Background and purpose:Chemotherapy plays an important role in the treatment of gastric cancer. It is becoming a direction that the choice of chemotherapeutic agent for the clinic will be based on the levels of some relative gene expressions in the tumor tissue. As the key enzymes of fluoropyrimidines, the expression levels of thymidylate synthase(TS) and thymidine phosphorylase(TP) may correlate with the response of tumor to chemotherapy. This study was to investigate the expression of TS and TP mRNA in gastric cancer tissues and their correlation with prognosis. Methods:The expression levels of TS,TP mRNA in 51 gastric adenocarcinoma tissues were detected by real-time quantitative reverse transcription polymerase chain reaction(RT-PCR).Results:The median expression levels of TS,TP mRNA were 0.94 and 21.20,respectively.There was significant difference in terms of disease-free and overall survival between the gastric cancer patients with high and low TS mRNA expression level(P0.05)but not overall survival(P0.05).Conclusions:The expression levels of TS,TP mRNA may serve as prognostic markers for gastric cancer patients treated with FU-based adjuvant chemotherapy.

Background Rare studies on the effect of statin on early stage of atherosclerosis have been repor- ted.Oxidative stress induced endothelial dysfunction may be the initiative factor for the development of atheroscle- rotic plague. Objective To investigate the mechanisms by which simvastatin, prevents atheroselerosis independ- ently of its lipid-lowering effect in Apolipoprotein E deficient mice. Methods Twenty-four 6 week old male apoE- deficient mice were randomly to receive placebo or simvastatin 5 mg/(kg?d)by gavage for 4 weeks.Total choles- terol(TC),super oxide dismutase(SOD),malondialdehyde(MDA)and serum nitric oxide(NO)were measured by biochemical analysis.Endothelium was observed by HE dyeing. The expression of caveolin-1 in aortic wall was detected by immunohistochemistry. Results There was no significant difference in serum TC between control and simvastatin treatment groups.Simvastatin caused less damaged endothelium(33.33% vs control's:75%,P
7. Mechanism of Shenluotong regulating NR3C2/SGK-1/Smad pathway in inhibiting renal interstitial fibrosis
Chinese Traditional and Herbal Drugs 2018;49(21):5149-5154
Objective To investigate the mechanism of Shenluotong inhibiting renal interstitial fibrosis by regulating NR3C2/SGK-1/Smad pathway. Methods A total of 48 Wistar rats were randomly divided into sham operation group, model group, Eplerenone group, and Shenluotong group (n = 12). Model group, Eplerenone group, and Shenluotong group used unilateral ureteral obstruction (UUO) method to establish rat renal interstitial fibrosis model. After the operation, the rats in the eplerenone group were treated with eplerenone at a dose of 100 mg/(kg∙d). Rats in the Shenluotong group were oral given Shenluotong decoction at a dose of 26 g/(kg∙d) and Sham operation group and model group were administrated equal volume of saline once daily for continuous 10 d. Laser confocal microscopy was used to detect mineralocorticoid receptor NR3C2 expression. The expressions of SGK-1, TGF-β1, Smad4, and Smad7 in renal tissues were detected by immunohistochemistry, Western Blot and Real time-PCR. Results In the sham operation group, NR3C2 was expressed in the cytoplasm of renal tubular epithelial cells and was not expressed in the nucleus. The expression of NR3C2 in the UUO rat was significantly up-regulated in cytoplasm and positive expression was observed in the nucleus. The expression of NR3C2 in the nucleus of cells in the Eplerenone group and Shenluotong group was significantly decreased when compared with the model group. Compared with the sham-operated group, the expression of SGK-1, TGF-β1, and Smad4 was significantly up-regulated and the expression of Smad7 was significantly decreased (P < 0.05, 0.01) in the other groups. Compared with model group, the expression range and intensity of TGF-β1 and Smad4 were significantly decreased in Eplerenone group and Shenluotong group (P < 0.05, 0.01), and the expression range and intensity of Smad7 were significantly increased (P < 0.01). Conclusion Shenluotong can inhibit renal interstitial fibrosis through blocking the activation of mineralocorticoid receptor, reducing the level of SGK-1, and regulating the Smads signal pathway to inhibit the overexpression of TGF-β1.

OBJECTIVE:
To explore the injury mechanism of the human knee in a traffic accident by establishing a 3D finite element (FE) model.
METHODS:
The FE model, composed of femur, tibia, fibula, patella, meniscus, knee ligaments and surrounding soft tissues, was reconstructed by CT scanning data from a male volunteer. Validation was performed by the lateral impact simulation, and the stress and strain results were obtained to be compared with those previously reported for injury prediction.
RESULTS:
The results derived from the FE model were found to be similar with those previously reported, most of the ligaments and meniscus wounded at 40 m/s collision, which was readily observed.
CONCLUSION
The simulation results generated by FE model can be effectively used for the injury mechanism analysis of initial contact.
