1.Expressions of Toll like receptor 3 (TLR3) and TLR9 in lesions of condyloma acuminatum with different prognosis

Objective To investigate the location of TLR3 and TLR9 protein as well as the expressions of TLR3 and TLR9 protein and mRNA in the lesions of condyloma acuminatum (CA) with different prognosis. Methods Immunocytochemical staining with streptavidin-peroxidase (SP) and reverse transcription (RT)-PCR were conducted to detect the expressions of TLR3 and TLR9 in tissue specimens from lesions of 10 patients with recurrent CA, 14 patients with non-recurrent CA as well as from the foreskin of 10 normal human controls. Results TLR3 and TLR9 were mainly distributed in spinous layer and granular layer in non-recurrent CA lesions, but in basal layer and spinous layer in recurrent CA lesions. A significant elevation was observed in the expression of TLR3 mRNA in non-recurrent CA lesions compared with the control tissue and recurrent CA lesions (P < 0.01, 0.05), and in the expression of TLR9 mRNA in non-recurrent CA lesions compared with the control tissue (P < 0.05). No significant difference was observed between the recurrent and non-recurrent CA lesions in the expression of TLR9 mRNA. Conclusions The changes in the distribution and expressions of TLR3 and TLR9 in epidermis may be associated with the prognosis of CA, and TLR3 may exert a greater impact.

3.The effects of glucosamine and chondroitin sulfate compound tablet on anti-inflammatory and analgesic

Purpose The glucosamine and chondroitin compound tablets were prepared. The anti-inflammatory and analgesic effects of glucosamine and chondrotin compound tablets were investigated. Methods The proportion of glucosamine and chondroitin was 5:4 to prepare compound tablets. The anti-inflammatory effect was investigated with carrageen-induced rats paw edema and cotton granuloma in rats. The analgesic effect was investigated using the pain models of mice which were induced by 0.6% acetic acid. Results Compared with control group the degree of paw edema in the low, middle and high dose group was decreased ( P < 0.05). Fourtreatment groups compared with control group respectively at the weight of granuloma were also markedly reduced ( P < 0.05) and the writhing number of mice induced by acetic acid was decreased ( P < 0.05 ) . Conclusion glucosamine and chondroitin compound tablets have anti-inflammatory effect and analgesic effect on models induced by acetic acid.
5.Diagnosis of CT Spectrum Curve Combined with Morphology in Central Lymph Node Metastasis of Papillary Thyroid Carcinoma

Purpose To investigate the diagnostic value of dual energy CT spectrum curve combined with CT morphology in central lymph node metastasis of papillary thyroid carcinoma (PTC).Materials and Methods Thirty-one PTC patients who accepted dual energy CT double-phase enhanced scan before surgery were analyzed.Central lymph nodes with short diameter > 5 mm were labelled using axial plus 3D positioning.Preoperative labelled lymph nodes were collected and marked during operation.The CT morphology of metastatic lymph nodes was analyzed.The spectrum curve slope (K) difference between metastatic and non-metastatic lymph nodes of arterial-phase and venous-phase primary lesion was compared.The critical K value and the diagnostic efficacy of K value combined with morphology were obtained according to receiver operating characteristic (ROC) curve.Results A total of 73 central lymph nodes were obtained from 31 patients,among which 51 were metastatic and 22 were non-metastatic.There was no significant difference in K value between metastatic and non-metastatic lymph nodes of arterial phase group (P>0.05).While,there was a significant difference in K value between metastatic and non-metastatic lymph nodes of venous phase group (P<0.05).For venous phase,the sensitivity and specificity of the K value in diagnosing central lymph node metastasis were 62.7% and 59.1%,respectively,and combined with morphology,the sensitivity and specificity reached 76.5% and 81.8%,respectively.Conclusion The K value of CT spectrum curve is of certain significance in predicting PTC central lymph node metastasis,and the K value combined with CT morphology can improve the accuracy of diagnosis.
6.Urethral stent implantation in treatment for detrusor-sphincter dyssynergia caused by neurogenic lower urinary tract dysfunction

ObjectiveTo observe the effect of urethral stent implantation on detrusor-sphincter dyssynergia caused by neurogenic lower urinary tract dysfunction.Methods13 patients with detrusor-sphincter dyssynergia caused by neurogenic lower urinary tract dysfunction were treated with the operation of the urethral stent implantation. Voiding function, renal function, residual urine volume and hytronephrosis were examined before and after the operation to evaluate the effect of this procedure.ResultsAfter operation, 7 patients normally emptied their bladders and 6 patients had urinary incontinence. Urethral stents were removed from 2 patients in this group due to the irritation symptoms, the second implantation was performed in a patient due to the voiding difficulty. The renal function of patients after the operation had a non-significant improvement, but the residual urine volume and hytronephrosis improved significantly.Conclusion Urethral stent implantation can decrease residual urine volume and hytronephrosis in patients with detrusor-sphincter dyssynergia caused by neurogenic lower urinary tract dysfunction.
7.Preparation and in vitro and in vivo release of ALA and HMME hydrogel suppository

