2.Mechanism and prevention of bacterial translocation arising from the damage to the barrier of colonic mucosal epithelial cells

The damage to the barrier of colonic mucosal epithelial cells is the fundamental cause of bacterial translocation.The tight junction among epithelial cells forms the most important constituent part of the mechanical barrier of the organism to prevent bacterial translocation from happening in the intestinal canal.The para-colonic-mucosal-epithelial-cell space is closed by protein complexes which are considered to be used for regulating the permeability between cells.ZO-1,Claudin-1 and Occludin are the most representative among the numerous protein complexes.They have become the important index to judge the damage degree of the barrier of colonic mucosal epithelial cells in test.The endotoxin is the product released by bacteria.Anti-endotoxin is one of the methods to protect the colonic mucosal barrier,to prevent and treat bacterial translocation.There are anti-endotoxin drugs.However,there has not been any drug widely recognized clinically up to now.Qingyi Decoction and Chaiqin Chengqi Decoction are the traditional Chinese medicine with the function of protecting the intestinal mucosal barrier and suppressing the transloeation of enteric bacteria.Oxymatrine has many pharmacological effects such as anti-bacterial,anti-inflammatory and immunoragulation.The recent animal experiments showed that oxymatrine has protective effect on the intestinal mucosa during severe infection.However,the mechanism of the drug action has not been completely known and needs to be studied in depth.Our traditional Chinese medicine applies extensively in clinical application of the prevention of the damage to the colonic mucosal epithelial cells during severe infection.
3.Detection of HPV-DNA and SCCAg in the Follow-up of Cervical Squamous Cell Carcinoma Patients

Objective To investigate the infection of human papilloma virus(HPV) and expression of serum squamous cell antigen (SCCAg) in the follow - up of 120 cervical squamous carcinoma patients who had received operation or operation combined with radiotherapy or only radiotherapy. Methods 120 cases of therapical cervical squamous carcinoma patients were detected HPV - DNA by HC - Ⅱ and serum SCCAg using immunohistochemical method. Results The positive rate of HPV - DNA and serum SCCAg was 49.17% and 17.50% , with a significant difference between them( P 0.05 ). Conclusion HPV - DNA test with HC - Ⅱ for follow - up of cervical squamous carcinoma patients was feasible. It was more sensitive than serum SCCAg. But it suggested that high risk type HPV -DNA test combined with serum SCCAg may be the independent prognostic factors.
4.Effect and Mechanism of Cortex Lycii Radicis Extracts on the Proliferation and Apoptosis of Pancreatic Beta Cell

Objective] Observing Cortex Lycii Radicis' effect(Cortex Lycii) on rat insulinoma cells(INS-1) proliferation and apoptosis. To explore the mechanism of Cortex Lycii on Pancreatic Beta Cell Proliferation and apoptosis.[ Methods] After primary culture, cells were randomly divided into blank control group:control,11.1mmol·L-1 glucose,HG,30mmol·L-1 glucose, HG+Cortex Lycii(1g·L-1), HG+Cortex Lycii(2g·L-1), HG+Cortex Lycii(4g·L-1), the survival rate of cells was observed by cell counting kit-8(CCK-8);the apoptosis rate was observed by Annexin V stammg. [Results] ①Compared with HG the better effect of cell proliferation groups of Cortex Lycii(P<0.01), the best is group of Cortex Lycii(2g·L-1)(P<0.05) ②Compared with HG group the higher survival rate is group of Cortex Lycii(P<0.01), the lower apoptosis rate of Cortex Lycii(2g·L-1) compared with Cortex Lycii(1g·L-1). [Conclusion] Cortex Lycii can promote the proliferation of pancreatic beta cell, inhibit the apoptosis to protect the pancreatic beta cell. The optimal concentration is 2g·L-1.
5.Molecular imaging of epidermal growth factor receptor in lung cancer

Epidermal growth factor receptor (EGFR) inhibitors are currently widely used for targeted lung cancer therapy. Target-ed therapy evaluation principally relies on genetic testing or rough clinical estimation. PET receptor imaging is highly sensitive and spe-cific. According to domestic and foreign literature, PET imaging using radiolabeled EGFR-targeting agents can be used to predict and monitor therapy response. This paper presents new evaluating methods for targeted lung cancer therapy.
6.Construction and verification of adenovirus containing the PUMA gene under the control of the hTER promoter

Objective To construt the adenovirus vectors of Ad-hTERT-PUMA-EGFP containing human telomerase reverse transcriptase(hTERT) ,PUMA and enhanced green fluorescent protein (EGFP) .Methods By a series of molecular biology tech-niques ,PUMA gene was inserted into downstream of the hTERT promoter .And then EGFP was inserted into downstream of the PUMA in adenovirus vectors .Finally ,recombinant adenovirus vector ,Ad-hTERT-PUMA-EGFP was assembled into the adenovi-rus .Results Correct adenovirus laoding vectors was verified by viral genome PCR technology .After 12 h of transfection ,HEK293 cells showed green fluorescence in 72 h .Conclusion The correct structure of the adenovirus vectors containing PUMA gene and EGFP gene was identified by restriction enzyme analysis .
7.Multi-slice Spiral CT Perfusion Imaging of Liver Cirrhosis

