1.A voxel model constructed for calculating mouse internal radiation dose using MC method

Objective To construct a voxel mouse model combining with Monte Carlo method used for radiation dose calculation.Methods A set of slice images obtained from a nude male mouse (28 g) was utilized, all slice images were registered,some organs or tissues were identified, segmented and filled with specific color, and finally the physical property was defined by MCNP application.Results A mouse model with a voxel size of 0.2 mm×0.2 mm×0.2 mm, consisting of 14 organs or tissues,was constructed, which could satisfy the requirement of precision for radiation dose distribution calculation and moderate computing time.Conclusion The voxel mouse model can be used to calculate the quantity of ionization radiation dosimetry in related areas including radiological medicine, nuclear medicine and space radiation medicine.

3.Clinical trial of ticagrelor in the treatment of acute myocardial infarction in patients with percutaneous coronary intervention after thrombolytic therapy

Objective To compare the clinical efficacy and safety of ti-cagrelor and clopidogrelin the treatment of acute myocardial infarction in patients with percutaneous coronary intervention ( PCI ) after thrombolytic therapy.Methods A total of 130 ST -segment myocardial infarction (STEM) patients were randomly divided into control group (n=64) and treatment group ( n=66 ).The patients of control group were given clopi-dogrel 300 mg, maintenance dose 75 mg qd.The patients of treatment group were given ticagrelor 180 mg, maintenance dose 90 mg bid.All the patients of two groups received selective PCI successfully after 8-10 d treatment.The change of thrombolysis in myocardial infarction ( TIMI ) flow grades after operation , left ventricular ejection fraction ( LVEF) and left ventricular end -diastolic dimension ( LVEDD ) , the incidence of no-reflow/slow flow and adverse drug reactions after operation were compared between two groups.Results After operation, the TIMI flow grades and LVEF in treatment group were significantly higher than those in control group ( P<0.01 ).After operation , the LVEDD in treatment group was lower than that in control group , with significant difference ( P<0.01 ).The incidences of no -reflow/slow flow in treatment group and control group were 3.03%and 14.06%respectively , there was statistically significant difference in two groups ( P<0.05 ).Only some minor bleeding events happened in the two groups , there was no statistically significant difference in two groups after operation 6 months ( P>0.05 ).Conclusion Ticagrelor could support better clinical efficacy than clopidogre in selective PCI after thrombolytic therapy for acute myocardial infarction , which has a stronger anti -platelet aggregation function without increasing the incidence of bleeding events.
4.Preparation of tissue-engineered skin grafts with hTERT cells carrying human papillomavirus type 6 genome in vitro:a preliminary study

Objective To establish a model for preparation of tissue-engineered skin grafts with hTERT cells carrying human papillomavirus type 6 (HPV 6) genome in vitro, so as to lay a foundation for studying HPV life cycle. Methods The full-length linear HPV6 genome and plasmid pEGFP-▲EGFP were electrophoretically cotransferred into hTERT cells. After selection using G418 resistance, Southern blotting was performed to determine the viral load of HPV6 in transfected cells. 3T3 J2 trophoblastic cells, type I rat-tail collagen and hTERT cells containing the full-length HPV6 genes (HPV6.hTERT cells)were mixed and cocultured on metal meshes to form skin graft-like structures. Hematoxylin and eosin (HE)staining was performed to observe the structure of formed skin grafts, an immunohistochemical assay to measure the expression of HPV6 L1 protein, and electron microscopy to observe virus particles in the skin grafts. Results The linear HPV6 gene was successfully transferred into hTERT cells, and Southern blotting showed the presence of HPV6 DNA in the transferred hTERT cells. The HPV6.hTERT cells, which were cocultured with 3T3 J2 trophoblastic cells and type I rat-tail collagen, proliferated and differentiated over time, and gradually formed skin grafts giving the appearance of verrucous hyperplasia. HE staining showed that the cocultured HPV6.hTERT cells could form typical stratified structure of skin after 7 days of cultivation, and histopathologic features of HPV infection, including obvious papillomatous hyperplasia, presence of vesicular cells, hyperkeratosis and parakeratosis, could be observed after 21 days. The immunohistochemical assay showed the expression of HPV6 L1 protein in the upper portion of skin grafts, and electron microscopy revealed the presence of HPV6 virus particles in skin grafts. Conclusions The established model for preparation of tissue-engineered skin grafts using HPV 6 genome-carrying cells provides a basis for biological studies of HPV, but its application is limited to some degree.
5.Expression of recombinant human zona pellucida-3 protein (rhZP3) in Pichia pastoris.

