1.Analysis about the relationship between thyroid hormone and leptin in Graves' disease of woman

Objective:To investigate the relationship of hyperthyroidism and the level of leptin and the role of thyroid hormone in leptin secretion.Methods:FT3,FT4,TSH,serum leptin and body weight were measured in 30 female Graves' patients and 30 female controls;the percentage of body fat and leptin were calculated,the changes in various index after 6-month treatment with methimazole were observed,the relationship between thyroid hormone and leptin,percentage of leptin analyzed,and multiple regressive analysis was made on these factors.Results:The BMI,percentage of body fat and leptin level were lower than controls,however,percentage of leptin was higher than controls.The leptin level elevated insignificantly but percentage of leptin decreased significantly after 6 months treatment with methimazole.Percentage of leptin showed positive correlation with FT3,FT4 and BMI percentage of body fat.Conclusion:The percentage of leptin is higher in female thyroid hyperfunction patients than controls and thyroid hormone increases the secretion of leptin even on the condition of thyroid hyperfunction.
2.Induction of apoptosis in thyroid cells by iodide excess and its mechanism

Objective:To explore the role of excessive iodide in inducing apoptosis in thyroid cells and its mechanism.Methods:Normal primary thyroid cells were cultured in NaI of different concentrations with or without IL-1? and methimazole.The morphological changes in thyroid cells were observed by light microscopy and apotosis rate was determined by flow cytometry techique.In the meantime Fas,FasL,p53 level were determined by histochemistry.Results:Thyroid cells treated with iodide excess underwent the morphological changes and apoptosis. There was a gradually increased apotosis rate and Fas/FasL protein expression accompanying increase in iodide concentrations.No changes in p53 protein expression was shown. IL-1? enhanced the effect of iodide cytotoxicity,whereas methimazole inhibited the role of iodide inducing apotosis.Conclusion:These data indicate that excess molecular iodide induces apoptosis in thyroid cell through Fas/FasL channel, but independent of p53.
3.Analysis on structural characteristics of poison system in acute poisoning of Guangxi

A total of 6004 cases of acute poisoning with 532 kinds of related poisons from 63hospitals were analyzed.According to the classification of pharmaceutical,pesticide,chemical,animal,plant and other poisons,the numbers of poison categories were 217,148,61,34,36 and 36 kinds accounting for 40.8%,27.8%,11.5%,6.4%,6.8%,6.7% of total number of poisons respectively.According to the case count,they were divided into three groups of low,medium and high morbidity and their poison numbers were 462,59 and 11 kinds accounting for 86.8%,11.1%,2.1% of total number of poisons respectively.According to the types of poisons,they were divided into five groups of poisoning 1,2,3,4,5 years and their poison numbers were 320,91,34,33 and 54 kinds accounting for 60.2%,17.1%,6.4%,6.2%,10.1% of total number of poisons respectively.According to the time of poisoning,they were divided into three groups of 1-year,discontinuous and perennial poisoning and their poison numbers were 320,158 and 54 kinds accounting for 60.2%,29.7%,10.1% of total number of poisons respectively.
4.Preparation and Identification of Anti-lactoferrin Single Chain Fv Antibody

Objective To prepare a specific anti-lactoferrin single chain variable fragment(ScFv).Methods Anti-lactoferrin clones were screened from a 'naive' phage antibody library against the immobilized lactoferrin antigen,then the clones were transformed to the E.coli HB2151 to give soluble expression of antibody fragments.The culture supernatant containing ScFv was purified by immobilized metal affinity chromatography,and then determined with SDS-PAGE and ELISA.Results The results demonstrated that ScFvs were specific;they did not react with transferrin,lysozyme and bovine serum albumin in ELISA.The SDS-PAGE showed that the ScFvs had high purity through affinity chromatography and the molecular weight of them was about 32 kD.Conclusions The successful generation of the ScFvs against lactoferrin provides a basis for further study and clinical applications for dry eye and other ocular diseases.

6.Effects of probucol on the expression of thioredoxin system in the kidney of type 2 diabetic rats

Objective To observe the expression of thioredoxin (Trx) and thioredoxin-interacting protein (Txnip) in the kidney of type 2 diabetic rats induced by streptozotocin and the effect of probucol treatment on thioredoxin system. Methods Thirty male SD rats were divided into control group( C, n = 10), diabetes group ( D, n =10), and probucol treated diabetic group ( P, n = 10). After eight weeks of probucol treatment, the expressions of Trx and Txnip in the kidney of three groups were measured by RT-PCR, Western blot, and immunohistochemistry. Body weight,24 h microalbuminuria( ALB), fasting plasma glucose( FPG), fasting insulin( HNS), blood urea nitrogen (BUN), creatinine (Cr), malondialdehyde ( MDA ), superoxide dismutase ( SOD ), and catalase (CAT) were determined. Results Compared with group C, Trx was markedly decreased in group P (0. 162 ±0. 008 vs 0. 239 ±0. 006, P<0.05 ), while Txnip was significantly increased (0. 159±0.003 vs 0. 091 ±0.016, P<0.05 ). Trx in group P was increased as compared with group D (0. 162 ±0. 008 vs 0. 108 ± 0. 013, P < 0. 05 ), while Txnip was lowered (0. 159±0.003 vs 0. 236±0.009 ,P<0.05 ). FPG, 24 h ALB, BUN, Cr,and MDA levels in group D were markedly increased as compared with group C (P<0. 05), while the activity of SOD, CAT, and FINS levels were decreased apparently (P<0.05). The above markers except for FPG in group P were ameliorated (P<0. 05 ).Conclusions Probucol attenuated oxidative stress by means of partially restoring Trx function and reducing Txnip expression, and thus played a major role in renoprotection of type 2 diabetic nephropathy.
7.Research on relativity between R353Q polymorphism of the coagulation factor Ⅶ gene and cerebral infarction

