1.Effect of RGD-insulin on activities of bone resorption and the possible mechanism in human osteoclast-like cells in vitro

Objective In this study, we explored the mechanism of anti-bone resorption of RGD-insulin using osteoclast-like cells (OLCs) from giant cell tumor of bone as an in vitro model. Methods The function of bone resorption was observed and the staining for tartrate-resistant acid phosphatase (TRAPase) was identified. The carbonic anhydrase Ⅱ (CAⅡ) gene expression, apoptosis and the adhesion of OLC that treated by RGD-insulin and echistatin were determined by in situ hybridization, TUNEL staining and adhesion test, respectively. Results OLCs were TRAP positive and had bone resorptive function. After OLCs were treated with 10 -5, 10 -6, 10 -7mol/L RGD-insulin and echistatin respectively, the numbers of TRAP-positive multinucleated cells were (179?3), (238?11), (351?10) per well that treated with echistatin and (229?20), (222?21), (265?17) per well that treat with RGD-insulin.The expression of CAⅡ mRNA decreased, the number of OLCs apoptosis increased in a dose-dependent manner (P
2.Application of China-made expanded polytetrafluoroethylene in plastic and aesthetic surgery

Objective To evaluate the clinical efficacy of China-made expanded polytetrafluoroethylene (ePTFE) in the treatment of facial defects and hollow deformities on the face. Methods Facial augmentation was performed in different sites as the forehead, temple, nose, chin, nosal base and maxilla, respectively, to correct the facial defects and hollow deformities, by using either China-made ePTFE (Experimental Group) or imported ePTFE (Control Group). Postoperative parameters between the two groups were compared. Results The Experimental Group included 16 patients (18 sites), in whom the postoperative follow-up was conducted for 6~9 months. In this group, a secondary infection (in the nose) occurred in 1 patient because the implant was placed too superficially and too close proximity to the incision, and the implant was removed out. Delayed healing of the incision (in the chin) with uncovered implant was found in 1 patient, who was cured by the change of dressing. In the rest of the patients, no obvious allergic, inflammatory or rejection reaction was seen and a good cosmetic result was achieved. The satisfactory rate of this group was 94.4%(17/18). The Control Group included 10 patients (20 sites). The implant was found bared and then removed in 1 patient (in the nose). The satisfactory rate of this group was 91.7%(11/12). There were no statistical differences between the two groups in the wound healing ( ? 2 =1 109, P =0 574), the adverse reaction ( P =1 000), and the clinical efficacy ( P =1 000). Conclusions China-made ePTFE gives histocompatibility as good as imported one. It is suitable for filling the soft tissue and can be used as a safe and economical alternative.
3.Capsule preservation of prosthesis to treat capsular contracture after augment ation mammoplasty

Objective To explore the treatment of capsular contr ac ture after augmentation mammaplasty with mammary prostheses. Method Through primary incision (axillary or inframammary) and minor capsu le incision, patients with capsular contracture after augmentation mammoplasty w ere treated by preserving the contractive capsule and repositing of the breast p rosthesis anterior to the primary capsule. Results Total ly 12 cases (14 breasts) of capsular contracture were achieved satisfactory res ults since 2001. Conclusion The method of capsule preser vation and reposting of the breast prosthesis anterior to the capsule present n umerous advantages: little trauma, less bleeding, simple surgical procedure, saf e, less possibility of recrudescence, and can be finished through a small primar y axillary incision. It is a preferable method to treat capsular contracture.

5.Survey of satisfaction in patient with modified subpectoral and subglandular breast augmentation

Objective To investigate the different procedure effects in patients with modified sub-pectoral and subglandular breast augmentation. Methods Patients with modified subpectoral breast aug-mentation (10 cases) and subglandular breast augmentation (10 cases) were randomly selected, and satis-factory surveys to the procedures were conducted in the two groups of patients, including breast touch-feeling, dynamic change, as well as self-feeling and spouse-feeling. Results By the comparison of sub-glandular with subpectoral breast augmentation, touch-feeling was excellent in the former group, with breast spreading out to bilateral side in horizontal position. The breasts had different changes with differ-ent posture and natural movement. Spouse was more satisfactory to the former than the latter group. Conclusion Subglandular breast augmentation has natural shape, dynamic changes and fidelity of the re-al, especially in horizontal position, which is more conformable to the standards of patients and their spouse, and better than subpectoral breast augmentation.