Human Zona Pellucida(ZP), which is a complex matrix surrounding oocytes,is comprised of three immunologically distinct glycoproteins(hZP1, hZP2 and hZP3). Because hZP3 possesses the sperm receptor activity and the acrosome-inducing activity, it has long been used as a candidate antigen to develop an immunocontraceptive vaccine. However, a large amount of native hZP3 protein is unavailable. It is an effective way to express hZP3 protein directly in vitro. Nevertheless, it had been reported that the rhZP3 protein produced in Pichia pastoris was not secreted but accumulated in the cells and could only be purified after being solubilized by strong denaturants. More unfortunately, after purification the final product required 6mol/L urea to maintain solubility. An improved project was advanced with the aim to express secreted and soluble rhZP3 protein in yeast. In this study, the fragment of hZP3 cDNA coding for aa 23 - 408, which the N-terminal leader was removed and most of the C-terminal transmembrane-like domain was reserved, was amplified by two PCR primers including EcoR I and Not I sites respectively and a His6 codon cassette was added to 5'-terminal. The hZP3 insert was incorporated into expression vector pPIC9K. The resulting recombinant yeast expression vector was designated pPIC9K-rhZP3. Linearized pPIC9K-rhZP3 was transformed into Pichia pastoris. After G418 selection, the recombinant Pichia pastoris strains were identified by PCR and the rhZP3 was expressed following the manufacturer' s protocol. Following induction with methanol, the rhZP3 protein was secreted and dissolved into the culture supernatant. SDS-PAGE and Western blot analyses showed that the apparent molecular weight of the expressed rhPZ3 proteins in yeast was smaller and a little size heterogeneity than native ones; after purified with Ni-chelating affinity chromatography, the final product's apparent molecular weight was about 32 - 34KD and their yield more than 20mg/L. We supposed that the C-terminal transmembrane-like domain be useful for secretion of rhZP3 into the culture supernatant and the expressed rhZP3 protein be incompletely digested by proteinases of Pichia into shorter fragments which all were glycosylated inhomogeneously. Fortunately, the fragments of rhZP3 protein can be recognized in Western blot by the polyclonal antibodies to porcine ZP3 which has showed a cross-reactivity with human ZP in vitro. It will be expected that the rhZP3 protein expressed in Pichia pastoris not only has immunogencity, say, it can rise antibodies in vivo to prevent spermatozoa-ovum binding, but also does not contain ovarian factors that might be the cause of undesired side effects, e.g. ovaritis and can be used as a safe immunogen in human antifertility vaccine research.

7.Endothelium-independent vasorelaxant effect of puerarin on rat thoracic aorta.

OBJECTIVETo investigate the vasorelaxant effect of puerarin in rat aortic rings and the mechanism.
METHODThe isolated thoracic aortic rings of male Sprague-Dawley rats were mounted on the organ bath and the contractile responses of the vessel were recorded.
RESULTPuerarin completely relaxed the contractions induced by phenylephrine in a concentration-dependent manner in endothelium-intact and endothelium-denuded rat aorta, but it had no effect on those preconstricted by a high concentration of potassium chloride (KCl, 60 mmol x L(-1)). The relaxant effect of puerarin was significantly inhibited by pretreatment of endothelium-denuded aorta with potassium channel antagonists tetraethylammonium, 4-aminopyridine but not glibenclamide.
CONCLUSIONPuerarin induces an endothelium-independent relaxation in rat aortic rings. The mechanisms may involve the reduction in Ca2+ influx through the calcium channels operated by alpha-adrenergic receptor and the activation of the potassium channels (Kv and BKca, but not KATP).

8.Induced differentiation of bone marrow stem cells in transplanted rat liver.

OBJECTIVETo investigate the differentiation of bone marrow stem cells in transplanted livers and its impact on the long-term survival of rats with orthotopic liver transplantation.
METHODSTwenty-four female recipient rats with orthotopic liver transplantation were randomized into blank-control group, D-hanks solution group, bone marrow stem cells group with postoperative infusion of stem cells, and the pathological changes of the liver grafts and survival time of the rats were observed. The differentiation of the bone marrow stem cells were assessed 60 days after transplantation using in situ hybridization histochemistry for Sry gene and alpha-fetoprotein (AFP) immunohistochemistry.
RESULTSIn rats with postoperative infusion of bone marrow stem cells through the portal vein, the median long-term graft survival time exceeded 180 days, significantly longer than that in the other two groups (P<0.05), and no obvious evidence of acute rejection was observed with positive Sry expression and AFP expression.
CONCLUSIONInfusion of bone marrow stem cells through the portal vein following liver transplantation may alleviate acute graft rejection and promote long-term liver graft survival and AFP expression.

