Objective To explore the expression and clinical significance of hypoxia-inducible factor 1α(HIF-1α)and glucose transporter-1(GLUT-1)in human lung adenocarcinoma cell line A549 and in elderly primary lung cancer tissue under hypoxia. Methods MTT assay was used to test the effect of CoCl2-induced hypoxia on the growth activity of lung adenocarcinoma cell line A549.Reverse transcription-polymerase chain reaction analysis(RT-PCR)and Western blot were used to determine the changes of HIF-1α and GLUT-1 in lung adenocarcinoma cell line A549 under hypoxia.The expressions of HIF-1α and GLUT-1 were detected by using immunohistochemical method and RT-PCR in 60 specimens of lung carcinoma and 26 benign lung tissues in elderly patients with pulmonary diseases.Their interrelations and their relationship with clinicopathologicaI characteristics of the tumors were analyzed. Results The growth activity of lung adenocarcinoma cell line A549 was increased under hypoxia.RT-PCR analysis indicated that the mRNA expressions of HIF-1α and GLUT-1 were 0.69±0.08,0.65±0.09 after hypoxia exposure for 24 hours,and 0.99±0.16,0.83±0.11 after hypoxia exposure for 48 hours,which were higher than those in control group (0.47±0.11,0.40±0.08,P＜0.01).The results from Western blot showed that the protein expressions of HIF-1α and GLUT-1 were 1.57±0.29,1.66±0.28 after hypoxia exposure for 24 hours,and 2.31±0.46,2.22±0.34 after hypoxia exposure for 48 hours,which were higher than those in control group(0.93±0.07,1.06±0.17,P＜0.01).In specimens of lung carcinoma,the positive expression rates of mRNA and proteins of HIF-1α and GLUT-1 were higher than those in benign lung tissues(all P＜0.01).Compared with the mRNA and protein levels of HIF-1a and GLUT-1 in lung carcinoma tissues of Ⅰ and Ⅱ stage,there were marked increases in Ⅲ and Ⅳ stage (all P＜0.05).The expressions of HIF-1α and GLUT-1 in lymph node metastasis group were higher than those in group without lymph node metastasis(all P＜0.05).There were positive correlations between the mRNA and protein expressions of HIF-1α and between the mRNA and protein expressions of GLUT-1(r1=0.527,r2=0.408,respectively,both P＜0.01). Conclusions CoCl2 can up-regulate the expressions of HIF-1α and GLUT-1 in lung adenocarcinoma cell line A549 by inducing hypoxia.The hypoxia-mediated effects on them have the potential to make lung cancer cells further grow and promote hypoxie tolerance.The overexpressions of HIF-1α and GLUT-1 are closely correlated with the progression of lung carcinoma.Combined detection of HIF-1α and GLUT-1 may be helpful for the diagnosis and prognosis prediction of primary lung cancer in elderly patients.

Objective To investigate the possible mechanisms of teratogenesis caused by passive smoking, temporospatial changes in surface carbohydrates of the embryonic neural epithelium and the mesenchyme around it were explored. Methods The pregnant golden hamsters were randomly divided into the treated group and the control groups. In the chamber made by ourself for passive smoking experiment, the animals of the treated group were exposed to smoke from embryonic day 4 to 7, 3 times per day for 4 consecutive days. At twelve o'clock and eighteen o'clock of embryonic day 8, eight o'clock of embryonic day 9 and eight o'clock of embryonic day 10 respectively, the pregnant golden hamsters of both treated group and control group were sacrificed, the uteri were removed and embedded routinely with paraffin, sectioned transversely in series. Objective To assess the value of transrectal echography(TRE) in diagnosing peri-rectal lesions. Methods The echography images of 287 cases of peri-rectal lesions were retrospectively analyzed,and contrasted with the results of postoperative pathologic examination and the final clinical diagnosis. Results 83 cases of peri-rectal recess lesions were found by TRE, and the occurate rate of diagnosis was 91%. 123 cases of peri-rectal viscera lesions were found by TRE, and the accurate rate of diagnosis was 94%. Conclusion TRE plays an role in the diagnosis of peri-rectal recess and viscera lesions, and in the dynamic and real-time detection of the obstructive causes of bladder orifice.

Objective To detect serum Vaspin level in subjects with normal glucose regulation(NGR)and newly-diagnosed type 2 diabetes mellitus (T2DM) ,and to investigate the association between serum Vaspin and body mass index (BMI), age, gender, glycometabolism and lipemetabolism and insulin sensitivity index. Methods The fasting serum levels of Vaspin were measured in 48 normal controls and 66 T2DM patients using enzyme linked immunosorbent assay ( ELISA ). BMI, waist-hip ratio (WHR) and % body fat were measured,as well as fasting blood glucose (FBG), HBA1C, lipids and insulin levels. Results The fast serum Vaspin concentrations in the obese T2DM patients was ( 1.13 ±0. 25 ) μg/L,which were significantly higher than that in the non-obese T2DM patients (0. 65 ± 0. 13 ) μg/L (P ＜ 0. 01 ). In the NGR group, the fast serum Vaspin concentrations in the obese was (0. 95 ± 0. 11 ) μg/L, which were significantly higher than that in the non-obese (0. 38 ± 0. 18) μg/L( P ＜ 0. 01 ). In the T2DM group, serum Vaspin concentration in females ( [0. 92 ± 0. 35]μg/L) was higher than in males ([0.76 ± 0. 22] μg/L) (P ＜ 0.01 ). In the NGR group, serum Vaspin concentration in females was also higher than that in males ( [1.05 ± 0. 21] μg/L vs [0. 48 ± 0. 14] μg/L) )(P＜0. 01 ). Serum Vaspin level was positively correlated with BMI,WHR,% Body Fat,fasting insulin(FINS)and insulin sensitivity index (ISI) (r = 0. 365,0. 214,0. 238,0. 183 and 0. 147, respectively, Ps ＜ 0. 05 ). The multiple stepwise regression analysis showed that gender( R2 = 0. 161, P ＜ 0. 01 ), ISI ( R2 = 0. 183, P ＜ 0. 01 )and WHR( R2 = 0. 216, P ＜ 0. 01 ) were independent variables associated with Vaspin. Conclusion Serum vaspin level significantly increases in subjects with obesity, and independently associated with gender, ISI,WHR. These findings suggest that serum Vaspin may be involved in the pathophysiology of insulin resistance syndrome.

