25 RA and 22 OA patients. The expression of CRT in synovial tissue was examined by immunohistology. In order to investigate the role of CRT on angiogenesis, human umbilical vein endothelial cells(HUVECs)were isolated and cultured for in vitro experiments. The proliferation, migration and tube formation of HUVECs following CRT stimulation were examined in vitro by MTT assay, scratch wound healing assay and tube formation assay. Results Our results showed a sig?nificantly higher concentration of CRT in serum [(6.4±3.1) μg/L] of RA patients compared to that of OA [(3.7±0.9) μg/L, P<0.01] and HC [(3.4±1.0) μg/L, P<0.01];and significantly higher CRT in synovial fluid [(6.9±3.4) μg/L] of RA vs OA [(3.9± 0.7) μg/L, P<0.01]. Increased CRT expression predominantly localized to vascular endothelial cells, inflammatory cells and perivascular areas in both the lining and sublining layers of RA synovial tissue. Furthermore, CRT significantly promoted the proliferation, migration and tube formation of HUVECs, as showed by MTT assay, scratch wound healing assay and tube for?mation assay. Conclusion These findings suggested that CRT may be involved in synovitis and pannus formation events via promoting angiogenesis in RA.
6.The Correlation between Serum Amyloid A and Disease Activity in Patients with Rheumatoid Arthritis

Objective To investigate the correlation between serum amyloid A (SAA) and disease activity (DAS28) in patients with rheumatoid arthritis (RA). Methods Forty-four patients with RA, 35 patients with systemic lupus erythe-matosus (SLE), 18 patients with osteoarthritis (OA) and 30 healthy controls (HC) were enrolled in this study. The levels of SAA were measured by ELISA. Erythrocyte sedimentation rate (ESR) was measured by the Westergren method. The value of serum C reactive protein (CRP) was examined by immunonephelometry assay. The correlation between SAA and DAS 28, ESR and CRP was assessed, respectively. Results The SAA levels were significantly higher in RA group than those of SLE, OA, and HC groups (P<0.05). The serum ESR and CRP levels were both higher in RA group than those of OA and HC groups (P>0.05), but there was no significant difference between RA group and SLE group. There was positive correlation between SAA and DAS28, ESR, and CRP levels (rs=0.790, P<0.001;rs=0.674, P<0.001;rs=0.679, P=0.004), respective-ly. Conclusion SAA may be a new serological marker to assess disease activity in RA.
7.Serum amyloid A-induced angiogenesis via scavenger receptor class B type 1 signaling pathway in rheumatoid arthritis

Objective To investigate the role of scavenger receptor class B type 1 (SR-B1) signaling pathway in serum amyloid A (SAA)-induced angiogenesis in rheumatoid arthritis (RA).Methods The expression and location of SR-B1 in RA and osteoarthritis (OA) synovial tissues were observed by immunohistochemistry.And SR-B1 expression in the resting human umbilical vein endothelial cells (HUVECs) was detected by immunoflourescence.Wound repair assessement and tube formation assessement were employed to evaluate the effect on cell migration and tube formation stimulated by SAA and/or anti-SR-B1 antibody.The t-test and one-way analysis of variance (ANOVA) were used for statistical analysis.Results ① SR-B1 was significantly highly expressed in RA tissue samples (A=6 788±819) when compared to the minimal expression in OA (A =31 849±6 977,t=3.567,P<0.01).Positive staining of SR-B1 was observed in RA synovial vascular endothelial cells and perivascular areas.② Strong staining for SR-B1 was observed in all HUVECs tested.③ Significant wound healing induced by SAA (MI=2.50±0.17) was found compared with the untreated controls (MI=1.00±0.09,q=14.38,P<0.01),and the effects were inhibited in the presence of anti-SR-B1 antibody (MI=1.16±0.14,q=13.02,P<0.01).④ Compared to the untreated group (branch point number:6.6±0.8),there was an enhanced formation of branched and capillary-hke tube structure followed by SAA stimulation (branch point number:19.0±1.1,q=25.04,P<0.01) after culturing for 72 h,whereas,tube formation decreased markedly upon pre-treated with anti-SR-B1 antibody (branch point number:7.6±1.3,vs SAA,q =23.32,P<0.01).Conclusion Our present study suggests that serum amyloid A may induce angiogenesis via SR-B1 signaling pathway in RA.