Objective To investigate the 16-slice spiral CT perfusion imaging features of liver cirrhosis.Methods CT perfusion imaging of liver was performed in 37 participants, including 25 patients with liver cirrhosis and 12 healthy adults as control group.The hepatic blood flow parameters were obtained and compared with Child classifications.Results As comparing to control group,blood flow(BF),blood volume(BV)decreased,and hepatic arterial fraction(HAF) increased gradually,but mean transit time(MTT) changed little with cirrhosis from Child A to C.Conclusion CT perfusion imaging is valuable in evaluating and grading liver cirrhosis early.
8.Analysis of treatment outcome and prognosis of elderly patients with advanced epithelial ovarian cancer

Background and purpose: At present, the treatment of elderly patients with ovarian cancer, espe-cially advanced ovarian cancer, tends to be conservative, and elderly patients often can not receive standard treatment. There are few treatment experiences and research data. This study aimed to investigate the treatment outcome, survival and prognostic factors of elderly patients with advanced epithelial ovarian cancer. Methods: This study retrospectively analyzed the clinical data for women older than 65 years diagnosed with stage Ⅲ and Ⅳ epithelial ovarian cancer between Apr. 2008 to Dec. 2012 in Chongqing Cancer Hospital. Results: Of the 181 women who met study criteria, 8.8% received no treatment, 12.7% underwent surgery alone, 15.5% received chemotherapy alone, 33.2% underwent non-standard treatment, 29.8% underwent standard treatment. Baseline comparison of different treatment methods showed that the proportion of patients with cardiovascular and cerebrovascular diseases was lower in the standard treatment group. After Correcting the effect of cardio-cerebrovascular diseases, the median survival time of standard treatment (48 months) and the median survival time of non-standard treatment (47 months) were significantly longer than that of the other treatment groups (P<0.001). The median survival time of chemotherapy alone (26 months) was significantly longer than that of the surgery alone and untreated group (P<0.001), while the median survival time of surgery alone was similar to untreated group (9 months vs 8 months, P=0.269). Multivariate analysis using Cox model showed that treatment, residual lesion size, cardio-cerebrovascular diseases significantly impacted on survival time. Conclusion: Standard treatment was still the key factor for the best survival of elderly women with advanced ovarian cancer. When this is not offered or possible, chemother-apy alone offers better survival than surgery alone. However, surgery alone does not improve prognosis.
9.Related factors of suicide ideation in military personnel

Objective:To explore the prevalence rate of suicidal ideation and its risk and protective factors in military personnel.Methods:A total of 2071 military personnel were investigated by stratified random cluster sampling in May to August of 2015.The Question 7 of Beck Depression Scale (BDI) was used to test suicide ideation (item 7 score ≥ 1 was dovetailed in suicide ideation group).Family,social demographic and occupational factors were assessed with the self-made general characteristics questionnaire.The data were analyzed with multiple stepwise logistic regression.Results:There were 382 (18.4%) military personnel whose scores of item 7 were ≥1.Family history of suicide (OR =42.97,95% CI =29.14-63.35),surrounding suicide (OR =2.46,95% CI =1.75-3.46),drinking (OR =1.64,95% CI =1.31-2.07) and level of education (OR =1.38,95% CI =1.01-1.90) were risk factors of suicidal ideation.Protective factors were (OR =0.60,95% CI =0.50-0.72) and station (OR =0.15,95% CI =0.03-0.64).Conclusion:It suggests that in suicidal prevention for military,more attention is deserved paying to those military personnel who have family history of suicide,surrounding suicide,drinking habit,and are stationed in remote areas and air force.
10.The association between body fat mass and diabetes mellitus, hypertension and dyslipidemia in an elderly population in China

Objective To identify the threshold of percentage of body fat (BF%) associated with diabetes mellitus,hypertension and dyslipidemia in an elderly Chinese population.Methods An individual was considered as a subject if they exhibited any one of the three cardiovascular disease risk factors:diabetes mellitus,hypertension or dyslipidemia.A total of 1 140 subjects were recruited from the Physical Examination Center of Tianjin Hospital,China (male 511,female 629,aged ≥60 years).Questionnaire interview,physical examination,body composition measurement,blood biochemistry measurement,muscle strength measurement and other measurements were performed for each subject.Receiver operating curves (ROC) were created to determine the optimal percentage of body fat cutoff points,with the presence of diabetes mellitus,hypertension or dyslipidemia as outcome variable.Results The percentage of subjects with at least one of the following conditions (diabetes mellitus,hypertension or dyslipidemia) was 62.8% for men and 73.9% for women.Increased percentage of body fat was associated with an increased occurrence of diabetes mellitus,hypertension or dyslipidemia in the elderly,particularly among women.The ROC curve shows that the threshold of fat percentage in men and women was 28.2% and 36.6%,respectively.Conclusion The risks of diabetes,hypertension and dyslipidemia is closely related to elevated body fat mass in the elderly.The percentage of body fat mass is an important indicator of obesity,threshold of BF % can provide reference for clinical obesity intervention in elderly patients.It also can provide reference for the treatment and prevention of cardiovascular disease.
