The electrochemical behaviors of parathion(PT) on nano-Al2O3 film modified electrode(nano-Al2O3/GCE/CME) were investigated. It was found that the redox peak currents of PT remarkably increased and the oxidation peak potential negatively shifted at the nano-Al2O3/GCE/CME. All the experimental parameters were optimized and a simple and sensitive electroanalytical method was developed for the determination of PT. The oxidation peak current was linear with the concentration of PT over the range from 2.5×10-9 to 1.0×10-7 mol/L and 1.0×10-7 to 1.0×10-5 mol/L. A low detection limit of 1.0×10-9 mol/L was obtained for 30 s accumulation at open circuit(S/N=3). The relative standard deviation(n=10) was 3.8% for 1×10-5 mol/L PT. The proposed method was used for the quantitative analysis of PT in some vegetables and the results were satisfactory.

Objective To study the effects of excessive iodine intake through the meal on the expression of mRNA of placental NIS in pregnant rat and breast NIS in lactating rats. Methods Wistar rats, weaning one month, were randomly divided into three groups according to the body weights, i.e., normal iodine (NI), ten fold high iodine(10HI), one hundred fold high iodine(100HI), the ratio of female and male was 2∶1. Iodine intake of the groups were about 6.15, 61.5 and 615.0 mg/d respectively. After 3 months of treatment, the urine iodine was determined by As-Ce-catalytic spectrophotometry. The rats mated and had offspring. Their placenta and breasts were taken on the seventeenth day of pregnancy and tenth day of lactation respectively. Then NIS mRNA expression was determined by RT-PCR. Results The urine iodine increased with the increase of the iodine intake. The urine iodine and iodine intake showed the parallel magnification. Compared with the NI group,AR value of the placental NIS mRNA in 100HI group and the breast NIS mRNA in 10HI and 100HI group significantly decreased. Conclusion Excessive iodine intake may down-regulate the expression of the placental and breast NIS, which presents a protective effect on offspring.

Objective To explore the feasibility of He-Ne laser irradiation in promoting orthodontic tooth movement and periodontal remodeling through observing the effect of low power laser on the expression and distribution of matrix metalloproteinases- 9(MMP- 9) during orthodontic tooth movement in rabbit. Methods Thirty rabbits were divided into 1d,3d,5d,7d,14d and 21d groups, each group were given a coil spring connected to the maxillary upper first molar to the upper incisor and fixed bilaterally by a ligature wire and set force to 80g. He-Ne laser irradiation was performed on the right side of the orthodontic tooth, and the left side served as control. Radiation time ranged from 1 day to 5 days, once a day, 15min each time. All of the tissue sections were proceeded with MMP- 9 immunohistochemical staining. MMP- 9 average gray scale in periodontal tissue of the orthodontic teeth was analyzed by a Computer Image Analyzing System. Results The expression level of MMP- 9 increased earlier and lasted for a longer time in the compression and tension area on the side of irradiation by He-Ne laser as compared to those on the control side. There was significant difference between the irradiation side and the control side in terms of the MMP- 9 expression gray scale in the tension zones at 5 and 7 days post-operation, and in the pressure zones at 3,5,7 day post-operation. Conclusion During the movement of orthodontic teeth, local irradiation with He-Ne laser could increase the expression of MMP-9. It was suggested that the irradiation with low power laser accelerate blood vessel and bone remodeling.

Seventy newly diagnosed diabetic patients were enrolled in the study, including 37patients aged over 60 (elderly-onset diabetes, EOD group) and 33 patients under 60 (adult-onset diabetes,AOD group) ; 30 elderly healthy subjects served as controls. Serum melatonin levels were determined with ELISA and the expression of inducible nitric oxide synthase (iNOS) mRNA in peripheral blood leucocytes (PBL) was detected by reverse transcription polymerase chain reaction (RT-PCR). The results showed that serum melatonin levels were significantly lower in EOD group than those in control group [ (6. 6 ± 1.3 )ng/L vs. (7.7 ± 1.4) ng/L, P <0. 01 ], the expression of iNOS mRNA was significantly higher than that in AOD group [ (36 ± 3 )% vs. (30 ± 4)%, P < 0. 01 ]; and i NOS mRNA was not detected in control group.Logistic regression analysis indicated that decreased serum melatonin levels and up-regulated iNOS mRNA might be independent risk factors for diabetes mellitus in the elderly.

Objective To investigate the effect of percutaneous vertebroplasty and kyphoplasty (PKP) on bone density and serum leptin in patients with osteoporotic thoracolumbar compression fractures. Methods One hundred and two patients with osteoporotic thoracolumbar vertebral compression fractures and 100 cases of healthy control were treated with PKP surgery. The serum leptin and adiponectin, and the bone mineral density of the lumbar spine were determined. Results patients Levels of the serum leptin and adiponectin in patients with osteoporotic thoracolumbar vertebral compression fracture were significantly higher than those with non osteoporotic fracture patients (P < 0.05). One month post-PVP, the levels of serum leptin and adiponectin in the two groups were significantly decreased (P < 0.05). postoperative group of The lumbar bone density at 1 month post-operation was is significantly higher than that before operation in each group (P < 0.05, respectively), with no significant difference between the two groups. The levels of serum leptin and adiponectin were negatively correlated with the bone mineral density (BMD) (P < 0.05). Conclusions tLevels of serum adiponectin and leptin were both increased in osteoporotic or non bone osteoporosis thoracolumbar vertebral compressor fractures. The percutaneous vertebroplasty forming operation could effectively reduce the serum level of leptin and adiponectin, and improve the bone mineral density of the lumbar vertebrae.

Objective To investigate the effects of glycyrrhizin on gene expression of transforming growth factor(TGF)-?signaling pathway of hepatic stellate cell(HSC)in mice by gene microarray tech- nique. Methods The HSCs were isolated from mice and cultured in vitro. Then the mice were divided into control group,TGF-?_1 group(5 ng/ml) or TGF-?_1(5 ng/ml)combined with glycyrrhizin(100?mol/L) group. The cells were collected after 10 hours to extract RNA.A cDNA microarray(GEArray~(TM) Q) targeting TGF-?/BMP signal transduction was used to screen the genes which showed significant changes in expression of TGF-?pathway of HSC by glycyrrhizin. Results The microarray analysis showed that 16 genes(16.7%),such as Smad2,Smad3,Smad7,CoL3A1,CoL1A2,and PAI-1,were upregulated by TGF-?_1 and then down-regulated by glycyrrhizin.Five genes(5.2%)(including BMP7,IGFbp3 and etc.)downregulated by TGF-?_1,were then up-regulated by glycyrrhizin.Finally,2 genes upregulated by TGF-?_1 were then up-regulated predominantly by glycyrrhizin in HSC(T?R2,betaglycan).Changes in some genes, such as Smad2 Smad3,Smad7,were further confirmed to be coincided with cDNA microarray by semi-quantitative RT-PCR.Conclusions The anti-fibrosis mechanisms of glycyrrhizin may be through to interference of TGF-?signaling pathway, decrease the synthesis and increase the degradation of collagen.