Objective Artificial atrial septum defect combining pulmonary artery banding to create a model of congenital heart defect with decreased pulmonary blood flow to explore the morphological changes of immature pulmonary vascular. Methods Choose twenty piglets with about one to two-month-old, which are exclusively for experiment used. The piglets were randomly divided into three groups: normal control group (group C, n = 6), Small incisions on the right chest, produced a transient reduction in pulmonary blood; low-medium pulmonary artery stenosis groups ( group T1, n = 7 ) : Did artificial room septostomy creation by self-dilators which were delivered into the surface of the right atrium and controlled Systolic trans pulmonary artery banding pressure (Trans-PABP) at pressure of 20 - 30 mmHg; severe pulmonary artery stenosis groups ( group T2, n = 7): T2 were the same surgical procedures with group T1 ,and controlled Trans-PABP ≥ 30 -50 mmHg. Monitored ultrasound after operation , carried out 64-slice computed tomography scanning after one month, to measure the proximal vessel diameter and TransPABP , after two month surgical exploration on the left chest. When the animals were sacrificed, the heart and lung tissue was cut to measure atrial septal defect, pulmonary artery and the banding diameter. By weihgt elastic fiber and van Cieson staining to observe the morphological pathological changes, three groups took lung tissue with right middle lobe lateral segment about 1.0 cm × 0.8 cm × 0.8cm at the end of surgery and 2-months after operation respectively. Results The models were all successfully in the survival animal of the two test groups. One pig died from tracheal intubation accident in the C group, there was one case died due to bowel obstruction in the T1 group, And there were two cases died result from acute right heart failure and chronic heart failure respectively in T2 group. 64-slice CT angiography showed that BD was significantly lower than the AOD in the two test groups, the proximal pulmonary vascular expansion result from stenosis, distal pulmonary vascular scarce. Histopathology showed that the pulmonary artery inside diameter of T1 and T2 was significantly higher than group C(P <0. 05,P < 0.01), and the NAPSC of two experimental groups were significantly lower than group C 2-month after operation( P <0.01).Conclusion This type of Piglet model is closer to clinical pathological and physiological ,64-slice spiral CT combined with lung histopathology observed for the evaluation of pulmonary vascular hypoplasia is a reliable method. Tunica media of pulmonary arterioles hypoplasia with the number reducing, with pulmonary artery banding increased,the degree of pulmonary arterioles hypoplasia gradually increased.
9.Application of Roy adaptation model in patients with adolescent insanity

Objective To investigate the effect of Roy Adaptation Model(RAM)in patients with adolescent insanity.Methods One hundred patients with adolescent insanity during October 2011 to March 2012 were randomized in equal number into two groups by random digit table:the study group and the control group.The former were intervened with RAM and the latter received routine care and health education.Seven weeks after intervention,Hamilton Depression Rating Scale(HAMD),Hamilton Anxiety Scale(HAMA) and Observation Scale(NOSIE)were used for the assessment. Results After intervention,the scores on HAMD and HAMA in the study group were significantly lower,compared to the control group(P<0.01).The scores on social function,social interest,subjective support and use of social support were all significantly higher than those of the control group(all P<0.01). Conclusions RAM can improve their ability of the patients with adolescent insanity to adapt to the environment.It may improve their mental state and their quality of life.
10.Effect of vitamin D insufficiency on the quality of life in the elderly

Objective To assess the effect of vitamin D insufficiency on the quality of life in the elderly.Methods A total of 205 cases in health examination center of our hospital were randomly divided into elderly group (aged ≥ 60 years) and middle-aged g roup (aged 45-60 years).The SF-36 Form was adopted to investigate status of body and mental health.Serum 25 (OH)D3 levels were measured by enzyme-linked immunosorbent assay (ELISA).Serum 25 (OH) Ds ≤ 49.9 nmol/L was defined as vitamin D insufficiency. ResultsThere were 55 cases (53.9%) with vitamin D insufficiency and 16 cases (15.7%) vitamin D sufficiency in the elderly.The scores of vitamin D insufficiency group in the elderly were lower than of vitamin D threshold and sufficiency groups in fourdimensions,including physical function,physical role,body pain and general health.The scores ofvitamin D insufficiency group in the elderly were also lower than of vitamin D threshold and sufficiency groups in three dimensions,including social functioning,vitality and mental health (F=10.28,9.20,6.60,all P<0.05).Serum 25(OH)D3 played more important role in scores of the quality of life in the elderly than other factors by stepwise multiple linear regression. ConclusionsThe vitamin D insufficient status widely exists in the elderly,and it might influence their quality of life.
