2.A study on antro-duodenal motility in irritable bowel syndrome

Objective To investigate the variety of antro duodenal motility in patients with diarrhea or constipation predominant irritable bowel syndrome (IBS) and to elucidate the pathophysiology of IBS. Methods After 6 hours fasting, 17 patients with IBS (diagnosed by Rome Ⅱ criteria), 8 with diarrhea predominant diarrhea(D IBS) and 9 with constipation predominant IBS(C IBS), were investigated by gastrointestinal manometry (STD Co., Sweden) for recording at least 2 intergrated interdigestive migrating motor complex (MMC) cycles and observing 2 hours of postprandial antro duodenal motility. Results The MMC periods was shorter, phase Ⅲ duration was longer and amplitude was higher in D IBS group than in controls, whereas the MMC periods was longer and the phase Ⅲ duration was shorter in C IBS group than in controls. The duration of discrete clustered contraction(DCC) in phase Ⅱ was longer in IBS groups than in health controls, but there was no significant difference between the C IBS group and the D IBS group. The motility index in late phase Ⅱ and phase Ⅲ was higher in IBS D group, but for the IBS C group the motility index had no statistical difference from that for controls. The postprandial motility pattern of duodenum had no remarkable difference between IBS patients and controls. Conclusions The gastrointestinal motility disorders are apparent in the IBS patients. There is close relation between gastrointestinal motility disorders and the symptoms of the IBS patients. DCCs waves may almost be independent of constipation or diarrhea of IBS.
3.Effect of self-made Shengji ointment on wound healing after anal carbuncle

Objective To study the clinical efficacy of self-made Shengji ointment on wound healing of postoperative perianal abscess. Methods 80 hospitalized patients with simple low anal abscess after surgery were randomly divided into control group and treatment group according to random digital table method, 40 cases in each group.The self-made Shengji ointment was smeared perianally and covered with gauze in treatment group, and Mayinglong shexiang zhichuang ointments was smeared perianally and covered with gauze in control group.VAS pain score and the granulation tissue scoring criteria were used to evaluate the growth status of the granulation.Results The VAS score in two groups were lower on the 3rd, 7th and 15th day than that on the 1st day (P<0.05);the VAS score on 1st day of two groups had no statisticall significant difference, but the VAS score in the treatment group were significantly lower than that in control group on the 3rd, 7th and 15th day, respectively (P<0.05).Duration of pain in treatment group was significantly lower than that of control group (P<0.05).The granulation tissue score in two groups on the 3rd, 7th and 15th day were lower than that on the 1st day,respectively (P <0.05); the granulation tissue score on the 1st day of two groups had no statistically significant difference, but the granulation tissue score in treatment group on the 3rd, 7th and 15th day were significantly lower than that in control group, respectively(P <0.05). Conclusion Self-made Shengji ointment could significantly relive the pain in wound, and promote the growth of granulation tissue, which has better effect in the treatment of wound healing of postoperative perianal abscess than Mayinglong shexiang zhichuang ointments.
4.Rapid europium-sensitized fluorescent determination of ulifloxacin, the active metabolite of prulifloxacin, in human serum and urine

A new fluorescent method was developed based on the ulifloxacin-europium (Ⅲ)-sodium dodecylbenzene sulfonate system for the determination of ulifloxacin,the active metabolite of prulifloxacin.Sodium dodecylbenzene sulfonate formed a ternary complex with ulifloxacin-europium (Ⅲ) and significantly enhanced the characteristic fluorescence of europium (Ⅲ).The enhanced fluorescence intensity showed a good linear relationship with the concentration of ulifloxacin in the range of 5.0× 10 8 -2.0× 10-6M with a detection limit of 2.0× 10-10 M (3σ).This method is rapid and sensitive,and has been successfully applied to the determination of ulifloxacin in human urine and serum samples.

Method The front-end display tool of OLAP is applied to the setup of the database. Result OLAP platform is established and convenient query, statistics can be realized. Conclusion A great deal of aid decision making information can be got. Thus, the hospital resource utilization and medical quality management are improved.

Objective To establish real-time fluorescence quantitative polymerase chain reaction(RFQ-PCR) for measurement of the expression level of a proliferation-inducing ligand(APRIL)and its receptors in children with non-Hodgkin′s lymphoma(NHL).Met-hods Specific primers and TaqMan probes of APRIL and its receptors had been designed,and fluorescence of the PCR products were detected continuously during amplification.According to the standard curves created by plasmid DNA,the expression level of target genes in clinical samples had been determined using software,and these results were presented as the ratios of target genes′ mRNA to ?2 microg-luobulin(?2M)′s mRNA.Results The detection range of RFQ-PCR was between 101-109 ng/L,the coefficient of variation values for both intra-experimental and inter-experimental reproducibility ranged from 1.68% to 5.97% and 6.40% to 10.58%,respectively.The results from 22 samples showed that the expression level of APRIL,B cell maturation antigen(BCMA) and transmembrane activator and calcium modulator and cyclophilin ligand(TACI) in peripheral blood of NHL were significantly higher than those in normal children(P
8.Protective effect of Chinese Medical Formula Decoction on anoxic damage in cultured hippocampal neuronal cells from newborn rats

