A microcapsule is a spherical, with a diameter that can be controlled in the range of 200 -1500 μm and biocompatible semipermeable membrane, which allows the bidirectional diffusion of nutrients, oxygen, secreted therapeutic product, and waste but prevents the penetration of high molecular weight substances from the microcapsule, such as antibodies and immunocytes. In comparison to monolayer culture and multicellular tumor spheroid model, orthotopic injection of microencapsulated tumor cells has uncomparable advantages in cell proliferation, mimicking the in vivo situation, making orthotopic tumor model and distant organ metastases model. Microencapsulated tumor assay has the potential of being widely used for in vitro anticancer drug screening and evaluation of the effects. This article mainly reviews the advantages of microencapsulated tumor assay and its application.

Objective To observe the best compatibility proportion and action mode of protective effects of combined baicalin and taurine in Alzheimer’s disease (AD) model. Methods Aβ1-42 was injected through lateral ventricles to establish AD rat models. SD rats were randomly divided into sham-operation group, model group, aricept group, combined baicalin and taurine in different proportions group. Administration groups were treated with different medicines, while sham-operation group and model group were treated with the same amount of normal saline for 40 days. The step-through latency and the times of mistakes were detected through step-through test;the escape latent and the times of crossing target quadrant were detected through Morris water maze. Furthermore, the human neuroblastoma SH-SY5Y cells were induced by 5 μmol/L Aβ1-42 for establishing a cellular AD model. The AD cellular model was treated with baicalin and taurine compatibility in different proportions for 48 hours. Then the rate of cell viability was detected by MTT assay, and the median effective concentration (EC50) and combination index were calculated. Results Baicalin and taurine in 2∶1 and 1∶1 proportion groups can significantly improve learning and memory ability in AD rats (P<0.05,P<0.01). The EC50 were 2.110 μmol/L and 0.422 μmol/L, respectively. The combination index was 0.308.Conclusion Baicalin combined with taurine exhibit synergetic protective effects in AD rats induced by Aβ1-42. The optimal compatibility proportion of baicalin and taurine is 2∶1, and EC50 is 0.279 μmol/L.

Background and objective The aim of this study is to determine the effcacy and feasibility of a novel folate receptor (FR)-based circulating tumor cell (CTC) detection method in the diagnosis of lung cancer. CTCs were col-lected from 3 mL of blood based on negative enrichment by immunomagnetic beads and then labeled by a conjugate of a tumor-speciifc ligand folate and an oligonucleotide. Methods Atfer washing off redundant conjugates, the bound conjugates were removed and analyzed by quantitative polymerase chain reaction. Results The CTC levels of 97 patients with lung cancer were signiifcantly higher than that of the controls (18 patients with benign lung diseases;P<0.001). With a threshold of 8.7 Folate units, the method showed a sensitivity of 82.5% and a speciifcity of 72.2% in the diagnosis of lung cancer, especially a sensitivity of 86.8% in stage I disease. Compared with the existing clinical biomarkers such as neuron-speciifc enolase (NSE), carcinoembryonic antigen (CEA), and CYFAR21-1, the method showed the highest diagnostic effciency in lung cancer (area under the curve, 0.859; 95%CI: 0.779-0.939) and stage I lung cancer (area under the curve, 0.912; 95%CI: 0.829-0.994). For future work, the CTC levels of 5 lung cancer patients higher than 8.7 Folate units/3 mL in their postoperation. Conclusion FR-positive CTCs were feasible diagnostic biomarkers in patients with lung cancer, as well as in early-stage tumors.
