1.Activation of Jak_1/STAT_3 signal pathway by TNF-? in mesangial cells is inhibited by lipoxin A_4

AIM: To find whether lipoxin A_4 (LXA_4) inhibits cell proliferation induced by TNF-? in rat mesangial cells, and to explore the molecular mechanisms of signal pathways of LXA_4 actions. METHODS: Cultured rat mesangial cells were growth-arrested and exposed to TNF-? with or without preincubation with LXA_4. Proliferation of mesangial cells was measured by MTT methods. Activities of STAT_3 were analyzed by electrophoretic mobility shift assay. Expression of cyclin E mRNA was assessed by RT-PCR. Cyclin E proteins were determined by Western blotting analysis. RESULTS: TNF-?-induced proliferation and increased mRNA and protein expression of cyclin E in mesangial cells were inhibited by LXA_4 in a dose-dependant manner. TNF-?-stimulation of the STAT_3-binding activities in mesangial cells was down-regulated by lipoxin A_4. CONCLUSION: Inhibitory effect of LXA_4 on TNF-?-induced mesangial cell proliferation is mediated by Jak_1/STAT_3 signal pathway.
2.Calpain 10 mediates lipoxin A_4-induced apoptosis in rat renal interstitial fibroblasts

AIM: To examine whether lipoxin A_4 (LXA_4) induces apoptosis of rat renal interstitial fibroblasts and explore the mechanisms. METHODS: Rat renal interstitial fibroblasts (NRK-49F cells) were incubated in RPMI-1640 medium supplemented with 5% fetal calf serum and exposed to LXA_4 at the concentration of 10 nmol/L, 100 nmol/L or 1 ?mol/L for 24 hours. Prior to experiment, some cells were transfected with calpain 10 antisense oligodeoxynucleotide. Apoptosis of cells was recognized by double staining using fluorescent dye acridine orange and ethidium bromide, observed under laser scanning confocal microscopy and counted by flow cytometry following propidium iodide and annexin staining. Activity of caspase-3 was measured by colorimetric assay. The expression of calpain 10 mRNA was determined by RT-PCR. RESULTS: LXA_4 at the concentration of 100 nmol/L or 1 ?mol/L induced 9.83% or 33.82% apoptosis of cells, respectively. Treatment of cells with LXA_4 up-regulated the expression of calpain 10 mRNA and increased the activity of caspase-3. The transfection of the cells with calpain 10 antisense oligodeoxynucleotide inhibited the LXA_4-induced apoptosis, activity of caspase-3 and expression of calpain 10. CONCLUSION: LXA_4 at high concentration induceds apoptosis in rat renal interstitial fibroblasts via up-regulating of calpain 10 mRNA expression.
3.Protective effect of triptolide on retinal ganglion cells in chronic ocular hypertension model

Background Glaucoma is the second leading cause of blindness,which is characterized by processing retinal ganglion cells (RGCs) loss and optic nerve dystrophy.Clinical study showed that lowing IOP can not arrest the glaucomatous damage of RGCs.To seek a neuroprotective drug is an urgent need.Objective This present study focused on the effect of triptolide,a natural biologically active compound extracted from Tripterygium wilfordii,on RGCs in glaucomatous eyes. Methods Glaucoma animal models were established in the right eyes of 80 clean Wistar rats by combination with aspiration of aqueous humor and phtocoagulation on anterior chamber angle.Wistar rats were assigned to two groups at random.Triptolide (5μg/kg) was intraperitoneally injected daily from three days before photocoagulation through scarification of animals (total 8 weeks),and same amount of physiologic saline solution was used at the same way.IOP was measured with a Tonopen XL tonometer at at 1day,3,5,7 days and weekly for 8-week duration after phtocoagulation.RGCs numbers was calculated by retinal Nissl staining.Morphology of retina in frozen section was examined under the light microscope.The experiment followed the Standard of Association for Research in Vision and Ophthalmology. Results The IOP was elevated in model eyes from 1 day through 3 weeks after operation with statistically different in comparison with before operation(P<0.05).No obvious differences in IOP changg was found hetween the triptolide group and the normal saline group at each time point(P>0.05).The numbers of RGCs of model eyes in normal saline group decreased gradually after operation,but no evident decline of numbers of RGCs in model eyes in triptolide group. RGCs in triptolide group were considerably more than those of normal saline group in various time points after operation ( P<0. 05). However,no obvious difference in RGCs numbers was found between model eyes and control eyes in Triptolide group. Conclusion Triptolide could protect RGCs in glaucomatous eyes,and its effect does not depend on IOP in chronic glaucoma model.

