1.MTHFR 677 C/T polymorphism-new ideas about depressive disorder treatment

It has been confirmed that genetic factor plays an im-portant role in the pathogenesis of depression. MTHFR is one of the key enzymes in folate and homocysteine ( Hcy ) metabolism which participates in Alzheimer’ s disease, depression and other mental illnesses. MTHFR-677C/T polymorphism causes the de-crease of enzyme activity and heat resistance, which will lead to lower folate and elevated plasmal Hcy concentration. All of these results put together will cause the central neuronal damage and microvascular damage and affect the synthesis of central neuro-transmitter and methylation of biogenic amines and phospholipids in the central nervous system, which will eventually induce vari-ous mental illnesses like depression, etc. This article reviews the research advancement in the relationship between MTHFR-677C/T polymorphism and depression in recent years on the basis of MTHFR gene mutation and function, lack of folic acid, elevated plasma homocysteine levels and the MTHFR-C677T polymor-phism. Based on which we hope to bring new ideas about treat-ment of depression.

3.Study on the Compatible Stability of Amphotericin B for Injection and 5%Glucose Injection from Different Manufacturers

OBJECTIVE:To study the factors affecting the compatible stability of amphotericin B(AmB)and 5%glucose injec-tion from different manufacturers,and to provide reliable evidence for the selection of solvent carrier of AmB. METHODS:After the combination of AmB and 5%glucose injection from different manufacturers(manafactarer A,B,C,D,E),it was shielded from light with room temperature(25℃)for 0,1,2,4,6 and 8 h. HPLC was conducted to determine the content changes of AmB in samples and observe the character,clarity and pH values. RESULTS:With the prolonging of time,insoluble impurities were detected in AmB and 5%glucose injection with the pH value less than 4.2 and the content of main drugs was decreased. Only 5%glucose injection from manufacturer A was in line with the requirements of AmB compatibility. CONCLUSIONS:The compatible stability of AmB and 5%glucose injection is closely related to pH values. It is suggested that manufacturers of infusions make clear descriptions of the pH value to select suitable infusions and safe drugs.
4.Determination of Chicoric Acid in Three Different Pharmaceutical Dosage Forms by RP-HPLC

OBJECTIVE:To determine the contents of chicoric acid in three different pharmaceutical dosage forms by RP-HPLC.METHODS:Samples were determined on VP-ODS C18,with the mobile phase consisted of methanol-1% acetic acid-tetrahydrofuran(33∶62∶5) with flow rate at 1.0 mL?min-1,UV detection wavelength at 327 nm,column temperature at 30 ℃ and sample size at 20 ?L.RESULTS:The linear range of chicoric acid was 0.398 72~3.987 2 ?g(r=0.999 8).The average recoveries of the oral liquid,capsules,and tablets were 99.85%(RSD=0.98%),102.50%(RSD=1.84%),and 100.50%(RSD=1.69%),respectively.CONCLUSION:This method is accurate,reproducible,specific,and suitable for the content determination of chicoric acid.

6.A feasibility study laparoscopic liver resections of hepatolithiasis

Objective To assess the feasibility of hepatolithiasis treatment using laparoscopic liver resections. Methods A prospective study of laparoscopic liver resections was undertaken in selected patients with left intrahepatic stone, and these patients were operated by laparoscopic left lobectomy. Results Two cases were operated successfully. Mean blood loss was 400~600 ml. Conclusion Laparoscopic resections are feasible in selected patients with left intrahepatic stone and bring up a new method for hepatolithiasis.
7.Effect of propofol anesthesia on electroconvulsive therapy-induced hyperphosphorylation of Tau protein in hippocampus in depressed rats