7.Expression and clinical significance of PI3K, Akt and E-cadherin in thyroid papillary carcinomas

Objective To investigate the expressions of phosphatidylinositol 3-kinase (PI3K), Akt and E-cadherin and their clinical significance in thyroid papillary carcinomas.Methods Expressions of PI3K, Akt, and E-cadherin were detected in 62 cases of thyroid papillary carcinomas,30 cases of thyroid goiter and 30 cases of normal thyroid by immunohistochemistry (EnVison),and simultaneously compared with age, sex, tumor size, clinical tumor node metastasis( TNM) stages, and lymph node metastasis in thy-roid papillary carcinomas.Results The expression rate of PI3K, Akt, and E-cadherin was 74.2%(46/62), 66.1%(41/62), 16.1%(10/62),respectively.Expressions of three proteins in thyroid papillary carcinomas were significantly different from those in thyroid goiter and normal thyroid tissues ( P <0.05). The lower positive rates of PI3K and Akt proteins were obtained in the group of stageⅠ~Ⅱthan that in the group of stageⅢ~Ⅳ(χ2 =4.976, P =0.026;χ2 =6.233, P =0.013).Higher positive rates of PI3K and Akt proteins were obtained in the group of lymph-node metastasis than that in group of non-lymph-node metastasis (χ2 =6.675, P =0.010;χ2 =7.511, P =0.006).Higher positive rate of E-cadherin protein was obtained in the group of stage Ⅰ~Ⅱ than that in the group of stage Ⅲ ~Ⅳ (χ2 =6.558, P =0.010 ) .Higher positive rate of E-cadherin proteins was obtained in the group of non-lymph-node metastasis than that in the group of lymph node metastasis(χ2 =5.678, P =0.017).There was significant positive correlation between expressions of PI3K and Akt through Spearman correlation analysis ( r =0.423, P <0.05).PI3K was negatively correlated with E-cadherin with Spearman correlation analysis ( r =-0.527, P <0.05).Akt was also negatively correlated with E-cadherin ( r =-0.417, P <0.05).Conclusions PI3K/Akt pathway might regulate thyroid papillary carcinoma cells proliferation, invasion and metastasis.
8.Effect of oral mucosa transplantation in the treatment of severe contracted conjunctival sac after ocular prosthesis implantation

AlM: To observe the clinic effect of oral mucosa transplantation in the treatment of severe contracted conjunctival sac after ocular prosthesis implantation .
METHODS: Thirty-three cases ( 33 eyes ) with globe disorders and severe contracted conjunctval sac were operated ocular prosthesis implantation firstly, and conjunctival sac plasty using oral mucosa after 6mo.
RESULTS: Thirty - one cases were successful, no complications appeared. One case had primary ptosis and 1 case had recurrent conjunctival sac contracture.
CONCLUSlON:lt is recognised that the methods of oral mucosa transplantation in severe contracted conjunctival sac after ocular prosthesis implantation are effective on those cases.
10.MicroRNA expression in hepatocytes with hydrogen peroxide-induced oxidative stress-injury and alleviating effect of mesenchymal stem cell-conditioned medium

AIM: To evaluate the changes of microRNA (miRNA) in hepatocytes during hydrogen peroxide-induced oxidative stress injury, and to observe the alleviating effect of mesenchymal stem cell-conditioned medium (MSC-CM) in this progress.METHODS: The hepatocyte oxidative stress injury model was established using hydrogen peroxide and human normal liver cell line L02.MSC-CM was prepared using centrifugation and filter.The effects of MSC-CM on hepatocyte injury were evaluated by apoptosis analysis, cell viability detection, cell cycle, and mitochondrial membrane po-tential (MMP).Twenty-one differentially expressed miRNAs were selected by gene chip hybridization, in which miR-143, miR-145, miR-301a and let-7a were confirmed by RT-qPCR.Bioinformatics software was utilized to predict target proteins of these miRNAs, and then the proteins were verified by Western blot.RESULTS: MSC-CM markedly attenuated hydrogen peroxide-induced oxidative stress injury by reducing apoptosis, promoting cell viability and regulating cell cycle.The ex-pression of miR-143, miR-145, miR-301a and let-7a, indentified by RT-qPCR, increased under the condition of oxidative stress injury, while decreased after MSC-CM treatment.The expression of miR-143 predicted target proteins, HK2 and ADRB1, decreased under the hydrogen peroxide-exposure, while increased after MSC-CM treatment, which is consistent with the regulatory trend of miR-143.CONCLUSION: MSC-CM might attenuate hydrogen peroxide induced oxidative stress injury via inhibiting apoptosis and regulating some miRNA expression.