5.Electroencephalogram Non-linear Analysis on Menopause Depression Subjects

ObjectiveTo investigate the changes of non-linear dynamics characteristics of EEG of the patients with menopause depression under eyes closed state.MethodsEEG were recorded in 18 patients with menopause depression and 18 healthy volunteers under eyes closed state. Approximate entropy (ApEn) was calculated for all subjects.ResultsApEn of the menopause depression group at eyes closed state increased significantly compared with that of the healthy control group.ConclusionDynamic non-linear analysis is more appropriate for the study of mental functions changes of menopause depression.
6.Application of EEG Non-linear Analysis in Vision Memory Study

ObjectiveTo investigate the changes of non-linear dynamics characteristics of EEG during picture memory masks, and the application of non-linear dynamic analysis for memory research.MethodsEEGs of 30 healthy old volunteers were recorded under the states of opened eyes and picture memory. Correlation dimension (D2) was calculated for all subjects.ResultsD2 increased significantly during memory tasks compared with the rest state with eyes opened (P<0.05). In different memory tasks, the complexity of mental functions and the regions involved in the memory task were different.ConclusionNon-linear analysis is appropriate for the study of functional changes and working mechanism of brain during memory.
7.Change of lipxin A4, leukotriene B4 and 15-1ipoxygenase in chiMren with acute poststreptococcal glomerulonephritis

ObjectiveTo investigate the change of lipoxin A4 (LXA4), leuotriene B4(LTB4) in blood and urine and leukocyte 15-lipoxygenase (15-LO) of the children with acute poststreptococcal glomendonephritis (APSGN) and to evaluate its significance. MethodsBlood and urinary levels of LXA4 and LTB4 were measured with ELISA within 3 days (acute phase), 10 to 14 days (early resolution phase) and 6 to 8 weeks (late resolution phase) respectively after onset of APSGN in 22 patients. In 8 children with APSGN, expression level of leukocyte 15-LO mRNA was examined with RT-PCR. Leukocyte LTB4 synthesis was assessed with ELISA. Chemotactic effect of LTB4, LXA4 and 15-S-hydroxyeicosatetraenoic acid (15-S-HETE) on neutrophils was determined by in vitro chemotaxis assay. Twenty-two healthy children were served as control. ResultsBlood and urinary levels of LXA4 and leukocyte 15-LO mRNA were up-regnlated in acute phase, further increased in early resolution phase, and decreased in late resolution phase of APSGN, which were stir higher than those in the controls (P<0.01). Blood and urinary levels of LTB4 were increased in acute phase (P<0.01) and then were decreased in early resolution phase and hte resolution phase of APSGN, which were still higher than those in the controls (P<0.01). Administration of 15-S-HETE or LXA4 in vitro inhibited LTB4-induced chemotactic effect on neutrophils of the patients,and inhibited the production of leukocyte LTB4. ConclusionsChanges of blood and urinary levels of LXA4 and LTB4 in early resolution phase of APSGN are contrary. 15-S-HETE and LXA4may play a role in anti-inflammation and resolution of APSGN via inhibiting LTB4.
8.Robots for the rehabilitation of upper limb motor function after stroke

Objective To observe the effect of robot-assisted upper limb training on the recovery of upper limb motor function after stroke.Methods One hundred patients within one month of suffering a stroke were randomly divided into a control group (50 cases) and a therapy group (50 cases).Both groups were given conventional drug therapy and rehabilitation training,while the therapy group also received robot-assisted training once a day,5 days per week for 12 weeks.The Fugl-Meyer assessment for the upper extremities (FMA-UE),the modified Ashworth scale (MAS) and the modified Barthel index (MBI) were used to evaluate the subjects before the experiment and after 4,8 and 12 weeks of treatment.Results During the treatment,the average FMA-UE scores and the average MBI of both groups increased gradually.The treatment group's averages were significantly better than those of the control group at each time point,except that there was no significant difference in elbow and wrist MAS scores.Conclusion Upper limb robots can be used with acute stroke patients in a clinical setting,and they may be beneficial for improving upper limb function and ability in the activities of daily living.

ObjectiveTo explore the regular pattern of TCM beauty prescription.MethodsBased on the ACCESS database platform,we established TCM beauty prescription database and analyzed the information in detailed.ResultsThe total number of beauty prescription in the database was 1821.We focused on analyzing the frequency of medicine application in ancient beauty prescriptions,medicinal diet,acne,chloasma and freckles prescriptions.Statistics showed that dahurian angelica root,indian bread,medlar,honey,black sesame,baical skullcap root,and Chinese angelica were the most commonly used seven kinds of Chinese herbs.ConclusionThe main parts of Chinese medicines for making beauty were those herbs which can nourish yin and blood,supplement yang and qi; and the main parts of Chinese medicine for treating skin diseases were those herbs which can clear heat and detoxicate.
10.Real-time Control and Information Management in Monitoring of Invasive Procedure

OBJECTIVE To use the software for hospital infection control in our hospital to improve monitoring of nosocomial infection in invasive nursing procedure and reduce incidence rate of nosocomial infections.METHODS The software for hospital infection control was used in this prospective investigation to collect common information of invasive nursing procedure,institute intervention measure to invasive nursing procedure,and observe the(occurrence) of nosocomial infection.RESULTS Element administration,process administration,and monitoring(administration) were used to real-time control in invasive nursing procedure,thus fasten information transfer and optimize(performance) flow-sheet of nosocomial infections administration.The software usage could accurately(provide) the(information) of nosocomial infection in-time,and feed-back rapidly.CONCLUSIONS Whole process(control) in invasive nursing procedure can discover and solve problems,thus improve efficiency and effectiveness for preventing and controlling nosocomial infection.
