The discussion could help to clearify the current situation of TCM trestment for IPF.
3.Construction and expression of eukaryotic expression vector for recombinant human plasminogen Kringle 5 gene

Objective To construct baculovirus expression system for the human plasminogen kringle 5(hK5) gene,and detect its expression in Spodoptera frugiperda 21(sf21). Methods Baculovirus Bacmid-rhK5 was prepared with bac-to-bac baculovirus expression system,and sf21 cells were infected.The expression of rhK5 in sf21 cells was determined by Western blot.The activity of purified protein was detected by vascular endothelial cell inhibition test. Results The pFastBac HTB-rhK5 was successfully constructed,and sf21 cells were transfected with the constructed vector.The output of rhK5 obtained was 90 ug/L,and the protein possessed the inhibitory activity of endothelial cell proliferation.The median effective dose(ED50) was 4 ug/mL. Conclusion The rhK5 baculovirus expression vector is highly expressed in sf21 cells,which lays a foundation for the the expression of rhK5 protein with biologic activities.
4.Expression of EZH2 and Ki-67 in salivary adenoid cystic carcinoma and their clinical significance

Objective:This study aimed to investigate the expressions of EZH2 and Ki-67 in the salivary adenoid cystic carcino-ma (SACC) of humans and their correlation. Methods:A total of 42 cases of SACC tumor tissues and 5 cases of normal tissues were considered to determine the expressions of EZH2 and Ki-67 by immunohistochemistry. The relationship and correlation of such expres-sions with the clinicopathological characteristics were also analyzed. Results:The expression of EZH2 was notably higher in SACC than in normal tissues (P<0.05). EZH2 expression was detected in 66.67%(28/42) of the tumor tissues. This expression was correlated with pathological grade and clinical stage. By contrast, EZH2 expression did not correlate with gender, age, and localization. EZH2 was not expressed in normal tissues. The incidence of EZH2 expression in the Ki-67 positive group was 75.76%(25/33) and the incidence in the Ki-67 negative group was 33.33%(3/9). The difference between the two groups was statistically significant (P<0.05). Conclu-sion:The increased expression of EZH2 in SACC was related to tumor proliferation. EZH2 may participate in tumor cell proliferation via cell cycle management.
5.Efficacy of adjunctive intravitreous injection with Lucentis for neovascular glaucoma

AIM: To observe the clinical efficacy of adjunctive intravitreous injection with Lucentis for the treatment of neovascular glaucoma ( NVG) .
METHODS:The retrospective case series study included 25 eyes of 25 patients who underwentintravitreous injection with Lucentis. Patients firstly received an intravitreous injection with Lucentis (0. 5mg/0. 05mL), after the regression of neovascularization of the iris, patients accepted different surgical treatments according to different etiopathogenesis condition. Iris, chamber angle neovascularization condition, intraocular pressure, and visual acuity were observed postoperatively. The follow-up duration was 3mo.
RESULTS:After 3-7d of intravitreous Lucentis injecting, iris and chamber angle neovascularization was totally faded in 20 cases (20 eyes) and was not completely faded in 5 cases ( 5 eyes ) . Additional treatments were compound trabeculectomy ( 14 cases, 14 eyes ) , vitrectomy ( 4 cases, 4 eyes ) . The patients' mean intraocular pressure was 43. 42 ± 10. 99mmHg before treatment, which decreased rapidly when they came out of the hospital (14. 26±7. 64mmHg, P<0. 05) and stabilized during the follow-up 3mo (18. 76±5. 96mmHg, P<0. 05). Follow-up at 3mo, visual acuity improved or remained in 20 cases ( 20 eyes ) and decreased in 5 cases ( 5 eyes ) . The complete success, qualified success and failure were 21 eyes, 3 eyes and 1 eye, respectively.
CONCLUSION:Intravitreous injection with Lucentis can be used as an assisted treatment of NVG. According to different etiopathogenesis condition, it is an effective treatment to combine with other treatment methods for NVG.
6.Mechanism of mesenchymal stromal cells in promoting islet graft revascularization

Objective To investigate the mechanisms of mesenchymal stromal cells (MSCs) in promoting the transplanted islets neovascularization.Methods We performed co-transplantation of allogeneic islet and MSCs into non-obese diabetic (NOD) mice and the lymphocyte chimeric rate was detected by flow cytometry.The MSCs migration,transformation and secretion of cytokines were detected by immunofluorescence.Results In mice of co-combination transplantation of islet with MSCs,donor lymphocytic chimerism in graft was increased.BM-MSCs can migrate to transplanted islet,differentiate into vascular smooth muscle cells,and also secrete vascular endothelial growth factor (VEGF).Conclusions MSCs can migrate to transplanted islet and promote neovascularization.Also,it enhanced allograft immune tolerance of islet grafts via increasing donor lymphocytic chimerism.
7.Contrast enhanced ultrasound in the evaluation of hepatocellular carcinoma ( HCC) after percutaneous radiofrequence ablation