9.The effects of metformin on glucolipid metabolic disorders and liver lipid deposition caused by clozapine in rats

Objective To study the effect of metformin on glucolipid metabolic disorders and liver lipid deposition caused by clozapine in rats.Methods From 1 d to 4 d,Clozapine 5 mg·kg -1 ·d -1 was gavaged,and the dose increased to 25 mg·kg -1 ·d -1 from the 5th day.Metformin 100 mg·kg -1 ·d -1 or 400 mg·kg -1 ·d -1 or simvastatin 1 mg· kg -1 ·d -1 was gavaged from the 15th day.The total period of dosing was 8 weeks.Body mass,fasting blood sugar (FBS) and postprandial 2 hours blood glucose (2hPBG)were measured at baseline,3 d,1 week,2 weeks,4 weeks,6 weeks and 8 weeks.At the end of the 8th week,serum cholesterol (TC),triglyceride (TG),low density lipoprotein (LDL -C), high density lipoprotein (HDL -C),fructosamine (FA)and insulin (IRS)were measured and liver HE staining was done.Results There were no significant differences of the measured indexes between control group and metformin group at the all test points.By the end of the 6th and 8th week,compared with control group,the body mass,FBS,2hPBG,IRS, FA,TC,TG and LDL -C were significantly increased in clozapine group (P <0.05 ),while HDL -C decreased in clozapine group (P <0.05).Compared with clozapine group,body mass,FBS,2hPBG,IRS,FA,TC,TG and LDL -C were significantly decreased by metformin or simvastatin administration (P <0.05),while HDL -C increased(P <0.05).Rat liver cells in clozapine group were not neat around the small blood vessels;there were more white fat cells and hepatocellular lipid calm far away from the blood vessels.However,in other groups,there were moderate white fat cells, and there were not much hepatocellular lipid calm far away from the blood vessels.Conclusion Metformin could effectively prevent and treat weight gain,glucolipid metabolic disorder and liver lipid deposition caused by clozapine.
10.An epidemiology study on hypertension in the Zhuang nationality village from the Guangxi Zhuang Autonomous Region

Objective To observe the risk factors and prevalence status of hypertension at the Guangxi Zhuang-rural region.Methods Through cross-sectional survey,Xinlan village (Ethnic-Zhuang tribe village) in Liangqing District of Nanning was chosen as survey site.Select Zhuang-ethnic villagers (age≥18 years,living time ≥ 6 months per year) as survey subjects.Questionnaire survey and examinations including blood pressure measurement,blood tests,and urine test were made by trained medical professionals.Multivariate logistic regression analysis was used to analyze the risk factors of hypertension.Results Complete data were obtained in 2036 residents.The hypertension prevalence rate was 11.6% (237/2036) [12.5% (110/883) for male,l1.0% (127/1153) for female],adjusted prevalence rate was 10.7% (11.3% for male,10.4% for female) based on the population of Guangxi in 2010.Hypertension awareness rate was 36.3% (86/237),therapy rate was 22.8% (54/237),and control rate was 11.0% (26/237).Multivariate logistic regression analysis showed that age (OR =1.06,95% CI:1.05-1.08,P < 0.01),alcohol drinking habit (OR =1.92,95% CI:1.25-2.95,P < 0.01),high salt diet(OR =1.48,95% CI:1.04-2.10,P < 0.05),diabetes (OR =7.03,95% CI:1.94-25.44,P <0.01),obesity (OR =1.20,95 % CI:1.15-1.25,P < 0.01) and hypertriglyceridemia (OR =1.12,95% CI:1.03-1.22,P <0.01) were risk factors for hypertension in this cohort.Conclusions Prevalence rate is slightly lower in adult of the Guangxi Zhuang-rural areas than national or Guangxi Zhuang Autonomous Region levels,but hypertension awareness,therapy and control rates in this cohort are not satisfactory.Targeted strategies are needed to improve the prevention and therapy of hypertension in this region.
