1.An experimental study on changes of tissue oxygen partial pressure in Zusanli (ST36) acupoint and in its related organs in rabbits with acute respiratory distress syndrome

Objective To observe the characteristics of changes in tissue oxygen partial pressure in Zusanli (ST36) acupoint and related organs in acute respiratory distress syndrome (ARDS), and to clarify the correlation between oxygenation of local tissue and systemic oxygenation and the correlation between oxygen metabolism of acupoint tissues and related organ tissues.Methods Twenty healthy New Zealand white rabbits were selected and divided into an ARDS model group and a control group, according to random number table method, 10 in each group. Oleic acid (0.08 - 0.1 mL/kg) intravenous injection was used to replicate the ARDS animal model. Only tracheotomy, mechanical ventilation, insertion of arterial/venous catheter, etc other manipulations were conducted, and no oleic acid was injected in the control group. Tissue oximeter was used to determine the fraction of inspired oxygen (FiO2), when FiO2 was 0.21 and 1.00 respectively, the tissue oxygen partial pressure (PtO2) in Zusanli acupoint, stomach and liver was measured. Meanwhile, the blood gas analyses of arterial blood and mixed venous blood were carried out to calculate the oxygen extraction rate (O2ER).Results When FiO2 was equal to 0.21, the levels of PtO2 in Zusanli acupoint, stomach and liver of the ARDS model group were significantly higher than those of the control group in the same period; the partial pressure of oxygen arterial blood (PaO2), partial pressure of oxygen in mixed venous blood (PvO2), arterial oxygen saturation (SaO2), mixed venous oxygen saturation (SvO2) and O2ER of the ARDS model group were significantly lower than those of the control group over the same period. When FiO2 was equal to 1.00, PtO2, PaO2, SaO2 and O2ER in Zusanli acupoint, stomach and liver were increased compared with those when FiO2 was 0.21 in both groups, and the increase of PtO2 and O2ER in Zusanli acupoint, stomach and liver was more significant in the ARDS model group [PtO2 (kPa): Zusanli acupoint: 16.75±2.12 vs. 13.80±1.83, stomach: 16.45±1.33 vs. 13.35±1.25, liver: 16.43±1.45 vs. 13.45±1.36, O2ER: (36.14±0.97)% vs. (30.81±1.01)%]; the increase of PaO2, SaO2 and SvO2 was more significant in the control group [PaO2 (mmHg, 1 mmHg = 0.133 kPa): 682.02±50.32 vs. 159.32±40.17, SaO2: 1.00±0.00 vs. 0.98±0.01, SvO2: 0.69±0.01 vs. 0.63±0.03, allP < 0.05]. The indexes under FiO2 0.21 compared to those under FiO2 1.00 in the same group, it was shown that when FiO2 1.00, PvO2 was increased in the ARDS model group, but decreased in the control group, the difference between the two groups being statistically significant (mmHg: 36.00±2.83 vs. 42.50±1.70,P < 0.05). Besides, PtO2 in Zusanli acupoint was positively correlated to that in stomach (r = 0.963,P < 0.001).Conclusions When ARDS is under the condition of systemic oxygen delivery disorder, the demand of organ tissues for oxygen is significantly increased, but the oxygen utilization is impaired obviously, which is possibly related to the cellular mitochondrial dysfunction. There is good correlation between the tissue oxygen partial pressure of acupoint tissue and related organ tissue. The detection of oxygen in acupoint tissue has important guidance significance for monitoring the oxygen metabolism of related organ tissues.
2.Correlation research on the MRI quantity of lumbar modic changes and low back pain.

OBJECTIVETo analyzed the relationship between lumbar endplate Modic area changes rate and low back pain by measuring MRI T2 sagittal image of lumbar endplate Modic area changes rate.
METHODSFrom December 2011 to June 2012,70 patients with low back pain in operation were evaluated on pain by VAS and function by JOA,and examined by MRI including 39 males and 31 females with an average age of (51.00 +/- 11.89) years ranging from 29 to 72 years old. Among them, 54 cases had lumbar endplate Modic changes involving 15 cases in types Modic I ,21 cases in type Modic II, 11 cases in type Modic III ,mixed type Modic in 7 cases (eliminated for too few cases). Modic area changes and corresponding vertebral area were measured on MRI T2 median sagittal. The areas of two ways were compared to yield the rate of changes for Modic, for multisegmental Modic changes to calculate the total ratios. A correlation was observed among JOA, VAS and the rate of Modic changes.
RESULTSThe correlation coefficient of change rate of Modic I with JOA score was r = -0.308, P = 0.048 < 0.05, there was a negative correlation;the correlation coefficient of change rate of Modic I with VAS scores was r = 0.428,P = 0.021 < 0.05, there was a positive correlation. The correlation coefficient of change rate of Modic II with JOA score was r = -0.375, P = 0.043 < 0.05, there was a negative correlation;the correlation coefficient of change rate of Modic II with VAS score was r = 0.352, P = 0.041 < 0.05, there was a positive correlation. The area change rate of Modic III had no significant correlation with low back pain degree (P > 0.05).
CONCLUSIONModic I and II area changes rate of of patients with low back pain is closely related to the degree of pain low back pain, Modic III area changes rate is not significant correlated to the degree of lower back pain.