Objective To evaluate the association between R353Q polymorphism of the coagulation factor Ⅶ gene and cerebral infarction in Chinese Han people. Methods Restriction fragment length polymorphism(PCR-RFLP) methods was adopted to detect FⅦR353Q genotype and R、Q allelomorphie gone frequency of 100 cerebral infarction(CI) patients and 106 healthy people. Results There was FⅦR353Q polymorphism in the CI patients and healthy subjects. RR and RQ genotype could be found in the control group and CI group. RR and RQ genotype dis-position consistented with Hardy-weinberg equilibrium. In CI group,RR genotype was in 91 cases,RQ was in 9 cases and QQ in 0 case(the distribution frequencies were 91.00% ,9.00% and 0), and in the healthy subjects, those are 94 cases, 12 cases and 0 case(88.70% ,11.30% and 0). In the control group and CI group,R allele genotype fre-quencies were 94.33% and 95.50%, and Q were 5.67% and 4.50%. There was not significantly different in the R353Q polymorphism (χ20.3027, P=0.5822);Q allele genotype was not significantly different between the cases of control group and CI patients(χ20.2865, P=0.5925). Conclusion There are the FⅦR353Q polymorphism in Han population,however,the idea that the Q allele is a protective factor in CI is not supported.
8.Placing subcutaneous drain and preseting triclosan-coated polyglactin 910 suture with delayed suturing to prevent typeⅢabdominal surgery incision infection

Objective To investigate the effect of placing subcutaneous drain tube and preseting triclosan-coated polyglactin 910 suture with delayed suturing to incision infection after typeⅢabdominal surgery. Methords Dividing 504 patients with typeⅢincisions undergone abdominal surgery into 3 groups. The number of group A patients with thoroughly incision washing and primary triclosan-coated polyglactin 910 suture after abdominal surgery was 143. The number of group B with closed anterior rectus sheath, opened skin and subcutaneous fat with preseted triclosan-coated polyglactin 910 suture was 190. The number of group C with subcutaneous drain tube after abdominal surgery was 171. We compared the incidence rates of incision infection and the second phase debridement suture rates among the 3 groups. Results For the group A, B, C, the number of incision infection people was 11, 4, 3 and the incision infection rate was 7.69%, 2.11%and 1.75%respectively. The difference of the 3 groups incision infection rate were statistically significant (P<0.05). The incision infection rate of the group B and group C were lower than that of group A and the difference were statistically significant (P<0.016 7). There were no significant differences in second phase debridement suture rates among the 3 groups. Conclusion Preseting triclosan-coated polyglactin 910 suture with delayed incision sutue and placing subcutaneous drain tube can decrease the incision infection rates for type Ⅲpatients after abdominal surgery, but can not decrease the second phase debridement suture rates of the infectious incision.
9.Iodine nutrition and thyroid autoantibodies in pregnancy and their relation to thyroid function

Electrochemiluminescence was used to determine thyroid function and thyroid autoantibodies in 140 pregnant women,who were then divided into normal group (n =117) and subclinical hypothyroidism group (n =23) based on the thyroid function.The urine iodine level in the pregnant women was detected by arsenic cerium catalytic spectrophotometric method.The awareness of past history of thyroid disease among the subjects with thyroid dysfunctions were investigated.The results showed that the prevalences of iodine deficiency were 50% and 57% in the normal group and the subclinical hypothyroidism group,respectively.The state of iodine level was not related to thyroid function.The levels of serum thyroid peroxidase antibody (TPOAb) and thyroglobulin antibody were markedly related to serum TSH(P<0.01),so was the level of serum TPOAb related to serum FT4 (P<0.05).Among the subclinical hypothyroidism women,70% did not undergo thyroid function and thyroid autoantibodies screening before pregnancy,8.7% denied past history of thyroid disease,and 21.7% suffered from hypothyroidism before pregnancy.Therefore,we advocate the screening of urine iodine and thyroid autoantibodies before or during the first trimester of pregnancy,aiming to correct iodine deficiency,avoid supplementing too much iodine,improve the outcome of perinatal stage,and reduce all the negative effects on the offsprings.
10.Clinical analyses of laparoscopic hepatectomy for liver neoplasms : a report of 21 cases

The clinical data were retrospectively analyzed for 21 cases of liver neoplasms undergoing laparoscopic hepatectomy from December 2007 to October 2012.Among 11 cases of borderline hepatocellular carcinoma (HCC) (1.0-9.0 cm),6 of them were of micro hepatocellular carcinoma (MHCC) with a diameter ≤2 cm.There were 10 cases of borderline hepatic benign tumor,including liver hemangioma (n =7),hepatic adenoma (n =1),liver lymphoma (n =1) and liver focal necrosis (n =1).According to Couinaud's liver segmentation method,neoplasm was located on segment Ⅲ (n =13),segment Ⅳ (n =6),segment Ⅴ (n =1) and segment Ⅵ (n =1).Laparoscopic hepatectomy was successful in all patients.There was neither conversion into open approach nor postoperative complications of bile leakage,air embolism or perioperative mortality,etc.The mean operative duration was (120 ± 30) minutes,average hemorrhagic volume (165-±79) ml and normal diet & ambulation at Day 1-2 post-operation.The average postoperative hospitalization stay was (16 ± 10)days and l-year survival rate 100%.The parameters of leucocyte,liver enzymes,albumin and bilirubin returned to normal at Week 1 post-operation.Once a reasonable surgical indication is selected,laparoscopic resection is both safe and effective for peripheral micro hepatocellular carcinoma.
