1.The effect of concentrate growth factor extract on MC3T3-E1 cells

Objective:To evaluate the effect of concentrate growth factor extract(CGFe)on the proliferation and differentiation of MC3T3-E1 osteoblasts.Methods:MC3T3-1 cells were cultured with and without CGFe respectively.At 1 ,3,5 d of culture the pro-liferation of the cells was detected by MTT assay,the activity of alkaline phosphatase (ALP)of the cells was examined by ALP kit;at 1 4,21 d of culture the calcium nodus formation was observed by Alizarin red staining;RUNX2 and OSX expression was quantified by real-time PCR.Results:With the extension of culture time,the proliferation,ALP activity,the calcium nodus and the level of RUNX2 and OSX mRNA of the experimental group were increased more than those of the control group.Conclusion:CGFe may pro-mote the proliferation and differentiation of osteoblasts.
2.Toxic Effect of Exhaust Particles Extracts of Diesel Vehicles on V79 Cell

Objective To observe the toxicity of diesel exhaust particles extracts(DEPE) on V79 cell in cell viability, membrane and oxidative stress. Methods Cell exposed different concentrations of DEPE for different time , the change of cell viability, the leakage of intracellular lactate dehydrogenase(LDH), intracellular glutathione(GSH) and glutathione peroxidase (GPX)content were determined respectively, and analyzed the relation between GSH and GPX. Results The results showed that the viability of V79 cell decreased and the leakage rate of lactate dehydrogenase increased gradually in according with the increasing dosage of DEPE,and that DEPE could impair V79 cell by oxidative stress, included intracellular GSH content decreased and the activity of GPX increased, indicating that GPX might play a important role in DEPE induced GSH depletion. Conclusion DEPE may impair cell viability and the cell membrane integrity, also impair cell by oxidative stress.
4.Prognostic analysis of patients with cerebral glioma treated with three-dimensional conformal radiotherapy

Objective To evaluate prognostic factors of patients with cerebral postoperative glioma treated with three-dimensional conformal radiotherapy (3DCRT). Methods62 patients with cerebral glioma who had residual tumor received 3DCRT. Factors such as gender, age, location of the glioma, pathological type, pathological grade, degree of surgical resection, combined chemotherapy, radiation dose, Karnofsky score before treatment were analyzed to show wether the impact on prognosis, Drawing survival curve with the Kaplan-Meier method, Log-rank test for univariate analysis and multivariate analysis were performed by using COX regression model.ResultsLast follow-up time was April 1, 2009. Median follow-up was 22 months, 43patients died. The 1-, 2- and 3-year overall survival rate were 65.7 %, 34.2 % and 20.5 %. In univariate analysis, the survival was related to age (P=0.008), pathological grade (P=0.016), degree of surgical resection (P=0.000), Karnofsky score before treatment (P =0.018). In COX multivariate modal, age (P =0.031), degree of surgical resection (P =0.000), Kamofsky score before treatment (P =0.035) were independent prognostic factors. ConclusionAge <40 years, Karnofsky score before treatment ≥70 and mostly resection are independent factors for predicting better survival of glioma patients.

6.Analysis of etiology and clinical features of hospital-acquired bloodstream infection in children

Objective To summarize the pathogenic spectrum and antimicrobial susceptibility of hospital-acquired bloodstream infections (HABSI) in children,and to provide evidence for clinical antiinfection treatment.Methods During January 2004 to December 2011,the clinical data and drug susceptibility results of 168 children who were diagnosed with HABSI based on positive results of pathogen tests were reviewed retrospectively in Yuying Children's Hospital affiliated to Wenzhou Medical College.Results A total of 171 strains were isolated from blood specimens of the 168 children.The majority of HABSI occurred in the intensive care unit (73.7 %),followed by department of hematology (22.6%).Gram-positive bacteria,gram-negative bacteria and fungi accounted for 53.8%,34.5% and 11.7%,respectively.The predominant pathogens were Stagphylococcus epidermidis (14.1%),followed by Klebsiella pneumonia (13.5 %),Staphylococcus haemolyticus (7.6%),Staphylococcus aureus (6.4%),Enterococcus faecium (6.4%) and Escherichia coli (6.4 %).Staphylococcus aureus and Staphylococcus epidermidis resistant to methicillin were detected in 9.1 % and 91.7 % of specimens,respectively.Staphylococcus and enterococcus which were resistant to vancomycin or linezolid were not detected.The isolation rates of Klebsiella pneumoniae and Escherichia coli with extended-spectrum beta-lactamases (ESBL) were 95.7% and 72.7%,respectively.One hundred and sixty cases (95.2 %) had underlying diseases including premature birth and low birth weight (60.7%).One hundred and twenty-six cases (75.0%) underwent invasive procedures.The peak incidence of HABSI occurred in children less than 3 months old (75.6%).Conclusions HABSI is most common in infants younger than 3 months old,with underlying diseases or undergone invasive procedures.The pathogens are mainly gram-positive bacteria.Very low birth weight infants can acquire unusual infection of Kodamaea ohmeri.Thus,it is essential to strengthen the prevention of hospital-acquired infection.