Objective To investigate the effect of propofol anesthesia on electroconvulsive therapy (ECT)-induced hyperphosphorylation of Tau protein in hippocampus in depressed rats.Methods Thirty-two female WYK rats in which the total score was 30-120 after Open-field test,aged 24 weeks,weighing 200-250 g,were randomly divided into 4 groups ( n =8 each):control group (group C),propofol group (group P),ECT group (group E)and propofol + ECT group (group PE).In groups C and E,the animals received intraperitoneal normal saline 5 ml,and in addition the animals received ECT 15 min later in group E.In groups P and PE,the animals received intraperitoneal 100 mg/kg propofol 5 ml,and in addition the animals received ECT 15 min later in group PE.The learning and memory function was assessed by Morris water maze test at 24 h after ECT.The animals were sacririced at 6 h after Morris water maze test and the hippocampal tissues were removed for determination of the expression of phosphorylated Tau protein.Results Compared with group C,the escape latency was significantly prolonged,the swimming time was significantly shortened in groups P,E and PE,the expression of phosphorylated Tau protein in hippocampus was down-regulated in group P,and the expression of phosphorylated Tau protein in hippocampus was up-regulated in group E ( P < 0.05).Compared with group E,the escape latency was significantly shortened,the swimming time was significantly prolonged,and the expression of phosphorylated Tau protein in hippocampus was down-regulated in group PE (P <0.05).Conclusion The mechanism by which propofol anesthesia improves cognitive impairment induced by ECT may be related to inhibition of hyperphosphorylation of Tau protein in hippocampus in depressed rats.
8.Investigation on Degree of Familiarity of GCP in Guang’anmen Hospital of China Academy of Chinese Medical Sciences

Objective To investigate the mastery degree of GCP (Good Clinical Practice) in Guang’anmen Hospital of China Academy of Chinese Medical Sciences (hereinafter referred to as“Guang’anmen Hospital”). Methods Totally 338 clinical researchers, covering 18 clinical professional sections and medical clinical trial institutions, in Guang’anmen Hospital with both qualification of clinical trials and GCP certificate were investigated in 23th-27th June 2014. The investigation was in the written answer sheet form, including four noun explanations and 36 multiple choice questions, which were related to GCP. Results 338 researchers could accurately explain the concepts of 4 nouns, and 57 (16.86%) researchers could do all multiple choice questions correctly. 141 (41.72%) researchers chose 1 wrong answer and 89 (26.33%) researchers chose 2 wrong answers. 39 (11.54%) researchers failed to answer 3-4 questions. Only 12 (3.55%) researchers failed to answer 5 or more questions. Conclusion The mastery degree of clinical researchers with qualification of clinical trials and GCP certificate in Guang’anmen Hospital is satisfied. In the future, pertinence and diverse training should be strengthened, in order to improve the overall level of the mastery degree of GCP.

10.Spectroscopic studies on binding of beta-elemene to human serum albumin.

Beta-Elemene is an antitumor drug which is isolated from the traditional Chinese medicinal herb Curcumae Phaeocaulis Rhizoma, it is the main component of elemene which is extracted from the plant and delivered via blood circulation after intravenous injection. The antitumor effect of beta-elemene in vitro and in vivo was definite, and beta-elemene could improve the patient immunity and no sever side effect, drug resistance or bone marrow suppression were found during the clinical studies. And human serum albumin (HSA) is a primary extracellular protein which has a high concentration distribution in blood plasma and has many characteristic physiological functions. Therefore, the binding of beta-elemene to protein may be very important for absorption, distribution, metabolism and elimination. Therefore, the study on the interaction of beta-elemene with drug-carrying protein is very important. In this work, molecular binding of beta-elemene to human serum albumin (HSA) was investigated by using spectrofluorometer. the binding constants suggested that a strong interaction and the formation of a complex between beta-elemene and HSA. This clearly implies that beta-elemene can be stored and removed by the proteins in the body. Furthermore, the fluorescence quenching results showed that the HSA fluorescence was quenched by beta-elemene through static quenching mechanism. Thermodynamic parameters showed that hydrophobic interactions play a role in the binding of beta-elemene to HSA. The negative deltaH(0) and positive deltaS(0) in case of beta-elemene therefore showed that electrostatic attraction play a role in the binding of beta-elemene to HSA.
