3.Roles of matrix metalloproteinases and tissue specific inhibitors of metalloproteinases in dentinogenic ghost cell tumor and ghost cell odontogenic carcinoma

Objective To investigate the roles of matrix metalloproteinases(MMP)and tissue inhibitors of metalloproteinases(TIMP)in dentinogenic ghost cell tumor(DGCT)and ghost cell odontogenic carcinoma(GCOC).Methods The expressions of MMP-2,MMP-9,MMP-14,TIMP-1 and TIMP-2 were examined in 15 DGCT cases and 9 GCOC cases by immunohistochemistry.Their mRNA expression in one DGCT case and one GCOC case were investigated by RT-PCT.MMP-2 and MMP-9 protein activities in the two csses were analyzed by gelatin zymography.Results MMP-9 and TIMP-1 expressions elevated greatly in GCOC,and there was a significant dliferenee(P<0.05)in TIMP-1 expression between GCOC and DGCT.Pro-MMP-9,MMP-9 activated form,pro-MMP-2,and MMP-2 activated forms were detected in the GCOC case.while pro-MMP-9 and MMP-9 activated form were very faint in the DGCT case.The mRNA level of MMP-9 elevated obviously in the GCOC case,which was similar to that of TIMP-1.Conclusions The elevated expression of MMP-9 and TIMP-1 may influence the behaviour of GCOC.

Cardiac hypertrophy is a common phenomenon in heart disea-ses, ultimately lead to heart failure.Hepatocyte growth factor (HGF), which is a pleiotropic cytokine.It has been considered as a promising candidate for cardiovascular therapy for several years .This review focus on these molecular mechanisms and signaling pathways of HGF targeted in cardiovascular system , and summarizes current research about HGF on cardiac hypertrophy , analyses the potential contribution or adverse effect of HGF on cardiac hypertrophy turns out in different heart diseases.
5.Effect of hepatocyte growth factor on the action potential duration of ventricular myocytes in rat

Objective To study the effect of hepatocyte growth factor ( HGF) on the action potential duration ( APD) of ventricular myocytes in rat.Methods Myocytes were isolated by collagenase and were randomly divided into control group ( HGF,0 ) and three concentration test groups (5, 10, 50 μg· mL-1 HGF).With isoprenaline ( ISO) as stimulating factor , patch clamp technique was used to record the APD 50 and APD90 in cells incubated by HGF.Results APD in control group were significantly prolonged by ISO.Compared with the control group , the prolongation of APD50 in small, middle and large test groups decreased 1.2%, 42.3% and 49.2%, while the one of APD90 decreased 19%, 41.1% and 48.8%, respectively as the increasing dose of HGF.The effect of HGF against ISO tends to be concentration dependent.Without ISO, APD90 in three test groups were shortened 6.8%, 13.3% and 28.2%respectively by HGF compared with control group , as the similar trend.Conclusion HGF could shorten APD and promote repolarization , which might have the potential to resist the electrical remodeling when cardiac hypertrophy that was helpful to the treatment of cardiac hypertrophy.
6.The role of matrix metalloproteinases and their tissue inhibitors in oral lichen planus.

OBJECTIVETo investigate the role of MMP-2, MT1-MMP and TIMP-2 in the carcinogenesis of oral lichen planus (OLP).
METHODSImmunohistochemistry was performed to examine the expression of OLP and compare with that of NOM.
RESULTSThe expression of these proteinases significantly increased from NOM, non-atrophic OLP, to atrophic OLP and OSCC. The expression of MMP-2 and MT1-MMP in atrophic OLP was significantly higher than in non-atrophic OLP. Furthermore, the expression of TIMP-2 consequently increased with the increasing of the MMP, but the increase of TIMP-2 was less than that of MMP.
CONCLUSIONSMMP may be useful marker to judge the possibility of malignant change of OLP.

8.MRI analysis of cerebral infarction of hematopathy patient

Objective To enhance the diagnosis of cerebral infarction of the hematopathy patient by analyzing its causes and MRI manifestations.Methods Totally 36 hematopathy patients with cerebral infarction from April 2010 to November 2015 had their data discussed retrospectively on involved vessel,anatomic distribution and MRI manifestations.Results Of the 36 patients,12 ones had only an artery involved in and 24 ones had multi arteries affected,31 ones had multi cerebral infarction foci and 5 ones had single focus.The foci had high or slighdy high signals in T2WI and very high signals in DWI,and there were no enhancement or only gyrus-like enhancement around the foci found by enhanced scan.Conchusion MRI gains advantages over CT when used to diagnose and find the hematopaty patient with cerebral infarction,when it has to be differentiated with cerebral early infection and parenchymal infiltration.It's suggested that the hematopathy patient has to undergo MRI examination in case central nervous system symptoms especially cerebral infarction occur.
9.Recombinant fibronectin polypeptide CH50 improves positive immune regulation in tumor microenvironment

Objective:To investigate the inhibitory effect of in vivo non-targeting transfection of recombinant fibronectin polypeptide CH50 against tumors and to study the related mechanisms.Methods:After inoculated with tumor cells, BALB/c mice were injected with CH50 plasmids,control plasmids,and normal saline separately.The growth of the tumor was observed;the expression of genes (such as B7-1,B7-H1 etc.) in tumor tissues was detected by RT-PCR;and the count of T lymphocytes in local tumor tissues was analyzed by flow cytometry.Results:The tumor growth was obviously suppressed by in vivo CH50 expression.The expression of genes (B7-1 and B7-H1) was up-regulated along with the growth of tumor.CH50 increased the ratios of B7-1/B7-H1 and B7-1/B7-DC and suppressed the up-regulation of IL-10 and TGF-?genes.The direct action of CH50 on H22 cells resulted in the down-regulatoin of TGF-?gene.The count of T lymphoeytes in tumor tissues of CH50 treatment group was significantly higher than that in other groups.Conclusion:Ex- pression of CH50 by non-targeting transfection can effectively inhibit the growth of tumor;the regulation of the immuno- regulatory genes in tumor mieroenvironment is an important part of the treatment mechanism of CH50.

Ion channels are widely distributed in the cardiomyocytes and they are the electrical bases of cardiac excitement-contraction coupling.Abnormal changes of ion channel can lead to a variety of cardiovascular diseases,such as arrhythmia and heart failure.Recent years,several research have focused on the miRNA and diseases.Some of them suggested that miRNAs involved in the occurrence and development of cardiovascular diseases,which were closely related with ion channels.The function of ion channels would change along with the change of the miRNAs.The following paper reviewed that some miRNAs regulated myocardial ion channels according to the existing studies,especially those potent targets of drugs.