Objective To investigate the effects of 15-deoxy-?12,14-prostaglandin J2(15d-PGJ2) and DDP on the growth of human pulmonary carcinoma PLA-801D cells and the mechanisms of apoptosis.Methods The human pulmonary carcinoma PLA-801D cells were selected and added to each well of 96-well place and cultivated for 24 h.Then the cells were treated with different concentrations of 15d-PGJ2(0,5,10,20,40 and 80 ?g?L-1) or 15d-PGJ2 combined with DDP(3 mg?L) for 24 h.0 ?g?L-1 15d-PGJ2 group was control group.The morphological changes of cells were observed under inverted microscope.Microculture tetrazolium(MTT)dye was applied to detect the proliferation of the human pulmonary carcinoma PLA-801D cells treated with 15d-PGJ2 and DDP.Diphenylamine assay(DPA) was used to evaluate the activation.Flow cytometry assay(FCM) was used to detect the apoptosis proportion and the changes of cell cycle.Results When the human pulmonary carcinoma PLA-801D cells were treated with low-concentration 15d-PGJ2 alone(5,10 and 20 ?g?L-1),no significant difference was observed in the inhibitory rate of cell growth and the apoptotic indexes such as the apoptosis proportion,the percent of DNA fragmentation and the activity of caspase-3 compared with control group(P
9.Isolation and structure determination of cypritibetquinone A and B, two new phenanthraquinones from Cypripedium tibeticum

Aim To study the chemical constituents of Cypripedium tibeticum. Methods Compounds were isolated by repeated silica gel chromatography and purified on Sephadex LH-20 and structures were determined by spectral analysis. Results Cypritibetquinones A and B were isolated from the ethyl acetate residue and their structures were determined as 7-hydroxy-2-methoxy-1, 4-phenanthraquinone ( 1 ) and 7-hydroxy-2,10-dimethoxy-1,4-phenanthraquinone ( 2 ), respectively, by extensive spectral analyses. Conclusion Cypritibetquinones A and B are two new phenanthraquinones.
10.A correlation between liver fibroindex and prognosis of hepatocellular carcinoma after hepatectomy

Objective To study the correlation between liver fibroindex and prognosis of hepatocellular carcinoma after hepatectomy.Methods The clinical data of 77 patients with hepatocellular carcinoma who underwent hepatectomy at the 5th Hospital Affiliated to Sun Yat-sen University from 2009 October to 2014 December were analyzed retrospectively.Using data from preoperative Fibroindex,these patients were divided into 2 groups,Group 1:Fibroindex ≤5.4,and Group 2:Fibroindex >5.4.The clinical pathological parameters and postoperative disease free survival rates in these groups were analyzed.The Cox ratio risk pattern analysis was used for factors correlating recurrence.Results The parameters in the 2 groups which included sex (35/0 vs 29/13),type of cirrhosis (23/12 vs 14/28),numbers of tumor (35/0 vs 26/16),maximal diameter of tumor (31/4 vs 23/19),vascular invasion status (32/3 vs 26/16),resection margin status (35/0 vs 36/6),hepatic vein/portal vein tumor thrombus status (35/0 vs 29/13),perioperative change of HBV DNA levels (23/12 vs 15/27)were correlated with postoperative recurrence,and they all showed significant differences (P < 0.05).Disease free survival rate in Group 2 was lower than Group 1 (71.4%,33.7%,22.4% vs 94.3%,46.2%,46.2%,P < 0.05).The Cox regression analysis showed Fibroindex > 5.4,AFP-L3% ≥ 10%,and number of tumor > 3 to be independent predictors of poor disease-free survival after hepatectomy for hepatocellular carcinoma (P < 0.05).Conclusions Preoperative Fibroindex > 5.4 was an independent adverse predictor of poor disease-free survival.It was valuable to predict postoperative recurrence in hepatocellular carcinoma patients.
