1.Effect of Effective Monomer of Kangxianling Prescription on Extracellular Matrix Components of 5/6 Nephrectomy Rat

Objective To observe the effect of effective monomer of Kangxianling prescription on renal function and extracellular matrix composition of 5/6 nephrectomy rats model, and provide basis for the screening of effective traditional Chinese medicine of anti-renal fibrosis. Methods Sixty SD rats were randomly divided into normal group, model group, chrysophanol group, salvianolate A group, oleanolic acid group and losartan group. The kidney fibrosis model was made by operation. Two months after intervented by correspong drugs, renal pathological changes of all groups were observed, the levels of renal function indexes were detected, and real-time PCR assay was used to detect laminin (LN), fibronectin (FN), type Ⅲ collagen (C-Ⅲ), type Ⅰ collagen (C-Ⅰ) mRNA expression in rat kidney tissue. Results SCr and BUN in model group increased significantly compared with the normal group (P<0.01). SCr and BUN in salvianolate A group decreased significantly compared with the model group (P<0.05, P<0.01). BUN in salvianolate A group decreased significantly compared with the losartan group (P<0.01). The expression of C- ,Ⅲ C-Ⅰand LN were reduced by effective monomer of Kangxianling prescription. Conclusion Effective monomer of Kangqianling prescription can inhibit renal fibrosis through reducing the expression of extracellular matrix components, thereby improve the renal function.
2.Expression and clinical pathological implications of carbonic anhydrase 9 and P glycoprotein in laryngeal squamous cell carcinoma.

OBJECTIVE:
To identify the difference of CA IX and P-gp expression level between laryngeal squamous cell carcinoma (LSCC) and benign tissues, evaluate the relationship of these two proteins in LSCC, and their correlation with clinical and pathological features.
METHOD:
Immunohistochemical detection of CA IX and P-gp were performed in 47 cases of LSCC and 20 cases of vocal cord polyps.
RESULT:
Overexpression of CA IX and P-gp both in LSCC and in vocal cord polyp (P < 0.05) were confirmed, with a correlation between the two proteins in LSCC (r = 0.324, P < 0.05). The expression of CA IX was related to clinical staging and lymph node metastasis in LSCC (P < 0.05). While P-gp was related to clinical staging and histological grading in LSCC (P < 0.05).
CONCLUSION
The overexpression of CA IX and P-gp may play a role in LSCC progression.

Objective To observe the effect of stress on the rapid component of delayed rectifier potassium current (IKr) in rat cardiomyocytes. Methods Forty male SD rats were randomly divided into four groups (10 each): control group (Ctrl), exhaustive group (ES), noise group (WN) and composite group (ES+WN). Stress animal models were prepared as follows: Rats in ES group were undergoing exhaustive swimming as the stress factor, in WN group undergoing white noise and in ES+WN group undergoing exhaustive swimming + white noise as the stress factor. Langendorff device was used to reversely perfuse collagenase for isolating the rat's ventricular myocytes. The effect of stress on IKr current and gating mechanism of single ventricular myocyte was recorded by whole-cell patch clamp technique. Results Compared with the Ctrl group, the tail current density of IKr (IKr,tail) of ventricular myocytes increased significantly in ES group and WN group (P<0.01). The IKr,tail current density of the ventricular myocytes in ES+WN group was significantly higher than that in ES group and WN group (P<0.01), and the effect was voltage dependent. Gating mechanism revealed that the half inactivation voltage of IKr,tail (V1/2,inact) can be shifted to the right in ES group, WN group and ES+WN group when compared with the Ctrl group, and the recovery time constant shortened after inactivation (P<0.01). However, the steady-state activation, fast inactivation constant and voltage dependence of IKr,tail were not statistically significant in ES group, WN group and ES+WN group when compared with the Ctrl group. Conclusion Stress increases the IKr current in rat cardiomyocytes, suggesting it be one of the electrophysiological bases of stress-induced arrhythmia.
6.MEDCHART application software in management of the smoking cessation