Background Glaucoma is the second leading cause of blindness,which is characterized by processing retinal ganglion cells (RGCs) loss and optic nerve dystrophy.Clinical study showed that lowing IOP can not arrest the glaucomatous damage of RGCs.To seek a neuroprotective drug is an urgent need.Objective This present study focused on the effect of triptolide,a natural biologically active compound extracted from Tripterygium wilfordii,on RGCs in glaucomatous eyes. Methods Glaucoma animal models were established in the right eyes of 80 clean Wistar rats by combination with aspiration of aqueous humor and phtocoagulation on anterior chamber angle.Wistar rats were assigned to two groups at random.Triptolide (5μg/kg) was intraperitoneally injected daily from three days before photocoagulation through scarification of animals (total 8 weeks),and same amount of physiologic saline solution was used at the same way.IOP was measured with a Tonopen XL tonometer at at 1day,3,5,7 days and weekly for 8-week duration after phtocoagulation.RGCs numbers was calculated by retinal Nissl staining.Morphology of retina in frozen section was examined under the light microscope.The experiment followed the Standard of Association for Research in Vision and Ophthalmology. Results The IOP was elevated in model eyes from 1 day through 3 weeks after operation with statistically different in comparison with before operation(P<0.05).No obvious differences in IOP changg was found hetween the triptolide group and the normal saline group at each time point(P>0.05).The numbers of RGCs of model eyes in normal saline group decreased gradually after operation,but no evident decline of numbers of RGCs in model eyes in triptolide group. RGCs in triptolide group were considerably more than those of normal saline group in various time points after operation ( P<0. 05). However,no obvious difference in RGCs numbers was found between model eyes and control eyes in Triptolide group. Conclusion Triptolide could protect RGCs in glaucomatous eyes,and its effect does not depend on IOP in chronic glaucoma model.

Objective To probe into the effects of fluoride on metal elements in the testis tissue of male rats, and provide experimental basis to further research for reproductive toxicity of fluoride. Methods Thirty-two healthy male Wistar rats, weighting 150 - 180 g, were randomly divided into 4 groups, normal sodium(control) by intragastrie administration for 90 days, and body weight was observed daily. After the last intragastric administration, all rats were killed by cervical dislocation. The contents of calcium(Ca), ferri(Fe), zincum(Zn),cuprum(Cu ) and magnesium(Mg) in the testis tissue were measured by atomic absorption speetrophotometry.Results After 30 days exposure, the difference of body weight between groups was statistically significant(F=3.884, P < 0.05). The body weight in low- and medium-dose groups[(235.00 :t: 14.56), (235.44 ± 24.99)g] were significant increased than high-dose group[(206.00 ± 18.16)g, all P < 0.05]. There was no significant difference of body weight between the groups at 0, 60 and 90 days(F = 0.501, 0.578, 1.893, all P > 0.05). The difference of Ca, Zn and Mg levels among four groups was statistically significant(F = 6.630, 6.844, 5.333, all P < 0.05). The content of Ca of the low-dose group[(56.15 + 4.21 )mg/kg] decreased than that of the control group[(77.57 ± 6.66)mg/kg, P < 0.05];the content of Zn of the low-, medium- and high-dose groups[(4.80 ± 0.55), (4.56 ± 0.33),(5.46 ± 0.79 )mg/kg] deceased than that of the control group [(7.16 ± 0.28 )mg/kg, all P < 0.05];the content of Mg of the high-dose group [(32.44 ± 1.53 ) mg/kg] decreased than that of the control group [(42.54 ± 8.07 ) mg/kg,all P < 0.05]. The difference of testis Fe and Cu between four groups was not statistically significant(F = 1.324,0.207, all P > 0.05). Conclusion Chronic fluorosis can affect the levels of metal elements in rat testis and damage the reproductive system.

〔Abstract〕 By use of the decision tree algorithm and diagnostic indexes , the paper sets up the discrimination rules to make differential diagnosis of Primary Hepatocelluar Carcinoma ( PHC) based on basic data of 95 patients with PHC and 190 patients with liver cirrhosis , in-cluding the CT diagnosis, testing results of imaging and serologic markers such as the HbsAg , AFP, CEA and AFU, sex and age, etc.As indicated by the results , the data mining technology represented by the decision tree can support the differential diagnosis of PHC .

Objective To investigate the curative effect of biofeedback treatment on attention-deficit/hyperactivity disorder ( ADHD ) patients using integrated visual and auditory continuous performance test ( IVA-CPT).Methods 23 ADHD patients undertaked 20 times of EEG biofeedback treatment using VBFB-3000A facility.Their executive and attention quotients were measured before and after treatment by IVA-CPT.Results The pre-therapy full scale of response control and attention quotients of ADHD were 58.26 ± 32.9 and 54.43 ±34.01 respectively.After the therapy, all the quotients got improved (response control 99.3 ± 12.6, attention 84.1 ± 15.9 ) except for stamina and speed quotients both in visual and in auditory.Conclusion Shortterm EEG biofeedback treatment is one of effective methods for ADHD patients,especially for hyperactive cases.
