1.Mechanism of thiocoraline combination with cisplatin on promoting apoptosis of cervical cancer cell line c4-1 and Hela

Objective:To investigate the apoptosis by thiocoraline in combination with cisplatin on cervical cell line c4-1 and Hela and its mechanism. Methods:Different concentrations of thiocoraline and cisplatin alone or in combination used for cultured c4-1 and Hela cells. The inhibition of cell proliferation was detected by MTT assay,and the apoptosis was detected by Hoechst 33342 and AO-EB staining. The expressions of the protein Bax,Bcl2,p53,p21 were detected by Western blot. The cell cycle of c4-1 and Hela cells were detected by Flow cytometry. Results: The cell proliferation activity of c4-1 and HeLa cells were inhibited obviously by different concentrations of thiocoraline and cisplatin alone or in combination. The inhibition effect was in a dose-dependent manner. Meanwhile, when the concentrations of thiocoraline and cisplatin were 10 nmol/L and 4 μmol/L, respectivity the inhibitory effect was most significant (P<0. 05);Hoechst 33342 and AO-EB staining showed that the cell apoptosis effect was more obvious on thiocoraline in combination with cisplatin. The Western blot showed that the expressions of the protein Bax,p53,p21 were significantly up-regulated, and the Bcl2 was significantly down-regulated. Conclusion:Thiocoraline combination with cisplatin could up-regulate the expression of the protein Bax,p53,p21 and down-regulation the expression of Bcl2 to inhibition the proliferation and promotion apoptosis of cervical cell line c4-1 and Hela.

3.EFFECT OF RETINOIC ACID ON DIFFERENTIATION,MATURATION AND FUNCTION OF INTESTINAL DENDRITIC CELL

Objective To investigate the influence of retinoic acid (RA) in differentiation,maturation and function of intestinal dendritic cells (DC),and probe into the effect of Vitamin A (VA) on intestinal mucosal immunity and pathway. Method Rat intestinal mucosa was cultured in vitro,supplemented with all-trans RA and/or retinoid acid receptor ? (RAR?) antagonist (Ro 41-5253). After cultured for 24h and 48h,the surface markers OX62、OX6 and CD86 on DCs were detected by flow cytometry,to observe the effect of RA on differentiation and maturation of DC. The cytokines and RAR? were measured in mRNA levels by RT-PCR to analyze the influence of RA on mucosal immunity and the pathway. Results RA promoted the maturation of DC in mucosa cultured in vitro,and up-regulated RAR? mRNA levels,the changes were marked at 24h. Moreover,when RA was present in the culture,IL-12 and IFN-? (Th1 cytokine) mRNA were reduced,and IL-10 was increased significantly compared with control . However,the actions of RA above can be inversed by Ro 41-5253. Conclusion The modulation of RA on DC could be one of the important mechanisms that VA influences intestinal mucosal immunity. RAR? participates in the regulation of RA on DC.

5.Longitudinal observations of the prevalence of nutritional risk and undernutrition in patients with head and neck cancer during radiotherapy

Objective To investigate the changes in prevalence of nutritional risk and undemutrition in patients with head and neck cancer during radiotherapy.Methods In this longitudinal observational study,a convenience sampling method was used to recruit patients with head and neck cancer who were receiving radiotherapy in Beijing Cancer Hospital.Nutritional Risk Screening 2002 (NRS 2002) was applied to assess the prevalence of nutritional risk in the patients,and Patient-Generated Subjective Global Assessment (PG-SGA) and body composition test to determine the prevalence of malnutrition (undernutrition) before,during and after radiotherapy.Results 56 patients finished the three follow-up exams.Along with the progress of radiotherapy,the scores of NRS 2002 increased gradually (1.64±1.09 vs.2.30 ±1.06 vs.3.14 ±1.07,x2 =46.639,P<0.001),and the prevalence of nutritional risk also increased gradually (21.43% vs.37.50% vs.71.43%,x2 =29.700,P <0.001);the total scores of PG-SGA [1 (1-13) vs.6 (1-15) vs.12 (1-18),x2 =63.206,P<0.001] and dimensions of weight [0 (0-4) vs.1 (0-4) vs.3 (0-6),x2 =40.798,P<0.001],intake [0 (0-2) vs.1 (0-2) vs.2 (0-4),x2=64.707,P<0.001] and symptoms [0 (0-7) vs.2 (0-10) vs.6 (0-11),x2 =61.562,P < 0.001] all increased gradually with statistical significance.The prevalence of malnutrition in different stage of radiotherapy were significantly different (x2 =64.999,P < 0.001).The body composition analysis in 40 patients showed that all the indicators of body composition decreased significantly along with the progress of radiotherapy.There was a great loss in patients' body weight during radiotherapy,especially the fat-free mass.Conclusions The prevalence of nutritional risk and undernutrition may increase in patients with head and neck cancer during radiotherapy.Lean body mass accounted for most of the weight loss.We should pay more attention to those patients' nutritional status during radiotherapy.

