1.Preparation and Quality Control of ZnO Cream

OBJECTIVE:To prepare ZnO cream and to establish its quality control method. METHODS: The O/W cream base was used to prepare ZnO cream; The ZnO cream was assayed by EDTA-Na2 titration method with C2H5OH-CHCl3 (1∶1) to dissolve cream base. RESULTS: The prepared ZnO cream was well-spread, and its property, identification and tests etc were all in conformity with the description of China Pharmacopeia (2005 edition). The content of the ZnO stood at 16%. CONCLUSION: The preparative procedure of ZnO cream was simple and practicable, its quality control method was reliable and its quality was stable.
2.Reliability and validity of Chinese Version of the Caregiver Preparedness Scale in caregivers of stroke survivors

Objective The research aimed to translate the English version of the Caregiver Preparedness Scale (CPS) into Chinese and to test the reliability and validity of the Chinese version of CPS in caregivers of stroke survivors. Methods Totally 176 stroke survivors and their caregivers were recruited and were investigated by the Chinese version of CPS. Results The internal consistency coefficient of the Chinese version of CPS was 0.925;the test-retest reliability was 0.807 (P<0.01). Factor analysis got one factor, which explained 61.647% of the total variance. The factor loading of each item was>0.4. Conclusions The Chinese version of CPS has been proved to be reliable and valid. It can be used as a valid tool for the measurement of caregiver preparedness in caregivers of stroke survivors.
3.Reliablity and validity of Chinese Version of the Exercise Self-Efficacy Scale in patients after stroke

Objective To translate the English version of the Exercise Self-Efficacy Scale (ESES) into Chinese and test the reliability and validity of the Chinese version of ESES in patients after stroke. Methods Totally 214 patients after stroke were recruited and were investigated by the Chinese version of ESES. Results The internal consistency coefficient of the Chinese version of ESES was 0.879. The split half reliability coefficient was 0.858(P<0.01). The test-retest reliability was 0.657. Item-total correlation coefficient ranged between 0.627 and 0.779 (P<0.01). Factor analysis got one factor, which explained 59.745%of the total variance, and the factor loading of each item was above 0.4. Using General Perceived Self-Efficacy Scale (GSES) as an external criterion, the Pearson correlation coefficient of ESES with GSES was 0.496. Conclusions The Chinese version of ESES has been proved to be reliable and valid. It can be used as a valid tool for the measurement of exercise self-efficacy in patients after stroke.
4.Reliability and validity of Chinese version of the Breastfeeding Self-efficacy Scale Short Form

Objective The research aimed to translate the English version of the Breastfeeding Self-efficacy Scale Short Form (BSES-SF) into Chinese and to test the reliability and validity of the Chinese version of BSES-SF. Methods Totally 308 mothers were recruited and were investigated by the Chinese version of BSES-SF. Results The internal consistency Alpha coefficient of the Chinese version of BSES-SF was 0.927, P<0.01, the half reliability coefficient was 0.853, P<0.01;the test-retest was 0.811, P<0.01. Factor analysis got one factor, which explained 52.498% of the total variance. Conclusions The Chinese version of BSES-SF has been proved to be reliable and valid. It can be used as a valid tool for the measurement of the breastfeeding self-efficacy among Chinese mothers.

6.Analysis of correlation between intestinal microflora and intestinal mucosa immune in the elderly

Objective To investigate the characteristics of intestinal microflora and its effect on fecal secretory immunoglobulin A (sIgA) in the elderly. Methods All 104 cases were assigned to two groups: the control group (healthy adults, age<60 years ,n=36) and the elderly group (healthy elderly persons, age ≥60 years, n=68). Feces samples of all cases were cultured under both aerobic and anaerobic conditions for the 6 bacteria (4 kinds of anaerobes and 2 kinds of aerobes) and the yeasts. The ratio of bifidobaeterium to Enterobacteria (the ratio of B/E) was calculated, which reflected personal colonization resistance. The fecal sIgA was also measured. Results Compared with control group, the number of fecal bifidobacterium and the ratio of B/E were significantly decreased in elderly group[(9.1±1.1)vs. (10.2±0.8)lgCFU/g,P<0. 01 ;(1. 02±0.14) vs. (1.24± 0. 18), P < 0. 01], and the number of enterobacteria and peptostreptococcus was significantly increased in elderly group [(9.2±1.3) vs. (8. 2±0.7) lgCFU/g, P < 0. 05 ; (8. 5 ±1.4) vs. (7.1 ± 1.6)lgCFU/g, P<0. 05]. The fecal slgA was much lower than that in control group [(652.9± 184.3) vs. (793. 5±150. 3)μg/ml, P<0. 05]. Moreover, the quantity change of bifidobacterium was correlated with the concentration of fecal sIgA(r=0. 562, P<0. 01). Conclusions The number of fecal bifidobacterium is significantly decreased, while the number of enterobacteria and peptostreptococcus is significantly increased in elderly persons. The colonization resistance is significantly decreased and the intestinal immuno-barrier function is injuried in elderly persons, which have influenced the intestinal microflora.
7.Effect of Yijing Recipe on apoptosis of testis spermatogenic cells and the expression of Bcl-2 and Bax protein in infertility rats.