3.Loaded CA4P and rapamycin acid-sensitive liposomes target blood vessels for the treatment of triple-negative breast cancer

Given the vital role of vasculature in solid tumors, the potential of vascular disrupting therapy in the treatment of triple-negative breast cancer (TNBC) is promising. In this study, we prepared the acid-sensitive liposome PPD/CA4P/Lip-Rap loaded with the vascular disrupting agent CA4P and the anti-angiogenic drug rapamycin (Rap) to explore the potential of the vascular disrupting strategy in TNBC. PPD/CA4P/Lip-Rap was characterized by 1H NMR, dynamic light scattering, and transmission electron microscopy. Its drug loading and acid sensitivity were determined. The particle size of PPD/CA4P/Lip-Rap is 161.53 ± 1.89 nm, the zeta potential is -20.03 ± 0.9 mV and it demonstrated good drug release on acidic sensitivity responses. CCK-8 experiments proved that Rap can enhance the ability of CA4P to destroy tumor vascular endothelial cells. Rap can kill marginal residual tumor cells, suppress tumor recurrence. Nanocarriers can further enhance the therapeutic effect. Western blot (WB) showed that Rap decreased the expression of hypoxia-inducible factor-1

5.Gender difference in the smile aesthetic features in aged Han Chinese and its clinical significance

Objective To investigate the gender difference in the smile aesthetic features in aged Han Chinese with normal occlusion and its clinical significance.Methods Fifty-six aged Han Chinese (male:28,female:28,aged 60-66 years)with normal occlusion were included in this study.Smiling images of the resting-position and mandibular postural position were obtained in an anteriorposterior view.The CoSmileMAA1.0 software was used to evaluate the indices associated with the smile-aesthetic features,and its clinical significance was analyzed.Results Significant differences were noted in the nasal ala width,upper lip length,lip clearance,angle oris width in resting-position/ smiling position together with the changes of lip clearance,angle oris width,distance of inferior margin of upper lip to cutting edge of the maxillary incisor teeth,distance of superior border of lower lip to cutting edge of the maxillary incisor teeth,smile line ratio and type of smile (all P<0.05).When the subjects were smiling,the changes of lip clearance was greater in females than in males [(10.7±1.9)mm vs.(11.3±1.6)mm,P<0.05],the changes of angle oris width was greater in males than in females [(14.1±1.6)mm vs.(13.4±1.3)mm,P<0.05],and the smile line ratio was less in males than in females [(0.9±0.2) vs.(1.1±0.5),P<0.05],which indicate that the females had more attractive smile in aged Han Chinese with normal occlusion than males.The type of smile was mainly high smile in elderly females and median smile in elderly males [53.6% (15 cases) and 60.7% (17 cases),x2 =6.43,P<0.05].Conclusions Significant gender difference is noted in the smile-aesthetic features in aged Han Chinese with normal occlusion.Modulation of maxillary incisor teeth length and smile line ratio can contribute to the aesthetic appearance of smiling.
6.Which patients are likely to have a persistent hypersplenism after liver transplantation for the treatment of dysfunctional liver cirrhosis and hypersplenism

Objective To evaluate the recovery of normal splenic function of hyperplenism in patients of liver transplantation. Methods In this study,93 liver transplant patients(all with pretransplant hypersplenism)were divided into group in which the platelet count become normal and the group in which the platelet count failed to recover. Results Hypersplenism disappeared after transplantation in 60 patients(64.5%),hypersplenism after transplantation persisted in 33 patients (35.5%);the portal vein pressure in new liver period,the splenic veinous blood flow after transplantation and the size of spleen were all significantly different between the two groups.The platelet count on three months after operation significantly correlates with pretransplant platelet count,the size of the spleen,the portal vein pressure in new liver phase and the posttransplant size of spleen.Conclusions The persistence of hypersplenism after liver transplantation is common,in may correlate with an unsatisfactory fall in portal pressure after transplantation,characteristics of splenomegaly and poor splenic venous blood flow.
7.Prevalence of hepatitis B virus infection and residual transmission risk among volunteer blood donors in Hangzhou City

