3.Effect of torasemide or furosemide combined with mannitol in treatment of acute cerebral hemorrhage patients with cerebral edema

Objective To compare the effect of torasemide or furosemide combined with mannitol in the treatment of acute cerebral hemorrhage patients with cerebral edema.Methods One hundred and sixty patients with cerebral hemorrhage were selected,and divided into torasemide group and furosemide group according to different treatment methods,40 cases in each.The torasemide group was treated with torasemide combined with 20% mannitol,the furosemide group was treated with furosemide combined with 20% mannitol.The curative effect,mannitol dosage,edema volume,24 h urine and adverse reactions in two groups were compared.Results The total effective in torasemide group was higher than that in furosemide group [97.5% (78/80) vs.77.5% (62/80)],mannitol dosage was less than that in furesemide group [(347.5 ±32.5) ml vs.(438.2 ±30.7) ml],the incidence of adverse reactions was lower than that in furosemide group [7.5%(6/80) vs.27.5%(22/80)],which reached statistical significance (P< 0.01 or < 0.05).The edema volume in the 7th,14th day in torasemide group was less than those in furosemide group [(21.1 ±3.4) ml vs.(23.3 ±4.8) ml,(17.6 ±4.5) ml vs.(22.4 ±5.6) ml],the 24 h urine in the 3re,7th,14th day were more than those in furosemide group [(3 684 ±528) ml vs.(3 429 ±592) ml,(3 854 ± 746) ml vs.(3 185 ±490) ml,(3 742 ±t658) ml vs.(2 251 ± 324) ml],which reached statistical significance (P < 0.05).Conclusions Torasemide in treatment of acute cerebral hemorrhage patients has better efficacy and safety.It is better than furosemide.

5.Expression and function of CD40 on rat peritoneal mesothelial cells

Objective To determine whether peritoneal mesothelial cells express CD40 and to investigate potential mechanism by which CD40-CD40 ligand(CD40L) interaction may be involved in the inflammation of peritoneal membrane. Methods Rat peritoneal mesothelial cells (MC) were harvested from peritoneal cavity and maintained under defined in vitro conditions. Expression of CD40 on MC under normal culture or stimulation with interferon-?(IFN-?), tumor necrosis factor-?(TNF-?), interleukin(IL)-1 was examined by RT-PCR and FACS analysis. After activation with CD40 mAb, the expression of intercellular adhesion molecule-1 (ICAM-1) on MC was analyzed by FCAS. Results MC cultured in vitro expressed CD40 constitutively. The expression of CD40mRNA and CD40 protein up-regulated markedly following the stimulations with IFN-?, but not with IL-1, TNF-?. The expression of ICAM-1 on MC was significantly increased after activation of CD40 with IFN-? and CD40mAb. Conclusions MC functionally expresses CD40. The interaction of CD40 on MC and CD40L+ cells in peritoneal cavity may play an important role in peritoneal local defense and may be involved in the inflammation process of peritoneum.

Polymerase chain reaction (PCR) is one of the common techniques in molecular biology, which can amplify nucleic acids through the cycle of denaturation, annealing and extension. Based on the principle of common PCR, rapid PCR is to realize the amplification of nucleic acids in less time without affecting the specificity, sensitivity and fidelity of the reaction. A lot of research work in this field has been going on in recent years. This article will make a review of the development of rapid PCR with emphases on the improvement of DNA polymerase, the choice of additives and the improvement of thermocyclers.
7.Value of Heparin Blocking Attachment of Respiratory Syncytial Virus to Human Airway Epithelial Cells

Objective To investigate the mechanisms of attachment of respiratory syncytial virus(RSV) to human airway epithelial cells.Methods Attachment of RSV to CFBE was quantitatively assessed by flow cytometry.Various polyclonal and monoclonal antibodies to RSV and heparin were pre-incubated with RSV.The blocking effects of these antibodies and heparin on attachment were evaluated.Results CFBE cells reduced capacity being bound by RSV.All the antibodies used were failed to block attachment of RSV on CFBE,whereas heparin blocked RSV attachment in a dose-response manner and the blokade by heparin was almost complete at the concentration of 0.4 mg/L.Conclusions Flow cytometry provides direct evaluation of attachment without growth of virus.Heparin-like molecules on cell surface of CFBE appears to be involved in attachment between RSV and human epithelial cells.
8.Advance in studies on anti-diabetic mechanism of Gardeniae Fructus and its active ingredient geniposide.

The diabetes is mainly treated by the oral administration of western medicines at present. Despite their rapid curative effect, there have been still many reports for the western medicines about their clinical adverse reactions, failure of effective prevention and treatment of complications and drug resistance. Hence, they are not suitable for long-term administration. Traditional Chinese medicines have a long history in treating diabetes mellitus (DM) , which is commonly known as Xiaokezheng in the theory of traditional Chinese medicines. In recent years, many scholars have taken extracts from traditional Chinese medicines or separated active constituents as the study objects in the expectation of developing new-type drugs for treating and preventing diabetes. Therefore, a large number of study reports have been emerged in this field. Due to their significant glucose-reducing effect and specific effect in treating complications of diabetes, traditional Chinese medicine Gardeniae Fructus and its iridoid component geniposide shall be given full attention. This paper summarized the advance in studies on the curative effect and action mechanism of Gardeniae Fructus and geniposide in preventing and treating diabetes.

10.The research progress of Aurora-B kinase and its inhibitors.

Aurora-B as an important kinase to adjust the cell normal mitosis is a potent target for cancer treatment. Aurora-B is overexpressed in a broad range of tumor and tumor cells are more sensitive while Aurora-B is inhibited. Due to the key role of the Aurora-B in cell mitosis, the development of its inhibitors is becoming more and more important. Several small molecules inhibit with a similar efficacy both Aurora-A and Aurora-B, however, in most cases the effects resemble Aurora-B disruption by genetic methods, indicating that Aurora-B represents an effective therapeutic target. There were several Aurora-B kinase inhibitors which had entered the clinics and displayed good antitumor activity. In this review, we will outline the functions of Aurora kinase B in normal cell division and in malignancy. We will focus on recent preclinical and clinical studies that have explored the mechanism of action and clinical effect of Aurora-B inhibitors in cancer treatment.
