1.Construction and transfection of neural stem cells of recombinant plasmid pLEGFP-mIFN?

Objective To construct a recombinant plasmid pLEGFP-mIFN? for developing the genetical therapy of tumor by stem cells.Methods PCR was used to amplify the mIFN? gene from plasmid pcDNA3.1-mIFN? in which the mIFN? was cloned.A new plasmid,pLEGFP-mIFN?,was then constructed by inserting the amplified mIFN? gene into pLEGFP-C1.Restriction analysis and sequencing were used to confirm the structure of pLEGFP-mIFN?.Neural stem cells were transfected by supernatant of PA317.Results A DNA fragment in size of 500 bp was amplified.Restriction analysis showed that the amplified gene was inserted into pLEGFP-C1 correctly.(Conclusion The) plasmid pLEGFP-mIFN? is constructed successfully,and it can transfect neural stem cells.

5.The study on effect of long-termed administration of mixed rare earth Changle on rat liver

Objective: To research effect of different doses of Changle on the structure and function of rat liver.Methods: 180 healthy Wistar rats were divided into 5 experimental groups according following doses: 0.1,0.2,2.0,10.0 and 20.0 mg*kg-1,respectively, and the control group given physiological saline for six months. The changes of liver structure were examined by means of normal histological chemistry and transmission electron microscope(TEM). Results: The body weight of animal was linearly increased with the decrease of administered doses, gradual reduction of glycogen in hepatocytes and infiltration of inflammatory cells in the portal area were found in the group of 20.0 mg*kg-1. Changes of ultrastructure showed there were dense bodies and lysosomes containing dense granules in Kupffer cell and hepatocyte,and they were increased along with doses adding. Nuclei deformed, ALP and GPT in serum were rose in the group of 20.0 mg*kg-1. Different doses of Changle could lead to distinct biological effects. Conclusion: Long-termed administration of 20.0 mg*kg-1 Changle can lead to damage of structure and function of rat liver.
6.Nitrogenous chemical constituents of Weiceng

Objective To investigate the nitrogenous chemical constituents of Weiceng .Methods Weiceng extract was subjected to various column chromatography and spectroscopic methods were used for the elucidation of compounds .Results Seventeen compounds were isolated and identified as cyclo-(Leu-Tyr)(1), cyclo-(Phe-Tyr) (2), cyclo-(Pro-Gly) (3), L-Pyroglutamic acid methyl ester (4), uracil (5), thymine (6), N-acetylphenylalanine (7), tyrosine (8), phenylala-nine (9), phenylalanine methyl ester (10), N-methyl leucine (11), isoleucine (12), valine (13), leucine (14), glutamic acid (15), glycine (16) and aspartic acid (17).Conclusion All the seventeen compounds are isolated from Weiceng for the first time .Before this study , cyclopeptides 1-3 have never been isolated from soy bean or its products .
9.Improving teaching quality of oral and maxillofacial surgery with diversified discussion teaching

To improve the students'! comprehensive quality and cultivate their innovative medical talents, we applied different forms of discussion teaching in different stages of the students. In the course of theory, the form of short discussion was adopted to improve the students'! interest and concentration. In preclinical practice stage, the method of problem centered discussion was used to consolidate the profes-sional knowledge of the students. Discussion in the clinical practice was based on the actual problem of diagnosis and treatment, in order to cultivate the students'!clinical thinking. Through the diversified practice of discussion teaching with positive teaching evaluation, students'!interest in oral and maxillofacial surgery and the quality of teaching were improved.
10.Comparison of the five models on experimental hyperlipidemia rats

Aim To select the ideal experimental rats model for research on prevention and treatment of Hyperlipidemia.Methods Five prescriptions on experimental hyperlipidemia-rats were selected and emulsion was made.At 10、20、30 d blood samples were taken from ocular orbit and content of CHO、 LDL、 HDL 、TG in the serum were measured.Results Four of the five prescriptions could lead to the disorder of lipid metabolism at different levels.Conclusion An ideal experimental rats model of hyperlipidemia can be made if bile salt and propylthiouracil are conbined to add to the fat emulsion of lard and cholesterol.