Objective To investigate the effectiveness of Varenicline combined with MEDCHART application software for health education and follow-up in patients at smoking cessation clinic.Methods A total of 104 cases who had smoking cessation counseling and intervention at smoking cessation clinic from February 2014 to January 2015 were divided randomly into MEDCHART APP group (n=51) and conventional group (n=53).Both groups were intervened by brief admonishment and given Varenicline.The 9-12 weeks and 26th week protracted continuous abstinence rates were observed.Results There were no significant difference in 9-12 week protracted withdrawal rate between 26th week APP and conventional groups (56.9% vs.45.3%),the 26th week abstinence rates in group MEDCHART APP (41.2%) were significantly higher than the conventional group (20.8%)(x2=5.088,P<0.05).There was significant difference in full course completion rates between the two groups;15 cases (29.4%) completed the course in group MEDCHART APP,whereas 5 cases (9.4%) did so in conventional group (x2=4.405,P<0.05).Conclusion MEDCHART APP can strengthen health education and follow-up,which helps extend the time for Varenicline,increase 26th week smoking cessation withdrawal rate.It is a useful tool for smoking cessation education and follow-up.
7.Correlation between serum vascular endothelial cells cadherin level and atherosclerosis in diabetic patients

To investigate the relationship among serum vascular endothelial cells(VE) -cadherin, advanced glycation end-products( AGE), and atherosclerotic lesion. 20 healthy subjects and 60 patients with diabetes mellitus,including 30 patients with carotid atherosclerosis (CI), were enrolled.Soluble VE-cadherin and AGE were determined using the enzyme-linked immunosorbent method (ELISA). The relationships among the concentration of soluble VE-cadherin, AGE, and the course of the disease, blood glucose, and blood lipid levels were analyzed with multivariant stepwise regression analysis. The levels of serum VE-cadherin and AGE in the patients with diabetes and CI were higher than those in control group( P<0. 05 ). There was a significant difference in VE-cadherin between the diabetes group and the CI group( P<0. 05 ). Serum VE-cadherin levels were positively correlated with serum AGE levels(r = 0. 69, P<0. 01 ). AGE levels were positively correlated with the diabetes duration ( r = 0. 31, P =0. 02 ). The levels of serum VE-cadherin in diabetic patients are positively correlated with their serum AGE levels. The VE-cadherin seems to play an important role in the development of atherosclerosis caused by AGE.
8.Diagnostic significance of ulnar/median compound muscle action potentials amplitude ratio in motor neuron disorders

Objective To investigate prospectively the diagnostic significance of ulnar/median nerve amplitude ratio in motor neuron disorders.Methods Patients referral to our department between May 2009 and February 2012,due to muscle weakness,inflexible,cramps and atrophy,were consecutively enrolled.Conventional nerve conduction studies of 4 extremities (using surface electrodes) and needle electromyography were performed in all patients with fixed examiner.The compound muscle action potentials (CMAP) were recorded from abductor digiti minimi (ADM) and abductor pollicis brevis (APB) with stimulation of ulnar and median nerve at wrist,respectively.Moreover,the ratio of CMAP amplitude between ADM and APB (ADM/APB) was calculated in the patients who met the diagnostic criteria for definite amyotrophic lateral sclerosis (ALS) and Hirayama disease (HD).The patients with Guillain-Barré syndrome (GBS) and other popyneuropathies (PN) were served as case-controls,and 34 healthy volunteers (aged (45.7 ± 16.3) years) as normal-controls.Results (1) There were 78 cases with ALS,25 HD,51 GBS and 140 other PN,with the mean age(years) of 54.7 ± 11.6,17.6 ± 2.2,41.3 ± 18.4 and 57.1 ± 14.3,respectively.(2) ADM/APB in the ALS subgroup was 2.28 ±2.87 (0.12-22.38),HD0.66±0.36 (0.05-1.34),GBS 1.42 ± 1.33 (0.25-9.85),other PN 1.36 ± 1.48 (0.08-14.44) and normal-controls 1.07 ± 0.28 (0.61-1.64,F =6.872,P =0.000),respectively.(3) The areas under receiver operator characteristic curve in patients with ALS was 0.830 (s-x =0.039) and HD 0.691 (sx =0.039,P =0.000) ; the diagnostic sensitivity and specificity for ALS patients were 36.7% and 93.3%,respectively,with cutoff value of ADM/APB =2; and the diagnostic sensitivity and specificity for HD patients were 53.6% and 89.0%,respectively,with cutoff value of ADM/APB =0.7.Conclusions The ulnar/median CMAP amplitude ratio increases in ALS,but decreases in HD,which may be served as a relatively specific electrophysiological index.ADM/APB amplitude ratio > 2.0 is suggested to be a diagnostic parameter for ALS and < 0.7 for HD.
9.Biocompatibility of osteoblast and demineralized dental matrix