OBJECTIVETo observe the effect of Yijing Recipe (YR) on the apoptosis of testis spermatogenic cells and the protein expression of Bcl-2/Bax in rats with adenine induced infertility.
METHODSTotally 75 Wistar rats were randomly divided into 5 groups, i.e., the blank control group, the model group, the high dose YR group, the middle dose YR group, and the low dose YR group, 15 in each group. Except those in the blank control group, rats in the rest groups were intragastrically administered with adenine for 10 successive days. From the 11th day, rats in the blank control group and the model group were fed with equal volume of normal saline. Rats in the YR groups were intragastrically administered with YR at different doses (3.38 g/100 g; 1.69 g/100 g; 0.85 g/100 g), once daily for 20 consecutive days. All rats were killed by the end of the experiment and their testes extracted. The apoptosis of spermatogenic cells and the expression of Bcl-2/Bax proteins were detected by terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling (TUNEL) and SABC method.
RESULTSCompared with the blank control group, the Bcl-2 protein expression decreased, the Bax protein expression increased, and the apoptosis index increased in the model group, showing statistical difference (P <0.01). Compared with the model group, the Bcl-2 protein expression increased in the three YR treated groups (P <0.01, P <0.05). The Bax protein expression level decreased in the high and middle dose YR groups (P <0. 01, P <0. 05). The apoptosis index decreased in the middle dose YR group (P <0.01).
CONCLUSIONYR could inhibit the apoptosis of spermatogenic cells through regulating the expression of Bcl-2 and Bax protein in the testis.

8.Reversed-phase ion pair high performance liquid chromatography in determination of berberine and palmatine in Phellodendron chinense Schneid.and its granules

Objective:To determine berberine and palmatine in Phellodendron chinense Schneid.and its granules.Methods: The reversed-phase ion pair high performance liquid chromatography(RP-HPLC) was used and the validation of the method was tested.The chromatography condition was with Lichrospher C18 column(4.6 mm?250 mm,5 ?m), mobile phase was ACN∶25 mmol/L NaH 2PO 4∶25 mmol/L SDS(2∶1∶1),flow speed was 1.0 ml/min,detection wavelength was 345 nm,and temperature of column was 25℃,Phellodendron chinense Schneid.and its granules were extracted with methanol solution of hydrochloric acid.Results: The theoretical plate number of berberine and palmatine were 14 906 and 14 847,the resolution were 2.33 and 2.86,the tailing factor were 1.09 and 1.06,respectively; all the parameters were suitable for determination.The calibration curves were linear in the range of 40-500 ng,Y=698 278X-3 846,r=1.000(berberine) and 20-250 ng, Y= 536 632X- 7 738, r=0.999 9,r=0.999 4(palmatine).The intra-day and inter-day precision(RSD) at low,middle and high injection amount was all less than 2.5%(berberine) and 1.5%(palmatine).The stability(RSD) was 0.66%(berberine) and 0.70% (palmatine) in 48 h.The recurrence(RSD,n=5) was 0.11%(berberine) and 0.12%(palmatine).The limits of detection was 2.0 ng(berberine) and 1.0 ng(palmatine).The recoveries were 100.4% (RSD=0.12%,n=3) for berberine and 99.80%(RSD=0.22%,n=3) for palmatine.The contents of berberine and palmatine in 3 batch of Phellodendron chinense Schneid.and 5 batch of its granules were determined.Conclusion: Our method can be used for determination of berberine and palmatine in Phellodendron chinense Schneid.and its granules, which is simple and reliable.
9.Investigation of effects of atorvastatin on anti-inflammatory Aβ1-42 induced Alzheimer's disease rat model

Objective To investigate the anti-inflammatory impacts of in the progression of Alzheimer' s disease (AD) in the rat model induced by β-amyloid 1-42 (Aβ1-42 ). Methods Sixty healthy male Wistar rats (weight 250--300 g) were randomly divided into 4 groups: control group, model group, statins control group and statins treatment group, 15 in each group. Rats model were established via intracerebroventricular injection of A13, and then atorvastatin (5 mg·kg-1·d-1) were given to the treatment group for 3 weeks, saline to the control group. Water Maze was used to observe learning and memory ability changes in rats, and expression of inflammatory eytokines IL-1β, IL-6 and TNF-α in the hippocampus were repectively detected by immunohistochemical technique. Furthermore, HE staining patterns, hippoeampns neurons and glial cells in the small ultra structural changes were observed under light microscope and electron microscope respectively. Results The model rats resulted in decreased learning and memory abihties ( the escaping latency: 12. 0 ± 1.2, 41.3 ± 3.4, t = 18. 0363, P < 0. 01 ) and increased secretion of the brain inflammatory factor compared with the controls with statistically significant difference (IL-1β:53.5 ± 2.4, 101.0 ± 3. 8, t = 23. 8246, P < 0. 01 ). Atorvastatin treatment group improved learning and memory performance ( the escaping latency: 25. 7 ± 1.6, 41.3 ± 3.4, t = 9. 1076, P < 0. 01 ), reduced the secretion of inflammatory factors in the hippocampus, compared with the model rats (IL-1β:60.0±3.4,101.0±3.8, t = 18.0231, P <0.01). There were less injured nerve cells and proliferated glial cells in the atorvastatin treatment group than in the model group. Conclusions Atorvastatin plays an anti-inflammatory role in the progression of Alzheimer's disease, reducing the nerve cell damage and improving learning and memory ability.
10.Analysis on content of polysaccharides of Semen Plantaginis

Object To determine and compare the content of polysaccharides in three species of Semen Plantaginis from different places of origin. Methods Spectrophotometry was used. Results The content of polysaccharides in the seed of Plantago major L. was higher than those in the seed of P. asiatica L. and P. depressa Willd. Conclusion The content of polysaccharides in Semen Plantaginis of different species or origins are obviously different.