Objective :
To investigate the prevalence of hepatitis B virus ( HBV ) infection among volunteer blood donors in Hangzhou City, and to evaluate the residual risk of transfusion-transmitted HBV infections.
Methods :
Data pertaining to volunteer blood donors in Hangzhou City from 2016 to 2019 were retrieved from the blood donor management system. Hepatitis B surface antigen ( HBsAg ) was detected by enzyme-linked immunosorbent assay ( ELISA ) and HBV DNA was detected using nucleic acid testing. The incidence/window period model was employed to assess the residual risk of HBV transmitted through transfusion from donors.
Results :
The prevalence of HBV infections was 0.56% among the 320 755 first-time donors and 0.13% among the 279 816 repeat donors in Hangzhou City from 2016 to 2019, and a higher prevalence of HBV infection was detected among first-time donors than among repeat donors ( P<0.05 ). The residual risks of transfusion-transmitted HBV infection were 296.38 per million person-times ( 95%CI: 277.57 to 315.19 per million person-times ) and 98.79 per million person-times ( 95%CI: 87.15 to 110.43 per million person-times ) among first-time and repeat donors with positive HBsAg, and were 86.79 per million person-times ( 95%CI: 76.60 to 96.98 per million person-times ) and 28.93 per million person-times ( 95%CI: 22.63 to 35.23 per million person-times ) among first-time and repeat donors tested positive for HBV DNA, respectively.
Conclusions
There is still a residual risk of HBV infection transmitted through transfusion from blood donors in Hangzhou City. Nucleic acid testing may remarkably reduce the residual risk of transfusion-transmitted HBV infection in blood donors.

Objective To analyze the role of continuous renal replacement therapy(CRRT) in the prevention of renal failure in perioperative patients undergoing orthotopic liver transplantation. OBJECTIVETo evaluate the effect of lateral supramalleolar artery descending branch antidromic flap for the repair of soft tissue defects in the foot and ankle.
METHODSFrom May 2009 to October 2013,12 patients with soft tissue defects combined with tendon and bone exposure in the foot and ankle were treated by lateral supramalleolar artery descending branch antidromic flap for the repair of soft tissue defects in the foot and ankle, including 9 males and 3 females with an average age of 37.5 years old ranging from 19 to 58 years. Ten cases had the soft tissue defects in the dorsum of foot and 2 in the ankle. The defect area of soft tissue was from 11 cm x 9 cm to 8 cm x 5 cm.
RESULTSTwelve patients were follow-up for 3 to 12 months (averaged 7.3 months). The flaps of 9 cases were survived,the flaps edges of the other 3 cases were necrosis,and healed after dressing change. The flaps were slightly swelling without ulcer occurrence.
CONCLUSIONLateral supramalleolar artery descending branch antidromic flap can repairing the damage by one-stage operation with advantage of dissection easy,rich blood supply without sacrifice of major artery.

10.Effects of long non-coding RNA-HOTAIR on the cell cycle and invasiveness of prostate cancer.

OBJECTIVETo investigate the expression of long non-coding RNA-HOTAIR in prostate cancer cells and its effects on the growth and metastasis of the cells.
METHODSUsing quantitative reverse-transcription PCR (qRT-PCR), we determined the relative expression of HOTAIR in the normal human prostate epithelial cell line RWPE-I and prostate cancer cell lines PC-3 and DU145. We detected the effects of HOTAIR on the cell cycle and invasiveness of prostate cancer cells by RNA interference, flow cytometry, and Transwell mitration assay.
RESULTSThe expressions of HOTAIR in the PC3 and DU145 cells were increased 3.2 and 5.7 times, respectively, as compared with that in the normal RWPE-1 cells. After si-HOTAIR interference, the prostate cancer cells were arrested in the G2 phase and downregulated in the G1 phase. The invasive ability of the prostate cancer cells was evidently inhibited, with the inhibition rates of 32% and 44% of the PC3 cells and 43% and 34% of the DU145 cells for si-HOTAIR1 and si-HOTAIR2, respectively.
CONCLUSIONIncRNA HOTAIR is highly expressed in prostate cancer, which is associated with the growth and invasiveness of prostate cancer cells. HOTAIR is potentially a novel marker for the diagnosis and prognosis of prostate cancer.
