3.Pathological Observation on 90 Cases of Bladder Transitional Cell Carcinoma and Its Prognosis
Journal of Third Military Medical University 1984;0(01):-
Ninety specimens of pathologically confirmed transitional cell carcinoma of the urinary bladder were studied. The patients consisted of 71 male and 19 female with a male/femai ratio of 3.5:1. Their age was ranged from 32 to 79. 12 specimens were studied with electron microscope. The histopathological and ultrastructural features of transitional cell carcinoma were described. Our fol-lowup data of the cases revealed that the larger a tumor, the lower its differentiation, the deeper its invasion, and the more extensive the involvement of lymphatic and blood vessels, the poorer the prognosis of the tumor. In addition the ultrastructural characteristics of transitional oell carcinoma were briefly discussed.
4.Histopathological and Ultrastructural Changes of Brain Tissues after Experimental Complete Cerebral Ischemia in Rats
Journal of Third Military Medical University 1984;0(01):-
The selective vulnerability of brain tissues after 30 minutes of complete ischemia was observed 5, 15, and 30 minutes after ischemia and 15, 60, and 180 minutes after reperfusion in rats under optical and electron microscopies. It was found that the cerebral cortex especially the third and fifth lamina, the H1 segment of the hippocampus, and the Purkinje cells of the cerebellum were most liable to be damaged after ischemia. After reperfusion, the alterations of membrane system, nuclei and organelles were significant. The membraneous structures which contained a large amount of lipids were more severely damaged. At the same time cerebral edema was found, the severity of which was correlated with the intensity of histopathological and ultrastructural changes of the brain and the degree of blood brain barrier disruption. The findings suggest that oxygen free radicals and lipid peroxides may play an important role in the development of the damages in the brain mentioned above after reperfusion.
5.Research progress in diagnosis of intensive care unit patients with invasive pulmonary aspergillosis
Chinese Journal of Postgraduates of Medicine 2016;(2):186-189
Invasive pulmonary aspergillosis is valued in intensive care unit (ICU) patients, but in ICU patients, the diagnosis standard of the European Organization for Research on the Treatment of Cancer/Mycosis Study Group (EORTC/MSG) could easily lead to misdiagnosis and missed diagnosis. It’s imaging manifestations are diverse: halo sign, nodules or micro nodules, nodular shadow, pulmonary consolidation, wedge-shaped consolidation, voids, air crescent sign, ground glass shadow, and so on, and with the relative characteristic features. The sensitivity and specificity of galactomannan detection in bronchoalveolar lavage fluid (BALF) are high for diagnosis of invasive pulmonary aspergillosis in ICU patients, but the significance of the cultivation of lower respiratory tract specimens is limited.
6.Suppressing effects of MMPs inhibitor GM6001, MMP-2/9 inhibitor Ⅰ ,MMP-2/9 inhibitor Ⅱ on migration of human lens epithelial cells
Chinese Journal of Experimental Ophthalmology 2015;33(9):811-815
Background The primary pathologic mechanism of posterior capsular opacification (PCO) is proliferation, migration and epithelial-mesenchymal transformation of residuary lens epithelial cells (LECs) following cataract surgery.Matrix metalloproteinases (MMPs) play a role during the migration of LECs.Researches showed that GM6001,a broad inhibitor of MMPs,can arrest the migration of LECs,but as specific inhibitors of MMPs,the efficacy and safety of MMP-2/9 inhibitor Ⅰ and Ⅱ on LECs migration remain unclear. Objective This study was to determine and compare the inhibitory efficacy among GM6001,MMP-2/9 inhibitor Ⅰ and Ⅱ on human LECs and search the clinical medication to prevent PCO.Methods Human LECs were cultured and passaged in vitro, and the cells of 3-4 generation were incubated in 6-well plates. Then the cells of 70% confluent monolayer were cultured in DMEM without fetal bovine serum for 12 hours.GM6001,MMP-2/9 inhibitor Ⅰ and Ⅱ at different concentrations (0.25,0.50,1.00,2.00,4.00,8.00,16.00,32.00,64.00,128.00 μmol/L) were added into the culture medium for 24 hours separately, and regularly cultured cells served as the control group.A bare area was made by a 200 μl sterile spear on the cell layer, and the migrated distance and inhibitory rate were calculated. The second or third generation of cells were incubated in 96-well plates at a density of 5×105/ml (200 μl/well).GM6001 (128.00 μmol/L),MMP-2/9 inhibitor Ⅰ (64.00 μmol/L) and Ⅱ (32.00 μmol/L) were added into the culture medium for 24 hours, and the cell viability was assayed by using MTT assay. Results Cultured cells grew well with irregular arrangement and presented the polygon in shape. The migrated distance was gradually reduced as the increase of concentrations of GM6001,MMP-2/9 inhibitor Ⅰ and Ⅱ,showing significant differences among the various concentration groups (GM6001:F=248.647,P<0.05;MMP-2/9 inhibitor Ⅰ:F=357.125,P<0.05;MP2/9 inhibitor Ⅱ:F=396.374,P< 0.05).The cell migrated distance in the control group was set to 1,the relative migrated distances were 0.478 ± 0.091,0.294±0.088 and 0.191 ±0.081 in the GM6001 group,MMP-2/9 inhibitor Ⅰ group and MMP-2/9 inhibitor Ⅱ group at the concentrations of 32.00 μmol/L,respectively,showing a significant difference among the groups (F =116.031,P<0.01),and cell migrated distance was obviously shorter in the MMP-2/9 inhibitor Ⅱ group than that in the GM6001 group or MMP-2/9 inhibitor Ⅰ group (all at P<0.01).The A values were 0.607±0.016,0.567±0.015,0.583±0.010 and 0.595 ±0.0138 in the control group,GM6001 group (128.00 μmol/L),MMP-2/9 inhibitor Ⅰ group (64.00 μmol/L) and MMP-2/9 inhibitor Ⅱ group (32.00 μmol/L),respectively,without significant difference among the groups (F=1.403,P>0.05).Conclusions GM6001,MMP-2/9 inhibitor Ⅰ and Ⅱ reduce the mobility of human LECs effectively but do not affect the viability of the cells in vitro.MMP-2/9 inhibitor Ⅱ appears to be most dominant in inhibiting migration of human LECs.
7.Advance in research on liver cell injury and its initiation in developing new drugs for treating liver diseases
Chinese Pharmacological Bulletin 1986;0(06):-
Liver cell death, including apoptosis and necrosis, is triggered by a number of insults. The apoptotic cascade involving the participation of proteolytic caspases is an important pathway of cell death. Mitochondria play a key role in liver cell death. TNF as an important mediator plays an crucial action in liver cell death. Reactive oxygen metabolites may act directly or through immunological mechanism as initiating factors and modulators in liver cell necrosis and apoptosis. Some intrinsic factors may act on the cascade of liver cell death. The extensive knowledge gained in recent years about the mechanisms of hepatocyte death will lead to new and exciting advances in the prevention and treatment of liver diseases.