BACKGROUND: Bone-induced protein and its carrier are widely used at present; however, the source is limited, and the preparation is complex. Demineralized dental matrix (DDM) is a natural compound containing many osteoinductional proteins and carriers, thus DDM is an ideal material as the substitute of allogenic bone transplantation.OBJECTIVE: By co-culture of MC-3T3 osteoblast and DDM, to evaluate the biocompatibility of DDM via measuring proliferation and alkaline phosphatase (ALP) activity of osteoblast.DESIGN, TIME AND SETTING: A randomized controlled experiment was performed in Stomatology Hospital of Lanzhou University and Stomatology Hospital of Liwan from November 2007 to May 2009.MATERIALS: DDM was supported by Shenzhen Chuangbo Biological Products Development Co., Ltd.; hydroxyapatite (HAP) was supported by Nanjing Emperor Nano Material Co., Ltd.METHODS: 0.1 g HAP and DDM were added in to a 24-well plate, three wells per samples, and the MC-3T3 osteoblasts were seeded onto the surface of samples. After culturing for 2, 4, and 6 days, the cell proliferation percentage was calculated according to MTT assay. ALP activity was evaluated by the quantitative ALP assay.MAIN OUTCOME MEASURES: The effect of DDM on the proliferation and ALP activity of osteoblasts.RESULTS: The proliferation of osteoblasts in DDM group was obviously higher than that in HAP group. With culture time increasing, the ALP activity of osteoblasts in two groups was all augmented, and DDM group was higher than HAP group. There was significant difference between the two groups (P < 0.05).CONCLUSION: DDM can promote adhesion and proliferation of osteoblasts, and promote osteoblastic growth, displaying a great biocompatibility.
10.The analysis of pharmacotherapy for benign prostatic hyperplastic in outpatients in geriatrics department of three hospitals in Changsha

Objective To understand the elementary information and pharmacotherapy for benign prostatic hyperplasia (BPH) in outpatients in geriatrics department of three hospitals. Methods The outpatients diagnosed as BPH were investigated by daily BPH diagnosis report form, BPH medical-care requirement questionnaire, international prostate symptom score (IPSS) questionnaire and BPH quality of life scale (BPHQLS) in odd months. Results There were 657 outpatients in the three hospitals, including 456 males and 201 females. 289 patients were diagnosed as BPH, accounting for 44% of all outpatients and 63.4% of male patients. The average age of BPH patients was (77.2±5.7)years, the mean volume of prostate was (41.44 ± 21.00)ml, the median value of prostate specific antigen (PSA) was 2.24 μg/L, the mean maximum flow rate was (12. 65± 5.74)ml/s, and the average of IPSS and BPHQLS were 14.8±8. 11, 2. 56±1.36, respectively. The percentage of pharmacotherapy was 66.21% (96/145), and the rates of a-receptor blocker monotherapy and 5α-reduetase inhibitor monotherapy were 6.90% and 8. 97%, respectively. The percentage of drug combination was 26.90%. Conclusions BPH outpatients in geriatrics department of the three hospitals are at high risk, and most of them receive drug therapy. The drug choice is reasonable.