ObjectiveTo study the protective effect of Chinese Medical Formula Decoction on anoxic damage in cultured hippocampal neuronal cells from newborn rats. MethodsThe sera with Chinese Medicine (SCM-1,2,3) were collected from rats fed on Chinese Medical Formula Decoction Ⅰ,Ⅱ,Ⅲ(CMFD-Ⅰ,Ⅱ,Ⅲ) for 3 days,while the sera of rats which were fed with normal saline was collected as control. Hippocampal neuronal cells were obtained from dissociated cerebrella of 2 day old Wister rats. The cells were maintained in 5% CO2 humidified atmosphere at 37℃. After 7 days, the cells were grown in culture media containing SCM-1,2,3 and normal saline for 24 hours and in anoxic atmosphere for 1 hour. After anoxia, the cells were cultured for 24 hours. Culture media were collected and Malonaldehyde (MDA) and Lactate Dehydrogenase (LDH) in culture media were detected. ResultsThe survival neuronal cell rate in SCM-1 and 2 groups were significantly higher than control group. LDH and MDA in culture media of SCM-1 and 2 groups were lowed than control group. Conclusions There is a protective effect of CMFD-1 and 2 on anoxic damage in cultured hippocampal neuronal cells from newborn rats.
9.Effect of Shuxuetong injection on microcirculation of patients with septic shock

Objective To analyze the effect of Shuxuetong injection on microcirculation in patients with septic shock and its therapeutic effect. Methods A prospective study was conducted. Eighty patients with septic shock treated in Department of Critical Care Medicine of Hebei Provincial Traditional Chinese Medical Hospital were randomly divided into a Shuxuetong group and a conventional therapy group according to random number table, 40 cases in each group. The conventional treatment in the two groups was energetically carried out in accord to the sepsis shock guidelines, such as positive fluid resuscitation, anti-infection, etc. In the Shuxuetong group, additionally Shuxuetong injection 6 mL in 5% glucose injection 250 mL intravenous drip was given once a day for 7 days. The levels of urine output, lactic acid (Lac), blood urea nitrogen (BUN), creatinine (Cr), aspartate aminotransferase (AST), alanine aminotransferase (ALT), left ventricular ejection fraction (LVEF), and cardiac index (CI) in the two groups were observed before and after treatment; the total dosages of dopamine, dobutamine, noradrenaline, etc. vascoactive agent used for the patients and 28-day mortality in the two groups were also recorded. Results The comparisons of levels of urine output, Lac, BUN, Cr, AST, ALT, LVEF, and CI before treatment between the two groups were of no statistical significant differences (all P>0.05). After treatment in the two groups, the urine output, LVEF, and CI were increased compared with those before treatment, whereas the Lac, BUN, Cr, AST, and ALT were significantly decreased, and the changes were more obvious in Shuxuetong group [urine output (mL/h):112.1±39.8 vs. 73.3±28.5, LVEF:0.49±0.15 vs. 0.44±0.14, CI (mL·s-1·m-2):66.2±5.7 vs. 54.2±6.2, Lac (mmol/L):3.83±1.65 vs. 4.72±2.25, BUN (mmol/L):7.1±2.7 vs. 9.3±3.5, Cr (μmol/L): 73.9±16.2 vs. 95.7±15.8, AST (U/L): 39.8±9.5 vs. 45.8±12.7, ALT (U/L):34.3±9.7 vs. 41.7±11.3, all P<0.05]. The total dosages of all kinds of vascoactive agent of Shuxuetong group were remarkably less than those in the conventional therapy group [dopamine (mg): 993.1±261.7 vs. 1 340.9±356.4, dobutamine (mg):776.2±281.0 vs. 1 049.2±364.3, noradrenaline (mg):56.4±34.6 vs. 107.6±51.3, all P<0.05]. The 28-day mortality of Shuxuetong group was obviously lower than that of conventional therapy group [40.0%(16/40) vs. 60.0%(24/40), P<0.05]. Conclusion Shuxuetong injection can improve the microcirculation perfusion in patients with septic shock and reduce their mortality.
10.Peptide tyrosine-tyrosine combined with its receptors exhibits an anti-cancer potential in pancreatic cancer MiaPaCa-2 cell

Background Pancreatic cancer is a common malignant tumor of the digestive system.It is the fourth major cause of tumor-related death and its morbidity is increasing,and hence it is imperative to develop effective forms of therapy for pancreatic cancer.Peptide tyrosine-tyrosine (PYY) is an important gastrointestinal peptide hormone.According to previous literatures,PYY has been shown to inhibit tumor proliferation in cellular and animal models,but there has been limited research on the detailed mechanism of PYY in pancreatic cancer.This study was to observe the effects of PYY on pancreatic cancer cell and investigate the possible mechanism.Methods The expression of Y1,Y2,and Y5 receptors on pancreatic cancer cell lines were detected by reverse transcription quantitative polymerase chain reaction (RT-qPCR).The cytotoxicity of PYY toward the MiaPaCa-2 cell was evaluated by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay; the cell morphology and structure changes were observed under inverted microscope and transmission electron microscope respectively.Apoptosis and cell cycle were evaluated by flow cytometry.The activity of caspase-3 was determined by activity assay kits and Western blotting.The expression of survivin,vascular endothelial growth factor (VEGF),and cyclooxygenase-2 (COX-2) were determined by RT-qPCR and Western blotting.Results Expression of Y2 receptor is the most abundant PYY receptor on pancreatic cancer cell.PYY inhibited MiaPaCa-2 cell proliferation,blocked it in G0/G1 phase,increased the proportion of apoptosis cells and caspase-3 activity,and reduced the expression of survivin,VEGF,and COX-2.Conclusions PYY weakened the ability of the pancreatic MiaPaCa-2 cell viability through cell cycle blocking and apoptosis inducing.The inhibition effect of PYY may be mediated by the Y2 receptor.The increased caspase-3 activity and reduced expression of survivin,VEGF,and COX-2 may serve as a novel mechanism in PYY inhibition effect on MiaPaCa-2 cell.