Chinese Journal of Interventional Cardiology 1993;0(03):-
Objective To investigate the effect of Tongxinluo on VSMC apoptosis and protein expression of Bcl-2 and Bax in rabbits after angioplasty. Methods Atherosclerotic and PTCA models were set up in 60 rabbits which were randomly divided into 3 groups, the control group the hypertipidemia group and the Tongxinluo group. Apoptosis was determined by TUNEL method and protein expressions of Bcl-2 and Bax were detected by S-P immunohistory chemistry staining. Results Apoptosis of VSMC in the hyperlipidemia group was less than that in the control group, but the level of apoptosis in the Tongxinluo group was even higher compared with the hyperlipidemia group. The level of Bcl-2 expression was found higher in the hyperlipodemia group than in the control group. The expression was mainly seen in endothelium and tunica media of the arteries. The expression of Bcl-2 in the Tongxinluo group is the least among the 3 groups studied. Bax expression, which can mainly be seen in the endothelia, is higher in the Tongxinluo group than that in the hyperlipidemia group. Conclusion Tongxinluo may enhance the apoptosis of VSMC, downregulating the expression of Bcl-2 protein and upregulating the expression of Bax protein.

Objective To understand the status of Toxoplasma gondii infection in women with adverse pregnancy outcomes in Longhua region,Hebei Province. Methods A total of 393 women with adverse pregnancy outcomes were chosen as respon-dents in the Longhua County Maternal and Child Health Care Center between January 2013 and June 2016 and were divided into an acute infection group,a previous infection group, and an active infection group according to the test results. Totally 256 wom-en without adverse pregnancy outcomes were selected as a control group. IgM and IgG antibodies to T. gondii were detected by using ELISA in each group. The risk factors of T. gondii infection were surveyed by questionnaires. Results The T. gondii infec-tion rate of the women with adverse pregnancy outcomes was 27.23%(107/393),which was significantly higher than 8.20%(21/256)in the control group(χ2=35.46,P<0.01). The rates of acute infection, previous infection, active infection in the wom-en with adverse pregnancy outcomes were 6.87%(27/393),18.58%(73/393),and 2.54%(10/393)respectively,which was significantly higher than those[1.17%(3/256),7.03%(18/256),0(0/256)]in the control group(χ2=11.43,17.15,7.90 re-spectively,P<0.01). The ratios of the career in contact with raw meat, feeding pets(dog and cat),tasting raw meat, chopping board regardless of uncooked or cooked food, frequently eating rinsing boiler or barbecue, frequently eating outside of the wom-en with T. gondii infection were significantly higher than those of the women without T. gondii infection(χ2 =12.08,29.23, 8.55,13.41,7.28,6.06 respectively,P<0.01 or 0.05). Conclusions T. gondii infection could lead to serious adverse preg-nancy outcomes. Therefore,to avoid contacting with pets, not eating undercooked food, and strengthening personal health pro-tection are the important keys to avoid T. gondii infection.

Bone is now regarded as an endocrine organ modulating energy metabolism. Osteocalcin, which was a traditional bone remodeling marker, especially in its undercarboxylated form, was believed to be a metabolic active molecule involving in glucose homeostasis. Bone can uptake glucose through glucose transporter 1 expressed on osteoblasts, and this process is crucial for osteoblasts differentiation and bone formation. In addition, the osteoblasts specific insulin resistance may also lead to the dysregulation of whole body glucose metabolism. Clinical investigations generally echo the findings from mice studies. More work, especially prospective clinical studies are needed to prove the clinical utility of osteocalcin and/or other bone turnover parameters as solid predictors of pancreaticβcell function and incident diabetes, as well as the potential use of osteocalcin and/or its undercarboxylated form as an anti-diabetic agent. In the 11th issue of Diabetes in 2016, a review paper entitled asRegulation of glucose handling by the skeleton:Insights from mouse and human studies was published [Diabetes, 2016,65(11):3225-3232]. With the permission of Diabetes and American Diabetes Association, we translated the most important part of this review into Chinese with a new title:The regulatory role of skeleton in maintaining glucose homeostasis.