Objective To investigate the roles of theaflavins(TFs) in airway mucus hypersecretion induced by human neutrophil elastase(HNE). Methods Human lung adenocarcinoma cell A549 was stimulated by HNE for mucus hypersecretion,and TF monomers(TF1,TF2 and TF3) were used for intervention.The effects of TF monomers on viability of A549 cells were examined by MTT method.After the effective doses of TFs were determined,A549 cells were divided into 4 groups for experiment.In control group,A549 cells were cultured with serum-free medium.In HNE treatment group,A549 cells were treated with HNE(50 nmol/L) for 24 h.In TF monomer intervention groups,A549 cells were pre-treated with TF1,TF2 or TF3(50,100 or 200 ?g/mL) for 24 h,and were then treated with HNE for another 24 h.In AG1478 intervention group,A549 cells were pre-treated with AG1478(5 ?mol/L),an epidermal growth factor receptor blocker for 30 min,and were then treated with HNE for another 24 h.The changes in mucin(MUC) after treatment by different doses of TF1,TF2 and TF3,and by TF3(200 ?g/mL) for different time(12 h,24 h and 36 h) were detected.The changes in MUC5AC mRNA expression and MUC5AC protein expression were detected by RT-PCR and ELISA,respectively. The hepatocellular carcinoma (HCC) is one of the most common malignant tumors.Patients with hepatobiliary cancer suffer from multiple symptoms,such as physical symptoms and psychology symptoms.In recent years,along with the further research of symptoms,the concept of symptom clusters occurred.Those two issues have always been the hot topics in recent years.Through the efforts of experts at home and abroad,there has been a part of progress in this area.However,the results are not the same.There are lots of disputes in the numbers、sequence、incidences of the symptom clusters.The paper will discuss this issue from symptom experience and symptom clusters in order to provide the reference and basis for symptom management.
9.Retrospective analysis of 91 patients with T cell non-Hodgkin's lymphoma

ObjectiveTo analyse treatments and prognostic factors of T cell non-Hodgkin lymphoma (T-NHL). MethodsNinety-one patients with T-NHL were retrospectively analyzed, and clinical features,histopathology, laboratory data were included in Kaplan-Meier and prognostic analysis. Results Median age was 38 years,58 (63.7 %) had high-intermediate and high risk by IPI,72 (79.1%) presented with advanced stage disease,extranodal disease was present in 64.8 % of patients.The overall response rate (ORR) for the whole group was 63.8 %,and the estimated 3,5-year ORR were 55.5 %,41.3 % respectively.Compared with CHOP-like regimen, the addition of etoposide could improve the survival of patients, meanwhile radiation therapy could improve the outcome of patients with NK/T cell lymphoma and mediastinal bulky disease, and consolidation chemotherapy with HSCT could improve the survival and reduce the recurrence of patients.Clinical stage,B symptoms,ECOG score,the level of LDH,extranodal involvment,anemia,initial treatment outcome, IPI score, the level of serum albumin and fibrinogen were predictive to overall survival. Cox multivariate analysis showed initial treatment outcome and B symptoms were independent prognostic factors.IPI and m-PIT were useful for stratified patients into different prognostic risk groups. Conclusion T-NHL is a heterogeneous group of malignancies with an inferior long term outcome. New treatment modality needs to be explored for these patients,and new drugs and HSCT may be good choices.
10.Relationship between HBx protein and epithelial-mesenchymal transition in hepatocellular carcinoma

Objective To investigate the existence of epithelial-mesenchymal transition(EMT)and its relation to the expression of HBx protein in hepatocellular carcinoma(HCC).Methods The expression of HBx protein,E-cadherin,β-catenin,N-cadherin and fibronectin were detected by immumnohistochemistry technique in 76 cases of HCC.Results Among 76 HCC samples,the loss of expressions of E-cadherin and β-catenin were 34%(26/76)and 20%(15/76),respectively.The positive expressions of HBx protein,N-cadherin and frbronectin were observed in 68%(52/76),55%(42/76),46%(35/76)of HCC samples,respectively.The loss of expression of E-cadherin significantly correlated with the positive expression of N-cadherin and HBx protein (P<0.01).The positive expression of N-cadherin and fibronectin significantly correlated with the loss of expression of β-catenin and the positive expression of HBx protein(P<0.05).Conclusions EMT exists in hepatocellular carcinoma,and the expression of HBx is significantly associated with EMT.