Objective To evaluate contrast-enhanced ultrasound (CEUS) for the diagnosis and follow-up of hepatocellular carcinoma ( HCC) patients after percutaneous radiofrequence ablation ( RFA). Methods The data of 46 HCC cases were reviewed in our study. Results of contrast enhanced computed tomography (CT) , ultrasound contrast agent sonography and grey ultrasound were compared with each other pre- and after percutaneous radiofrequence ablation. Results Contrast agent ultrasound imaging had the same value with CT in preoperative diagnosis of HCC, with postive likelihood ratio of 1.26 and 1.31, respectively,sensitivity of 97% and 91% and specificity of 23% and 31% respectively. For grey ultrasound the positive likelihood ratio was 0.99, sensitivity and specificity was 76% and 23% respectively. The sensitivity and specificity of contrast-enhanced ultrasound after percutaneous radiofrequence ablation was 60% and 90% respectively, and the postive likelihood ratio and negative likelihood ratio was 6. 18 and 0. 44 respectively. The difference was significant while compared with CT (P = 0. 012). All cases were followed-up from 1 to 9 months, and all were alive with recurrence found by contrast-enhanced ultrasound in 3 cases. Conclusions Contrast agent ultrasound imaging is sensitive and effective in the diagnosis of HCC and follow-up after percutaneous radiofrequence ablation.
8.Antibacterial and Ultrastructure Effect of RES on Staphylococcus Aureus Standard Strains ATCC 2 5 9 2 3

Objective To study the antibacterial effect and mechanism of Resveratrol(RES)on Staphylococcus aureus stand-ard strains (ATCC 25923).Methods Using the microdilution protocol,to obtain minimal inhibitory concentrations(MIC)of RES on Staphylococcusaureus standard strains.Transmission electron microscope (TEM)and scanning electron microscope (SEM)were tested to obtain the effect of Staphylococcusaureus on Ultrastructure.Results The MIC of RES to Staphylo-coccusaureus standard strains ATCC 25923 were 0.256 mg/ml.Compared with control group,transmission electron and scanning electron microscope observation showed that after the act of RES on S.au standard strains,the cell morphology of S.au standard strains changed serious deformation,cells in all shapes and size,cell boundary roughness,cell wall defected and ruptured,and osteoporosis of edgeo,looseness of its structure,the nuclear membrane rupture,local showed vacuolar characteristics.Cell ultrastructure was obvious damaged and inhibition.Conclusion Rseveratrol had obvious inhibition and destruction to the standard strains of staphylococcus aureus,and the inhibition mechanism may be related to the destruction of cell membrane permeability.
9.Diagnostic value of spectral CT imaging on complications after breast augmentations with siliconeim plants

Objective To evaluate the diagnostic value of the spectral CT imaging on the complications after breast augmentations with silicone implants.Methods A lot of 22 cases with breast implants were scanned by the Gemstone spectral imaging (GSI) CT.The original data were loaded in processed workstation, analyzed by GSI analysis software, and then stored the series we needed.The features of the complications after breast augmentations with silicone implants were reviewed retrospectively on the spectral CT images.Results A lot of 44 breast implants scanned in our study and there were 23 breast implants with complications, including 7 breast implants with capsular rupture (6 breast implants with endometrial rupture detected by spectral CT, including 4 breast implants with endometrial rupture, 3 breast implants with outer membrane rupture and leakage), 13 breast implants with capsular contracture (classification according to Baker's classification, 6 in grade Ⅰand 7 in grade Ⅱ), 8 breast implants were moved and 2 breasts had foci of calcification.Conclusions The spectral CT imaging play important role in detecting the complications after breast augmentations with silicone implants by GSI analysis software.
10.Metabolic profiling of liver in the rats with chronic heart failure

Objective To establish a rat model of liver metabolism profile in chronic heart failure (CHF), to explore the dynamics of liver metabolism in CHF from the point of view of metabolism, and to find the characteristic metabolites valuable for the molecular mechanism and management of CHF.Methods Twenty male Wistar rats were assigned to the CHF group to receive aortic coarctation or to the control group to receive sham surgery, and were bred for 24 weeks following surgery.The metabolic profiling of the rat liver tissues was analyzed on a metabonomics research platform. Orthogonal partial least squares-discriminant analysis ( OPLS-DA) model and principal component analysis ( PCA) model were established for liver tissues of the CHF rats, and the characteristic metabolites were finally derived by data processing with SPSS 19.0 software.Results The PAC and OPLS-DA models were established successfully.Ten characteristic metabolites with significant differences between the CHF and control groups, including lysophosphatidyl choline, lysophosphatidyl ethanolamine, oleic acid, glycocholic acid, and dehydroepiandrosterone sulfate, were screened and identified from the models.Conclusions The metabolic disorders in CHF rats are well fitted to the established metabolic profile models, and these identified characteristic metabolites may provide reference for the pathophysiological molecular mechanism and management, etc., of chronic heart failure.