Objective To prepare 5-aminolevulinic acid (ALA) and hematoporphyrin monomethyl ether (HMME) hydrogel suppositories and to evaluate their photosensitizer transfer efficiencies in rectal tumor tissue.Methods The BALB/c mice implanted SW837 rectal cancer cells subcutaneously were randomly divided into four groups:intrarectal suppository administration group,cutaneous administration group,intratumoral injection group and intravenous injection group.Fluorescence spectrophotometry was used to measure the concentration of protoporphyrin Ⅸ (PpⅨ) and HMME in rectal wall,skin and tumor tissue.The distribution of photosensitizer was determined by a fluorescence spectroscopy system.Results The concentration of PpⅨ in the ALA suppository administration group was 9.76 times (1 h) and 5.80 times (3 h) higher than that in the cutaneous administration group,and the differences were statistically significant (all P<0.05).The maximal penetration depth of ALA in tumor tissue was about 3-6 mm at 2 h after the cutaneous administration.After the HMME suppository administration,the concentration of HMME in the rectal wall was very low.The maximal penetration depth of HMME in tumor tissue was less than 2 mm after the cutaneous administration.Conclusions ALA is more likely to penetrate mucosal barrier compared to skin tissue.The hydrogel suppository based rectal administration is expected to be a new administration method for the rectal cancer photodynamic therapy using ALA.
8.Paradoxical devation of senna adiponectin and its relationship with the level of serum leptin in preeclampsia

Objective To investiagte the serum adiponectin concentration in preeclampsia and its relationship with serum leptin and soluble leptin receptor levels.Method The level of adiponectin,leptin and soluble leptin receptor in serum were measured by enzyme-linked immunosorbent assay(ELISA)in 38 patients with preeclampsia and 42 patients as control.The relationship of free leptin index(leptin/soluble leptin receptor) to preeclampsia was analyzed.Results There were no significant differences in maternal age,gestational age and body mass index(BMI)between two groups.But the gestational age and birth weight were significantly lower in preeclampsia than in control.The patients with preeclampsia had significantly higher levels of serum adiponectin, leptin and free leptin index(1691.7?g/ml,37.5 ng/ml and 0.95 respectively)than the control(689.4?g/ml, 19.3 ag/ml and 0.49,respectively).But there was no significant difference in serum level of soluble leptin receptor between the groups(35.0 ng/ml vs 42.2 ng/ml).Serum adiponectin was not significantly correlated with the level of leptin,soluble leptin receptor and free leptin index.Area of serum adiponectin,leptin and free leptin index in preeclampsia under the ROC curve were less than 0.5.Conclusions The patients with preeclampsia have paradoxical higher serum levels of adiponecfin and more bioavailability of leptin,suggesting these may be important factom of this complication.
9.Role of PI3K-Akt-eNOS signaling pathway in attenuation of myocardial ischemia-reperfusion injury by sevoflurane postconditioning in rats

Objective To investigate the role of PI3-kinase-Akt-endothelial nitric oxide synthase (PI3KAkt-eNOS) signaling pathway in the attenuation of myocardial ischemia-reperfusion (I/R) injury by sevoflurane postconditioning in rats.Methods Fifty healthy male Wistar rats weighing 250-280 g aged 2-3 months were randomly divided into 5 groups ( n =10 each):sham operation group (group S),I/R group,sevoflurane postconditioning group (group Spo),sevoflurane postconditioning + dimethyl sulfoxide (DMSO) group (group Spo + D),and sevoflurane postconditioning + LY294002 (a specific PI3K inhibitor) group (group Spo+ L).I/R was produced by occlusion of anterior descending branch of left coronary artery for 30 min followed by 120 min reperfusion in anesthetized rats.In group Spo,sevoflurane was inhaled for 5 min after the end-tidal concentration reached 2.5%-3.0% at 1 min before reperfusion.In group Spo + L,LY294002 0.3 mg/kg in 0.02% DMSO was injected intravenously at 5 min before reperfusion,and then sevoflurane postconditioning was performed.In group Spo + D,0.02% DMSO equal to the volume of LY294002 was injected intravenously at 5 min before reperfusion,and then sevoflurane postconditioning was performed.Arterial blood samples were taken at 120 min of reperfusion for determination of the levels of creatine kinase isoenzyme MB (CK-MB),lactate dehydrogenase (LDH) and cardiac Troponin Ⅰ (cTnI).The myocardial infarct size (IS) and area at risk (AAR) were measured and IS/AAR ratio was calculated.The rats were sacrificed at 120 min of reperfusion and the myocardial tissues in the area at risk were taken for determination of the expression of Akt,phosphorylated Akt (p-Akt),eNOS and phosphorylated eNOS (peNOS) by Western blot.The ratios of p-Akt/Akt and p-eNOS/eNOS were calculated.Results Compared with group S,the levels of CK-MB,LDH and cTnI,IS/AAR ratio,p-Akt/Akt ratio and p-eNOS/eNOS ratio were significantly increased in the other groups ( P < 0.05 or 0.01 ).Compared with group I/R,no significant change was found in the parameters mentioned above in group Spo+ L (P > 0.05),and the levels of CK-MB,LDH and cTnI and IS/AAR ratio were significantly decreased,and the ratios of p-Akt/Akt and p-eNOS/eNOS were significantly increased in groups Spo and Spo + D ( P < 0.05 or 0.01 ).There was no significant difference in the parameters mentioned above between group Spo and group Spo + D (P > 0.05).Conclusion PI3K-Akt-eNOS signaling pathway mediates the attenuation of myocardial I/R injury by sevoflurane postconditioning in rats.
10.Anti-inflammatory and Analgesic Effects of Combined Application of Glucosamine and Chondroitin Sulfate

Objective To investigate the anti-inflammatory and analgesic effects of combined application of glucosamine(GS)and chondroitin sulfate(CS).Methods The acute and chronic anti-inflammatory effects of combined application of GS and CS were observed respectively through carrageen-induced paw edema and cotton-induced inguinal granuloma of rats.The analgesic effect of combined application of GS and CS was investigated by the mouse pain model induced by 0.6% acetic acid Results As compared with the control group,the degree of paw edema and the weight of granuloma in combined application of GS and CS group were significantly reduced(P<0.05 and P<0.01,respectively);and the writhing number of mice decreased significantly(P<0.05).Conclusion Combined application of glucosamine and chondroitin sulfate demonstrates obviously anti-inflammatory and analgesic effects.
